1.Assessment of effectiveness of individual rehabilitation therapy in nonspecific low back pain

Objective To explore the effectiveness of individual rehabilitation therapy in patients with nonspecific low back pain.Methods A total of 50 patients with nonspecific low back pain ( NLBP) between January 2013 and January 2015 were enrolled in this study.Individual rehabilitation therapy was performed based on their different pathogenesis and risk factor of low back pain.Visual analogue score (VAS),Oswestry disability index ( ODI) and Japanese orthopaedic association ( JOA) were used for the assessment of effectiveness of individual rehabilitation therapy.Results Compared with pre-rehabilitation, VAS and ODI scores were significantly decreased at two-week after rehabilitation(P<0.05),and were further decreased at six-week after rehabilitation(P<0.01).The mean score of JOA of 50 patients before rehabilitation was (10.83 ±2.94) score,which was decreased to (24.37±7.55 )score at six-week after rehabilitation(P<0.05),with 12 patients were cured,32 were improved obviously,6 were improved moderately.The rate of efficacy was 100%.Conclusion Individual rehabilitation therapy was an effective option for NLBP,with significant decrease of pain.
2.Relationship between personality. positive psychological resource and depression in elderly:a path model

ObjectiveTo explore the relation between personality and depression in elderly,particularly to investigate mediating effect of positive psychological resource.MethodsThe eysenck personality questionaire-revised short scale for Chinese(EPQ-RSC),self-rating depression scale(SDS) and scales for positive psychological resource were performed by 270 elderly in community of Tianjin.Then correlation analysis,regression analysis and path analysis were accomplished to explore association between those variables.Results Neuroticism and Psychoticism personality correlate positively with depression ( r=0.45,0.48) and contribute about 30% variance of it (F=49.95 ).Positive psychological resource such as optimism,resilience and self-efficacy correlated negatively with depression ( r =- 0.41,- 0.39,- 0.20 ) and contributed 19% variance of it (F =30.02 ).The results of path analysis showed positive psychological resource mediated the relation between personality and depression also.ConclusionNeuroticism and Psychoticism are important risk factors of depression in elderly.Positive psychological resource such as optimism,resilience and self-efficacy are key protecting factors for depression and they mediate the relation between vulnerable personality and depression.
3.External quality assessment for the quantitative detection of HCV RNA in China

Objective Anti-RNase virus-like particles containing HCV RNA 5'-UTR were used as positive samples in national external quality assessment ( EQA ) to evaluate the competency of clinical Laboratories for the quantification of HCV RNA and analyze the possible problems of domestic kits. Methods The quality control samples with target values in EQA panels were distributed nationally twice by National Center of Clinical Laboratory (NCCL) to participating laboratories for the quantification of HCV RNA in 2008 and 2009. Each panel consisted of 5 samples. All participants were required to carry out the detection and to return results in expected time. Positive samples were virus-like particles which had been calibrated against the WHO HCV International Standard (NIBSC96/798)and the results of positive samples from participants should be in the range of target value of logarithm ± 0. 5. The 2nd panel in 2008 contained the common HCV genotypes and the 2nd panel in 2009 contained serial diluted samples of genotype 1b. The results of positive samples detected with 3 different lots reagent (21001,21078 and 21097) from the 2nd EQA in 2009 were statistically analyzed using the analysis of variance, then Dunnett'S T3 and Tamhane'S T2 were used if heterogeneity of variance was found. Results There was 390 participating laboratories in 2008 and 428/426 in 2009. The percentages of laboratories within the range of target value of logarithm ± 0. 5 for varied genotypes were different. The percentages of laboratories for 1b were more than 91%, for 2a were 93.7% and 74. 2% ,for6 were 83.3% and 80. 3%. The CVfor the low-level sample was higher than that for the high-level sample in the same year. The numbers of laboratories reporting false-negative samples in 2008and the 2nd in 2009 were 5, 1 and 10 respectively. Statistical differences were found among the results of four quality control serum samples using 3 different reagents( F = 288.23, 324. 79, 291.98 and 261.16,P <0. 01 ). Conclusion The competency for detecting low concentration samples and samples with genotype 2a or 6 needs to be improved.
4.Systematic Evaluation onYupingfeng Granules Combined with Western Medicine in the Treatment of Children with Recurrent Respiratory Tract Infections

