1.Progress in the studies of P_2 purinoceptors

P 2 purinoceptors were first subdivided into P 2X and P 2Y subtypes, and later this classification was broadened to include P 2T , P 2Z and P 2D subtypes. In the 1990s, a new kind of receptors was found, which respond to UTP, ATP and ATP?S, but not to 2 MeSATP or ?,? MeATP, this finding led to the definition of the so called “P 2U ” or “nucleotide” receptor. Most recent evidence demonstrated the existence of a pyrimidine receptor responding to UTP but not to ATP. For this case, IUPHAR (International Union of Pharmacology) committee recommended that P 2 purinoceptors be subdivided into P2X and P2Y subtypes, any subtypes of intrinsic ion channel be termed P2Xn, and any subtypes of G protein coupled receptor be termed P2Yn purinoceptors. With the development and application of the molecular biologic technique and the cloning and expression of the receptors, the classification was strongly confirmed.
2.Effects of adenosine triphosphate and adenosine on the proliferation of Chinese esophageal carcinoma Eca-109 and hepatoma SMMC-7721 cells

Aim To study the effects of adenosine triphosphate(ATP) on the proliferation of Chinese esophageal carcinoma Eca-109 and hepatoma SMMC-7721 cells.Methods MTT assay was used to determine the inhibition of the proliferation of the two cultured tumor cell 1ines in vitro by ATP,adenosine(ADO) and uridine triphosphate(UTP).Morphological changes of the two cell lines induced by ATP were observed under light microscope.Results ATP(0.03~0.3 mmol?L~(-1)) and ADO(0.1~0.3 mmol?L~(1)) had inhibitory effects on Eca-109 and SMMC-7721 cells concentration-dependently,and the inhibition by ATP was stronger than that by ADO in both cell lines.For Eca-109 cell line,the maximal inhibition rate of the proliferation by ATP and ADO was 86.36% and 29.88%,and the IC_(50) was 0.056 and 0.823 mmol?L~(-1),respectively.For SMMC-7721 cell line,the maximal inhibition rate of the proliferation by ATP and ADO was 82.06% and 52.84%,and the IC_(50) was 0.218 and 0.517 mmol?L~(-1),respectively.UTP had a very weak inhibitory effect on Eca-109 cell line,with the maximal inhibition rate of 18.27%,and did not significantly affect SMMC-7721 cell line.Exposed to higher concentration(0.3 mmol?L~(-1)) of ATP for 72 h,SMMC-7721 cells displayed morphological changes of apoptosis,but Eca-109 cells did not show the characteristics of apoptosis markedly.Conclusion ATP has a strong inhibitory effect on the proliferation of Eca-109 cell line,which is mainly induced by ATP per se,and a metabolite ADO also has weaker effects.For SMMC-7721 cell line,however,ATP inhibits the cell proliferation mainly via its degradation to ADO.

4.Research on characteristic psychological capital of doctors in public hospitals

Objective To study the state of doctors' psychological capital in public hospitals,and to analyze the characteristics of such capital,so as to better leverage the capital as a hospital management tool.Methods A quantitative and qualitative research was made on the state of doctors' psychological capital.Results The doctors' task-oriented psychological capital scored 4.25,and guanxi-oriented psychological capital 4.41; “modesty and prudence”scored the highest in all dimensions of the doctors' psychological capital,“optimism and hope”lowest; the doctors with different genders,ages,seniority,and monthly average income have a difference in psychological capital.However,no significant difference was found with varying titles,hospital levels,and employment models.Conclusion The doctors were found with a strong psychological capital to cope with stress.Better psychological capital will raise quality of care in practice.
5.Application of BODE grading system in evaluating clinical prognosis of chronic obstructive pulmonary disease

