1.Effects of ketamine on expression of nitric oxide synthase in medial septal nuclei and vertical diagonal band of rats

AIM: To explore the effects in different dosage of ketamine on the expression of nitric oxide synthase (NOS) in medial septal nuclei (MS) and vertical diagonal band (VDB) of rats. METHODS: 40 SD rats were randomized into normal control group,normal saline(NS) group, low, middle and high dosage of ketamine(25,50,100 mg?kg~ -1 ,ip) group.Each group was maintained intraperitoneally by given daily dosage once a day for a period up to 7 days. That was done at 2100 hour each night, and by 2 h of the last administration, poured, fixed and sliced were carried out. Method of nicotinamide adenosine dinucleotide phosphate diaphorase (NADPH-d) was used to show the expression of NOS in MS and VDB pars dorsalis and ventro. RESULTS: Compared with normal control and NS group, the number of NOS positive neurons were decreased in MS and VDB pars dorsalis and ventro in ketamine groups.There were statistical meaing in different-dosage groups of ketamine(P
2.Investigation into G6PD deficiency gene frequency in Tujia nationality in Jiangkou,Guizhou.

Objective The frequency of G6PD deficiency in the Tujia nationality in Jiangkou,Guizhou was investigated.Methods In Oct.2002,227 male subjects were selected randomly from the local people,and NBT qualitative and G6PD/6PGD quantitative methods were used to detect G6PD deficiency in them.Results Among the 227 subjects,17 cases of G6PD deficiency were found.The gene frequency of G6PD deficiency was 0.0749.Conclusion There is a high incidence of G6PD deficiency in Tujia nationality in Jiangkou,Guizhou.The investigation will illustrate the distribution of G6PD deficiency in Guizhou,and provide some useful data for the preventing G6PD deficiency,directing clinical management,improving minority population diathesis and studying the origin of Tujia nationality.

OBJECTIVE To study the distributive characteristics of the pathogens of the bacterial infectious diseases in Wuzhou,and to provide the basis for prevention and therapy of the infectious diseases.METHODS The VITEK-ATB system,BDPhoenix100 system,API system and CHROMagar were used to appraise the pathogens,which were from the bring up of doubtful infectious disease specimens.RESULTS The positive rate of 4 878 pathogenic test was 32.10%.The Gram-negative bacilli were the major(63.96%) from the 1 623 strains of pathogens isolated.The main composition of pathogen spectrum was Escherichia coli,Klebsiella pneumoniae,Staphylococcus aureus,Pseudomonas aeruginosa,Acinetobacter baumannii,Candida albicans,Enterococcus faecalis,Staphylococcus haemolyticus,Enterobacter cloacae,and Staphylococcus epidermidis.The distribution of main pathogens had different characteristics in different season and different infectious diseases.CONCLUSIONS Strengthening the test of pathogens in the doubtful infectious diseases and understanding the characteristics of local pathogen spectrum and the transitional current of infections in time,significantly influence on prevention and therapy of the infectious diseases.
4.Synergistic protective effects of sesamin in combination withvitamin E on nephropathy in rats with metabolic syndrome

AIM: To study the renoprotective and synergistic effects of sesamin in combination with vitamin E on nephropathy in rats with the metabolic syndrome.METHODS: Nephropathy of metabolic syndrome in rats was induced by high-fat and high-carbohydrate diet.Sesamin(30 mg?kg-1?d-1),sesamin +vitamin E [(15+20) mg?kg-1?d-1),sesamin +vitamin E(30+20) mg?kg-1?d-1] and vitamin E(20 mg?kg-1?d-1) were given to rats with metabolic syndrome at 9th week and lasted for 16 weeks.After 24 weeks,the body weight,left kidney humid weight,blood glucose(BG),blood lipids(BL),systolic blood pressure(SBP), renal function,and indexes of oxygenation and antioxygenation for renal cortex were measured.Pathological changes,collagen deposition and iNOS protein and nitrotyrosine expression of kidney were observed by HE-stain,Masson-stain and immunohistochemical method,respectively.RESULTS:(1)In high-fat and refined-sugar diet group(HFS): the levels of BG,BL,SBP,and MDA,OH and NO2-/NO3-in renal cortex were increased;the activities of T-SOD, CAT and GSH-Px in renal cortex were decreased;the expressions of iNOS protein and nitrotyrosine in kidney were up-regulated;the inflammatory cell infiltration and collagen deposition in renal glomerulus and renal interstitium were severe;the glomerular sclerosis and renal interstitial fibrosis were obvious;the renal function was deteriorated.(2)In sesamin +vitamin E [(30+20) mg?kg-1?d-1] group: the decreases in BG,BL,SBP,MDA,OH-and NO2/NO3-in renal cortex were obviously observed;the activities of T-SOD,CAT and GSH-Px in renal cortex were increased;the expressions of iNOS protein and nitrotyrosine in kidney were down-regulated;the inflammatory cell infiltration and collagen deposition in renal glomerulus and renal interstitium were improved;the glomerular sclerosis and renal interstitial fibrosis were reversed;the renal function was ameliorated.The therapeutic effect of sesamin +vitamin E[ (30+20) mg?kg-1?d-1] surpassed that of using sesamin or vitamin E alone(P

