1.Treatment for the type Ⅱ-Ⅲ pilon fractures by external fixation with limited open reduction and internal fixation

Object:To observe the clinical effect of the treatment for the type Ⅱ-Ⅲ Pilon fracture by external fixation with limited open reduction and internal fixation. Methods:from year 2000 to year 2006,17 patients suffered from type Ⅱ-Ⅲ Pilon fractures were treated by external fixation with limited open reduction and internal fixation. Among these patients,11 are males and 6 are females,aged from 20 to 54 years old with an average of 36.5 years old. According to Rueeli、ALLgower classification:3 cases were type Ⅱ,14 cases were type Ⅲ. Results:All patients were followed up 6 to 24 months with mean 11 months,14 cases were excellent and 3 cases were well evaluated by Hefet evaluation system. Conclusion:Treatment for the type Ⅱ-Ⅲ type Pilon fractures by external fixation with limited open reduction and internal fixation is a favorable method which is characterized by its simple skills,reliable fixation and satisfactory clinical effect.

Objective To investigate the fluorescence quantitative polymerase chain reaction(FQ-PCR)in the giant cell viral hepatitis in value.Methods 50 cases of serum IgM HCMV-positive patients with hepatitis and 25 cases of healthy people to do the control group respectively serum FQ-PCR detection of its content HCMVDNA its detection rate compared with the liver function and the relationship.Results Of 50 cases of serum IgM HCMV-positive patients with hepatitis HCMVDNA>103Copies/ml those 35 cases,HCMVDNA positive to the negative group than HCMVDNA serum total bilirubin(TBiL)alanine aminotransferase(ALT)aspartate aminotransferase(AST)to the high statistics P <0.05 the difference was significant.25 cases of healthy control group and HCMVDNA HCMV-IgM were negative.Conclusions FQ-PCR in cytomegalovirus infection with hepatitis reliable early diagnosis of the characteristics through HCMVDNA content of the clinical evaluation of drug efficacy has certain clinical value.
3.Relationship between cyclic adenosine monophosphate and auqaporin 8 expression and distribution in human amnion-derived cells

Objective To study the effect of cyclic adenosine monophosphate on aquaporin 8 (AQP8) expression and distribution in human amnion-derived WISH cells.Methods Human amnionderived WISH ceils were cultured.The cells were divided into control group and study group at random.The study group was established by exposure to various concentrations of 8-Br-cAMP.Western analysis was used to quantify AQP8 expression levels. RT-PCR was used to quantify AQP8 mRNA expression levels.Immunofluorescence was used to determine the localization of AQP8 in WISH cells.Results With increase of cAMP,AQP8 mRNA and protein expressions significantly increased in WISH cells in vitro.When the concentration of cAMP reached 200 μmol/L,AQP8 mRNA and protein expressions were highest.Incubation with cAMP (200 μmol/L) for 2 hours resulted in a 2-fold increase in AQP8 mRNA level,and incubation for 8 hours resulted in a peak.After incubation for 16 hours AQP8 mRNA level began to descend,and after 24 hours it decreased to baseline.Incubation with cAMP for 8 hours AQP8 protein level began to increase.Incubation for 24 hours resulted in a peak in AQP8 protein level,and after incubation for 48 hours it began to decline.By immunofluorescence microscopy after incubation with cAMP (200 μmol/L) cells,AQP8 labeling in plasma membrane was enhanced and intracellular AQP8 labeling was decreased.Conclusion The cAMP triggers translocation of AQP8 from cytosel to the plasma membrane via vesicle-transporting related protein instead of AQP8 itself,cAMP may upregulate the transcription of target gene protein kinase A.The cAMP may be the critical regulatory medium of AQP8 in WISH cells.

