1.Correlation between oxidative stress and trace elements in blood of patients with cancer

BACKGROUND: The occurrence and development of tumor may result from the reaction of a series of factors. Up to now, earlier detection, monitoring treatment, and prognosis assessment for tumor are very difficult. It can provide the theoretical foundation of rehabilitation and survival for cancer patients to study the mechanisms of occurrence and development of tumors, or investigate the changes of blood level and tissue distribution.OBJECTIVE: To investigate the changes of levels of some trace elements and serous prooxidant-antioxidant system of cancer patients.DESIGN: A controlled observation trialSETTING: Department of Laboratory Medicine, Mianyang CentralHospital.PARTICIPANTS: From September 1999 to December 2000,111 cancer patients were selected from the Third People's Hospital of Zigong, Yibin First People's Hospital of Sichuan Province, and Longchang County people's Hospital of Neijiang City, Sichuan Province.Of them, 21 cases were of liver cancer, 16 of gastric carcinoma, 15 of colorectal cancer, 11 of breast cancer, 13 of lung cancer, 13 of esophageal cancer, 7 of brain cancer and 15 of other cancers. At the same time; 36 control subjects were recruited from those who came to the hospital for health examination.METHODS :Blood samples of subjects after overnight fasting were pre pared by collecting venous blood in 5-mL Vacutainer tubes (Becton Dickinson, American). After 2-3 hours, the blood samples were centrifuged at 3 000 r/min for 15 minutes. 2.0 to 3.0 mL of serum was separated. The sera were stored in the refrigerator until analysis. Serum glutathione(GSH)level was determined by dithiobis-2-nitrobenzoic acid colorimetry. Xanthine oxidase (XOD) concentration was determined with xanthine-nitrotetrazole colorimetry. Glutahione oxidase (GSHPx) activity was determined with dinitrobenzoic acid colorimetry. Malondialdehyde(MDA) concentration in serum was measured with thiobarbituric acid reaction. Vitamin C (Vit C),vitamin E(Vit E) and total antioxidant power(TAOP) levels were measured with phenanthroline colorimetry. Albumin concentration was determined with bromcresol purple. Serum transferrin (Trf) and ceruloplasmin (CER)concentrations were determined with immunonephelometry. The concentrations of Cu, Zn, Fe, and Se were measured with flame atomic absorption spectroscopy.MAIN OUTCOME MEASURES: MDA (as lipid peroxidation marker),trace elements copper, zinc, iron, and selenium and their transport proteins (albumin, transferrin and ceruloplasmin); antioxidation status markers: the concentrations of XOD, GSH, GSHPx, vitamin C, and vitamin E, and total antioxidant power levels.RESULTS: It was shown that lipid peroxidation ( measured as MDA)was significantly higher in cancer patients than in healthy controls [(5.21±1.05) nmol/L vs (4.04±0.68) nmol/L,P < 0.001], and the TAOP level was significantly decreased in cancer patients than in the health controls[(4.34±0.980) U/L vs (5.87±0.93) U/L,P < 0.001]. There were not obvious changes of antioxidation components (XOD, GSH, GSHPx, vitamin C,and vitamin E). Serum albumin concentration was found to be significantly lower in the cancer patients than in the health controls [(34.19±6.94)g/L vs (42.34±4.89) g/L ,P < 0.001], and serum ceruloplasmin concentration was found to be significantly higher in the cancer patients than in the health controls [(0.371 ±0.031) g/L vs (0.346±0.026) g/L,P < 0.05] butserum transferrin concentration remained unaltered (P > 0.05). As com pared with the healthy controls, serum copper level was significantly increased[(19.27±4.74) μmol/L vs (14.29±2.71) μmol/L, P < 0.001], serum selenium levels was significantly decreased[(1.175±0.333 0 μmol/L vs (1.413±0.446) μmol/L,P < 0.001)]. However, the concentrations of zinc and iron remained unchanged. Correlation was observed between copper and MDA levels (r=0.281, P=0.003) in the cancer patients but not in the healthy controls. Moreover, a correlation was also observed between serum iron and MDA levels in the patients with liver cancer (r=0.680,P=0.001).CONCLUSION: The presence of an association between oxidative stress and some trace elements was found in cancer patients; however, the results are possibly inconsisteut because of different cancer types, cancer grades,or other characteristics of the patients engaged in the test.
2.Effect of propolis flavone on expression of matrix metalloproteinase-9 in human umbilical vein endothelial cell

