Caveolin,a membrane protein of 21-24 k,is the main structural component of caveolae,it plays quite a diverse functional role in maintaining vital activities of cells like endocytosis,cholesterol transport,cell membrane composition,and transduction of transmembrane signal to some virus involved in the processes of virus infection.Caveolin-1 also is a key regulator of multiple steps of oncogenesis and tumor development.Caveolin-1 can also serve as a negative regulator in proliferation,apoptosis,invasion and metastasis in varieties of cancer cells.There are some conflicting roles in different tumors,this review summarized the recent advances in the function of caveolin in cancers.

3.Influence of Heat Shock Treatment upon the Expression of Membrane HSP70 and Its mRNA of H22 Cell

To identify the best inducing condition, we studied the expression of membrane protein HSP70 and mRNA of H22 cell at various temperature. Using MTT,RT-PCR, immunofluorescence and FCM techniques, we observed H22 cell survival rate, the expression of HSP70 mRNA and membrane HSP70. No effects of H22 cell survival rate under 42 ~ 43℃ was observed, but cell survival rate declined with increasing stress time at 44 ~ 45 ℃; the level of HSP70 mRNA decreased initially (0.5~4.0) hours but gradually resumed and increased from 8 to 12 hours at 42℃. 4.Synthesis and Quality Evaluation of Mitoxantrone

OBJECTIVE: To synthetize the synthesis of mitoxantrone and evaluate its quality. METHODS: Crude product of mitoxantrone was prepared by slow oxidation of 1,4,5,8-tetrahydro-anthraquinone with N-(2-hydroxyethyl) ethylenediamine in water bath at 50 ℃ for 2 h under argon protection and in dry air for 4 h. The crude product was crystallized by ethanol-n-hexane (4 ∶ 1,V/V) mixture solution, which was cooled overnight and then washed by ethanol-n-hexane mixture for many times. The melting range, pH value of solution, ultraviolet-visible absorption spectrum, infrared structural characteristics, drying weight loss (water loss rate) and critical relative humidity (CRH) of the purified products (4 batches) were investigated. HPLC method was used to determine the contents of mitoxantrone. RESULTS: The mitoxantrone was prepared successfully, and synthetic yield of mitoxantrone was 34.3%; the melting point ranged from 159.1-163.6 ℃. The aqueous solution was alkaline (pH 7.63-9.54); there was a maximum ultraviolet absorption peak at 235-245 nm; there was a maximum absorption peak of visible light at 590-600 nm; the infrared characteristics were consistent with those described of mitoxantrone in the 2015 edition of the Infrared Spectrum Collection of Drugs; water loss rate were -0.83%-2.36%; CRH value was 54.7%, and the average content of the product was 78.1%(n=4) by HPLC method. CONCLUSIONS: The mitoxantrone is synthesized under mild, non-toxic and harmless experiment conditions. The synthesis step is simple, the cost is low and the yield is high. The quality of products meets the quality requirements.
5.Effect of intermittent theta burst transcranial magnetic stimulation on non-fluent aphasia after stroke

ObjectiveTo explore the therapeutic effect and mechanism of intermittent theta burst transcranial magnetic stimulation (iTBS) on non-fluent aphasia after stroke. MethodsFrom August, 2021 to August, 2022, 50 patients with non-fluent aphasia after stroke in the First People's Hospital of Lianyungang were randomly divided into sham stimulation group (n = 25) and iTBS group (n = 25). Both groups accepted speach training. iTBS group accepted iTBS, and the sham stimulation group received sham iTBS, for four weeks. The serum brain-derived neurotrophic factor (BDNF) was measured, and they were assessed by China Rehabilitation Research Center Standard Aphasia Examination (CRRCAE) and Boston Diagnostic Aphasia Examination before and after treatment. ResultsTwo cases in the sham stimulation group and three cases in iTBS group dropped down. The BDNF level, and listen and understand, repeat, name, read aloud, and total score of CRRCAE improved in two groups after treatment (|t| > 5.012, P < 0.001); and they were better in iTBS group than in the sham stimulation group (|t| > 3.968, P < 0.001). The total effective rate was more in iTBS group than in the sham stimulation group (χ2 = 8.835, P < 0.05). ConclusioniTBS can improve speech function in patients with non-fluent aphasia after stroke, which may associate with the promotion of BDNF.
6.Angiotensin-(1-7) improves endothelium-dependent vasodilation in rats with monocrotaline-induced pulmonary arterial hypertension.