Objective The aim of this study is to evaluate clinical application of BODE (body mass index, airflow obstruction, dyspnea, and exercise capacity) index in patients with chronic obstructive pulmonary disease (COPD). Methods Forty patients with stable COPD in Xuanwu Hospital of Beijing during September 2003 to December 2007, were recruited in the study, including 25 men and 15 women, aged 38 to 85 years (with a mean of 62 ± 13 years). BODE index was measured for each patient, one year and three years after recruitment, respectively. Severity of COPD, including number and duration of episodes, length of each hospital stay and times of hospitalization due to acute exacerbation of COPD (AECOPD), were recorded. All data were statistically analyzed with SPSS 13.0 software for windows. Results ①There was a remarkable difference in COPD severity between patients with BODE score equal to or greater than five and these with less than five (P <0.01 ). ①BODE score inversely correlated with forced expired volume at the first second ( FEV1 ) in COPD patients (P < 0.01 for those with BODE score equal to or greater than five and P <0.05 for these with BODE score less than five, respectively). ③BODE score positively correlated with number and duration of acute exacerbation, length of hospital stay and times of hospitalization due to AECOPD (P <0. 01 for those with BODE score equal to or greater than five and P < 0. 05 for those with BODE score less than five, respectively). For patients with COPD, their BODE score in the first year positively correlated with that in the third year ( r = 0.834, P < 0.01 ). Conclusions BODE index is a better indicator to evaluate their condition severity in patients with COPD.
6.Relationship between the expression of Toll-like receptor 4 on mononuclear cells in peripheral blood and changes of serum vitamin D level in children with bronchiolitis

Objective To investigate the relationship between the expression of Toll-like receptor 4 (TLR4) on mononuclear cells in peripheral blood and changes of serum vitamin D level in children with bronchiolitis.Methods The children who were diagnosed as bronchiolitis and received treatment in the Second Affiliated Hospital of Harbin Medical University from October 2013 to January 2014 were chosen as the pre-treatment group,and then divided them into moderate group and severe group according to the clinical symptoms,20 cases for each group.Then the cases in pre-treatment group who recovered after treatment were recruited as the after-treatment group,and the children who were healthy and medical examination in the Second Affiliated Hospital of Harbin Medical University in the same period were recruited as the healthy control group.The expressions of TLR4 on CD14 labeled mononuclear cells in the periphera were measured by flow cytometry.The level of 25 (OH) D in serum was detected by enzyme linked immunosorbent assay (ELISA).Results (1) The expression level in children with bronchiolitis of TLR4:the mode-rate group [(18.98 ±2.29)%] and severe group [(30.13 ±2.13)%] increased significantly (P <0.05) compared with control group [(1.17 ± 0.57) %].And the expression level of moderate group [(2.02 ± 0.48) %] and severe group [(11.43 ± 1.52) %] decreased significantly after treatment (P <0.05).(2) Serum vitamin D level in children with bronchiolitis of the moderate group[(17.16 ± 3.34) μg/L] and severe group [(6.56 ± 2.28) μg/L] were lower than healthy control group [(53.69 ± 20.18) μg/L] before treatment (P < 0.05),especially the severe group [(6.56 ±2.28) μg/L].The level of moderate group [(9.59 ± 2.31) μg/L] and severe group [(4.70 ± 0.67) μg/L] became lower after treatment (P < 0.05).(3) Both severe group (r =-0.491,P < 0.05) before treatment and moderate group (r =-0.436,P < 0.05) after treatment showed negative correlation between TLR4 on mononuclear cells in peripheral blood and serum 25 (OH)D level in children with bronchiolitis.And no correlation was found among healthy control group,moderate group before treatment and severe group after treatment (P > 0.05).Conclusions The conditions of children with bronchiolitis was positively correlated with the expression level of TLR4,and negatively correlated with the vitamin D level.The serum 25 (OH) D decreased steadily during the treatment.The expression of TLR4 in monocytes has a certain correlation with the level of vitamin D in children with bronchiolitis.
7.Effects of extracellular ATP on apoptosis of human esophageal carcinoma Eca-109 and hepatoma SMMC-7721 cells

