1.Computational Analysis of miRNA and Target mRNA Interactions: Combined Effects of The Quantity and Quality of Their Binding Sites

MicroRNAs (miRNAs) act by binding to complementary sites on target messenger RNA (mRNA) to induce mRNA degradation and/or translational repression. To investigate the influence of miRNAs at transcript levels, two human miRNAs (miR-1 and miR-124) were transfected into HeLa cells and microarrays used to examine changes in the mRNA profile showed that many genes were downregulated and that the fold decreases in levels of these target mRNAs differed remarkably. Features depicting interactions between miRNAs and their respective target mRNAs, such as the number of putative binding sites, the strength of complementary matches and the degree of stabilization of the binding duplex, were extracted and analyzed. It was found that, for a given target mRNA, both the quality and quantity of miRNA binding sites significantly affected its degree of destabilization. To delineate these types of interactions, a simple statistical model was proposed, which considers the combined effects of both the quality and quantity of miRNA binding sites on the degradation levels of target mRNAs. The analysis provides insights into how any animal miRNA might interact with its target mRNA. It will help us in designing more accurate methods for predicting miRNA targets and should improve understanding of the origins of miRNAs.

3.The Analysis on allocation status and problem of the large medical equipment

Objective: To analyze the problems of large medical equipment allocation in our country, put forward the method and basis of scientific configuration. Methods: The current situation of large medical equipment allocation was analyzed by the methods of literature, statistics and analysis, in order to understand the operating costs and benefits and provide a scientific configuration tool when bringing in medical technology. Results: The configuration of large medical equipment has some problems, such as the uneven distribution of medical resources, equipment shortage, blind purchase of high-end equipment, low localization rate and so on. These problems will directly lead to the unreasonable allocation and the waste of medical resources. Conclusion:The configuration of large medical equipment must comply with the relevant provisions of the state by the HTA and other scientific methods, so as to improve the level of investment decision-making. It will make the use of medical resources efficiently and reasonably.
5.Research on Precise Location Algorithm for QRS in ECG

Objective To research the precise location algorithm of onset and end of one wave for QRS in ECG. Methods During monitoring the ambulatory electrocardiograph (AECG),it was necessary to design new automatic analysis algorithms. The windows formed by filtering and integrating process could be considered as the fiducially mark. From the window,the apex of Q wave and S wave could be recognized. Results Introducing an auxiliary line from the apex,we could judge the boundary of the Q and S wave with the distance from ECG curve to the line. Conclusion Many cases studies inelicate that the algorithm is of low level locating error,about one sample point,and the accuracy of judgement of S wave boundary is raised obviously compared with the same kind of software in the world.

7.Relationship between gastrin expression,and development of colorectal neoplasm and cell proliferation

Objective To investigate the relationship between gastrin expression and the development of coloreetal neoplasm and cell proliferation.Methods 15 samples of normal coloreetal mucosal tissue,21 samples of coloreetal adenomas and 70 samples of colorectal cancer were selected for evaluation of the expression of gastrin and Ki67 with immunohistochemieal method.Results In normal colorectal mucosal tissues.colorectal adenomas and coloreetal cancer,the positive rates of gastrin expression were 0.28.6%and 25.7%,respectively-which were signff-ieantly hisher in coloreetal cancer and adenomas groups than in the normal colorectal mucosal tissues(P<0.05).But there was no significance between colorectal cancer and adenomas(P>0.05).The positive expression rate of Ki67 with the obove samples were 40.0%,76.2%and 82.8%-respectively,with statistical results identical with results of gastrin(P<0.05).There was significant correlation between gastrin and Ki67[P<0.05).Conclusion Gastrin and Ki67 are highly expressed in eolorectal neoplasm.Gastrin miight play an important role in the carcinogen-esis and development of colorectal neoplasm,and might promote tumor cell proliferation.
9.Cryopreservation study on seeds and embryos in Dalbergia odorifera.

The mature seeds and excised embryos of Dalbergia odorifera were used as materials to study the effect of moisture content on their survival, as well as the effect of rapid freezing and vitrification freezing method on seeds and in vitro embryos cryopreservation. The results showed that the germination rate and vigor decreased from 82.67%, 85% to 18.35%, 25% respectively, when the seed moisture content decreased from 15.04% to 8.14%; and the germination rate decreased from 82.67% to 37.50%, 25.37% respectively by vitrification freezing method and rapid freezing method, when the seed moisture content decreased from 15.04% to 9.37%. Among all the moisture content gradient, 12.35% moisture reached the maximal germination rate, which were 63.58% and 50.45% respectively by vitrification freezing and rapid freezing; and when the embryo moisture content was 26.32%, the germination rate decreased from 95.67% to 58.31% and 33.82% respectively by vitrification freezing and rapid freezing. And when the moisture content was in the range of 14.17% -21.34%, the germination rate was a bit of decrease. The experiment results showed that the optimum conditions of seed cryopreservation were: moisture content 12.35%, vitrification freezing; and the optimum conditions of in vitro embryo cryopreservation were: moisture 15.04%, vitrification freezing. In conclusion, the effects of moisture content on germination rate after cryopreservation in D. odorifera seeds and embryo were significant, and vitrification freezing method is much better than rapid freezing method.
