2.Effect of Different Methods on Surface Disinfection after Non-infected Surgery

OBJECTIVE To discuss the effect of the different methods on the surface disinfection after the non-infected surgery.METHODS The surface of the objects in the operating room after non-infected surgery was monitored which were divided randomly into group A(n:30) and group B(n:30).The samples of group A which wiping with water and of group B which spraying with Dian′er Kang spray disinfectant were collected.RESULTS After using water to wipe the surface of the surgery mattress,the body position mattress,the arm-support,the instrument table,and the standing stool,the numbers of the fall bacteria were(8.96?3.41),(6.87?2.39),(5.75?2.30),(6.80?2.31) and(7.94?3.40)CFU/cm2,respectively.After the method of spraying by Dian′er Kang spray disinfectant,the numbers were(0.56?1.22),(0.65?1.96),(0.50?1.20),(0.70?1.51) and(0.75?1.30)CFU/cm2.There was statistically significant difference between them(P
3.Short-term and long-term efficacy of laparoscopic versus open liver resection in the treatment of hepatocel-lular carcinoma

Objective To explore the short-term and long-term efficacy of laparoscopic versus open liver resection(LLR)in the treatment of hepatocellular carcinoma(HCC). Methods The perioperative and follow-up data ofLLR(n = 43)and OLR(n = 73)for the treatment of HCC at the same period were analyzed respectively. Results There were no significant differences in the age,gender,Child-Pugh classification,liver cirrhosis, AFP,and hepatic virus affection between the 2 groups. 1 case in the LLR group was converted for the open surgery and another one case in the OLR group died in the perioperative time. However ,compared with OLR group ,the intraoperative blood loss of LLR group(P < 0.05)was larger and the mean operative time(P < 0.05)was longer. The liver function indicators of the regular hepatectomy subgroup were recovered more quickly(P<0.05),but the indicators of remaining subgroups were not significantly different. The postoperative ambulation time (P < 0.05), the postoperative eating time(P < 0.05),the postoperative hospital stay(P < 0.05)were all shorter. The VAS score was lower(P<0.05). There was no difference in the incidence and distribution of complications and the inci-dence of severe complications between the two groups(all P>0.05). The first and third year disease-free survival rates were 82.2%,53.8%for LLR and 91.5%,75.9%for OLR,respectively(χ2=0.55,P=0.46). The first and third 5 year overall survival rates were 94.2%,79.3%for LLR and 94.9%,47.3%for OLR,respectively(χ2=1.06, P=0.30). Conclusion LLR for HCC treatment is a safe and effective way with the advantages of minimal opera-tive trauma,quick recovery and significant short-term efficacy.
4.Preliminary study for integrating DPRA with h-CLAT to predict skin sensitizers

Objective To establish a detection method integrating DPRA ( direct peptide reactivity assay) with h?CLAT ( human cell line activation test) to screen the skin sensitization potency of chemicals and plant extracts. Methods 12 chemicals and 7 plant extracts were chosen as the test substances. Firstly, the test substances were incubated together with two different peptides ( cysteine and lysine) respectively for reaction for 24 h. The peptide consumptions were analyzed by HPLC. Simultaneously, THP?1 cells were cultured in vitro and then exposed to different concentrations of test sub?stances for 24 h to examine the cell viability, cell surface markers CD54 and CD86 were assessed by flow cytometry. The predicting results were compared further between DPRA and h?CLAT. Results 12 chemicals were distinguished correctly by DPRA classified as 2 non?sensitizers and 10 sensitizers. The results of DPRA were in accordance with h?CLAT. Predic?ting the sensitization potency of plant extracts by DPRA showed that 6 plant extracts were determined as suspected sensiti?zers except for green tea extract. But using the method of h?CLAT, 4 plant extracts were examined as suspected sensitizers except for green tea extract, herba portulacae extract and ginseng fruit extract. The coherence of DPRA and h?CLAT was 0?57. Conclusion This detection method integrating DPRA with h?CLAT can predict single compound accurately. As for complex compound, it can achieve preliminary prediction and need other integrating methods to make a further identifica?tion.
5.Modified liver hanging maneuver in the application of hemihepatectomy

