1.STUDIES ON RADIATION EFFECTS OF NEUTRON AND ?-RAY TO THE DRUGS

Seventy two kinds of drugs were irradiated by neutron (1?1013cm-2) and ?-ray (1?104R) to evalute their radioactivity, potency, content and some other physicohe-mical properties at intervals of 48 hrs and 3 months after exposure. LD50 of 36 kinds of drugs were measured and main pharmacological effect of 40 kinds of drugs were observed .The induced radioactivities of drugs, the 5% glucose in normal saline and the injection KC1 were found to be about 1? 10-1 ?Ci/ml, the rest were either about equal to or less than 1?10-2 ?Ci/ml(g.).No radiative effdcts were observed among these drugs with the exception of VitB12, ether and 0.1% citric acid The content of VitB12 was dropped after the radiation. Its decrease was found to be proportional to the dose of radiation.Slight changes in color and content were found in 10 of these drugs after radiation, but their extents were within the normal ranges according to the Chinese Pharmacopoeia.
2.Advance in reseach on the diagnosis for pancreatic carcinoma by spiral CT

Carcinoma of pancreas is the most common primar y pancreatic malignancy. Symptoms usually are nonspecific and include abdominal pain, jaundice, loss of appetite, weight loss, nausea, and vomiting. CT usually demonstrates a mass with ill-defined margins but also solid components and dil atation of the main pancreatic duct. This tumor is usually smaller and virtuall y never contains calcification. Following contrast material administration,all masses were enhanced less than the surrounding pancreas. Tumors were oval, rou nd or lobular,and is almost always locally invasive. These features can be used to differentiate carcinoma of pancreas from other pancreatic tumors.
3.Isolation and Identification of Dioscin and Pseudoprodioscin from Di'ao Xinxuekang

OBJECTIVE:To isolate and identify the major constituents-steroidal saponins from Di'ao xinxuekang.METHODS:The constituents were separated and purified using normal-pressured and pressurized silica gel column chromatography technology,and the structures were identified based on the physico-chemical property and spectral analysis.RESULTS:Two steroidal saponins were obtained from Di'ao xinxuekang and their structures were identified as dioscin(1)and pseudoprodioscin(2).CONCLUSION:Compound 1 and 2 were demonstrated to be two major constituents in Di'ao xinxuekang.This finding is of great importance for the quality control and evaluation of Di'ao xinxuekang(Chinese Medicine).
4.Comparison of corilagin content in Erodium stephanianum from different collection month

AIM: To determine the contents of corilagin in Erodium stephanianum Willd. from different collection month and to define the best collection time for this herb. METHODS: The contents of corilagin in each samples of E. stephanianum were determined by HPLC. RESULTS: The linear regression of corilagin was obtained in the range of 0.22~1.10?g. The average recovery was 98.6% with a RSD of 0.63%. CONCLUSION: The content of corilagin in E. stephanianum collected in August is the highest among the samples throughout the collection months.
5.Efficacy of deproteinised cancellous bone combined with BMP and fibrin in healing of rabbit segmental radial defect

[Objective]To repair segmental bone defect with the composite of deproteinised cancellous bone,bone morphogenetic protein(BMP) and fibrin.[Method] Bone defects were created in 36 New Zealand rabbits on the bilateral radii and treated with three kinds of implantations:A,composite of deproteinised cancellous bone,BMP and fibrin;B,of deproteinised cancellous bone;C,blank control.The defects were observed by entgenography,optical microscopy,and electron-microscopy at 2,4,8,12,16 weeks.[Result] The defects treated with A implantation regenerated much more new bone and bridged earlier than the defects treated with B implantation and got completely repaired 16 weeks after operation.The defects with B and C implantation couldn't get osseous tissue healing.[Conclusion]The composite of deproteinised cancellous bone,bone morphogenetic protein(BMP) and fibrin can be effectively used to repair segmental bone defect.
6.Adhesion, proliferation and differentiation of human adipose mesenchymal stem cells on PLGA

Objective To study the ability of adhesion, proliferation and differentiation of human adipose mesenchymal stern cells,which were cultured in PLGA,and thus to provide a basis for their in vivo implantation.Methods PLA was mixed with PGA in a ratio of 1∶ 1,and PLGA was obtained by insert moldling with organic solvents.In vitro, human adipose mesenchymal stem cells were adhered to PLGA.The attachment and the proliferation of human adipose mesenchymal stem cells in PLGA were analysed by scanning electron microscope,and the morphological change of the cells was investigated before and after induction.Results Human adipose mesenchymal stem cells were able to attach, proliferate and secrete extracellular matrix in porous PLGA (pore size:100~400 pm) with porosity of 85 %, and to differentiate into round lipoblast-like cells under the induction of adipose medium.After 14 days, the particulate of PLGA was overlaid by extracellular matrix .Conclusion Human adipose mesenchymal stem cells have the capacity to adhere to PLGA and proliferate, indicating that PLGA can be used as a carrier for human adipose mesenchymal stem cells in adipose tissue engineering.
7.Analysis of the influencing factors of operative effect in complex acetabular fracture