In this study, we used a rat model of pulmonary arterial hypertension (PAH) induced by monocrotaline (MCT) to investigate the role and mechanism of angiotensin (Ang)-(1-7) in regulating pulmonary artery diastolic function. Three weeks after subcutaneous injection of MCT or normal saline, the right ventricular systolic pressure (RVSP) and right ventricular hypertrophy index (RVHI) of rats were detected using a right heart catheter. Vascular endothelium-dependent relaxation was evaluated by acetylcholine (ACh)-induced vasodilation. The relaxation function of vascular smooth muscle was evaluated by sodium nitroprusside (SNP)-induced vasodilation. Human pulmonary artery endothelial cells (HPAECs) were incubated with Ang-(1-7) to measure nitric oxide (NO) release levels. The results showed that compared with control rats, RVSP and RVHI were significantly increased in the MCT-PAH rats, and both ACh or SNP-induced vasodilation were worsened. Incubation of pulmonary artery of MCT-PAH rats with Ang-(1-7) (1 × 10-9-1 × 10-4 mol/L) caused significant vaso-relaxation. Pre-incubation of Ang-(1-7) in the pulmonary artery of MCT-PAH rats significantly improved ACh-induced endothelium-dependent relaxation, but had no significant effect on SNP-induced endothelium-independent relaxation. In addition, Ang-(1-7) treatment significantly increased NO levels in HPAECs. The Mas receptor antagonist A-779 inhibited the effects of Ang-(1-7) on endothelium-dependent relaxation and NO release from endothelial cells. The above results demonstrate that Ang-(1-7) promotes the release of NO from endothelial cells by activating Mas receptor, thereby improving the endothelium-dependent relaxation function of PAH pulmonary arteries.

7.Influence of 17AAG on proliferation and invasion of gastric cancer cell and its mechanism.

OBJECTIVETo investigate the effect of 17-allylamino-demethoxygeldanamycin (17AAG) on the proliferative and invasive ability of gastric cancer cells and associated mechanism.
METHODSThe proliferative ability was tested by MTT method and the cell cycle was detected by flow cytometry(FCM) when 17AAG was used to treat gastric cancer cell SGC7901. Apoptosis was detected by FCM and PI-Annexin V double staining. The invasive ability was tested by transwell method. Expression of HSP90, HSP70, c-met and AKT was detected by Western blot.
RESULTSThe growth of SGC7901 cells was inhibited after the administration of 17AAG, and the inhibitation was dose- and time-dependent. The cell cycle was blocked at the G0/G1 phase. The apoptotic ratio in 17AAG group was much higher than that in blank group and DMSO group (P<0.01). The cellular invasive ability decreased significantly (P<0.01). The expression of HSP70 was elevated by 17AAG, and the expression of c-met and AKT was down-regulated, but no change of HSP90 was observed.
CONCLUSION17AAG can inhibit the proliferative and invasive ability of SGC7901 cells, and induces apoptosis through down-regulating the expression of HSP90 client proteins instead of the target HSP90 itself.

8.Warming-needle moxibustion with different lengths of moxa stick for asthenospermia with kidney deficiency and liver depression: a randomized controlled trial.

OBJECTIVE:
To explore the clinical effect of warming-needle moxibustion with different lengths of moxa stick for asthenospermia with kidney deficiency and liver depression.
METHODS:
A total of 240 patients with asthenospermia of kidney deficiency and liver depression were randomly divided into a 4-cm group (moxibustion with 4-cm moxa stick, 60 cases, 3 cases dropped off), a 3-cm group (moxibustion with 3-cm moxa stick, 60 cases, 4 cases dropped off), a 2-cm group (moxibustion with 2-cm moxa stick, 60 cases, 2 cases dropped off) and an acupuncture group (60 cases, 3 cases dropped off). All patients were treated with warming-needle moxibustion with different lengths of moxa stick or conventicnal acupuncture at Zhongwan (CV 12), Guanyuan (CV 4), Zhongji (CV 3), Guilai (ST 29), Yaoyangguan (GV 3), Guanyuanshu (BL 26), etc., once a day, five times a week; 4-week treatment was taken as one course, a total of two courses of treatment were given. The semen routine indexes, seminal plasma biochemical indexes, sex hormone levels and TCM syndrome score were compared before and after treatment among the 4 groups.
RESULTS:
After treatment, the sperm density, sperm viability, ratio of grade A sperm, ratio of grade A and B sperm, seminal plasma fructose and neutral α-glucosidase were higher than those before treatment (P<0.05, P<0.01), and the sperm deformity rates were lower than those before treatment in the 4-cm group and the 3-cm group (P<0.01, P<0.05). The ratio of grade A sperm, ratio of grade A and B sperm, seminal plasma fructose and neutral α-glucosidase in the 4-cm group were higher than the other three groups (P<0.05, P<0.01), and the sperm deformity rate was lower than the other three groups (P<0.05). After treatment, except for dizziness and tinnitus score, each-domain score and total scores of TCM syndrome scale in the 4-cm group and the 3-cm group were lower than those before treatment (P<0.01, P<0.05). The each-domain score of depression, weak waist and knees, low sexual function and total score in the 4-cm group were lower than those in the other three groups (P<0.05, P<0.01). The total effective rate was 87.7% (50/57) in the 4-cm group, which was higher than 78.6% (44/56) in the 3-cm group, 77.6% (45/58) in the 2-cm group and 70.2% (40/57) in the acupuncture group (P<0.05, P<0.01).
CONCLUSION
The warming-needle moxibustion with 4-cm moxa stick could effectively improve quality and motility of sperm and clinical symptoms in patients with asthenospermia of kidney deficiency and liver depression, which is superior to moxibustion with 3-cm, 2-cm moxa sticks and conventional acupuncture.

