3.Effect of microparticles on echinocandin B production by Aspergillus nidulans.

Anidulafungin is an effective antifungal medicine, which can inhibit activities of candida in vitro and in vivo. Echinocandin B (ECB) is the key precursor of Anidulafungin, thus the price and market prospect of Anidulafungin is directly due to the fermentation titer of ECB. In this study, Aspergillus nidulans was used for ECB fermentation, and the influence of adding microparticles on ECB fermentation was studied, such as talcum powder, Al2O3, and glass beads. The particle size and concentration were the key factors for mycelium morphology and ECB production, and ECB production could reach 1 262.9 mg/L and 1 344.1 mg/L by adding talcum powder of 20 g/L (d50 = 14.2 μm) and 7 glass beads (6 mm), an increase by 33.2% and 41.7%, respectively. The results indicated that the mycelium morphology of filamentous microorganisms and the product yield of fermentation could be improved by adding microparticles remarkably, and it provide an important method for the fermentative optimization of filamentous microorganisms.

4.CpG Island Methylation Regulates BNIP3 Gene Expression in Gastric Cancer Cells

Objective To detect the methylation status of the promoter of BNIP3 gene in gastric cancer cell lines MKN1,and to explore the mecha?nism of DNA methylation regulating the expression of BNIP3 in gastric cancer cells. Methods The methylation status of BNIP3 promoter was de?tected by bisulfate sequencing PCR. Reverse transcription PCR was used to evaluate BNIP3 mRNA expression. MKN1 cells were treated with 5?Aza?2′?deoxycytidine(5?Aza?CdR),and after the treatment,the methylation status and BNIP3 mRNA expression were observed. Chromatin immuno?precipitation(ChIP)was used to determine the combination of BNIP3 with DNA methyltransferase 1(DNMT1). Results The promoter DNA of BNIP3 in MKN1 cells was in state of hypermethylation. Compared to the control group,methylation status and mRNA expression of BNIP3 in the drug treatment group(the 5?Aza?CdR concentration was 10μmol/L)were reversed,which showed statistical differences(P<0.05). 5?Aza?CdR inhibited the combination of BNIP3 with DNMT1. Conclusion CpG island methylation regulates BNIP3 gene expression in MKN1 cells. DNA methylation is related with the binding between the promoter of BNIP3 and DNMT1.
5.Effect of adipose tissue extract of greater omentum on human fibroblasts in vitro

Objective To study effect of the adipose tissue extract of greater omentum on human fibroblasts. Method The effect of the adipose tissue extract of greater omentum on human fibroblasts was observed by inverted microscope, MTT, flow cytometry, transmission electron microscope. Results The growth of fibroblasts was inhibited by the adipose tissue extract of greater omenturn. The apoptosis of fibroblasts was not inhibited, and the cell function of fibroblasts was inhibited and the mitosis of fibroblasts was affected. Conclusions The growth of fibroblasts can be inhibited by the adipose tissue extract of greater omentum. The formation and development of biliary cicatricial constriction may be affected by the adipose tissue extract of greater omentum.
6.A COMPARITIVE STUDY ON IODINE NUTRITIONAL STATUS OF STUDENTS IN ZHOUSHAN ISLANDS

Objective: To measure the iodine nutritional status of students in Zhoushan Islands and evaluate the advantages and disadvantages of iodized salt prophylaxis. Methods: A comparative study was carried out in 12 classes which were selected from Dinghai(iodized salt district) and Daishan(non-iodized salt district) of Zhoushan Islands by random sampling. Mann-Whitney test was used to compare the urinary iodine concentration and dietary iodine intake of the two groups. Spearman correlation was used to look for the correlation between urinary iodine concentration and dietary iodine intake in the two groups. Ordinal regression was used to analyze the factors that affect the urinary iodine concentration. Poisson regression was used to analyze the risk factors of goiter.Results: The urinary iodine concentration in non-iodized salt district was 129 ?g/L,which was lower than 169 ?g/L in iodized salt district(u=6.302,P=0.000) , whereas the daily iodine intake in the two groups was 105 ?g/d and 184 ?g/d respectively, the difference was also significant(u=4.710,P=0.000). The influence of sex and iodized-salt intake was significant in the final regression model (P0.05). D xylopyranosyl] 2,7 octadienoyl} acacic acid 28 O ? D glucopyranosyl (1→3) [? L arabinofuranosyl (1→4)] ? L rhamnopyranosyl (1→2) ? D glucopyranosyl ester (1) was separated and identified. 8.Advances and Perspectives on Genetic Modification of Hevea brasiliensis

As the major commercial source of natural rubber,Hevea brasiliensis attracts much attention.However,the heterozygous nature,long breeding cycle are strong limitations for conventional breeding.While genetic engineering,which can be used to widen the germplasm base and produce desirable agronomic traits quickly and efficiently,offers a viable alternative approach to complement traditional breeding.Comprehensive analysis indicates that in the past two decades,with calli derived from immature anther or integumental tissues of immature fruit as receptors,both biolistic and Agrobacterium-mediated transformation methods were employed for developping rubber genotypes with improved latex yield,tolerance to tapping panel dryness syndrome,producing high-value recombinant proteins,etc.Being recalcitrant to tissue culture,the transformation efficiency of Hevea is comparatively low,and the procedures are still needed to optimize.Finally,breeding objectives and strategies to improve transformation efficiency were also proposed in the review.
9.Characteristics of the Children Abandoned Treatment after Cerebral Palsy

ObjectiveTo investigate the time, causes and modes of the cerebral palsy children abandoned treatment. MethodsThe information of 50 cerebral palsy children who abandoned treatment were analysed retrospectively. Results and ConclusionCerebral palsy children abandoned their treatment in 3~9 months after treatment mostly. The main causes included their parents had little cognition and confidence to the treatment, or were in financial problem. 10 cases were without any treatment, 18 cases tried unofficial treatment, 19 cases accepted consolatory treatment, and 3 cases fell back on superstition.
10.A new labdane diterpenoid glycoside from Nervilia fordii.

To study the chemical constituents of Nervilia fordii (Hance) Schltr., various chromatographic methods were used, including D101 macroporous resin, silica gel, ODS and preparative HPLC chromatographic techniques. A new labdane diterpenoid glycoside named as nervilifordoside A was isolated from 60% EtOH extract of Nerviliafordii. The structure of compound 1 was elucidated as 12, 17-epoxy-3-hydroxy-17-methoxy-labdan-13-en-16, 15-olide 3-O-alpha-rhamnopyranosyl-(1 --> 2)-O-beta-glucopyranoside on the basis of HR-MS, 1D and 2D NMR spectroscopic data as well as chemical methods.
