1.Determination of Plasma Ozagrel Concentration by RP-HPLC

OBJECTIVE:To determine the concentration of ozagrel in human plasma by RP-HPLC.METHODS:The sample separation was performed on a SinoChrom OPS-AP column.The mobile phase consisted of 0.025 mol?L-1 potassium dihydrogen phosphate buffer solution-acetonitrile(85∶15) with a flow rate of 0.8 mL?min-1 and a detection wavelength of 274 nm.The internal standard was levodropropizine.RESULTS:The linear range of ozagrel was 0.7~21.3 ?g?mL-1,(r=0.999 6).The extraction recovery of ozagrel was 83.78%~89.14% and the methodological recovery of it was 101.95%~106.68%.The intra-day RSD and inter-day RSD were 1.33%~3.27% and 2.72%~5.97%,respectively.CONCLUSION:This method is simple,rapid,accurate and reproducible,and it is applicable for the pharmacokinetic study of ozagrel in human plasma.
2.Clinical features of recurrent biliary pancreatitis and its predisposing factors

Objective Toinvestigatetheclinicalfeaturesofrecurrentbiliarypancreatitisandrelatedpredisposingfactors.Methods Ato-tal of 272 patients with biliary pancreatitis who were admitted and treated in Jiangbei District People′s Hospital from January 2008 to Decem-ber 2014 were enrolled and divided into recurrence group (56 patients with recurrent biliary pancreatitis)and primary group (216 patients with primary biliary pancreatitis).Clinical features and predisposing factors were compared between the two groups.Comparison of continu-ousdatabetweenthetwogroupswasmadebyttestandcomparisonofcategoricaldatewasmadebychi-squaretest.Results Therewere no significant differences in age and mortality between the two groups (both P>0.05 ).Compared with the primary group,the recurrence group had a significantly higher proportion of males,a significantly increased APACHE-Ⅱscore,significantly increased incidence rates of moderate-to-severe pancreatitis and common bile duct stones,significantly higher rates of stenosis of the common bile duct orifice and hy-perlipidemia,and a significantly higher rate of surgical treatment (all P<0.05).In the recurrence group,the type of pancreatitis differed significantly between patients with 2 times of recurrence and those with >2 times of recurrence (recurrence for more than 2 times)(P=0.040).Compared with the primary group,the recurrence group had a significantly higher proportion of patients with known predisposing factors (80.36%vs 58.33%,P=0.002),as well as significantly higher rates of high-fat diet and drinking (both P<0.05).However, the rates of biliary tract infection and oral administration of drugs showed no significant differences between the two groups (both P>0.05). Conclusion Recurrentbiliarypancreatitisiscommonandsevereinmales,andisoftencomplicatedbycommonbileductstones,stenosisof the common bile duct orifice,and hyperlipidemia,with a high rate of surgical treatment.High-fat diet and drinking are important predispo-sing factors for recurrent biliary pancreatitis.
3.A evaluation research for the optimization and utilization of medical equipment in pediatric hospital

Objective:To provide more details and guide the management and usage as well as purchasing for medical equipment, so as to realize the optimization and utilization of the medical resources and impel Precision Medicine Initiative.Methods:The cost-effective analysis and the rate of return analysis of two large-scale precision equipments (MRI and Fundus camera usage) during January 2015 and June 2016 were performed. Besides, the equipment cost was divides into several more detailed projects in the research.Results: The rate of return of MRI in the first half year of 2016 was 8.5%, which was obviously higher than the one of 2015 which was 4.1%; the rate of return of Fundus camera usage in the first half year of 2016 was 25.9%, which was also obviously higher than the one of 2015 that was 15.0%. In the analysis, the depreciation of equipment was the main cost.Conclusion: It was critical and necessary to strengthen the standardized operation and routine maintenance in operation, in order to improve their safety usage rate.
4. Determination of ergosterol in Lasiosphaera Seu Calvatia in various species from different habitats by HPLC

Objective: The HPLC method was established for assessment of quality of Lasiosphaera seu Calvatia (LSC) based on the determined indicator of ergosterol. The contents of ergosterol in 42 batches of LSC were comparatively analyzed. Methods: The contents of ergosterol in LSC from different origins were determined by HPLC. The conditions were as follows: BDS Hypersil C18 column (250 mm × 4.6 mm, 5 μm); flow rate: 1.0 mL/min; detection wavelength: 282 nm; column temperature: 30 ℃; injection volume: 20 μL; mobile phase: acetonitrile (A): 0.1% formic acid (B) as the mobile phase gradient elution: 0-15 min, 95% A, 15-50 min, 100% A. Results: The HPLC method to determine the ergosterol content of puffball was established.The ranges of ergosterol contents among different samples was 0.009 9%-0.150 3%. Conclusion: The contents of ergosterol in different LSC are obviously different, and the quality of commercial LSC is not coincident. So the quality control of LSC a needs further normalization and standardization. The data in this paper would provide a reasonable reference for the quality control of LSC.