ObjectiveTo investigate the influence factors of operative effect in complex acetabular fracture. MethodsSixty-seven patients with complex acetabular fractures were selected. All patients were used surgical treatment,the clinical curative effect by Merle d'Aubigne-Postel hip scoring function to evaluate,set the age,sex, operation time,operative approach,the quality of fracture reduction,complex dislocation of the hip fracture and type of complex acetabular fractures with comparative index,the results were statistically analyzed. ResultsPatients were followed up for 6-24 months,the mean time was 14.6 months. Univariate analysis showed that the fine efficacy of surgery taken in no more than 2 weeks [84.0% (42/50)]was better than more than 2 weeks [64.7% ( 11/17 )](P < 0.05 ). The high-quality of reduction [86.4% ( 19/22 )]was better than low-quality of reduction [44.4% (4/9)] (P < 0.01 ). The compare of age,sex,operative approach,complex dislocation of the hip fracture were no statistically different (P >0.05).Complex acetabular fractures of T-shaped fracture healing rate was 85.7% (12/14),the excellent rate was significantly higher than posterior column with posterior wall fractures[77.8% (7/9)],transverse with posterior wall fractures [75.0% (15/20)], anterior column with half of transverse fractures [66.7% (2/3)],double-column fractures[71.4% (15/21)],the results were statistically different. Conclusion The necessary conditions for ensuring fracture prognosis is the quality of reduction,timing of surgery,and type of fracture.
8.A Comparative Study of Latanoprost and Timolol in Treating Patients With Glaucoma or Ocular Hypertension

OBJECTIVE: To confirm the efficacy and safety of latanoprost in treating glaucoma.METHODS: In a double blind, randomized control clinical trial, we compared the efficacy and adverse drug reactions of once daily topically applied 0.005% latanoprost with those of twice daily 0.5% timolol for 12 weeks in patients with open angle glaucoma or ocular hypertension.RESULTS: The study included 46 patients(22 pts.randomized to latanoprost treatment, 24 pts.to timolol) , 46 patients remained at the end of the study.Comparing with baseline diurnal intraocular pressure(IOP) , the IOP reduction(mean±standard deviation) achieved with latanoprost(7.86±2.39) mmHg, (31.1%, P<0.001),and timolol(6.24±2.58)mmHg (24.9%,P<0.001),the difference between the two groups(1.62mmHg) being significant(P<0.01). Two patients treated with latanoprost had foreign body sensation. No other ocular and systemic adverse reactions related to the two drugs were found. CONCLUSION: The results of this study demonstrated that 0.005% latanoprost applied once daily is well tolerated and more effective in reducing IOP than 0.5% timolol applied twice daily. Thus, latanoprost has the potential for becoming one of the ideal antiglaucoma drugs.
9.Establishment of Mice Model of Reproductive Hypofunction with Kidney Deficiency Caused by Tripterygium Glucosides

Objective To study the pharmacological mechanism of glucosides of tripterygium(GTW)inducing reproductive hypofunction in female mature mice and to establish female mice model of reproductive hypofunction.Methods Female Kunming mice aged 7~ 8 weeks were randomly divided into 6 groups,including GTW groups of A,B,C,D and E(60,40,30,20,15 mg? kg-1? d-1)and normal group.Estrous cycle of five GTW groups was observed successively.To study the effect of GTW on reproductive function in different period,three batches of mice were killed when estrous cycle of some mices was prolonged,estrous cycle of all the mice was prolonged or became irregular,or estrous cycle of all the mice was totally disturbed or even disappeared.Serum E2 and P levels were measured by radioimmunoassay.Ovary and uterus index were measured by tissue-weight-assay,and tectology observation on ovary and uterus was conducted.The numbers of follicle and corpus luteum as well as thickness of endometrium and myometrium were also measured.Results The mice were killed in batches after administration for 5,7,10 weeks.After 10-week treatment,uterus index and the numbers of follicle and luteum were decreased significantly,and endometrium and myometrium also became obviously thinner in groups A,B,and C.E2 level in groups A and B was decreased.P level and ovary index of all treatment groups were not decreased after 10-week treatment.Conclusion GTW at the dosages of 60,40 and 30 mg? kg-1? d-1 can damage female mice' s reproductive function and can induce mice reproductive hypofunction with kidney deficiency after 10-week successive treatment.GTW at the dosage of 40 mg? kg-1? d-1 for 10 weeks is recommended to establish female mice model of reproductive hypofunction with kidney deficiency.
10.Effects of osthol on bone marrow stromal stem cell differentiation and proliferation in vitro

Aim To investigate the effects of osthol on bone marrow stromal stem cells in vitro under the con-ditions of the ability to differentiate into osteoblasts and the case of proliferation. Methods The rat bone marrow sample was obtained,and the all bone marrow cell culture methods were used to separate and collect the stratum of mononuclear cells. The cells were cultured in DMEM containing 10% fetal bovine serum. Three days later the culture medium was changed for the first time. Nine days later,serial subcultivation proceeded. The final concentration of osthol was 1 ? 10 -4,1 ? 10 -5,1 ? 10 -6,1 ? 10 -7 mol?L -1 respectively. MTT method was adopted in proliferation analysis. Under the induced condition,the Alkaline phosphatase activity, calcium salt sediment yield and osteocalcin were meas-ured on 4th,8th,12th,16th day. On the fifteenth day,his-tochemistry dyeing proceeded for calcified tubercle. Total RNA was isolated and the gene expression of bFGF, IGF-1,Osterix and Runx-2 was investigated by RT-Real Time PCR. Results The BMSCs proliferation was refrained by osthol dose-dependently. But it evidently led to osteogenesis. The ALP activity,calcium salt sediment yield and osteocalcin were raised,and calcified tubercle amount was increased. Besides,it also could enhance the mRNA level of bFGF,IGF-1,Osterix and Runx-2. Conclusion The osthol with final concentra-tion of 1 ? 10 -5 mol?L -1 can markedly promote BM-SCs differentiation to osteogenesis. which proves osthol is an active constituent of the traditional Chinese medi-cine Common Cnidium Fruit.
