1.Effect of thymosin α1 on immunologic function and cytokines in Patients with severe infection

Objective To investigate the effect of thymosin α1 on immun ologic function and cytokines in the abdominal sepsis patients.and naddiscuss the therapeutic function of thymoin α1 in sepsis.Methods Thirty-eight sepsis patients admitted between May 2006 and May 2007 in the Hospital of Weifang Medical University.Patients were divided into the treatment group(20 cases)and control group(18 cases)randomly.Thymosin α1 was used in the treatment group on the basis of infection controlled.The concentration of cytokines(TNF-α,IL-6,IL-10)and the percent age of CD3,CD4,NK cell were observed.Results The concentrations of TNF-α and IL-6 were increased significantly in bothgoups.But it was more prominently in the control group.The concentration of IL-10 and the percent age of CD3,CD4,NK cell increased more significantly in thymosinod treatment group than those in the control group.Conclusions Thymosincd canmodulate the release of TNF-α,IL-6,IL-10 and mitigate the inflammatory reaction,which may improve the immunologic function.
2.Relationship between CD117 expression and chemotherapy effect on the patients with acute nonlymphoblastic leukemia

Background and Purpose:It has been proved that CD117 may be used as an immunology marker for diagnosis of leukemia of myeloid origin.The relationship between CD117 expression and effect of chemotherapy on the patients with Acute Nonlymphoblastic Leukemia(ANLL) remains unclear.This study is to investigate the relationship between CD117 expression and the response of patients with ANLL to chemotherapy.Methods:Flow cytometery(FCM) was used to detect the positive rate and the levels of CD117 expression of the bone marrow mononuclear cell(BMMNC) from 38 patients with acute lymphoblastic leukemia(ALL) and 81 patients with ANLL,respectively.All-trans Retinoic Acid(ATRA) was taken to treat M_(3) type of ANLL and protocol DA and/or HA was used to treat the other types.ANLL was divided into two groups: positive(+) and negative(-) expression of CD117.At the same time we compared the difference of rates of complete remission(CR) between CD117(+) and CD117(-) groups from ANLL after chemotherapy.Results:Positive percentage of expression of CD117 in ALL and ANLL groups were 13% and 70% respectively(P=0.000).Positive levels of CD117 decreased successively as follows: M_(3)/ M_(1)、M_(2)/ M_(6) / M_(4)、M_(5).CR rates of CD117(+) and CD117(-) groups of ANLL after chemotherapy were 51%(29/57) and 67%(16/24)(P=0.192),respectively.Conclusions:CD117 may serve as an immunology marker for the diagnosis of ANLL,but positive or negative expression of CD117 in ANLL was not associated with the response of the patients with ANLL to chemotherapy.
3.Upregulation of proto-oncogene c-mpl by interleukin-13 in Dami cells

AIM: To investigate the effects of recombinant human interleukin-13 (rhIL-13) on the expression of proto-oncogene c-mpl in Dami cells, a human megakaryobiastic leukemia cell line. METHODS: The expression of c-mpl mRNA in Dami cells was investigated with RT-PCR. The expression of membrane-bound protein c-mpl on Dami cells was investigated with ligand combination experiment. RESULTS: In RT-PCR experiment, we found the quantitis of expression of c-mpl mRNA in 25 ?g/L rhIL-13 group increased by 24.8%. In ligand combination experiment, we found quantitis of expression of membrane-bound protein c-mpl in 100 ?g/L rhIL-13 group increased by 28.5% ( P
