1.Cloning and purification of paired immunoglobin-like type 2 receptor beta and preparation of its polyclonal antibody

Objective:To express and purify paired immunoglobin-like type 2 receptor beta(PILR?) and prepare the polyclonal antibody against its protein.Methods: The PILR? cDNA was amplified from the total RNAs extracted from human peripheral blood cells by RT-PCR and was cloned into pET-32a vector.The vector harboring PILR? gene was then expressed in E.coli by IPTG induction and the product was purified by Ni~(2+)-NTA affinity chromatography.Polyclonal antibody was prepared by immunizing rabbits with the purified protein.The titer and specificity of the antibody were examined by ELISA and Western blot,respectively.Results: Highly purified PILR? protein(Mr 42 000) was obtained.The prepared polyclonal antibody was highly specific and had a titer of 110 000.Conclusion: PILR? gene is successfully cloned and the purified PILR? protein is expressed.Polyclonal antibody against PILR?(with high titer and specificity) is successfully obtained,which provides a basis for further study on PILR?.
2.The research progress of microRNA on regulation of drug disposition in vivo

The disposition of drug in vivo is subjected to a series of biotransformation and transport, depending on the involvement of drug-metabolizing enzymes and transporters.However, the individual capacity varies when metabolizing and transporting the same drug, and pharmacogenomics has trouble in completely explaining the differences.microRNA, a key aspect of epigenetic modifications, is a powerful complement to traditional genetics.Emerging evidences have confirmed that drug-metabolizing enzymes and transporters be controlled by different microRNAs, and the same microRNA also regulates several drug-metabolizing enzymes or/and transporters simultaneously.All of these researches infer that microRNAs are likely to realize the comprehensive macro-regulation of gene expression.The further study of microRNAs maybe a suitable point to research the interindividual variability in disposition of drugs, and it provides a theoretical basis for rational use of drug and individualized medicine.
3.Diagnostic accuracy of magnetic resonance imaging enhancement pattern in knee bone,meniscus,ligaments,and articular cartilage injury: Comparison with routine magnetic resonance imaging scanning

BACKGROUND: Routine magnetic resonance imaging(MRI)scans can comparatively show intra-articular soft tissue structures,but there are still some limitations in the diagnosis of intra-articular lesions and injury due to deficiency in contrast of imaging.Enhanced MRI scan has been applied to the liver,brain and other parts,which effectively improves the diagnostic accuracy,but there are few reports on the enhanced scans of knee.OBJECTIVE: To evaluate clinical application value of MRI enhancement pattern in diagnosis of knee injury.METHODS: Totally 21 patients with knee injury(22 knees)were routinely scanned by 0.5T Signa Contour MRI scanner(GE company,USA).Then enhancer was injected into knee joint by the same physician and MRI enhancement pattern was performed in these patients.Film-reading and comparison were completed by two associate chief physicians.RESULTS AND CONCLUSION: Different levels of joint structural damage,such as bone,meniscus,ligaments,and articular cartilage and so on,were found in all patients by routine MRI scan.The extent and scope of its damage had been more clearly displayed by enhanced MRI scan.Meanwhile,misdiagnosis of meniscal injury by routine MRI scan was confirmed by enhanced MRI scan in 2 patients.Some damaged parts that were not found by routine MRI scan were Confirmed by enhanced MRI scan.All these demonstrated that positive rate of knee injury in MRI enhancement pattern was higher than conventional MRI scan.There was significant difference in meniscus injury between two patterns(P = 0.035).MRI enhancement pattern in comparison with conventional MRI can further improve the diagnostic accuracy rate of knee injury.
4.Detection of receptors of Streptococcus mutants in human whole salivary components