6.Formulation Optimization of Asiaticoside Cationic Liposomes by Using Central Composite Design-response Surface Method

OBJECTIVE:To optimize the formulation of Asiaticoside cationic liposomes,and to investigate the characteristics of drug release in vitro. METHODS:The thin film dispersion method was used to prepare liposome;using encapsulation efficiency and drug-loading amount as index,the formulation of Asiaticoside liposomes was optimized by central composite design-response surface method with the ratio of drug to lipid(X1),the ratio of cholesterol to lipid(X2)and the concentration of D-mannose(X3) as factors. Using sodium lauryl sulfate as medium,in vitro release characteristics of cationic liposomes prepared with 1%octadecyl-amine was investigated by bag filter method,and compared with those of Asiaticoside solution and common liposome. RESULTS:The optimal formulation was X1 0.07,X2 0.17 and X3 0.03 g/ml. The encapsulation efficiency was (75.529 ± 1.071)%,and drug-loading amount was(2.539±0.029)%(n=3);the deviation from the predicted values were -0.217% and 0.205%;1% oc-tadecylamine was add into formulation to obtain cationic liposomes,and the Zeta potential had changed from -5.6 mV to 20.8 mV. in vitro accumulative release rates of Asiaticoside solution,common liposomes and cationic liposomes were 89.13%(12 h), 87.58%(72 h) and 94.46%(72 h),and the latter was in line with Weibull model. CONCLUSIONS:Asiaticoside cationic lipo-somes have high encapsulation efficiency,and can releases for 72 h.
7.Detection of Specific DNA Sequence with a Non-labeled Molecular Beacon and a Nucleic Acid Dye Hoechst 33258

A highly sensitive and selective method for specific DNA sequence detection is developed using a non-labeled molecular beacon (MB) and a nucleic acid dye Hoechst 33258. It is demonstrated by a specific DNA sequence of wild-type HBV as a model system. In this strategy, the stem of MB is completely designed as C/G base pairs. In the absence of target DNA, the interaction between Hoechst 33258 and the MBs is very weak,and the fluorescence signals of Hoechst 33258 is very low. In the presence of target DNA, the MBs hybridize with the target DNA and form double-stranded structure. Hoechst 33258 binds to dsDNA, and the fluorescence intensity is significantly enhanced. Under the optimum conditions, the fluorescence intensity of Hoechst 33258 exhibits good linear dependence on target DNA concentration in the range of 2 × 10-10-2 × 10-8 mol/L. The fitted regression equation is △I=3. 3439C(10-10 mol/L) ﹢18. 6949(R2=0. 9982) with a correlation coefficient of 0. 9982 (R2), and the detection limit is 9 × 10-11 mol/L (3σ). The proposed method has good precision, simple operation, fast detection speed, low detection limit, high accuracy and high sensitivity.
9.Antibacterial mechanism and safety of zinc oxide

BACKGROUND:Zinc oxide has shown extraordinary antibacterial effect. But the antibacterial mechanisms of zinc oxide have not been wel developed, and the safety of zinc oxide has not yet been confirmed.
OBJECTIVE:To summarize the antibacterial mechanism and safety of zinc oxide based on the current research situation of zinc oxide.
METHODS:The CNKI database and PubMed database (2005-01/2013-08) were used to search the related articles about zinc oxide antibacterial mechanism and safety. The retrieval keywords were“zinc oxide or ZnO, antibacterial”,“zinc oxide or ZnO, safety”both in English and Chinese.
RESULTS AND CONCLUSTION:The antibacterial mechanism of zinc oxide is complex. Zinc oxide reveals its antibacterial power by photo catalysis, zinc ion dissolve out, and active oxygen. But the relationship between photo catalysis and active oxygen is different based on different studies. The safety of zinc oxide is not confirmed, and different studies have different results. Especial y for the nano-zinc oxide, some studies have showed that the zinc oxide is toxic. Al of these mean much more researches are needed to verify the safety of zinc oxide.

OBJECTIVE:To develop effective and systematical software to monitor the application of essential medicines and promote rational use of essential medicines in hospitals at various levels. METHODS:Using original data of hospital information system(HIS) ,the five modules of Usage Statistics and Management System for Essential Medicines were designed,such as acquiring,inquiry,statistics,analysis and management of data,in order to develop relevance software. RESULTS&CONCLUSION:This system can carry out the real-time analysis and the monitory of the application of essential medicines and can master the consumption pattern and characteristic of essential medicines while provide an efficient way to evaluate and monitor the rational use of essential medicines.
