1.ULTRASTRUCTURAL OBSERVATION OF MOUSE MAMMARY ADENOCARCINOMA CELL LINE (MA782/5S-8102)

The ultrastructural characteristics of the mouse mammary adenocarcinoma cell line (MA782/5S-8102) have been studied by transmission electron microscope. The results show that the cells were irregular in the shape, with abundant microvilli on cell surface. The nucleus was abnormal in shape with the nuclear membrane sunk or swollen, the nucleus/cytoplasmic ratio (N/C) was high. There were many free ribosomes, abundant mitochondrias, dilated rough endoplasmic reticulum in the cytoplasm. Golgi apparatus, lysosomes and lipid droplets were present. Moreover, many cytoplasmic type-A particles and a few immatural type-B particles at the surface of the cell were observed.
2.The application of MRI in gluteal muscle contracture

Objective To evaluate the MRI findings and its diagnostic value in gluteal muscle contracture (GMC). Methods Eleven clinic or operation confirmed GMC patients were examined by plain X-ray and MRI. Conventional T 1WI and T 2WI MR imaging were performed and FFE-T 2WI (fast field echo-T 2WI) was also scanned. CT scan was conducted in 5 cases. Results 11 GMC patients were all diagnosed by MRI. Conventional T 1WI and T 2WI could only show the atrophy of gluteal muscles, while FFE-T 2WI could directly show the fibrous band of gluteal muscle and its fascia, and the fibrous band appeared as low signal intensity on FFE-T 2WI sequence. Conclusions MRI is the efficient modality in imaging the fibrous band for GMC patients, and FFE-T 2WI is the most valuable sequence. MRI is very helpful in the diagnosis and treatment of GMC.
3.Magnetic resonance imaging monitoring for osteonecrosis of the knee joints in recovered patients with severe acute respiratory syndrome after administration of glucocorticoids

Objective To evaluate the magnetic resonance imaging (MRI) monitoring for osteonecrosis of the knee joints in recovered patients with severe acute respiratory syndrome (SARS) after the administration of glucocorticoids and its characteristics. Methods One hundred and ninety-eight knee joints in 99 recovered SARS patients after administration of glucocorticoids,22 males and 77 females,were examined by MRI and interpreted independently by four senior radiologists for determining osteonecosis. Results Osteonecrosis were found in 56 knee joints (28%,56/198) in 31 of 99 patients (31%,31/99),25 with bilateral lesions (81%,25/31) and six unilateral ones (19%,6/31). The lesions were symmetric (100%,25/25) in all the patients with bilateral osteonecosis,involving weight-bearing zone in 27,non-weight-bearing zone in 29 and subchondral zone in 47 knee joints (84%,47/56). The anterior zone of the medial femoral condyle was involved in 14 (8%) lesions,the posterior zone of the medial femoral condyle in 27 (16%) ,the anterior zone of the lateral femoral condyle in 29 (17%),the posterior of the lateral condyle in 44 (27%),the metaphysis of femur in 18 (11%),the anterior zone of the medial tibial plateau in three (2%),the posterior zone of the medial tibial plateaus in 13 (8%),the anterior zone of the lateral tibial plateaus in two (1%),the posterior zone of the lateral tibial plateau in seven (4%),the metaphsis of the tibia in nine (5%),and no lesion was found in the patella. Conclusions MRI can preferably be used to reveal changes of ischemia and necrosis in the knee joints. So,MRI for the knee joints should be earlier performed in the patients with glucocorticoids for diagnosis and therapy for them.
4.Changes and clinical significance of intestinal Bacteroides in obese children