9.Analysis on the linkage of 415 generic medicines passing consistency evaluation and essential medicine list , medical insurance list and centralized procurement list from the perspective of medicine accessibility

OBJECTIV E To in vestigate the situation ,achievements and proble ms of consistency evaluation policy of generic medicines in China. METHODS The descriptive analysis was performed after collecting and sorting out the information of generic medicine passing consistency evaluation (GMPCE) published on the official website of the National Medical Products Administration. The basic information ,the distribution and changes of GMPCE were analyzed statistically in National Essential Medicine List (hereinafter refer to as “essential medicine list ”),Medicine List for National Basic Medical Insurance ,Industrial Injury Insurance and Maternity Insurance (hereinafter refer to as “medical insurance list ”)and the result of the successful selection of centralized medicine procurement organized by the state (hereinafter refer to as “centralized procurement list ”). RESULTS From 2017 to 2021,415 chemical generic drugs had passed consistency evaluation in China ,including 309 varieties,1 822 specifications, 6 dosage forms ,and 17 pharmacological mechanisms ,basically belonging to 30 provinces,and 492 drug manufacturers (except 12 products had not been found the manufacturers );the proportion of GMPCE in essential medicine list increased from 0.96% in 2012 edition to 25.40% in 2018 edition;that of GMPCE in medical insurance list increased f rom 2.13% in 2017 edition to 11.68% in 2021 edition;in the first 5 batches of centralized procurement list,GMPCE accounted for 81.65%,and the maximum price drop after entering the list was 97.52%. CONCLUSIONS The policy linkage has been achieved with the continual increase of the number of GMPCE and their total amount in three lists in China. The accessibility and affordability of related medicines have been improved with the apparent decrease of the price of those medicines. H owever,total number of GMPCE is a little small,with the higher repetition rate of variety and the low proportion in the three lists ;the guarantee measures of those medicine supply need to be strengthened.
10.Analysis of the Problems in the Naming of Commercially Available Chinese Patent Medicine in China and Improvement Suggestions Based on the Naming Technical Guidelines for Generic Name of Chinese Patent Medicines

OBJECTIVE: To investigate the situation and existing problems of the naming of commercially available Chinese patent medicine (CPM) in China, and to put forward the improvement suggestions. METHODS: Announced in Dec. 31, 2017 by CFDA, there were totally 169 601 kinds of national coded drugs. Total of 35 513 kinds of CPM with drug approval number “Z” and 68 kinds of imported CPM with importing registration certificates “Z” was screened by using Excel 2013 software. Based on Naming Technical Guidelines for Generic Name of Chinese Patent Medicines (hereinafter referred to as the Guidelines), the unqualified situation of domestic CPM were summarized, and unqualified domestic Chinese patent medicine were analyzed statistically in respects of dosage form, special population medication and area. RESULTS: There were 5 091 kinds of drugs named nonconformity with the “Guidelines”, accounting for 14.34% of domestic CPM. The problems of naming mainly focused on exaggerated naming (1 723 kinds, 33.84%), dosage form un-located after naming (1 118 kinds, 21.96%), naming by endangered protected animals and plants (851 kinds, 16.72%), naming by pharmacology and other related terms (848 kinds, 16.66%). Names emboding “traditional cultural features” (1 324 kinds, 4.35%) took the lower proportion. Top 5 dosage forms of domestic CPM with unqualified naming were tablets (1 203 kinds, 23.63%), capsules (821 kinds, 16.13%), mixture (802 kinds, 15.75%), pills (706 kinds, 13.87%), granules (448 kinds, 8.80%). The problems of special population medication for domestic CPM with unqualified naming concentrated on children’s medication (608 kinds, 11.94%). The main manufacturers of domestic CPM with unqualified naming came from Guangdong (613 kinds, 12.04%), Guangxi Zhuang Autonomous Region (371 kinds, 7.29%), Jilin (265 kinds, 5.21%), Shaanxi (245 kinds, 4.81%) and Beijing (233 kinds, 4.58%). CONCLUSIONS: There are many problems in naming of commercially available domestic CPM in China, mainly reflecting as naming type, dosage form, pediatric drugs, etc. There are fewer names emboding “traditional cultural features”. It is suggested that we should further standardize the naming of CPM by strengthening and perfecting the management and examination system of CPM, highlighting the naming principles of Chinese traditional culture and protecting traditional classical brands.