Objective To evaluate a modified liver hanging maneuver(retrohepatic tunnel of the IVC) in patients undergoing hemihepatectomy.Methods Twenty-four patients undergoing hemihepatectomy were divided into two groups:modified liver hanging maneuver group(n=12)and Pringle's maneuver group(n=12).The amount of intraoperative bleeding,operation time,postoperative liver function,liver function recovery and complications were compared between the two groups.Reset All operation were performed successfully and there were no difference in the time of operation etween the two groups.There was a difierence in the amount of mean intraoperative blood loss between the two groups.It was(160±40)ml in liver hanging group and(560±120)ml in Pringle's group(P<0.01).Liver function recovery measured on postoperative day 3 and day 7 was better in liver hanging groupthan that in Pringle's group(P<0.01).The volume of postoperative peritoneal serous fluid dranage was significantly less in liver hanging group(P<0.01).Conclusion The modified liver hanging maneuver is useful for hemihepatectomy.
6.Construction of an immortalized rat astrocyte strain with inducible enkephalin expression

Objective To construct an immortalized rat astrocyte strain expressing enkephalin which can be regulated by doxycycline.Methods pRevTet-On and recombinant plasmid pRevTRE/hPPE were transfected into packaging cell PT67 respectively by standard lipofectamine methods.The supernatant containing RevTet-On virus was used to infect immortalized rat astrocyte strain(IAST)and RevTRE/hPPE virus was used to infect the IAST/Tet-On cell.Immortalized rat astrocyte strain that stably expressed Tet-regulated enkephalin was established. hPPE gene expression level of IAST/Tet-On/hPPE cell strain was detected by real time-PCR,immuno- cytochemistry and radio-immunoassay.Results Real time-PCR,immuno-cytochemistry and radio-immunoassay confirmed the level of enkephalin expression was regulated by doxycycline in a dose-dependent manner.Conclusion An immortalized rat astrocyte strain which secrets enkephalin as controlled by doxycycline has been constructed successfully.
7.Treatment of osteosarcoma by targeting transportation of pHSP70-shPLK1/DOX compounds with bacterial magnetosomes

BACKGROUND:In an additional changing magnetic field, magnetosomes can invade the tumor cells and inhibit their proliferation.OBJECTIVE: To study the effect of targeted delivery of pHSP70-shPLK1/DOX compounds with magnetosome of magnetotactic bacteria on the treatment of osteosarcoma.METHODS: Human osteosarcoma cell U2OS was divided into four groups: group A: magnetosomes of magnetotactic bacteria+pHSP70-shPLK1/DOX complex; group B: magnetosomes of magnetotactic bacteria; group C: magnetosomes of magnetotactic bacteria+pHSP70-shPLK1/DOX complex, combined with magnetic intervention; group D: magnetosomes of magnetotactic bacteria, combined with magnetic intervention. After 12, 24, 48 and 72 hours of culture, the uptake rate of osteosarcoma cells was observed; cell cycle was evaluated by flow cytometry; PLK1 mRNA and protein expression levels were detected by RT-PCR and western blot, respectively; cell proliferation was explored by MTT assay; cell adhesion and invasion were checked by adhesion assay and Transwel chamber assay, respectively; and the apoptosis rate was measured by apoptosis detection Kit.RESULTS AND CONCLUSION: (1) After culture of 12 and 24 hours, the uptake rate was the highest in group D; meanwhile, the uptake rate was the highest in group B after 48 and 72 hours of culture. (2) The ratio of G2/M phase decreased and the ratio of G0/G1 increased gradualy in group A, group C and group D; the G2/M phase ratio of C group was the lowest at each time point, followed by group D, and that of group A was the highest; and the G2/M phase ratio increased gradualy in group B. (3) Folowing 12 and 24 hours of culture, the expression of PLK1 in group B and group D was higher than that in group A and group C (P < 0.05); in addition, the expression of PLK1 in group B was higher than that in the other three groups after 48 and 72 hours of culture (P < 0.05), and the PLK1 level in group D was higher than that in group A and group C (P < 0.05). (4) The proliferation, adhesion and invasion ability in group B were the highest at different time points, but the apoptosis rate was the lowest. Furthermore, the proliferation, adhesion and invasion abilities in group C were the lowest at different time points, and the apoptosis rate was the highest. To conclude, our experimental results show that the targeted delivery of pHSP70-shPLK1/DOX complex by magnetosomes of magnetotactic bacteria can enhance the apoptosis of osteosarcoma cells and inhibit their proliferation and invasion.
8.Status of anxiety and depression on clinical decision-making participation of laryngeal cancer patients

