1.Antagonistic action of Jiunaoning injection aganist oxygen/glucose-deprived and reperfusion injury-induced cultured rat cortical neurons

AIM: To determine whether Jiunaoning injection has protective effects on oxygen/glucose-deprived and reperfusion injury-induced neurons. METHODS: Various concentrations of Jiunaoning injection (0.5-5 mL/L) were used to observe their effects on cultured rat cortical neurons induced by oxygen/glucose-deprived and reperfusion injury in various time points. The neuronal metabolic rate and viability were assessed by using 3-(4,5-dimethylthiazol)-2, 5-diphenyl-tetra zoliumbromide (MTT) and lactate dehydrogenase (LDH) assay. RESULTS: Jiunaoning injection enhanced the neuronal metabolic rate in a dose-dependent manner in the range from 0.5 to 5 mL/L, and Jiunaoning injection (1.5-2.5 mL/L) enhanced the neuronal metabolic rate, decreased the cell death rate and depressed LDH leak rate significantly. CONCLUSION: Jiunaoning injection has an affirmative protective effect on oxygen/glucose-deprived and reperfusion-induced neuronal injury.
2.The effect of infrared spectral analysis in analyzing of the chemical composition of renal staghorn ;calculi and its relationship with urinary tract infections

Objective To explore the effect of infrared spectral analysis in analyzing of the chemical composition of renal staghorn calculi and its relationship with urinary tract infections. Methods From June 2014 to June 2016, the clinical data of 186 patients with renal staghorn calculi were collected. The stone composition were analyzed by infrared spectroscopy and traditional chemical titration, and the stones infection were detected by microbial analysis system. The relation between stones infection, urinary tract infection and stone composition were analyzed. Results The results of infrared spectroscopy and traditional chemical titration in detecting renal staghorn calculi ingredient had no significant differences (P>0.05). In 186 patients, 56 patients (30.11%) was in infected group, and 130 patients (69.89%)was in non-infected group. The abnormal urine rate, urinary tract infection rate, medistream urine positive infection rate and cotton swabs positive infection rate in infected group were was significantly higher than those in non-infected group: 73.21%(41/56) vs. 50.77%(66/130), 19.64%(11/56) vs. 3.85%(5/130), 50.00%(28/56) vs. 6.15%(8/130), 67.86%(38/56) vs. 8.46%(11/130), P<0.01. The carbonate apatite stones rate and six water magnesium ammonium phosphate rate in infected group were significantly higher than those in non-infected group: 21.43%(12/56) vs. 5.37%(7/130), 57.14%(32/56) vs. 2.31%(3/130), P<0.01. The calcium oxalate rate and uric acid rate in non-infected group were significantly higher than those in infected group:50.00%(65/130) vs. 5.36%(3/56), 24.62%(32/130) vs. 1.79%(1/56), P<0.01. Conclusions Analysis of staghorn calculi ingredient caused by urinary bacterial infection with infrared spectroscopy is simple, reliable and easy to operate. It is important for postoperative infection prevention.
3.Effect of Zhongfeng Nao De Ping on delayed neuronal death in hippocampus following cerebral ischemia/reperfusion in rats

AIM: To investigate the effect of Zhongfeng Nao De Ping, a traditional chinese medicine, on the cerebral ischemia/reperfusion injury in rats. METHODS: Delayed neuronal death (DND) model was established by bilateral carotid arteries occlusion for l0 min followed by 1 day and 7 days reperfusion. Effects of sodium glutamate, ketamine and Zhongfeng Nao De Ping on DND in the hippocampus was examined by counting neuronal density in CAl Sector, and the contents of Ca 2+ and amino acids in the hippocampus were also measured. RESULTS: The contents of the glutamate and aspartate in the hippocampus were increased significantly at l0 min after transient cerebral ischemia. Sodium glutamate markedly decreased the neuronal density in hippocampal CAl sector, while ketamine and Zhongfeng Nao De Ping increased it significantly in the same sector. The level of Ca 2+ increased on day 1, day 7 after the reperfusion, while Zhongfeng Nao De Ping inhibited the increase in Ca 2+ concentration in hippocampus. CONCLUSION: These results suggested that Zhongfeng Nao De Ping has an effective protection against DND in hippocampus following cerebral ischemia/reperfusion in rats.
4.Influence of Xiaoke keli Ji on glycemia and serum lipoprotein levels in diabetic nephropathy rats

AIM: To observe the changes of glycemia and serum cholesterol and triglyceride in diabetic nephropathy rats and therapeutic effects of Xiaoke Keli Ji. METHODS: 3/4 nephrectomy was adopted firstly, three weeks later streptozotocin(STZ) was administered intraperitoneally to establish diabetic nephropathy model in rats. Animals were divided into four groups:model group, Xiaoke Keli Ji treatment group, positive control group and sham group. Changes of serum sugar and serum creatinine, cholesterol and triglyceride were examined at 1, 2, 3, 4, 5 weeks after STZ injection. Renal tissue samples were adopted at 6th week and studied by light microscopy. RESULTS: Model group demonstrated different degree of glomerular sclerosis. Lesions in treatment group were alleviated. 5.Effect of "Shen Yan Yi Qi Solution" on renal function and histopathology of rats suffering from an experimental nephrotoxic serum nephritis

AIM: To investigate the therapeutic effects of "Shen Yan Yi Qi Solution",a chinese Medicine,on nephrotoxic serum nephritis in the rat. METHODS: Nephrotoxic serum nephritis model of the rat was set up by the Clarke's method. RESULTS: Proteinuria and renal insufficiency occurred in all rats after nephrotoxic serum (NTS) injection,but symptoms in treatment group were reduced. At 2nd week,the typical linear deposition of IgG along the glomerular basement membrane (GBM) was observed by fluorescence microscopy. Mesangial hyperplasia and crescent formation in glomeruli were also observed by light microscopy. In treatment group,all the above lesions were alleviated. CONCLUSION: It demonstrated that "Shen Yan Yi Qi Solution" might alleviate the pathological change of glomeruli and improve renal function.
