2.Clinical significance of detecting ET-1 ,TNF-α and ICAM-1 in peripheral blood of patients with chronic obstructive pulmonary disease

ObjectiveTo investigate the clinical significance of Endothelin-1,Tumor necrosis factor-α and Intercellular adhesion molecule-1 in the patients with chronic obstructive pulmonary disease(COPD).MethodsThe serum samples were prepared from 80 cases with COPD and 30 healthy persons.The levels of ET-1,TNF-α and ICAM-1 were measured by enzyme-linked immunosorbent assay ( ELISA ).Results Levels of ET-1,TNF-α and ICAM-1 in the COPD patients were higher than thosein the healthy cases ( t =4.896,4.621,4.231,all P < 0.05 ).Levels of ET-1,TNF-α and ICAM-1 in the period of AECOPD were higher than those in the cases of the COPD stable time( t =3.346,3.525,3.269,all P <0.05 ).ConclusionET-1,TNF-αt and ICAM-1 are involved in airway inflam mation,ET-1,TNF-α and ICAM-1 not only take part in the acute inflammation in COPD,but also closely connected with clinic stages.
3.The influence of fluvastatin on the levels of IL-6 ,MMP-1 and APN in patients with coronary heart diseases

ObjectiveTo explore the influence of fluvastatin on the levels of IL-6,MMP-1 and APN in patients with coronary heart diseases.Methods60 patients with coronary heart diseases were randomly divided into 2groups:40mg group and 80mg group treated by fluvastatin and 30 healthy persons were selected as control group,serum levels of IL-6,MMP-1 and APN were measured before and after treatmead.ResultsSerum levels of IL-6,MMP-1 were significantly increasd in the coronary heart diseases group (t =4.896,4.231,all P < 0.05 ).Serum level of APN was significantly decreasd in the coronary heart diseases group( t =4.352,P < 0.05 ).The serum levels of IL-6and M MP-1 were significantly decreasd after therapy (4.156,4.121、4.553,all P < 0.05 ).The decrease of serum levels of IL-6,MMP-1 in 80mg group was more significant than those of 40mg group( t =3.786,3.690,4.10,all P <0.05).ConclusionSerum concentrations of IL-6 and MMP-1 increase,but APN decrease in patients with coronary heart diseases.Serum concentrations of IL-6,MMP-1 and APN had relationships with coronary plaque extent.Fluvastatin could decrease serum levels of IL-6 and MMP-1,increase serum level of APN in patients with coronary heart diseases,and have a beneficial effect on steadying the plaque.

Retinol binding protein 4 (RBP4), synthesized in the liver, is an adipocytokine existing in the circulation. It is closely related to the incidence of macrovascular complications of diabetes. It also promotes the occurrence and development of diabetic microangiopathy. This paper summarized the progress in the field about the structure and function of RBP4, RBP4 and coronary heart disease and atherosclerosis, as well as RBP4 and diabetic nephropathy and diabetic retinopathy.

6.Effects of Lipo-PGE1 on the Lower Limb Blood Vessel in Diabetic Patients.

Objective To evaluate the effects of Lipo prostaglandin E1(Lipo-PGE1) on the lower limb blood vessel in diabetic patients.Methods The study was performed in 36 patients with diabetes related lower limb vascular disease.Before and after Lipo-PGE1 treatment for 28 days,vascular diameter and blood flow velocity of femoral artery,popliteal artery,posterior tibial artery and dorsal pedal artery were measured by Doppler ultrasonography examination.Results After the treatment with Lipo-PGE1,the vascular diameters of femoral artery,popliteal artery,posterior tibial artery and dorsal pedal artery were dilated;and there were significant differences in posterior tibial artery and dorsal pedal artery.After the treatment with Lipo-PGE1,blood flow velocity of femoral artery,popliteal artery,posterior tibial artery and dorsal pedal artery were increased,however,there were no significantly statistical differences.After the treatment with Lipo-PGE1,all the patients feel well.Painless walking distance and maximal walking distance of 18 patients with intermittent claudication were increased.Conclusion Lipo-PGE1 causes a significant improvement of diabetes related lower limb vascular disease.Furthermore,the results demonstrated the major effects on posterior tibial artery and dorsal pedal artery.Lipo-PGE1 can dilate the smaller artery,which is important and helpful for patients to improve symptoms and signs.
7.Effect of valsartan on platelet and endothelial function in chronic rapid atrial pacing dogs