BACKGROUND: It has been proved that matrix metalloproteinase-9 (MMP-9) has a correlation with stability of atherosclerotic plaque, and propolis flavone plays a role in anti-atherosclerosis. OBJECTIVE: To investigate the effect of propolis flavone on expression of MMP-9 in human umbilical vein endothelial cell (HUVEC). DESIGN, TIME AND SETTING: The comparative observation was performed at the Institute of Life Science, Taishan Medical University from January to August 2008. MATERIALS: The propolis flavone was extracted by the Institute of Life Science, Taishan Medical University. The neonatal umbilical cord with 1 hour was supplied by Tai’an Central Hospital, and the informed consent was obtained from the puerperant. METHODS: HUVEC was cultured and identified in vitro, followed by treated with propolis flavoneat with different concentrations for 24 hours. The blank was used as the control group. MAIN OUTCOME MEASURES: The effect of propolis flavone on expression of MMP-9 in HUVEC was detected by cytoimmunochemical staining and ELISA. RESULTS: Propolis flavonecan with all concentrations can restrain the expression of MMP-9 after co-culture with the cells for 24 hours, which showed in a dose-dependent manner. CONCLUSION: The propolis flavone can obvious depress the expression of matrix metalloproteinase-9 in HUVEC.
3.Clinical analysis of emergency coronary after interventional reperfusion arrhythmia

Objective To explore the characteristics of the reperfusion arrhythmia after acute myocardial infarction (AMI) pa-tients underwent emergency coronary interventional therapy ,in order to reduce the incidence of serious arrhythmia ,and improve the prognosis and quality of life .Methods Clinical data of 147 patients from January 2009 to May 2012 with acute myocardial infarction occurrence reperfusion arrhythmia after underwent emergency coronary interventional treatment were analyzed .Results In 147 ca-ses ,62 cases of reperfusion arrhythmia occurred .Infarct-related artery for the left anterior descending branch (LAD) 33 patients (LAD group ) ,left circumflex artery (LCX) 7 cases (LCX group ) after surgery ,the right coronary artery(RCA) 22 cases (RCA group) .The tachyarrhythmia in LAD group was higher than RCA group(P<0 .05);the bradyarrhythmia in RCA group was higher than LAD group (P<0 .05) .Opening of the infarct-related artery<6 h arrhythmia had 43 cases occurred ,and >6 h arrhythmia had 19 cases occurred ;Arrhythmia after acute myocardial infarction occurred mainly in infarct related artery opened earlier .Conclusion AMI occurrence reperfusion arrhythmia after underwent emergency coronary interventional treatment have high incidence ,there is a correlation between the arrhythmia type and infarct-related artery ,also has a correlation between incidence and reperfusion time .
4.Effects of quercetin on proliferation and expression of P38MAPK and HMGB1 protein in neonatal rat cardiac fibroblasts induced by TNF-α

Objective To study the effect of quercetin on proliferation and expression of P38MAPK and HMGB1 protein in neonatal rat cardiac fibroblasts induced by TNF-α.Methods Purified cardiac fibroblasts were obtained by trypsin digestion and differential adherence method.The proliferation of cardiac fibroblasts was detected by MTT assay.The expression of P38MAPK and HMGB1 protein was detected by Western blot.Results Quercetin had no effect on the proliferation of cardiac fibroblasts in the basal state but inhibited the proliferation of cardiac fibroblasts induced by TNF-α,and the A value of each group was increased with the increase of quereetin concentration(P<0.05).The exspression of P38MAPK、HMGB1 were decresed with the icrease of quercetin.Conclusion Quercetin may inhibit the proliferation of cardiac fibroblasts induced by TNF-α.The mechanism may be inhibit the expression of HMGB1 through P38MAPK signaling pathway.

6.Application of laser scanning 3D technology in rhinoplastic prothesis

Objective To investigate the effect of application of 3D printing technology in the rhinoplastic prothesis with the help of the laser scanning 3D technology.Methods Before the rhinoplastic prothesis,we got the whole facial morphology of patients by laser scanning technology,and printed out the facial model which was 1:1 with the real and then discussed with patients to confirm the ideal nose three-dimensional effect.In the process of operation,doctors were guided by the data for the procedure.Results Among 33 patients who were followed up postoperatively,only one patient's nosal tip dropped,then were repaired with her ear cartilage,both of us found satisfactory effect after repair,and without postoperative complications.The rest 32 patients were satisfied with their nose.Conclusions The laser scanning 3D technology is worth trying and using in plastic surgery.
7.Difference of morphology and proliferation ability between diabetes mellitus-derived ADSCs and normal ADSCs