Objective To study the effects of adenosine triphosphate (ATP) on proliferation of human squamous esophageal carcinoma Eca-109 and hepatoma SMMC-7721 cells in vitro and the underlying mechanism. MethodsMTT assay was used to determine the proliferation of tumor cells. The AO/EB double stained cells were observed under fluorescence microscope. The effects of ATP on the cell cycle, apoptotic rate and apoptosis-related protein were detected by flow cytometry. Results ATP showed inhibitory effects on Eca-109 and SMMC-7721 cells at the concentration of 0.01~0.3 mmol/L. Exposure to 0.3 mmol/L ATP for 72 h, some of SMMC-7721 cells displayed morphological changes of apoptosis, but Eca-109 cells did not show the characteristics of apoptosis markedly. There was no significant change in the apoptotic rate and apoptosis-related protein of the two tumor cell lines treated with ATP 0.03, 0.1 and 0.3 mmol/L for 72 h respectively. The proportion of Eca-109 cells in G0/G1-phase of cellcycle was significantly increased, meanwhile the proportion of Eca-109 cells in S-phase and proliferation index value was significantly decreased by treatment with 0.3 mmol/L ATP. Conclusion ATP inhibited Eca-109 cell proliferation by changing the distribution of cell cycle phase, and its mechanism might not related to apoptosis, but for SMMC-7721 cell, the inhibition of cell proliferation induced by ATP was not related to the change in cell cycle.
8.SENTINEL NODE BIOPSY BY TWO KINDS OF BLUE DYES IN PATIENTS WITH BREAST CANCER

Objective To evaluate the identification successful rate of sentinel lymph node (SLN) with breast cancer and the accuracy to predict axillary lymph node status in different vital blue dyes.Methods 94 patients with breast cancer were recruited for the study between Oct. 1999 and Apr. 2001, of whom 32 and 62, respectively, were injected 0.028mmol*L-1 Methylene blue and 0.018mmol*L-1 Patent blue violet in breast parenchyma surrounding the primary tumor to identify SLN.All 94 patients underwent the axillary lymph node dissection.Results For Methylene blue group and Patent blue violet group, SLN identification successful rates were 65.6% (21/32) and 88.7% (55/62) (P=0.012) and accuracy rate to predict axillary lymph node status were 90.5% (19/21) and 98.2% (54/55) (P=0.183), respectively.Conclusion In identifying SLN,Patent blue violet is more ideal vital blue dye than Methylene blue, whereas the accuracy rate to predict axillary lymph node status had no significant difference.
9.The evaluation of cognitive function in patients with obstructive sleep apnea hypopnea syndrome by expansive scale of dementia

Objective To asscess cognitive changes of patients with Obstructive Sleep Apnea-Hypopnea Syndrome(OSAHS)by the Expansive Scale of Dementia(ESD),and to compare ESD to Mini Mental Status Examenation(MMSE)in cognitive function.Methods 90 OSAHS patients were investigated in our study.All patients were divided into three groups,according to apnea-hypopnea index(AHI),including mild group,moderate group and severe group.The cognitive function was evaluated by MMSE and ESD.Results The parameters,including AHI,Longest apnea time(LAT)and percentage of time spend when oxygen saturation lower than 0.9(SLT 90%) in severe OSAHS group were significantly higher than those in mild and moderate groups(P0.05).The total score of MMSE and subtest scores of MMSE in memory,attention,calculation and short-and long-term memory were significantly lower in severe OSAHS group than those in mild and moderate groups(P0.05).Conclusion There was the obvious decrease in cognitive function in patients with severe OSAHS,especially in leaning,memory,calculation and constructive function.Both ESD and MMSE could be used to evaluate the cognitive function in OSAHS patients,especially ESD.
10.Advances in cardiovascular effects of tanshinone II(A).

Cardiovascular diseases, like coronary heart disease and myocardial infarction, are the most common cause of death worldwide. Chinese medicines have demonstrated rich cardioprotective activities for clinical applications. Salvia miltiorrhiza, a very important component of traditional Chinese medicine, can promote blood circulation and relieve blood stasis. Salvia miltiorrhiza is widely used in treatment of cardiovascular and cerebrovascular disease such as coronary heart disease and cerebral infarction ( CI). Tanshinone II(A), the major lipophilic components extracted from the root of S. miltiorrhiza, possesses anti-atherosclerosis, anti-cardiac hypertrophy, anti-oxidant, anti-arrhythmia and so on. This paper discusses current research status of tanshinone II(A) in cardioprotective effects.