5.Expression of aquaporin-8 in the hepatocytes of pregnant rats with intrahepatic cholestasis

Objective To investigate the alteration on the expression levels of protein and messenger RNA(mRNA)of aquaporin-8(AQP8)in the hepatocytes of pregnant rats with intrahepatic cholestasis induced by ethinylestradiol.Methods A total number of 20 15-day pregnant rats were randomly divided into two groups:control group,intrahepatic cholestasis of pregnancy(ICP)group.In ICP group,rats were subcutaneously injected with ethinylestradiol(2.5 mg·kg-1·d-1)for 5 days to make the model of ICP.In control group,rats received subcutaneous injection of appropriate volume of propylene glycol for 5 days.The protein expression and subcellular localization of AQP8 in the hepatocytes of pregnant rats were detected using immunohistochemical methods.The mRNA expression of AQP8 in the hepatocytes was assayed by RT-PCR method.Results The model of ICP of pregnant ats was made successfully.Expressions of AQP8 protein and mRNA were detected in two groups.AQP8 protein level in ICP group was 10.8±2.4,significantly decreased than in control group 17.1±2.2(P<0.05).AQP8 mRNA level in ICP group was 1.07±0.11,significantly higher than in control group 0.80±0.11(P<0.05).Conclusion Down-regulated AQP8 protein expression and up-regulated AQP8 mRNA expression in the hepatocytes of pregnant rats with intrahepatic cholestasis may contribute to the pathogenesis of ICP.

9.The Relationship between the Expression of Bone Morphogenetic Protein-2 and Matrix Metalllopro Teinase-9 in Fetal Membrane and Premature Rupture of Membranes

Objective:To study the correlation between the bone morphogenetic proteins (BMP-2) and matrix metalloproteinase-9(MMP-9) in fetal membrane and premature rupture of membranes. Methods: 18 samples were selected separately from each of the following groups: puerpera with term pregnancy in labor, puerpera with term premature rupture of membranes(PROM), puerpera in preterm labor, puerpera with pre-tenm PROM and puerpera with term pregnancy not in labor (control group). After labor, fetal membranes in the rupture site were taken out. Immunobistochemistry and pathologic image multimedia operation system were adopted to semiquantitative analyze the area integrated optical density (AIOD) of the positive expression of BMP-2 and MMP-9. Results:①The AIOD of BMP-2 and MMP-9 positive expression in fetal membranes was higher in term pregnancy in labor group, term PROM group, preterm labor group and PROM group than that in control group. There was statistic difference ( P < 0. 0071). ②The AIOD of BMP-2 and MMP-9 in term PROM group was higher than that in term pregnancy in labor group(P=0.0065,P=0.0069), which was higher in PROM group than that in preterm labor group (P=0.0069, P = 0.0052) and term PROM group P=0.0037, P =0.0065),③There was obviously positive correlation between BMP-2 and MMP-9 expression in the fetal membranes in five groups. ( r_s =0.76159,P<0.0001). Conclusions: The expression of MMP-9 and BMP-2 is closely relevant to the repture of membranes. There is a correlation between BMP-2 and MMP-9 expression in fetal membranes.
10.ANALYZING HLA HAPLOTYPE OF THE LOCI HLA-A, -B, AND -DRB1 IN MONGOLIA ETHNIC GROUP

Objective To investigate HLA-A,-B and -DRB1 allele and HLA-A-B-DRB1 haplotype frequencies in Mongolia ethnic group. Methods HLA-A, -B, -DRB1 allele and haplotype in the Mongolia ethnic group were investigated based on 93 individuals by PCR- sequence-based typing (SBT) method. Results Twenty-one alleles were detected for HLA-A, 44 for HLA-B, and 26 for HLA-DRB1. The most frequent alleles were HLA-A*2402(0.2097), HLA-B*1302(0.0699), and HLA-DRB1*0701(0.1237). The most common HLA-A-B-DRB1 haplotype were A*3001-B*1302-DRB1*0701, A*0101-B*3701-DRB1*1001, followed by the A*0201-B*4601-DRB1*0901, A*2402-B*4801-DRB1*1101, A*2402-B*5201-DRB1*1501, A*3201-B*3503-DRB1*1301, and A*3303-B*5801-DRB1*0301, which were also presented in Chinese populations. Conclusion The data can be used in forensic and paternity tests to estimate the frequency of a DNA profile or anthropologic research. The characteristics of the distribution of HLA alleles revealed that Mongolia ethnic group is characterized by northern Mongolian Chinese.