Objective To investigate the morphological characteristics and proliferation ability of diabetes mellitus-derived adipose-derived stem cells (ADSCs) by comparing with normal-derived ADSCs.Methods The ADSCs could be achieved from diabetes mellitus' adipose tissue by the similar method of normal ADSCs isolation and culture,and then the differnce was compared between the two groups about their morphologies under microscope,and the proliferation ability of two groups was determined by CCK-8.Results ADSCs in patients with diabetes were obvious hypertrophy and irregular in morphology.A values of ADSCs in diabetes and non-diabetes were 0.210+0.002 and 0.390+0.006 in 1st day;0.250+0.015 and 0.443+0.023 in 2nd day;0.368+0.014 and 0.726+0.033 in 3rd day;0.368+0.014 and 0.726+0.033 in 4th day;1.767+0.072 and 3.153+0.067 in 5th day and 1.810+0.072and 3.170+0.021 in 6th day,respectively.The difference was statstically significant at the beginning from 4th day (P<0.05).Conclusions There is obvious difference of the microscopic morphology between the diabetes mellitus-derived ADSCs and normal-derived ADSCs.The proliferation ability of the diabetes mellitus-derived ADSCs is lower than normal-derived ADSCs.These difference could be closely related to ulcerous non-healing wounds in patients with diabetes mellitus.
8.Predictive value of preoperative and postoperative serum tumor markers CEA, CA19-9, and CA72-4 in the diagnosis of gastric cancer recurrence at different stages

Objective:To investigate the predictive value of preoperative and postoperative serum tumor markers, namely, carcinoem-bryonic antigen (CEA), carbohydrate antigen 19-9 (CA19-9), and CA72-4, in the diagnosis of gastric cancer recurrence at different stag-es. Methods:Analysis was performed in 564 patients who underwent curative resection for gastric cancer between January 2002 and March 2007, received no chemotherapy at our hospital, and received complete follow-up according to the schedule determined pro-spectively. The values of CEA, CA19-9, and CA72-4 were evaluated before and after surgery. Results:In the pTNM-Ⅰ and pTNM-Ⅱ stage groups, patients with positive preoperative serum CEA, CA19-9, and CA72-4 levels showed recurrence rates of 50.0%, 24.1%, and 22.6%, respectively. Similarly, the recurrence rates of patients with positive postoperative serum CEA, CA72-4, and CA19-9 levels were 42.9%, 21.7%, and 14.3%, respectively. Multivariate analysis showed that the positive preoperative serum CEA level could be an inde-pendent factor of recurrence. In the pTNM-Ⅲ stage group, the recurrence rates of patients with positive preoperative serum CEA, CA19-9, and CA72-4 levels were 50.0%, 55.2%, and 47.6%, respectively. The recurrence rates of patients with positive postoperative se-rum CEA, CA19-9, and CA72-4 levels were 75.0%, 66.7%, and 66.7%, respectively. Multivariate analysis showed that high postoperative serum CA72-4 levels could be an independent factor of gastric cancer recurrence. Conclusion:Serum tumor markers exhibited differ-ent predictive values in different pTNM stages. Preoperative CEA level could be used to predict recurrence in patients with pTNM-Ⅰ and pTNM-Ⅱ stages of gastric cancer. Moreover, postoperative CA72-4 level could be used to predict recurrence in patients with pTNM-Ⅲ stage gastric cancer.
9.Clinical Outcome of Foley Catheter Balloon Versus Prostaglandins in Labor Induction of Late Pregnancy:A Meta Analysis

Objective To compare the efficacy and safety of Foley catheter (FC) and prostaglandins (PG) for cervical ripening used in labor induction of late pregnant women with unfavorable cervix. Methods Several databases including Cochrane Library,Embase,PubMed,Elsevier,Wanfang Medical Database, CNKI, VIP were searched for collecting the related literatures according to criteria for inclusion and exclusion.Meta analysis was performed by applying software Review Manager 5.2. Results Thirty randomized controlled trials were included.The FC group presented significantly lower risk of excessive uterine contraction [OR= 0.38,95%CI(0.19,0.75),P<0.05] than the PG group.There was no significant difference between the two groups in terms of Cesarean section rate, vaginal delivery rate, Bishop score improvement, duration of induction to delivery, neonatal birth weight, or in neonatal outcome. Conclusion This study shows that FC has good promoting effect of cervical maturity and maternal ̄ neonatal outcomes, and rarely produces excessive uterine contraction.It is safe and equally effective as PG does.The approach is easy to operate and worth to apply in clinical use.
10.Effect of propolis water extract on apoptosis of human umbilical vein endothelial cells in vitro
