2.Blindness from age-related macular degeneration in Changning District of Shanghai from 2003 to 2008

Total 1049 new cases of blindness were detected by visual acuity survey in a population aged 68 or above in Changning District of Shanghai from 2003 to 2008. Among all eases 21.5% were age-related macular degeneration (AMD) blindness, with a trend of ascending from 17.7% in 2003 to 29. 9% in 2008. Most of the cases were classified as grade 3 (56. 9% ) or grade 4 (41.3%) of blindness. AMD has become one of the leading causes of blindness at present and should be the key eye disease for prevention of blindness.
3.Quantitative Determination of Two Components in Isoniazid and Vitamin B6 Tablets by HPLC

Objective:To establish a determination method for isoniazid and vitamin B6 tablets by HPLC. Methods: The proce-dure was performed on an Agilent Eclipse Plus C18 (250 mm × 4. 6 mm,5 μm) column at 30℃ and the detection wavelength was 291 nm. The mobile phase was composed of 0. 1% triethylamine [ in which 20 mg sodium 1-octanesulfonate was added and the pH value was adjusted to (3. 0 ± 0. 05) by phosphoric acid] and methanol (70∶30) at a flow rate of 1. 0 ml·min-1 , and the injection volume was 10 μl. Results:The linear range of isoniazid and vitamin B6 was within the range of 2. 535-50. 700 μg(r=1. 000 0)and 0. 137-2. 735 μg(r=1. 000 0), respectively. The mean recovery was 99. 57%(RSD=0. 89%) and 99. 59%(RSD=0. 43%) (n=9), re-spectively. Conclusion:The method is specific, accurate, sensitive and simple, which is applicable in the quality control of isoniazid and vitamin B6 tablets.
4.Study on coronoid process of ulna osteotomy in the treatment of humerus condyle comminuted fracture in the elderly

Objective To observe the clinical effects of coronoid process of ulna osteotomy and bilateral anatomic locking plate on humerus condyle comminuted fracture in the elderly.Methods 17 elderly patients with humerus condyle comminuted fracture were collected in department of orthopaedics in our hospital from July 2005 to December 2013 and graded into C2 (6 cases) and C3 (11 cases) according to the standard of the American society for the prevention of bone trauma.All patients were treated by coronoid process of ulna osteotomy and bilateral anatomic locking plate and accessed by Mayo elbow performance score (MEPS) at the last follow-up.Results Postoperative incision in all patients were healed,and X-ray showed fracture healing occurred at 10 to 24 weeks after coronoid process of ulna osteotomy (average of 14 weeks).No iatrogenic ulnar nerve injury and nonunion were found at the follow-up.The MEPS were excellent in 13 cases,well in 3 cases,ordinary in 1 case,and the excellent and good rate was 94.1%.Conclusions Coronoid process of ulna osteotomy and bilateral anatomic locking plate in the treatment of humerus condyle comminuted fracture in the elderly have good clinical effects with many advantages such as exposure clearly,early elbow joint function exercise and avoiding the aggravation of elbow joint osteoarthritis.
5.Protection of adenosine A1 receptor agonist delayed preconditioning for myocardial ischemia reperfusion injury in rabbits

Objective:To investigate the protection mechanism of adenosine A1 receptor agonist(2-chloro-N6-cyclopentyladenosine, CCPA)delayed preconditioning for rabbit myocardium during ischemia reperfusion.Methods:Thirty New Zealand male white rabbits were randomly assigned to Sham group,I/R group and CCPA group,with ten in each.Group CCPA was given CCPA 0.1 mg/kg before myocardial ischemia.Twenty-four hours later the I/R and CCPA underwent 40 min of coronary occlusion followed by 2 h of reperfusion.Blood samples were taken from arterial line at 20 min before occlusion(T)1,20 min after occlusion(T2),40 min after occlusion(T)3,1 h after reperfusion(T)4and 2 h after reperfusion(T)5for determination of the plasma levels of IL-10.At the end of the reperfusion,infarct size(IS)and area at risk(AAR)were defined by Evans blue and TTC staining.The heart was harvested and levels of the HSP70 were determined by Western blot analysis,and ultrastructures were observed under the electron microscopy.Results:The IL-10 and HSP70 levels of group CCPA was higher than those of group I/R.The CCPA(P
6.Analysis of the effect of pcDNA3.1 on house keeping gene expression

Objective:To investigate the effect of pcDNA3.1 vector on house keeping gene expression and set guide to further study work in using pcDNA3.1. Methods:5 cell lines were transfected with pcDNA3.1,48 h post transfection total RNA was extracted and applied for real time PCR to detect the RNA level of GAPDH、 ?-actin、18sRNA difference.Meanwhile,extract total protein and analyze the GAPDH and?-actin expression by Western blot.Results:RNA and protein levels all show of a decrease in 5 cell lines after transfection,with the most obviously ?-actin.Conclusion:pcDNA3.1 might have non specifinhibition effect to cell,when using pcDNA3. 1 for gene function research,it is not recommend to use ?- actin as reference gene.

8.Monitoring of the difference of pressure of arterial oxygen between the carotid artery and the femoral artery and clinical significance in patients with head injury

Objective:To study difference in arterial oxygen(PO 2) pressure between the carotid artery and the femoral artery and its relation with hypoxia of brain after head injury.Methods:Forty patients with acute head injury were divided into four groups according to the GCS(Ⅰ:6 patients for GCS 3-5score;Ⅱ:16 patients for GCS 6-8 score;Ⅲ:8 patients for GCS 9-12 score;Ⅳ:10 patients for GCS 13-15score).The PO 2 of the carotid artery and the femoral artery was monitored for every patient.At the same time,26 patients from all groups were monitored using transcranial dopplergraphy.Additional,22 patients with severe head injury were continuously monitored on PO 2 of the carotid artery and the femoral artery for 5 days.Results:(1)The difference in PO 2 between the carotid artery and the femoral artery was identified in Ⅰ and Ⅱ groups.The PO 2 of carotid artery is much less than that of femoral artery(P
9.Application of high-throughput sequencing technology in lung cancer research

Over the past decade,the treatment of lung cancer gradually develops to individulized molecule targeted therapy for certain driver gene from chemotherapy based on histological features.High-throughput sequencing (HTS) technology has many advantages,such as higher throughput,shorter running time,less cost and so on.Further mining analysis to the massive data generated by HTS can reveal the mechanism of the development of lung cancer at genome level and seek new driver gene as the target of targeted therapy,it also can identify the gene alternatives that confer resistance to certain targeted therapy.
10.The review of chemotherapies containing fluorouracil and their side effects
