Objective To investigate effects of Livin antisense oligodeoxynucleotides(ASODN) on the proliferation and apoptosis of human leukemia(HL60) cells.Methods Livin protein on HL60 cells was examined by immunohistochemistry.Specific phosphorothioate antisense oligodeoxynucleotides and missense oligodeoxynucleotides target Livin mRNA were synthesized and transfected into HL60 cells following cationic liposome.The proliferation inhibition of HL60 cells was assessed by MTT.The expression of Livin mRNA was detected by RT-PCR.Transmission electron microscope and TUNEL technology were used to detect the apoptosis and morphologic change.ResultsASODN of 600 nmol/L inhibited the HL60 cell proliferation and the expressions of Livin mRNA.The percentage of apoptosis detected by TUNEL was 38.48%?4.37%.cellar ultrastructure was markedly destroyed by Livin ASODN.A significant difference was found when compared with the control group(P

Objective This study is to explore the infratentoriat-supracerebellar keyhole approach for microsurgical treatment of pineal region tumors and to evaluate its curative efficacy and safety.Methods According to hi-fi quality MRI images,individual operation schemes were designed.Microsurgical infratentori- al-supracerebellar keyhole approach was used to resect lesions in 7 consecutive patients with prone postition.A 2.0 cm?2.5 cm surgical bone window was performed with its superior margin reaching to the inferior margin of the transverse sinus and confluence sinus.Results Among the 7 pineal region tumors,there were 2 ger- minomas,2 pineocytomas,1 pineoblastoma,1 glioma and 1 chlesteatoma.All cases were re-examined with MRI after operation and it was found that 6 lesions were totally removed and 1 was subtotally removed.The outcome of the treatment was satisfying.There was no infection,bleeding or death after surgery.The follow-up result in the near future was good.Conclusion The infratentorial-supracerebellar keyhole approach for the excision of pineal region tumors was proved to be a satisfactory means with a total removal rate,an excellent curative effect and small surgical trauma.

Objective To analyze the dermal irritation characteristics of 8 types of cosmetics.Methods The selected 917 cosmetics samples of 8 types,which were underwent the health safety test in China during 2005-2007,were assessed in the acute dermal irritation according to the related standards and the data were statistically analyzed using CMH method.Results The dermal-irritant samples were detected in different levels in 8 types of cosmetics.In terms of the irritation of cosmetics and the dermal damage caused by cosmetics,the cosmetics for hair,for face clean and for bath showed a significant higher proportion compared with the other types of cosmetics and the dermal damage could last for more than 14 days.Conclusion The acute dermal-irritability is different in 8 types of cosmetics,the cosmetics for hair,for face clean and for bath can cause the irritation and damage in the skin in degrees.

Tsutsugamushi disease is an acute infectious rickettsial disease caused by the intracellular parasite Orientia tsutsugamushi. Due to its variety of clinical signs, this disease is often misdiagnosed. This article examines a total of 4 patients who visited our clinics with fever and sore throat. 3 of them had body temperature of 39.5 Celsius degrees when admitted. The characteristic black eschar occurred on 4 of them. Lymphadenopathy occurred on 2 of them. Cough occurred on 1 of them. Lab tests showed that 3 of them had Leukocytosis, 1 of them had increased bronchovascular markings, and 3 of them had Weil-Felix test positive. After admission, all patients, who were confirmed of diagnosis of tsutsugamushi disease instead of tonsillitis, received the comprehensive treatment and cured afterwards.