Objective:To analyze the diversity of Bacteroides in simple obesity children aged 7-14 and the difference from that of normal individuals.And to explore the influence of children's simple obesity on intestinal advantage bacterium of the genus.Methods:We have collected fecal samples of 30 cases with simple obesity aged 7 to 14 years old and 30 healthy children in the Central Hospital of Shan County.All bacterial DNAs were extracted from the feces, and then Bacteroides 16s DNA specific primers was used in PCR amplification, separated by DGGE, the intestinal Bacteroides molecular fingerprints of obese children and healthy children were obtained, and the diversity and similarity were analyzed.At the same time, the weight, height, BMI, blood pressure, blood lipid, leptin, plasma adiponectin in the obesity and normal children were measured, the relationship between the obese children and the advantage of intestinal bacteria genera and its clinical significance were investigated.Results:The BMI[(25.71±3.28)kg/m 2], SBP[(121±12.4)mmHg], TG[(1.15±0.40)mmol/L], TC[(4.20±0.73)mmol/L], LEP(18.94±1.68) in the obesity group were higher than those in the normal group [BMI(17.38±3.54)kg/m 2, SBP(104±7.3)mmHg, TG(0.87±0.31)mmol/L, TC(3.37±0.86)mmol/L, LEP(11.29±2.41)](all P<0.05). The levels of HDL[(1.01±0.28)mmol/L] and ADP(4.10±2.32) in the obesity group were lower than those in the normal group [HDL(1.40±0.26)mmol/L, ADP(5.84±3.42)]( P<0.05). There were no statistically significant differences in age, DBP and LDL levels between the two groups (all P>0.05). The stripe number of Bacteroides DGGE profiles (9.33±1.37) and H index(2.07±0.16) in the obesity group were lower than those in the normal group [(17.33±2.80), (2.67±0.20)]. Cluster analysis results showed that the similarities of obese individuals in the group and compared with normal group were reduced.The S and H index of DGGE profiles in Bacteroides were negatively correlated with BMI and LEP ( P<0.01), and positively correlated with ADP( P<0.05). S, H index of the normal group were negatively correlated with LEP ( P<0.05), and had no correlation with BMI, ADP( P>0.05). S and H index of the obesity group were negatively correlated with BMI and LEP, and had no correlation with ADP ( P>0.05). Conclusion:Human intestinal flora of Bacteroides changes is related to childhood obesity, the diversity and similarity of obese children's intestinal Bacteroides species are decreased.The diversity of obesity children's Bacteroides is associated with BMI, ADP and LEP.
5.Correlations between image quality and radiation dose in prospective and retrospective ECG-gated CT coronary angiography in patients with different heart rates

Objective To explore the correlations between image quality of prospective and retrospective electrocardiogram(ECG)-gated CT coronary angiogram and radiation dose in patients with different heart rates.Methods A total of 135 patients undergoing 256-slice spiral CT coronary angiography were enrolled in the study.Among them,66 cases received prospective ECG-gated scanning(prospective ECG-gated group)and further divided into two subgroups with heart rate≤80 beats/min(prospective ECG-gated+low heart rate subgroup,n=39)and>80 beats/min(prospective ECG-gated+high heart rate subgroup,n=27).The other 69 cases underwent retrospective ECG-gated scanning(retrospective ECG-gated group),including 45 cases with heart rate≤80 beats/min(retrospective ECG-gated+low heart rate subgroup)and 24 with heart rate>80 beats/min(retrospective ECG-gated+high heart rate subgroup).The baseline data,image quality[mean CT value,image noise,signal-to-noise ratio(SNR),subjective image quality score]and radiation dos[CT volume dose index(CTDIvol),dose length product(DLP),effective dose(ED)]were compared among 4 subgroups.The correlations of image quality with heart rate and radiation dose in prospective and retrospective ECG-gated groups were analyzed.Results The heart rates in prospective and retrospective ECG-gated+low heart rate subgroups were lower than those in prospective and retrospective ECG-gated+high heart rate subgroups(P<0.05).When comparing the mean CT value,image noise,SNR and subjective image quality score among 4 subgroups,no statistically significant differences were observed(P>0.05).The CTDIvol,DLP and ED in prospective ECG-gated+low heart rate subgroup were significantly lower than those in the other 3 subgroups(P<0.05),and the indicators in prospective ECG-gated+high heart rate subgroup were lower than those in retrospective ECG-gated group(including low and high heart rate subgroups)(P<0.05).Pearson correlation coefficient analysis revealed that the mean CT value,image noise,SNR,subjective image quality score had no significant correlation with heart rate,CTDIvol,DLP and ED in prospective and retrospective ECG-gated groups(P>0.05).Conclusion The subjective and objective image quality of 256-slice spiral CT coronary angiography is not correlated with radiation dose.Prospective ECG-gated scanning can reduce the radiation dose and ensure the image quality as compared with retrospective ECG-gated scanning.This holds true for eligible patients with high heart rate,and the former can effectively reduce radiation exposure.Therefore,prospective ECG-gated scanning is worthy to be promoted in clinic.
