1.Effect of PPARγ and glucocorticoid receptor on hepatic glucose metabolism

HepG2 cells were incubated with dexamethasone,glucocorticoid receptor (GR) antagonist RU486,and pioglitazone for 24 or 48 h.Glucose concentration in culture medium was detected.The results showed that 10 μmol/L RU486 blocked the surge of glucose concentration by 48 h of dexamethasone treatment and the glucose concentration in RU486 + pioglitazone group was higher than pioglitazone group( P<0.05 ),suggesting that the cross talk between PPARγ and GR may exist in hepatic glucose metabolism.

3.The mechanism study of EGCG on improvement of cognitive function and alleviation of hippocampal insulin resistance in APP/PS 1 transgenetic mice

Objective To explore mechanism of epigallocatechin-3-gallate (EGCG) on improvement of cognitive function and alleviation of hippocampal insulin resistance in APP/PS 1 transgenetic mice. Method 12 months old female APP/PS 1 mice were randomly divided into 3 groups:model group(Tg), EGCG low dose group (Tg/EGCG-L), high dose group(Tg/EGCG-H). C 57 BL/6 J mice were utilized as control. learning and memory ability in 4 group mice were detected by morris water maze test(MWM). The hippocampal TNF-α/JNK signal and IRS-1 pSer 312 expression were detected by Western blot and immunohistochemical staining. Results Compared with NT mice, Tg mice showed a marked prolongation of the escape latency and swimming distance in the MWM test(P<0.05);Abnormal activation of TNF-α/JNK signaling and increased IRS-1 pSer 312 expression in the hippocampus of Tg mice(P<0.05). EGCG-treated Tg mice showed significantly improvement of all these abnormal changes(P<0.05). Conclusion EGCG treatment is able to alleviate hippocampal insulin resistance and improve cognitive function in the APP/PS 1 mice. which may be partly attributed to the reduction of TNF-α/JNK signaling activity in this AD mouse model.
4.Case Report of Neonatal Teeth in Cleft Lip and Palate.

The teeth present at birth are called natal teeth, while the teeth that appear within the first thirty days following birth are called neonatal teeth. Approximately one in 2,000 to 3,500 infants experiences the eruption of teeth at birth or within the first few weeks postpartum. Many contributing factors have been suggested for the premature eruption of primary teeth. It has been suggested that upon eruption these teeth are of normal structure according to their developmental age, but after eruption, lateral movement in the cervical area leads to disturbance of the dentin and premature degeneration of Hertwig's root sheath. In most cases, they have immature, aborted structures consisting only of caps of enamel, dentin, or a combination of both. A radiograph can show the presence or absence of enamel, dentin, or root structures, as well as supernumerary teeth. These teeth may be aspirated or swallowed during feeding. Discomfort from mobile and erupting teeth often causes the infant to refuse to feed. In addition, a nursing mother's nipples may become lacerated and infected. Extraction is the indicated treatment if these neonatal teeth cause discomfort or demonstrate mobility or incomplete, immature development. Treatment decisions must be made on an individual basis from both clinical and radiographic findings. We report four cases of neonatal teeth in complete cleft lip and palate patients.

5.A sduty on changes of hepatic energy metabolism and their management in acute obstructive cholangitis in rats

:Mitochondrial respiratory rate,respiratory control rate(RCR),oxidative phospho-rylation efficiency(ADP/O),and hepatic ATP content were determined in rats under 5 conditions including acute obstructive cholangitis(AOC),bile duct ligation(BDL),verapamil pretreatment in AOC(VET),dexamethasone pretreatment in AOC(DET).and sham operation(SO).It was found that rnitochondrial respiratory rate especially its state 4 phase was significantly increased and RCR,ADP/O,and ATP were significantly decreased at 6th hour after AOC and at 24th after BDL.These changes occurred earlier and more dramatically in AOC than in BDL.Pretreatment with verapamil or dexamethasone in AOC could minimize the changes of RCR,ADP/O and ATP in varying intervals.The findings indicate that AOC and BDL can both result in severe functional impairment of the mitochondria of hepatocytes and verapamil and dexamethasone exert effective protection on the rnitochondrial functions of respiration and energy metablism.
6.The role of mononuclear phagocyte system in the pathogenesis of lung injury during acute obstructive cholangitis