Objective To select the acceptor ingredients of Streptococcus mutans protein in the whole saliva through the interaction between adhesion and saliva protein of Streptococcus mutans. Methods After activation and liquid culture of Streptococcus mutans strains of international standards, we took Su-bonding from liquid culture of bacteria secretion in the process of liquid culture by using chilled ethanol extraction. Then su-bonded components underwent electrophoresis analysis, content determination and antigen detection. We collected 15 with high carries and 15 with no carries. Two hours after eating, the whole non-irritating saliva was collected, electrophoresis samples were prepared after freeze-drying, and then stains were separated with SDSPAGE. Western blot detection was used for adhesive protein and Streptococcus mutans' antibody composition. Results The ingredients of Su-bonding secreted on the surface of Streptococcus mutans were obtained in the cultured saliva. There were at least six kinds of components in the extraction of Western blot detection, not limited to the three known kinds of proline-rich protein. Conclusion The adhesion of Streptococcus mutants can be bound with the proteins of saliva. It may be the material basis in Streptococcus mutants adhered to tooth surface.
5.A study on the adaptive mechanism of peripheral nerve to gradual limb lengthening: observation on the perineurial barrier and vascular permeability

Objective To evaluate the effect of limb lengthening on the perineurial barrier and vascular permeability of peripheral nerve. Methods Thirty-two rabbits with limb lengthening of 10% , 20% , 30% and 40% were used in this study; 8 animals were allocated in each group. Sixteen were employed for the electron microscopic(EM) observation and the others for the investigation of perineurial barrier and vascular permeability of the tibial nerves. Results Even if the elongation reached 40% of the original length of the tibia, both the perineurium and inner membrane of capillary was still intact, the perineurial barrier and vascular permeability maintained normal; however, EM examination revealed increase of a large number of vacuoles and the increase of the number of mitochondria in the cell of the perineurial membrane. Brain metastases (BM) are one of the main cause of death in patients with non small cell lung cancer ( NSCLC ).The prognosis is still poor, despite standard therapy based on surgery and radiotherapy.In recent years,with the deepening understanding of molecular mechanisms in lung cancer, epidermal growth factor receptor (EGFR) mutation might be considered as an effective therapeutic target for NSCLC with BM.Therefore,we review the current knowledge about the correlation between EGFR mutation with prevalence and prognosis in BM.
7.The diagnosis and treatment of localized high-risk prostate cancer: a case report and regimen prescription strategy

Objective To exploie the choices and timing of different treatment regimens of prostate cancer.Methods The complete clinical data of one case of localized high-risk prostate cancer was reported.The patient,aged 69 years old,was admitted to hospital with urinary frequency and dysuria for 2 years.Serum total PSA was 36.8ng/ml and prostate biopsy dignosed as prostate cancer.The Gleason score was 4 + 3,and the clinical stage was T2b N0M0.Results The patient underwent radical prostatectomy,salvage radiotherapy + androgen deprivation therapy,abiraterone,enzalutamide,Radium-223,docetaxel + prednisone chemotherapy and mitoxantrone + prednisone chemotherapy,which included almost all of the current domestic treatment regimens.The patient lived for 57 months from the initiation of treatment.Conclusions For the patients with high-risk prostate cancer,especially for elderly patients,they should firstly be recommended the treatment of radical radiotherapy + 2-3 years of androgen deprivation therapy.For young patients,radical prostatectomy + extended pelvic lymph node dissection as part of multi-modal therapies is considered to be a positive choice.However,for the patients with risk factors or recurrence after radical prostatectomy,the postoperative radiotherapy should be performed in order to reduce biochemical recurrence and improve local control.Currently,the order of choice of abiraterone,enzalutamide or docetaxel + prednisone chemotherapy,is based on the patient's situation,past history of treatment,symptoms,side effects as well as other related clinical features.

Conformal radiotherapy is an important therapeutic strategy for primary hepatic carcinoma,which has high local control rate.However,the frequency of intra- and extra-hepatic recurrences is very high.Sorafenib is the only drug that has been demonstrated effectiveness to increase overall survival in advanced or metastatic hepatocellular carcinoma.The combination about radiotherapy and sorafenib can increase the radiosensitiveness,and inhibit the angiogenesis,and relieve the hypoxia,as well as synchronize the cell cycle.A lot of biological studies of sorafenib combined with radiotherapy have been camied out,and the phase Ⅰ -Ⅱ clinical trials are is in progress.