ObjectiveTo evaluate clinical efficacy ofYupingfeng Granules combined with western medicine for children with recurrent respiratory tract infection (RRTI).Methods CNKI, VIP, WANFANG DATA, SinoMed, PubMed, Embase, and Cochrane Library were searched for the randomized controlled trials (RCTs) that investigated the clinical and immune effects ofYupingfeng Granules in children with RRTI from inception to October 2015. Two researchers independently screened studies according to the inclusion and exclusion criteria, extracted data and evaluated the methodological quality of included studies. Then Meta analysis was performed through the software RevMan 5.3.Results A total of 8 articles involving 550 patients were included. The results of Meta analysis showed effective rate ofYupingfeng Granules combined with western medicine for children with RRTI [OR=4.16, 95%CI (2.49, 6.95),P<0.000 01], IgA level [MD=0.52, 95%CI (0.26, 0.78),P<0.000 1], IgG level [MD=2.75, 95%CI (1.25, 4.25),P=0.000 3], CD4+ level [MD=6.15, 95%CI (4.76, 7.54),P<0.000 01], CD4+/CD8+ level [MD=0.36, 95%CI (0.25, 0.47),P<0.000 01]. All of the above were superior to single western medicine treatment. There was no statistical significance in levels of IgM and CD8+(P>0.05). ConclusionYupingfeng Granules combined with western medicine can improve effective rate, antibody level and T-cell level in the treatment of children with RRTI. The overall clinical efficacy is better than single western medicine treatment. However, RCTs with large samples, multiple centers and high quality are needed for further verification.
5.External quality assessment of autoantibodies detection in clinical laboratories during 2006-2011 in China

ObjectiveTo evaluate the performance of antinuclear antibody (ANA) detection in clinical laboratories.Methods There were 2 external quality assessments (EQA) scheme for nuclear antibody detection.The panel consisting of 5 samples was distributed.Each participant laborotory of the EQA program was required to report the ANA qualitative results,patterns,titers and anti-double strain DNA (dsDNA) antibody,anti-extractable nuclear antigen(ENA) antibody,the percent agreements of which were calculated respectively.ResultsThe number of laboratories performing ANA test with IIF increased from 77.6% ( 149/192 )in 2006 to 82.2% (342/416) in 2011,while the number of laboratories performing ANA test with ELISA was in the range of 14.5% ( 53/365 ) and 16.0% ( 52/326 ).The positive percent agreements of IIF was over 98%.The positive percent agreement of ELISA were all over 90%.IIF showed more satisfying positive percent agreements than ELISA every year.Over 90% of the laboratories reported correct results for samples with granular ANA pattern except 0613 and 0624.Over 95% of the laboratories reported correct results for samples with homogeneous ANA pattern.Two samples with centromere pattern were correctly detected by 88.5% ( 161/182 ),79.0% ( 147/186 ) of the laboratories in 2007,while the sample with centromere pattern was correctly detected by 98.4% (299/304), which indicated an improvement in the detection of centromere pattern.In ANA positive results,the lowest percentage of the laboratories reporting the median result was 36% (94/261),while the highest percentage was only 85.5%(224/262).The satisfied results of anti-ENA antibody were over 90%.And those of anti-dsDNA antibody was over 85%.ConclusionsIIF is the most common method for ANA screening in clinical laboratories.ELISA is also used in some laboratories.The two methods reported satisfying results in ANA test.The detection of anticentromere antibodies is improved.But the results of ANA titer reported are unsatisfactory. ANA detection in routine practice needs to be improved by standardization.
6.ARTERIO-VENOUS ANASTOMOSES (AVA) OF THE RABBIT'S EAR AS DEMONSTRATED BY THE METHOD FOR ACETYLCHOLINESTERASE

Fresh subcutaneous tissue from the rabbit's ear was processed with copper ferrocyanide method (Karnovsky and Roots, 1964) for demonstrating acetylcholinesterase. The arterio-venous anastomoses were revealed by the abundant "cholinergic" nerve fibers present around its arterial and intermediate segments. These nerves formed a dense network around these segments of AVA which gives AVA a conspicuous and outstanding picture under the microscope. 262 AVA were identified and their length and outer diameter of the intermediate segment were measured. The longest was 620.6 ?m, the shortest 81.9 ?m, and the averaging length was 254.8 ?m. Their outer diameters fell between 109.2~20?m, with an average of 52.2 ?m. The authors condered the measurements of the intermediate segment of the AVA give more significant information than the total length of the AVA.The AVA were classified into the following types according their morphological characteristics: the simple type (including the long and thin type, and short and thick type) and the complex type (including bifurcate, trifurcate, quarifurcate, compound, and circular types). This method of demonstrating AVA morphology was compared with former observations made directly in vivo or by histological methods and it serves as an new tool for the investigation of AVA.

Objective To observe the effects of statins on ATP-binding cassette transporter 1(ABCA1) gene expression in neurons and astrocytes.Methods Primary human astrocytes and neuron cell line HCN-2 were incubated with simvastatin and pravastatin at different concentrations and under different conditions.Using RNA isolated from the cultured cells,we performed quantitative PCR to measure the ABCA1 gene expression in astrocytes and neurons.The protein expression of ABCA1 was measured by Western blot.Results The two statins significantly decreased the ABCA1 gene and protein expressions.Compared with pravastatin,simvastatin significantly reduced the expression of ABCA1 in neurons and astrocytes by 95% and 75%,respectively(both P
8. Mesoporous silica nanocapsules loaded liposomes negatively enhance magnetic resonance T2 weighted imaging

