1.HIGH-PERFORMANCE LIQUID CHROMATOGRATHY ASSAY FOR NIFEDIPINE IN BIOLOGICAL FLUIDS AND KINETICS OF NIFEDIPINE METABOLISM BY HUMAN LIVER MICROSOMES

A high-performance liquid chromatographic method was developed to determine nifedipine in biological fluids, by using ultraviolet detection at 240 nm.Phenacetin was chosen as internal standard.A Nova-Pak C18(4 um, 3.9?150mm) was employed with methanol-water (68:32) as mobile phase. Nifedipine was extracted by ethyl acetate with a percentage of recovery about 95.3?1.0%. The limit of detection of this method for nifedipine was 4 ng for an injection volume of 20 ul. One major metabolite was formed from nifedipine after incubation with human liver microsom-es, the NADPH generating system was needed in this metabolic ptocess. Km and Vmax measured by nifedipine disappearance from roicrosomal incubation mixture were 58.0 umol/L and 1.51 nmol/mg/min, respectively
2.BRAFV600E-dependent Mcl-1 stabilization leads to everolimus resistance in colon cancer cells

mTOR is commonly activated in human cancer and an attractive therapeutic target.Its activation is caused primarily by oncogenic mutations in RAS/RAF/MAPK and PI3K/AKT pathways, which cooperate to promote cancer progression and therapeutic resistance. mTOR inhibitors induce growth suppression and death receptor/FADD-dependent apoptosis in colon cancer cells and xeno-grafts. Using a panel of BRAFV600E and WT colorectal cancer cell lines and in vitro selected resistant culture, and xenograft models, we demonstrate here that BRAFV600E confers resistance to mTOR inhibitors. Everolimus treatment disrupts the S6K1-IRS-2/PI3K negative feedback loop, leading to BRAFV600E-dependent activation of ERK and Mcl-1 stabilization in colon cancer cells, which in turn blocks the crosstalk from the death receptor to mitochondria. Co-treatment with inhibitors to Mcl-1, PI3K, RAF or MEK restores mTOR inhibitor-induced apoptosis by antagonizing Mcl-1 or abrogating pERK/Mcl-1 elevation in BRAFV600E cells. Our findings provide a rationale for genotype-guided patient stratification and potential drug combinations to prevent or mitigate undesired activation of survival pathways induced by mTOR inhibitors.
3.Ontogeny of drug metabolism enzymes and epigentic regulation

Great changes in drug metabolizing enzyme (DME) expression occur in the fetus and child during development. Individual hepatic DME ontogeny can be categorized into one of three groups based on developmental trajectories.Some enzymes such as CYP3A7,are expressed at highest level in the fetus dur-ing the first trimester and either remain elevated or slightly de-crease during gestation,but are silenced or reduced to relatively low levels within one to two years after birth.SULT1 A1 is an ex-ample of the second group of DME.These enzymes are ex-pressed at relatively constant levels throughout gestation and into adulthood.CYP3A4 belongs to the third DME group .These en-zymes are expressed at negligible or low levels in the fetus.Sig-nificant increases in enzyme levels are exhibited within the first one to two years after birth.The epigenetic regulation refers to genomic modifications that do not involve changes in DNA se-quence and include DNA methylation,histone modifications, and non-coding RNAs.The epigenetic regulation mechanisms are responsible for the developmental expression of DME genes dur-ing liver maturation.This review will provide a summary of DME developmental expression profiles and reveal epigenetic mecha-nisms underlying variable drug metabolism and drug response. Thus,knowledge regarding DME ontogeny has permitted im-proved capability to predict drug disposition in pediatric pa-tients,which is crucial for improving drug dosing leading to opti-mal safety and efficacy in children.
4.Explicit physical self and its correlation with their implicit physical self in college students

Objective:To investigate the relationship between explicit and implicit physical self among contemporary college students.Methods:A total of 485 college students were recruited,with 264 male students,221 female students,266 junior students,219 senior students,189 students in the urban areas and 296 students in the rural areas.The Adolescents'Physical Self Scale (APS) was used to measure their explicit physical self.The Implicit Association Tests were designed to examine implicit physical self of 50 college students,who were selected from 485 college students by isometric random sampling.Results:The difference were significant among the five dimensions of college students' APS scores (P < 0.001).The average scores of college students' appearance,sexual attractiveness were the highest,while the average score of body flaws was the lowest.Male students scored higher in five APS dimensions than female students (Ps < 0.05).Senior students scored higher in the appearance,movement characteristics,figure and body flaws than junior students (Ps <0.05).Students in the urban areas scored higher in the appearance,movement characteristics,figure and body flaws than the students in the rural areas(Ps < 0.05).There was no significant correlation between explicit and implicit physical self (P > 0.05).The scores of relative separation index of movement characteristics were lower in male students than in female students (Z =2.45,P < 0.05).The scores of relative separation index of body flaws were lower in the urban ones than in the rural ones (Z =3.14,P < 0.01).Conclusion:It suggests that separation phenomenon exists between explicit and implicit physical self and the separation phenomena is distinct among the students of different genders or from different areas.
5.Discussing of Mental Rehabilitation of Children with Disability under ICF Theory and Practice