OBJECTIVETo explore the nerve regeneration mechanism of electroacupuncture (EA) combined with polysaccharide of gastrodia elate blume (PGB) for secondary thalamic damage of focal cerebral ischemia.
METHODSForty Sprague-Dawley adult rats were randomly divided into a normal control group, a model group, an EA group, a PGB group and an EA + PGB group, 8 rats in each group. The rat model of right middle cerebral artery occlusion was prepared by suture-occluded method. Two weeks after model establishment, rats in the normal control group and model group received no treatment; rats in the EA group were treated with EA at "Baihui" (GV 20) and left "Zusanli" (ST 36), 30 min per treatment, once a day for 14 successive days; rats in the PGB group were treated with intragastric administration of PGB (100 mg/kg) , once a day for 14 days; rats in the EA + PGB group were treated with EA and PGB treatment, once a day for totally 14 days. The expressions of nestin and stem cell factor (SCF) in thalamic ventroposterolateral nucleus (VPL) were detected by immunohistochemical method.
RESULTSThere were positive cells of nestin in ischemia VPL in the model group, and the number of SCF positive cells was increased compared with that in the normal control group (P<0.05). The number of positive cells of nestin and SCF in ischemia VPL in the EA group, PGB group and the EA + PGB group was increased compared with that in the model group (all P<0.05), and the average gray value of immune positive product was all reduced (all P<0.05). The number of positive cells of nestin and SCF in the EA + PGB group was higher than that in the EA group or the PGB group (all P<0.05).
CONCLUSIONEA combined with PGB can significantly increase the SCF expression in ischemia VPL and promote the proliferation of neural stem cells, which is likely to be one of the nerve regeneration mechanism of acupuncture and medication tor secondary thalamic damage of local cerebral isctemia.

6.The Association of FGFR2 rs1219648 Polymorphism with Susceptibility of Breast Cancer in Han Population in Guizhou Province

Objective:To investigate the association between polymorphisms of FGFR2 and the susceptibility of breast cancer in Han population in Guizhou province.Methods:Genotyping was performed using PCR-sequence-specific primers(PCR-SSP)in 106 histologically confirmed breast cancer cases and 116 cancer-free controls.Results:The genotype frequencies of rs1219648 TT,TC,and CC were 50%,25.47%.and 24.53% in breast cancer cases and 29.31%,48.28%,and 22.41% in the controls.The gene frequencies of T in breast cancer cases and the controls were 62.74% and 53.45%.respectively.The gene frequencies of C were 37.26% and 46.55%.respectively.The distribution of allele and genotype frequencies of FGFR2 rs1219648 was statistically different between breast cancer cases and the controls(P<0.05).Conclusion:FGFR2 rs1219648 polymorphism influences the susceptibility of breast cancer.TT genotype might serve as a risk factor for breast cancer.
7.A study of mRNA expressi on and transcription regulation in the promoter region of myeloperoxidase gene from a population living in the area with coal-burning endemic fluorosis in Guizhou Province

Objective To explore the correlation between myeloperoxidase (MPO) genetic variation and coal-burning endemic fluorosis, and to understand the influence of integrated intervention including stove changes and health education on people’s health in the area. Methods In 2007, coal-burning endemic fluorosis disease areas were selected in Bijie City, Guizhou Province. No stove changes in Yachi Town, 150 patients with dental fluorosis were selected as fluorosis non-intervention group, and the intervention group was 150 patients in Changchun Town where the stoves were changed 2 years ago. The population in control group was selected in an area with non-endemic fluorosis in Changshun County. The mRNA expressions of MPO in leukoxytes were detected by real-time PCR. HepG2 cells were cultured in vitro and divided into four groups: pGL3-A group, pGL3-G group, pGL3-Control group and pGL3-Basic group. pGL3-A and pGL3-G were recombinant plasmid, while pGL3-Basic as a blank control and pGL3-Control as a positive one. The internal reference plasmid pRL-TK co-transfected the HepG2 cells with pGL3-G, pGL3-A, pGL3-Basic and pGL3-Control, respectively. The influence of sudden change of MPO gene promoter on the gene transfection activity was evaluated by a dual luciferasereporter gene system. Results The expression level of MPO mRNA in peripheral blood leukocytes in non-intervention group(0.054 ± 0 . 003 ) were higher than control and intervention groups (0.019 ± 0.004,0.019 ± 0.003, all P<0.05), and no significant change was found between intervention group and control group(P>0.05). After the MPO-463G/A locus genetic variation occured, the luciferase reporter gene expression level of the recombinant plasmid pGL3-G(0.753 4 ± 0.086 6) was higher than that of the pGL3-A(0.490 0 ± 0.022 3, P < 0.05). Conclusions The study on MPO gene promoter-463G/A locus has prompted that MPO gene allele may be a protective factor to coal-burning fluorosis. The integrated interventions have a role in the prevention and treatment of endemic fluorosis.

