1.Antiproliferative effect of curcumin on human breast cancer of MCF-7 cells
Journal of Third Military Medical University 2003;0(18):-
Objective To investigate the effect of curcumin on the proliferation, cell cycle distribution and gene expression of human breast cancer MCF-7 cells. Methods MCF-7 cells were treated with 10,20,40,80 ?mol/L curcumin for 24,48 or 72 h.MTT was used to assay the biologic activities of curcumin and its antiproliferative effect.FCM was used to detect cell cycle distribution and apoptosis. The expression levels of bcl-2,bax,PCNA mRNA were analyzed by RT-PCR.Results The curcumin IC_(50)value for MCF-7 cells was(18.5 ?mol/L.) The inhibitory effect of curcumin on cell growth was in a time-and dose-dependent manner.MCF-7 cells treated with 20 ?mol/L or(40 ?mol/L) curcumin for 24 h were arrested in G_0/G_1 phase and induced apoptosis. The expression levels of bcl-2,PCNA mRNA were decreased in MCF-7 cells by 10,20 and 40 ?mol/L curcumin treatment for 24 h,and bax mRNA was increased. Conclusion Curcumin is of inhibitory effect on MCF-7 cell growth, and it can arrest MCF-7 cells in G_0/G_1 phase, inhibit the proliferation and induce apoptosis.Curcumin down-regulates bcl-2,PCNA mRNA expression in MCF-7 cells, but upregulates bax mRNA expression.

Objective To determine the role of angiogenesis in the prognosis of breast carcinoma and its relationship with p53 expression and ER statue in 59 cases of breast cacinoma. Methods Factor Ⅷ related antigen (FⅧ RA) and p53 were evaluated by immunohistochemical staining, and the microvessel endothelial area (MEA) of tumor microvessels was detected by computer aided image analysis system. Results (1) The expression of MEA significantly increased with tumor size and histologic grade(P

Each woman of childbearing age has the right to enjoy reproduction, including cancer patients. Breast cancer is the most common malignant tumor in women. Many factors led to the significant increase of young patients with fertility problems, and the majority of doctors neglected their reproductive requirements in clinical work. Given the comprehensive treatment, almost every young patient suffers from fertility decline and even loss. Patients who chose to get pregnant also harbor a variety of concerns. This paper reviews the effects of comprehensive treatment on fertility, preservation strategies, pregnancy and prognosis, optimal time for pregnancy, and other aspects and provides guidance for young breast cancer patients in their reproduction choices.

Objective To study the effects of artesunate(ART)on estrogen receptor negative breast cancer cell line MDA-MB-231 and its mechanisms. Methods Treated with ART for 3 days,MDA-MB-231 cells proliferation was examined by MTT assay. The morphological and uhrastructural changes of MDA-MB-231 cells were observed under microscope and electronic microscope.Immunocytochemistry was used to detect the expression of Bax,mn23,Bcl-2,and P21 WAFl/CIPl.Results ART treatment led to a dose dependent inhibition of MDA-MB-231 cells.ART could change the morphology and ultrastructure of MDA-MB-231 cens. After treatment with ART(2μmol/L)for 72 hours, immunocytochemical staining showed that the expression of Bax,nm23,and P21WAF1/CIP1 was upregulated in comparison to the control group(P<0.05)while the expression of Bcl-2 in MDA-MB-231 cells didn't have a significant change(P>0.05).Conclusions ART shows an anti-proliferative effect on MDA-MB-231 cells. The mechanisms may be related to upregnlation of Bax,nm23 and P21WAF1/CIP1 expression.

Postoperative adjuvant therapy for primary invasive breast cancer patients with synchronous ax-illary metastases is mainly based on the characteristics of biomarker of the primary tumor. Recently, some studies have showed the discordance and clinical significance of estrogen receptor (ER), progesterone receptor (PR) and human epidermal growth factor receptor 2 (HER2) status between primary breast cancer and synchronous axillary lymph node metastases. As local metastasis, the synchronous axillary metastases may represent the potentially metastatic breast cancer cells much better than the primary tumor. Hence, determination of biomarkers status should be performed in synchronous axillary metastasis, together with primary tumor, to guide therapy management and evaluate the prognosis of primary invasive breast cancer patients with synchronous axillary metastases.

Objective To summarize the clinical characters, diagnosis and surgical treatment of primary hyperparathyoidism (PHPT).Methods The diagnosis, treatment and efficacy of 35 cases of PHPT were retrospectively reviewed. Results PHPT presented a variety of clinical manifestations, and occoured to different age groups without significant gender differences. All the 35 cases had elevated blood calcium and PTH.All patients underwent preoperative ultrasonography and 99Tcm-MIB imaging, and the positive rates were 68.6% and 97.1% respectively.34 patients received surgical treatment, among whom 30 cases had parathyroid adenoma,1 case had parathyroid hyperplasia and 3 cases had parathyroid carcinoma.Through operation,31 cases were cured,2 cases improved, and 1 case of parathyroid carcinoma suffered from lung metastasls. Conclusions PHPT can be diagnosed according to co-elevated serum calcium and PTH.Ultrasonography combined with 99Tcm-MIB imaging should be recommend for preoperative localization. The main cause of PHPT is solitary parathyroid adenoma.PTPT can be cured by surgery.Minimally invasive parathyroidectomy with accurate location is an efficient surgical strategy, and the prognosis is favorable.

Breast cancer is one of the most common malignant tumors in females.Recent years, surgery, chemotherapy as well as other systemic therapy had greatly improved the prognosis of the patients. However, damage of ovarian function by chemotherapy lowered life quality, especially for young females. At present, there are several methods to protect the ovarian function of female patients undergoing chemotherapy, such as administration of a gonadotropin-releasing hormone (GnRH) analogs, ovarian cryopreservation, unfertilized ova cryopreservation, embryo cryopreservation, inhibitors of apoptosis, etc. Each method has its advantage, disadvantage and indications.Issues related to ovarian protection are reviewed here.

Chinese Journal of Endocrine Surgery 2017;11(2):89-91,96
Breast cancer and dyslipidemia are common diseases in women. It is reported that breast cancer and dyslipidemia have some relevance.Breast cancer patients have a relatively high incidence of dyslipidemia, which is easily undiagnosed for lacking of obvious clinical manifestations and will affect the treatment and prognosis of breast cancer. In breast cancer patients, screening and diagnosis of dyslipidemia through blood lipid detection should be strengthened.