Objective To investigate the effects of valsartan on platelet and endothelial function in atrial fibrillation dogs induced by chronic rapid atrial pacing.Methods From Sep.2001 to Jun.2003,14 dogs paced at 400 bpm in the right atrium for 6 weeks were randomly divided into control group and valsartan group.The dogs in valsartan group received valsartan(30 mg?kg~ -1 ?d~ -1 )1 week before rapid atrial pacing until pacing stop.The plasma von Willebrand factor(vWF),endothelin-1(ET-1),angiotensin Ⅱ(AngⅡ)levels and blood platelet aggregation rate were measured before and after rapid atrial pacing.Results (1)Plasma AngⅡ level elevated significantly in both groups,indicating activation of renin-angiotensin system.(2)The maximam platelet aggregation rates by four different agonists(Adenosine diphosphate,Epinephrine,Collagen,Arachidonic acid)increased significantly in control group dogs.To some extent,valsartan could inhibit platelet activation caused by chronic rapid atrial pacing.(3)There were significant elevations in the plasma levels of vWF and ET-1 in control group dogs.Valsartan could meliorate endothelial function and decrease the concentrations of vWF and ET-1.Conclusion Valsartan can inhibit platelet activation and meliorate endothelial function in chronic rapid atrial pacing dogs.
8.The Investigation of Risk Factors of Hypertension among Adult Residents in Hebei Province

Objective To understand hypertension risk factors among adult residents in Hebei province.Methods The adult residents were selected by multistage stratified sampling,and investigated with the same questionnaires,body measurement,blood pressure measurement,blood sample analysis,to analyze risk factors of hypertension by ?2 with simple factor and by logistic regression with mult-factors.Results The prevalence of hypertension in 4200 adult residents(18-69 ages) was 39.6% and age-adjusted rate was 28.5%.The single factors analysis indicated that the prevalence of hypertension was higher in men than in women,in countryside than in city,and increased with age rising.Its prevalence is correlated with education level,marriage status,occupation,income,smoking,passive smoking,manual activity,eating habits,BMI,waist,diabetes,blood fat,status of spirit and health.Multiple logistic regression analysis indicated that city,female,educational level of senior high school and above that,farm working,eating less brined vegetables is protective factors for hypertension,and their OR(95%CI) were 0.777(0.644~0.937),0.674(0.551~0.823),0.654(0.534~0.801),0.826(0.684~0.997),0.844(0.729~0.977) respectively,and more than 35 ages,not usually eating vegetable,bad status of health by self-sensations,high serum TC and TG,overweight and obesity were the main risk factors for hypertension,and their OR(95%CI) were 4.145(3.263~5.266),4.460(1.049~18.969),1.234(1.068~1.425),1.400(1.200~1.634),1.420(1.074~1.878),1.478(1.235~1.768),1.728(1.448~2.061) respectively.Conclusion The prevalence of hypertension in adult residents in Hebei province was correlated with multi-factors,so complicated prevention and control in hypertension should be reinforced.

Objective To investigate the feasibility of Chelex DNA extraction from thin blood smears for genetic analysis, and to develop smear-based nested polymerase chain reaction (PCR) of the 18S RNA of Plasrnodium falciparum. Methods Chelex-100 which was chelating ion exchange resin was used to extract DNA from Giemsa-stained or unstained thin blood smears of different concentrations of Plasmodium falciparum. With the extracted DNA as the template, 18S RNA gene was amplified by nested PCR to test the susceptibility of Chelex method. Results Positive band of 205bp appeared in nested PCR with DNA extracted from Giemsa-stained or unstained thin blood smears of patient with falciparum malaria. Using the Chelex method, the detection limits of the smear-based nested PCR were 1.5 × 101 parasite/μL blood for Giemsa-stained and 1.5×10-1 parasite/μL blood for unstained thin blood smears. Conclusions Chelex DNA extraction is a simple and efficient method for extracting trace amount of DNA from thin blood smear. The smear-based nested PCR developed in this study is feasible to identify the gene from reserved thin blood smears and will provide a new approach for clinical diagnosis and study of molecular epidemiology.