Objective To study the status of anxiety and depression on clinical decision-making participation of laryngeal cancer patients.Methods A total of 82 laryngeal cancer patients participated in the investigation by questionnaire to study the clinical decision-making participation by control preferences scale.The degree of anxiety and depression was investigated by self-rating anxiety scale and self-rating depression scale.Results During the clinical decision-making process,40.24% of the patients were passive,29.27% had their families as absolute representatives,20.73% took part collaborating with their families and only 9.76% were active.47.56% had anxiety and 64.63% depression,and the anxiety and depression influenced their participation in clinical decision-making postively and passively relatively.Conclusions Patients with laryngeal cancer are passive in clinical decision-making and most of them have anxiety and depression.Doctors should enhance the communication with patients,which can help to eliminate adverse psychological emotions and make satisfying clinical decision-making.
9.Study on the change of migration ability of dendritic cells induced by intermittent hypoxia and the mechanism of its pathway

Objective By simulating the intermittent hypoxia(IH ) environment of human obstructive sleep apnea syndrome (OSAS) ,to reveal the effect of IH on migratory ability of human peripheral blood derived dendritic cells(DCs) ,and through the in‐tervention of RelB ,p38 expression in order to explore the possible mechanism of the change of DCs migration ability induced by IH . Methods DCs were divided into RelB ,p38 siRNA interfering and non interfering plasmid group before cultivation .The environment of hypoxia was created by a intermittent hypoxia cabin ,among them ,oxygen concentration was 0 .5% ,1 .5% ,5 .0% ,10 .0% ,hypox‐ia/reoxygenation time ratio was set as 1∶1 ,1∶3 ,1∶5 and 1∶9 ,while sustained oxygen was supplied to the contrast at a normal concentration of 21 .0% .The content of RelB and p38 was tested by Western blotting after culture in vitro ,migration ability of DCs was detected by invasion chamber .Results Compared with normoxia ,DCs under IH tended to have declined migratory ability , which was confirmed to be correlated with the average oxygen partial pressure level under IH .IH could promote the expression of RelB and p38 in DCs ,while the migratory ability of DCs was not reversed after intervening the expression of RelB and p38 .Conclu‐sion IH in vitro could cause a decline in migratory ability of DCs ,which may not be induced by activation of RelB or p38 in DCs .
10.Analysis of prognostic factors of non-small cell lung cancer in patients under 40 years of age.

OBJECTIVETo investigate the prognostic factors for non-small cell lung cancer(NSCLC)in patients under 40 years of age.
METHODSThe clinicopathological data of 148 young patients with NSCLC were retrospectively analyzed. Kaplan-Meier and Cox regression analyses were used to analyze the relationship between prognostic factors and survival time.
RESULTSThe patients were followed-up for 6 - 148 months, and the follow-up rate was 100%. In the whole group, 122 patients died and 26 cases were surviving. The 1-, 3- and 5-year survival rates were 54.7%, 10.4% and 5.6%, respectively. The median survival time (MST) was 14.7 months. Kaplan-Meier analysis showed that Karnofsky performance status (KPS), clinical stage, treatment modality and serum CEA were related with prognosis (P < 0.05). Multivariate analysis indicated that KPS, clinical stage, treatment modality and serum CEA were independent prognostic factors (P < 0.05).
CONCLUSIONSKPS, CEA, clinical stage and treatment modalities are independent prognostic factors in young NSCLC patients.