Objective To study the efficacy of complement depletion with CVF in preventing a-cute rejection after heart allotransplantation in inbred strain rats. Methods Inbred male Lewis rats were used as recipients and Brown-Norway rats as donors, and the heterotopic heart transplantation model was established. The allografts were divided into 2 groups (n=8 in each group). After infusing low-dose CVF 20?g/kg to the CVF-treated group, cardiac allograft survival time was observed on 4 rats of each group. Objective To compare and evaluate the usefulness of diagnostic tests of IFA with HEp-2 cell substrate and ELISA coated with purified nuclear antigens for ANA in SLE.Methods Sera derived from 226 SLE cases and 183 healthy controls were tested for ANA and all parameters were compared such as sensi- tivity,specificity,accuracy,positive predictive value,negative predictive value,positive likelihood ratio,nega- tive likelihood ratio,result consistency,rank correlation coefficient and kappa of ANA detected by IFA and ELISA.Accuracy was evaluated by ROC for two methods.All 36 samples with different results from two meth- ods were detected for ENA.The correlation of titer to A ratio of different patterns as studied.Results The sensitivity of IFA and ELISA was 91.15% and 92.04% respectively for SLE patients,specificity was 96.17% and 92.90%,accuracy was 93.40% and 92.42%,positive predictive value was 96.71% and 94.12%,negative predictive value was 89.80% and 90.43%,no significant difference was found between the two methods (P＞0.05).No significant difference was found in accuracy of both methods by ROC (P=0.409).Good agreement was found between two methods with rank correlation coefficient (R=0.823) and kappa (k=0.825).All of 36 samples with different ANA results from two methods were detected for ENA.In 14 cases with IFA positive and ELISA negative,the titer of one case was up to 1:1000 and the pattern was Golgi by IFA,the titers of the rest were about cutoff level and the pattern were granular and nucleolus mostly.In 22 cases with IFA nega- tive and ELISA positive,11 cases of them had the A ratio ranged from 2.67～30.5.Positive rate of ENA was 14.29% in 14 cases with IFA positive and ELISA negative,68.18% in 22 eases with IFA negative and ELISA positive and the difference was significant (P＜0.01).Poor correlation of titer to A ratio for granular pattern samples (R=0.083),but good correlation for homogeneous pattern was found (R=0.595).Conclusion IFA as the recommended detecting method for ANA is intuitive and can provide more information by different pattem than ELISA but it needs fluorescence microscope and experienced technician.While ELISA is very simple and the concentration of ANA can be evaluated by A ratio value.ELISA can be a substitute method for ANA be- cause both IFA and ELISA have high sensitivity,specificity,accuracy,agreement rate,kappa and rank correla- tion coefficient.In addition,ELISA is more accessable for screen test because of low rate of false negative re- sult.Result of ELISA is more accurate if new and uncommon antigens are coated such as Golgi and nucleolus. The new work flow in which ELISA is used to screen out the positive ANA samples and IFA is used then to detect the nuclear pattern of ANA can save time,cost,and in turn improve work efficiency.

Objective: To establish a method for determination of isofraxidin in Xiaoerkaiwei Oral Liquid (Radix Acanthopanacis Senticosi, Endothelium Corneum Gigeriae Galli, Semen Alpiniae Katsumadai, Semen Pharbitidis, etc.) by RP HPLC. Methods: The HPLC system consisted of Symmetry C 18 column(3.9m?150mm, 5?m), methanol water (0.1% formic acid) (30∶70) mixture as a mobile phase, detection wavelength at 342nm, 0.8mL?min -1 of flow rate and column temperature at 30℃.Results: The calibration curves of the isofraxidin were linear ( r =0.9993). The precision were perfect ( RSD =1.87). The samples were stable in 6 h. The average recovery of Xiaoerkaiwei Oral Liquid was 102.4% with RSD of 1.76%.Conclusion: This method is simple, quick and specific, suitable for the quality control.

Objective To investigate the variety of antro duodenal motility in patients with diarrhea or constipation predominant irritable bowel syndrome (IBS) and to elucidate the pathophysiology of IBS. Methods After 6 hours fasting, 17 patients with IBS (diagnosed by Rome Ⅱ criteria), 8 with diarrhea predominant diarrhea(D IBS) and 9 with constipation predominant IBS(C IBS), were investigated by gastrointestinal manometry (STD Co., Sweden) for recording at least 2 intergrated interdigestive migrating motor complex (MMC) cycles and observing 2 hours of postprandial antro duodenal motility. Results The MMC periods was shorter, phase Ⅲ duration was longer and amplitude was higher in D IBS group than in controls, whereas the MMC periods was longer and the phase Ⅲ duration was shorter in C IBS group than in controls. The duration of discrete clustered contraction(DCC) in phase Ⅱ was longer in IBS groups than in health controls, but there was no significant difference between the C IBS group and the D IBS group. The motility index in late phase Ⅱ and phase Ⅲ was higher in IBS D group, but for the IBS C group the motility index had no statistical difference from that for controls. The postprandial motility pattern of duodenum had no remarkable difference between IBS patients and controls. Conclusions The gastrointestinal motility disorders are apparent in the IBS patients. There is close relation between gastrointestinal motility disorders and the symptoms of the IBS patients. DCCs waves may almost be independent of constipation or diarrhea of IBS.

Conclusion:By the standard analysis model, the same item of the same drug can be compared to provide the reference for data collecting of experts. Meanwhile, formulate standard can be calculated to provide the reference for the standard improvement without drug standards.