The relationship between the behavior of the mononuclear phagocyte system(MPS)and acute pulmonary injury during acute obstructive cholangitis(AOC)was investigated in 90 Wistar rats.After the operation to induce actue obstructive cholangitis,the animals were killed in the Oth,6th,12th,24th and 48th hour,and the phagocytic function of Kupffer's cells,the number of alveolar macrophages in the lung lavage fluid,and the extravascular water content of the lungs were measured.And the pulmonary levels of lipid peroxidase and supperoxide dismutase were determined as well.It was found that when the phagocytic function of Kupffer's cells was suppressed,the alveolar macrophages were activated by the invading bacteria to release some inflammatory mediators such as free radicals resulting in acute pulmonary injury.It seems that there is a close relationship between the fuctional status of mononuclear phagocyte system and the development of pulmonary injury,and the functional failure of mononuclear phagocyte system plays an important role in the pathogenesis of multiple organ failure especially of acute pulmonary injury.
7.Protective effect of trigonelline on rat cardiomyocytes during hypoxia and reoxygenation

Objective To investigate the protective effects of trigonelline in neonatal rat cardiomyocytes during hypoxia /reoxygenation and its mechanism .Methods The neonatal rat cardiomyocytes was randomly divided into the normal control group , the hypoxia/reoxygenation group and the trigonelline group .Flow cytometry was used to determine the apoptosis and mitochondrial membrane potential .The levels of SOD and MDA were measured in different groups .Western blot method was used to measure the procaspase‐9 ,cleaved caspase‐9 ,procaspase‐3 and cleaved caspase‐3 protein level .Results Trigonelline could inhibit apoptosis (P<0 .05) ,enhanced activity of SOD (P<0 .05) ,reduce production of MDA (P<0 .05) ,stabilize the mitochondrial membrane potential (P<0 .05) and activate caspase‐9 and caspase‐3 in neonatal rat cardiomyocytes during hypoxia/reoxygenation .Conclusion Trigo‐nelline could protect myocardial cells from injury caused by hypoxia and reoxygenation ,and the mechanism may be associated with anti‐lipid peroxidation and stabilizing mitochondria membrane potential .
8.Application of Humanoid Robot in Intervention for Autistic Children

Objective To investigate the effect of humanoid robot on social skills, concentration and mobility in children with autism. Methods 10 autistic children were asked to interact with humanoid robot NAO in a teaching environment that involves active participation of the robot. The eye-contact and social interaction, as well as performances in accomplishing learning objectives were observed. Results The 10 children exhibited more concentrated gazing and significantly longer periods of silence. The actual number of interaction of the be-havior Gazing of 9 children exceeded 50%of the theoretical one, the success rate of which reaching 50%. For the behavior Waving, 5 chil-dren were able to interact for more than 50%of the theoretical times, with an accuracy of performance of 50%in 7 children. For Say Hello, 7 children were able to interact for more than 50%of the theoretical times, with an accuracy of performance of 50%in 4 children. Conclu-sion Application of humanoid robots NAO may improve the social skills, concentration and mobility in autistic children.
9.Clinical Characteristics of Recurrent Endometrial Carcinoma: Report of 22 Cases

Objective To summarize the clinical characteristics of recurrent endometrial carcinoma.Methods A total of 22 cases of recurrent endometrial carcinoma from January 2006 to December 2015 were enrolled.Primary diagnosis of endometrial carcinoma were made in our hospital,with complete clinical data and standard NCCN staging operation.The recurrence of endometrial cancer was confirmed by pathological diagnosis after second surgery or biopsy.Clinical data were retrospectively analyzed.Results The median age of primary diagnosis was 52 years old (rang,44-63 years old).And the most common clinical symptom was postmenopausal vaginal bleeding (54.5%,12/22).There were 18 cases (81.8%) of endometrial adenocarcinoma,3 cases (13.6%) of serous adenocarcinoma,and 1 case (4.5%) of clear cell carcinoma.As to the muscular layer infiltration,there were 14 cases (63.6%) < 1/2,7 cases (31.8%) > 1/2 and 1 case (4.5%) involving the whole layer.There were 4 cases (18.2%) of positive pelvic lymph node and no para-aortic lymph nodes metastasis.As to the postoperative clinical pathologic stage,there were 12 cases (54.5%) of FIGO Ⅰ A,5 cases (22.7%) of FIGO Ⅰ B,1 case (4.5%) of FIGO Ⅱ,and4 cases (18.2%) of FIGO ⅢC1.The median recurrent time was 4 years (rang,1.5-7 years),without obvious self-conscious symptoms,including serum CA125 elevated in 12 cases (54.5%) and positive ultrasound,CT or X-ray examination findings in 10 cases (45.5%).As to the recurrence location,14 cases (63.6%) were in the pelvic cavity,6 cases (27.3%) of retroperitoneal lymph nodes,1 case (4.5%) of anterior abdominal wall accompaning lung metastasis,and 1 case (4.5%) of lung metastasis.Three patients accept Lynch syndrome screening,of which 2 cases were positive.Conclusions Muscular layer infiltration is common in recurrence of endometrial cancer,with no obvious clinical symptoms.Pelvic cavity is the most common location of recurrence.