Objective In the present study, we investigated the effects of advanced oxidation protein products(AOPP) on reactive oxygen species(ROS) production in murine osteoblastic MC3T3-E1 cells by NADPH oxidase enzymes pathway. Methods Experiments were divided into three groups, including control group, rats albumin(RSA) group, and AOPP group. Different concentrations of AOPP were added to the osteoblastic MC3T3-E1 cells culture medium. The production of ROS in MC3T3-E1 cells was measured by the fluorescence intensity of intracellular fluoroprobe ( DCFD ) . In order to verify the effect of enzyme of the production of ROS, the specific inhibitors of corresponding enzymes were added in the MC3T3-E1 cells which were cultured in the medium with AOPP. Finally, western blot and immunofluorescence were used to observe the changes of NADPH oxidase enzymes subunits. Results Different concentrations of AOPP (50,100,200μg/ml) induced MC3T3-E1 cells to produce different amount of ROS. The higher concentrations of AOPP were added, the more ROS were produced. Furthermore,200μg/ml AOPP induced the maximum amount of ROS production(P<0. 05). Meanwhile, AOPP induced MC3T3-E1 cells to produce different amount of ROS with a time-dependent manner. The peak amount of ROS production in MC3T3-E1 cells was observed in 3h when AOPP were added (P<0. 05). In addition, when specific inhibitors of corresponding enzymes were added in the MC3T3-E1 cells, the production of ROS were significantly suppressed by C-SOD, DPI, and apocynin(P<0. 05). On the other hand, AOPP can up-regulate the expression of Nox4 protein of the MC3T3-E1 cells, which is one of the subunits of NADPH oxidase enzymes. Meanwhile, AOPP can also induce the membrane migration of p47phox subunit. Conclusion AOPP induces osteoblastic MC3T3-E1 cells to produce ROS by NADPH oxidase enzymes pathway, and which may be one of the pathogenesis of AOPP involved in osteoporosis.

OBJECTIVETo study the genetic polymorphisms of methylenetetrahydrofolate reductase (MTHFR) among the Han, Buyi and Miao populations in Guizhou and to provide genetic data for establishment of the genetic polymorphism bank of Guizhou Minorities.
METHODSThe technique of polymerase chain reaction and restriction fragment length polymorphism (PCR-RFLP) was used to detect the genotype and allele frequencies at two mononucleotide sites (677 and 1298) of MTHFR among the Han population in Libo county, the Buyi population in Libo county and the Miao population in Leishan county.
RESULTSAt the site of 677, the T allele frequencies were found to be 22.8%, 16.1%, 10.6%, for the Han, Buyi, Miao populations respectively. At the site of 1298, the C allele frequencies were 28.9%, 39.1%, 48.7% for the Han, Buyi, Miao populations respectively. The frequencies for the combined heterozygote of 677CT/1298AC were 16.66%, 22.7%, 11.1% for the three populations respectively. Moreover, one case with combined homozygote of 677TT/1298CC was seen in the Miao population.
CONCLUSIONThe polymorphisms of the two mononucleotide sites (677 and 1298) of MTHFR are diverse in different populations. The C allele frequencies at the site of MTHFR 1298 of the Miao population in Leishan county and the Buyi population in Libo county are high, and the C allele frequency in the Miao population is higher than those hitherto reported in literature.

10.Protection of indocyanine green fluorescence imaging on parathyroid gland in gasless unilateral axillary approach endoscopic thyroidectomy surgery

Objective:To evaluate the feasibility of indocyanine green fluorescence imaging technology applied in thyroid surgery to identify parathyroid gland.Methods:From Oct 2021 to May 2022, data of 42 patients undergoing thyroidectomy via gasless unilateral axillary approach in Zhejiang Provincial Hospital of Traditional Chinese Medicine were retrospectively analyzed. Cases using intraoperative fluorescence imaging technology(42 cases) were compared with conventional laparocopic approach.Results:The number of parathyroid glands dissected in the study group 1.57±0.61 was higher than that in the control group 0.56± 0.59 ( t=-5.472, P<0.05). The PTH value of the study group was (2.88±1.23)pmol/L on the first day after operation, which was higher than that of the control group (2.16±0.10)pmol/L ( t=-1.844, P<0.05). The blood parathyroid hormone value on the third day(3.22±1.31)pmol/L was higher than that of the control group (2.55±0.81) pmol/L ( t=-2.041, P<0.05). There were 2 cases of hypoparathyroidism in the study group, less than 5 cases in the control group, but there was no significant difference between the two groups( χ2=0.942, P>0.05). There was 1 case of hypocalcemia in the study group and 3 cases in the control group ( χ2=0.731, P>0.05). Conclusion:Using indocyanine green fluorescence imaging technology to identify parathyroid gland is feasible, simple, fast, safe and effective.