Objective To discuss the mental rehabilitation of disabled children under International Classification of Functioning, Disabil-ity and Health (ICF). Methods The target, the organization of work team, environment support and the core problems of disabled children's mental rehabilitation were discussed based on the concept of ICF. Results The goal of mental rehabilitation was to recover the mental func-tion of disabled children. The multidisciplinary team should include medicine, psychology, education, social work and so on. Supportive en-vironments such as professional workers, families, communities, partner and barrier-free environment were the effective means of disabled children's mental rehabilitation. Respect and equality were the precondition of disabled children's mental rehabilitation, and positive atten-tion should be the working attitude. Conclusion ICF-CY can provide a new perspective and framework for the mental rehabilitation of dis-abled children.
6.Sequence pattern mining of doctor's advice for Chinese traditional medicine in HIS

This paper analyzes some common information of doctor's advice for medicine,mines the doctor's advice for medicine by using sequence pattern mining method,and explains the mining results.
7.Strengthening Management of Hospital's Network by Means of Active Directory

Objective To solve the problems in the network management of the hospital's information construction.Methods The client workstations and the users of the hospital network were designed and organized by means of active directory.They are deployed and managed by means of setting the type of users and issuing the group policy.Results The application of active directory could manage the hospital's network uniformly,agilely and safely.Conclusion The management level is enhanced by means of active directory.It will have broad prospects in the hospital's network management.
8.Development of an superparamagnetic immunochromatographic test strip for rapid quantification of CA72-4

Objective To establish a simple and practical superparamagnetic immunochromatographic test strip for rapidly monitoring human serum level of CA72-4. Methods Water-soluble carboxylated super-paramagnetic nanoparticles were prepared with a modified one-step hydrothermal synthesis method. Magnetic probe was prepared by immobilizing specific antibody (mAb1) onto the surface of nanoparticles. Following with optimization and assembly of the test strip , we evaluated sensitivity , specificity , stability of this method for serum CA72-4 detection. Results The optimized test strip provided not only the qualitative results, but also the high sensitivity quantitative detection through stable magnetic signal. The detection limit was 0.83 IU/mL. One hundred clinical samples ( 70 positive and 30 negative ) were measured to assess these test strips with high sensitivity (99%) and high specificity (93%). The test strip and magnetic signal possessed high stability. Conclusion A rapid and quantitative detection of CA72-4 by the test strip was accomplished. This method is rapid, sensitive and quantitative, possessing great potential in large sample screening or in-home testing.
9.The system of the intellectual property of Chinese medicine studied on the current situation of the protection of Chinese medicine

The literature analysis and case analysis were used to analyze the current situation and problems of the patent protection of Chinese medicine.The patent of artemisinin was registered abroad as the failure case, while Tasly developed the construction of the patent net overseas as a successful case. The intellectual property right of traditional Chinese medicine has started late in China, so the Chinese pharmaceutical enterprises lacked awareness of the intellectual property protection of Chinese medicine, and it is difficult for us to have comprehensive intellectual propertyright protection in traditional Chinese medicine. The intellectual property system of Chinese medicine in China needs improvement.
10.Screening of homoacetogen mixed culture converting H2/CO2 to acetate.

Homoacetogens are a group of microorganisms with application potential to produce chemicals and biofuels by the bioconversion of synthesis gas. In this study, we collected waste activated sludge samples to screen homoacetogens by Hungate anaerobic technique, and studied the effect of pH on acetate and alcohol production from H2/CO2 gas. The mixed culture contained Clostridium ljungdahlii, Lysinibacillus fusiformis and Bacillus cereus. Acetate concentration achieved 31.69 mmol/L when the initial pH was 7. The mixed culture containing homoacetogen could converting H2/CO2 to acetate, which provides an efficient microbial resource for the bioconversion of synthesis gas.