Objective To detect the relaxation property of mesoporous silicananocapsules loaded liposomes (MSN-LIPO), and to explore their functions of enhancing MRI T2 weighted imaging. Methods In vitro, MSN-LIPO with different ironconcentrations were scanned by MRI system to detect their relaxation properties. In vivo, six BALB/c nude mouse models with subcutaneous malignant glioma were established and randomly divided into two groups, which were injected with the same concentration of MSN-LIPO intratumorally and intravenously, respectively. The mice in the two groups were scanned by MRI before and after injection, and the image characteristicswere studied and the signal intensity of tumors was statistically analyzed. Results In vitro MRI imaging showed that the signal intensities of the images were gradually decreased with the increase of iron concentrations in MSN-LIPO, and 1/T2 had a good linear relationship with the concentration within a certain range, with the relaxation rate (r2) being 413.7 mmoL-1 s-1. MRI imaging of BALB/ c nude mouse models with malignant glioma showed that the signal intensity of the tumor after intratumor injection of MSN-LIPO was significantly lower than that before intra-tumor injection (15.34±1.24 vs 211.44 ± 5.34, t=36.38, P < 05), and the signal intensity of the tumor after intravenous injection of MSN-LIPO was significantly lower than that before intravenous injection (179.00 ± 4.35 vs 235.99 ± 5.17, t=14.34, P <0.05). Conclusion MSN-LIPO has good magnetic relaxation property and good MRI T2 weighted contrast enhancement function in vivo, suggesting that it can be used as a MRI T2 contrast agent.
9.The relationship between microRNA-7 and phosphatase and tensin homolog in Sj?gren's syndrome

Objective:To study the expression of miRNA-7 in B lymphocytes of primary Sj?gren's syndrome (pSS) and its relationship with phosphatase andtensin homolog deleted (PETN) and disease activity.Methods:Twenty newly diagnosed outpatient and inpatient pSS patients were used as case group collected from January 2017 to December 2019 of Qinghai Provincial People's Hospital. Twentyhealthy persons were used as the control group. Disease-related indicators of the case group were collected. Quantitative polymerase chain reaction (RT-qPCR)was used to detect miRNA-7 and PETN mRNA expression in B lymphocytes of the two groups and the consistency between miRNA-7 expression in the plasma and B lymphocytes of the case group was analyzed. Western Blotting method was used to detect the PETN protein in B lymphocytes of the two groups. Correlation analysis was used to analyze the relationship between miRNA-7 expression in B lymphocytes and disease activity in the case group. Linear regression analysis was performed between miRNA-7 and PETN mRNA.Results:The expression of miRNA-7 (0.53±0.17) in the B cells increased and the expression of PTEN mRNA (0.88± 0.24) and protein (0.51±0.12) in the case group were reduced compared with that of miRNA-7(0.39±0.11), PTEN mRNA(2.32±0.30) and protein(1.03±0.21) of the control group. The above differences were statistically significant ( t=2.990, P<0.05; t=16.98, P<0.05; t=8.41, P<0.05). Linear regression showedthat PTEN miRNAwas negatively correlated with miRNA-7 ( b=-0.78, P<0.01), the expression of miRNA-7 in the case group was positively related with EULAR Sj?gren′s syndrome Disease Activity Index (ESSDAI), IgG, IgA, anti-SSB and was negatively correlated with C4 and WBC. Conclusion:There is a certain relationship between miRNA-7 and disease activity. MiRNA-7 may participate in the pathogenesis of pSS byregulating PETN in B cells of pSS. MiRNA-7 has certain clinical value for disease activity evaluation.
10.A study of serum cartilage oligomeric matrixprotein and matrix metalloproteinase-3 concentration in osteoarthritic rabbit models

Objectiye To study the levels of cartilage oligomeric matrixprotein (COMP) and matrix metalloproteinase-3 (MMP-3) in the serum fluid of osteoarthritic rabbit models and their relationships with the severity of pathological changes, so as to investigate their correlation with osteoarthritis(OA). Methods The osteoarthritic animal models were get from immobilizing the right knees of 18 rabbits in full extension using plaster cast. Knee joint pathological changes of 2,6 weeks were examined for pathological severity of OA; ELISA sandwich method was used to measure the levels of COMP and MMP-3 in serum before and after modeling( at 2, 6 weeks respectively); X ray of model keens was also obtained in different period.Correlation analysis was performed to demonstrate the relationship between the levels of COMP, MMP-3 in the serum and the pathological severity of OA. Results ( 1 ) Morphological observations: immobilizing the right knees of rabbits in full extension using plaster cast was a reliable methed for osteoarthritic animal models and the typical histopathologic character was seen; the severity of osteoarthritisgradually increased with time extended. (2) The levels of COMP[(3.64 ±0. 18)μg/L], MMP-3 [(1.99 ±0. 81 ) μg/L]in the serum of 2 weeks osteoarthritic animal models were higher than those before immobilizing with plaster cast [COMP(3.35 ±0. 20) μg/L,MMP-3( 1.61 ±0. 71 ) μg/L]. The levels of COMP[(3.96 ±0. 44) μg/L],MMP-3[(3.44 ±0. 91) μg/L] of 6 weeks were much higher,with a significant difference(P <0.05). The levels of COMP, MMP-3 in serum had a linear correlation with the pathological severity of OA (r >0. 710,and P < 0. 05 ). Conclusion The levels of COMP and MMP-3 in serum can help to predict and evaluate the progression of OA.
