8.Role of tumor microvascular destruction in tumoricidal effect of hyperthermia

Objective To study the relationship between the microvessel damage and the death rate of tumor cells and determine role of microvessel damage in the tumoricidal effect of hyperthermia.Methods H22 hepatoma cells were inoculated in the right hind legs of KM mice with immunosuppression.Local hyperthermia was administered to these mice which were divided randomly into 10 groups (8 in each group),which included groups of 3,6,12,24,48,72 and 96 h after hyperthermia for 30 min and groups of hyperthermia for 15,30 and 45 min.The tumor tissues were analyzed by flow cytometry(FCM)and immunohistochemistry.The cultured H22 cells were also given hyperthermia for 15,30,45 min and analyzed by FCM.Results The apoptotic rate(0.115±0.008)was higher in the initiation phase of hyperthermia in vitro,then the necrotic rates rose gradually.The difference between in vivo and in vitro was significant in hyperthermia of the same time about death rate(P<0.05).A strong positive linear correlation(r=0.844)was observed between the death rate of tumor cells and MVD.Conclusion Microvessel damage plays the cardinal role in tumoricidal effect of hyperthermia.
9.Erythromycin and alendronate inhibit aseptic loosening in murine osteolysis model

[Objective]To observe whether erythromycin and alendronate are effective in treating aseptic loosening in a murine osteolysis model. [Method]Air pouches were established in the back of BALB/c mice by injecting sterile air subcutaneously. After the air pouches were mature, grafts of calvaria from syngeneic mouse donors were implanted in the air pouches and polyethylene debris was injected into the air pouches. Mice were divided into 4 groups. In the first group, saline was injected into the air pouches.In the other three groups, polyethylene debris was used. Saline,erythromycin and alendronate were injected intraperitoneally separately.Pouch tissues were collected at 14 days after polyethylene debris administration for molecular and histologic analysis.[Result] Both erythromycin and alendronate inhibited osteoclastogenesis and bone resorption, but alendronate was not as effective as erythromycin in anti-inflammation.[Conclusion]Both erythromycin and alendronate can be promising drugs for the treatment of aseptic loosening.
10.Evaluation of life quality of the elderly undergoing internal fixation and arthroplasty for the displaced femoral neck fractures

Objective To evaluate the life quality in the elderly who underwent internal fixation with cannulated screws and bipolar hemiarthroplasty for the displaced femoral neck fractures. MethodsA retrospective study was performed on 141 cases older than 60 years treated from 1993 to 2004. All cases were followed up for mean 40.2 months to evaluate the differences in regard of pain and daily living at one and three years. Results The daily living in hemiarthroplasty group possessed better outcome than that in internal fixation group at one year, with statistical difference but without statistical difference at three years. There was no statistical differences in postoperative pain relief between both groups. Conclusions Both internal fixation and hemiarthroplasty can relieve pain and revive the life quality of the elderly.
