1.THE PHARMACOLOGICOL STUDY ABOUT ANTIDEPRESSANT EFFECTS OF THE 7903

The antidepressant effects of the N-Isosuccinyl-dichlorocinnamo-ylamide ( 7903 ) , an analogue of piperine, upon 5 animal models were studied. The 7903 was found to have antidepressant effects on the models with doses less than TD50. The 7903 with the single dose, which did not increase the spontaneous motor activity of animals, could significantly decrease the duration of immobility of mice and rats in the forced-swimming test and that of mice in the tail-suspension test. The duration of the longest permanent immobility indaced by cornea electro-stimulation in mice was also decreased with the chronic administration ( 2 weeks ) of the 7903. And the hypothermia induced by reserpine could be antagonized by the 7903.

The influences of the N -Isosuccinyl - dichloro - cinnamoylamide (7903) on the NA and 5 -HT systems in the brain were studied. Acute administration of 7903 could significantly enhance the level of NA and 5 -HT in mice brain and cortex of rats, and the level of 5 -HT in the hyporthalamus of rats was tending to be enhanced. However, chronic administration of 7903 (two weeks ) could decrease the level of NA and the ra-tio of 5-HIAA/5-HT in the cortex of rats; Chronic administration of 7903 could also decrease the turnover of NA and 5- HT in mice brain, but had no effects on the special binding of ? - receptor to 3-H - DHA and that of 5 -HT receptor to H - ketanserine in the cortex of rats.

5.Invasive ductal carcinoma of the breast is prevalence of loss of heterozygosity at 8p22,11p15 and 17p13

Objective:To investigate the LOH at regions on chromosomal arm 8p22,11p15,17p13 and its relationship with clinicopathological parameters. Methods:Thirty-four paraffin-embedded tumor and corresponding noncancerous tissues were analysed. PCR was used to amplified three microsatellite markers D8S136,D11S988 and TP53 located at these chromosomal regions. PCR products were electrophoresed on 6%polyacrylamide gel and detected using silver staining. The P53, c-erBb-2,PR,ER status were determined by immunohistochemistry. Results:Of the three markers we studied, D8S136 was detected LOH at a frequency of 12 in 34 tumors(35.29%). D11S988 and TP53 were detected LOH at a frequency of 5 in 34 tumors(14.71%). There were no obvious associations between LOH at D11S988、TP53 and clinicopathological parameters, but the tumors with LOH at D8S136 were significant larger than that without LOH(P=0.0049). Conclusion: Invasive ductal carcinoma of the breast has a frequent LOH on chromosome 8p22. The loss or inactivation of putative tumor suppressor genes on 8p22 may contribute to the excessive growth of the tumors.
6.Role of NO in reduction of myocardial ischemia-reperfusion injury by ginsenoside Rb1 preconditioning in diabetic rats

Objective To evaluate the role of by NO in reduction of myocardial ischemia-reperfusion (IR)injury by ginsenoside Rb1 preconditioning in diabetic rats. Methods Forty healthy adult male SD rats weighing 220-280 g were used in this study. Diabetes mellitus was induced by intraperitoneal streptozotocin 65 mg/kg and confirmed by fasting blood glucose > 16.7 mmol/L. The animals were randomly divided into 4 groups ( n = 10each): sham operation group (group S), group IR, ginsenoside Rb1 group (group R) and L-NAME + ginsenoside Rb1 group (group LR). IR was produced by occlusion of the anterior descending branch of left coronary artery (LAD) for 30 min followed by 120 min reperfusion in group IR, R and LR. In group S, LAD was exposed but not occluded. In group LR, L-NAME 10 mg/kg was injected iv 25 min before ischemia. In group R and LR, ginsenoside Rb1 40 mg/kg was injected iv 10 min before ischemia. In group S and IR, eaqual volume of normal saline was injected instead of ginsenoside Rb1. The blood sample was taken from carotid artery at 120 min of reperfusion for determination of serum activities of creatine kinase (CK) and lactate dehydrogenase (LDH). Then the animals were sacrificed and myocadial tissues were obtained for determination of infarct size, endothelial nitric oxide synthase (eNOS) expression, MDA and NO contents, SOD activity and microscopic examination. Results The serum activities of CK and LDH were significantly increased and the myocardial infarct size was enlarged in group IR, R and LR, and eNOS expression was significantly down-regulated, MDA content was increased, and SOD activity and NO content was significantly decreased in group IR and LR compared with group S ( P < 0.05). The serum activities of CK and LDH, and MDA content were significantly decreased, the myocardial infarct size was reduced, the expression of eNOS was up-regulated and the activity of SOD was increased in group R compared with group IR and LR ( P < 0.05). There was no significant difference in the indices mentioned above between group IR and LR ( P> 0.05). Conclusion Ginsenoside Rb1 preconditioning can attenuate myocardial IR injury in diabetic rats via activation of eNOS, increase in NO production, and inhibition of the lipid peroxidation reaction.
