1.Effect evaluation of prescription of dispersing stagnated liver and strengthening spleen combined with psychological care in treatment of functional dyspepsia

Objective To investigate the effect of prescription of dispersing stagnated liver and strengthening spleen combined with psychological care in treatment of functional dyspepsia.Methods 60 patients with functional dyspepsia were randomly divided into the nursing group and the control group with 30 patients in each group.The control group was treated with oral intake of non-boiled prescription of dispersing stagnated liver and strengthening spleen.The nursing group was treated with psychological care on the basis of the control group.Both groups were assessed with SCL-90,HAMD,HAMA and symptom scales before treatment and 4 weeks after treatment.Results HAMD,HAMA,SCL-90 scores and symptom score were improved after treatment in both groups,the treatment effect of the nursing group improved more significantly than the control group.Conclusions To give patients with functional dyspepsia the prescription of dispersing stagnated liver and strengthening spleen combined with psychological care can significantly improve symptoms of functional dyspepsia and psychological symptoms.
2.Evaluation of effect of community health education on type 2 diabetes mellitus patients

Objective To probe into the effect of community health education in comprehensive treatment of type 2 diabetes mellltus(T2DM)patients.Methods 120 cases of T2DM patients were claasified randomly into the intervention group(55 cases)and the control group (65 cases).After systematic diabetes knowledge education was given to the intervention group,individualized intensive education was accepted by each patient of the intervention group.General diabetic education was taken by the control group.The levels of plasma glucose,urinary glucoee,glycosylated hemoglobin and blood-fat were tested before and 3,6 months after education.Results Six months after education,the level of self care ability,self care technique,self care sense of responsibility and health knowledge were higher than those before education in both groups.And six months after education the levels of HbAlc,TC and TG in the intervention group and the control group improved obviously.All indicators in the intervention group were better than those in the control group.Conctusions Community health education can effectively improve self care ability,self care technique,self care sense of responsibility and health knowledge of diabetes patients,and can improve their glucolipide metabolic level as well.
3.Optimization of Purification Technology for Total Flavonoids from Callicarpa nudiflora Hook. et Arn. by Macroporous Adsorption Resin

Objective To optimize purification technology of total flavonoids in Callicarpa nudiflora Hook. et Arm. by macroporous adsorption resin. Methods Macroporous resin models including AB-8, D-101, HP-20, HP2MG, were optimized by static adsorption and desorption experiments regarding to adsorption rate and desorption rate of total flavonoids. Purification technology parameters of total flavonoids were optimized by single factor test. Results HP-20 macroporous resin presented the best purification efficiency,the optimum purification conditions were that taking 4. 46 mg·mL-1 of total flavonoidsat pH 3. 0, loading at 3 BV·h-1, washed with 3BV of water at 3 BV·h-1,then eluted with 4 BV 75% ethanol at 2 BV·h-1, finally obtaining the total flavonoids from the dry extract of Callicarpa nudiflora Hook. et Arn. with the purity of 47. 4%. Conclusion HP-20 macroporous resin is suitable for preliminary purification of total flavonoids in Callicarpa nudiflora Hook. et Arn.
4.Comparison of effects of kaempferide and anhydroicaritin on biomineralization of cultured osteoblasts.

This study is to compare the effects of kaempferide and anhydroicaritin on biomineralization of rat osteoblasts (ROB) in vitro. Calvarias were dissected aseptically from newborn SD rats, the osteoblasts were obtained by enzyme digestion and were cultured in MEM containing 10% FBS. The medium was changed every three days, and serial subculture was performed when cells covered with 90% of the dish. Kaempferide and anhydroicaritin were separately added with final concentrations of 1 x 10(-4), 1 x 10(-5), 1 x 10(-6) and 1 x 10(-7) mol x L(-1) under the conditions of osteogenic differentiation. The proliferation was measured by MTT, and the optimal concentration was detected by the ALP activity at the 9th day after osteogenic induction culture. The osteogenic indexes of kaempferide, anhydroicaritin and control group with the optimal concentration were compared. The result showed that the anhydroicaritin at concentration of 1 x 10(-5) mol x L(-1) had significantly promoted the activity of ALP, calcium content and osteocalcin content, increased the number of CFU-F(ALP) and mineralized nodules, enhanced the mRNA level of BMP-2, OSX and Runx-2, which are key genes of osteogenic differentiation, and raised the protein content of collagen-I. However, the kaempferide group had not significantly represented the ability that promoted osteogenic differentiation of ROB. The difference of osteogenic differentiation on ROB between kaempferide and anhydroicaritin was caused by the prenyl group on C-8 of icariin.
5.Inhibitory effect of 8-prenylnaringenin on osteoclastogensis of bone marrow cells and bone resorption activity.

This study is to investigate the effect of 8-prenylnaringenin (8-PNG) on osteoclastogensis of bone marrow cells and bone resorption activity of osteoclasts. Osteoclasts were separated from long bone marrow of newborn rabbits and cultured in alpha-MEM containing 10% FBS. 8-PNG was added into culture media at 1 x 10(-7), 1 x 10(-6), 1 x 10(-5) mol xL(-1), separately. 17beta-Estradiol (E2, 1 x 10(-7) mol x L(-7)) was used as positive control. T RAP staining and TRAP activity measurement were performed after 5 days, and the bone resorption pits were analyzed after 7 days. Annexin V staining for the detection of apoptotic osteoclasts was performed after 2, 4, 8, 12, 24, 36 and 48 h separately. The mRNA expression level of TRAP and cathepsin K (CTSK) was measured by real-time RT-PCR. 8-PNG significantly reduced the number of osteoclasts which was TRAP staining positive and with more than three nucleus, the area and number of bone resorption pits decreased obviously in 8-PNG-supplemented groups. The apoptosis rate peaked earlier in the 8-PNG-supplemented groups and the mRNA expression level of TRAP and CTSK decreased significantly. All these inhibitory effects were in a dose dependent manner, the highest effect was obtained by 1 x 10(-5) mol x L(-1) 8-PNG. 8-PNG inhibits bone resorption activity of osteoclasts by inducing osteoclast apoptosis and inhibiting the gene expression and enzyme activity including TRAP and CTSK, and restrains bone marrow cells to osteoclast differentiation.
6.Effect of genistein on rat femoral bone metabolic activity in vitro.

This study is to investigate effects of genistein on rat femoral bone metabolic in vitro. Rat femoral tissues was isolated and randomly divided into two groups including control group and genistein (1 x 10(-5) mol x(-1)) group. Determinations of alkaline phosphatase (ALP) activity, calcium content and osteoprotegerin (OPG), type I-collagen (Collagen-I), RANKL, Runx-2 and bone morphogenetic protein (BMP-2) mRNA expression were done by real-time PCR. The results showed that 1 x 10(-5) mol x L(-1) genistein could increase the activity of ALP and contents of Ca, regulate bone metabolism activity of OPG, RANKL, BMP-2, Collagen-I and Runx-2 mRNA expression level. Genistein can significantly modulate bone metabolism related gene expression level of rat femoral tissue in vitro, and can increase calcium content and the activity of ALP.
