1.Biodegradable materials combined with osteogenic factor in orthopaedics

BACKGROUND:Biodegradable implants cannot only rebuild bone defect site, moreover, with the gradual degradation of the materials, new bone tissue can completely replace the graft materials to fil bone defects. OBJECTIVE:To summarize the research progress of biodegradable materials combined with osteogenic factor in orthopedics. METHODS: We took the “biodegradable materials, factors, cel active factor, bone tissue engineering” as the search terms in Chinese and English, respectively, to retrieve the related literatures from PubMed, Wanfang and CNKI database during 2000 to 2015 by computer. RESULTS AND CONCLUSION: Biodegradable medical polymer materials can be divided into natural polymer materials and synthetic biodegradable materials. Natural polymeric materials have good biocompatibility, but poor mechanical strength. The mechanical strength of synthetic biodegradable materials is higher than that of natural polymer materials, but the synthetic biodegradable materials are likely to cause local accumulation of acidic substances, produce local inflammation. The biodegradable medical polymer materials combined with osteogenic factor can improve the mechanical strength and osteoinductive ability of materials, but as a bone repair material, it stil has many problems to be solved.

4.Value of anti-cyclic peptide containing citrulline antibody for the early diagnosis of rheumatoid arthritis

Objective To investigate the value of detection of anti-cyclic peptide containing citrulline (anti-CCP) antibody, rheumatoid factor (RF) and the combined detection of anti-CCP antibody and RF in rheumatoid arthritis (RA) patients. Methods From 2012 to 2014,1 961 patients were divided into three groups:diagnosed RA group(509 patients), not RA group(1 028 patients) and firstly was not diagnosed RA but later was diagnosed RA group (424 patients). The levels of RF and anti-CCP antibody were separately measured by rate nephelometry method and the electrochemical luminescence method. Results The sensitivity of detecting by anti-CCP antibody alone or anti-CCP antibody in combination with RF between RA group and not RA group was not significant difference ( P>0.05), but the specificity between two groups (88.6%vs. 60.4%) was significant difference ( P<0.05). There were significant differences in the sensitivity (81.7% vs. 74.3%) and specificity (88.6% vs. 66.0%) between by using anti-CCP antibody alone and RF alone.In firstly was not diagnosed RA but later was diagnosed RA group, there were significantly difference in sensitivity (98.3%vs. 82.1%) and specificity (91.6%vs. 81.5%) by using anti-CCP antibody alone and RF alone. Conclusion There is important clinical value by using anti-CCP antibody alone for the early diagnosis of RA.
5.ADR Cases Report of Our Hospital in 2004~2006:An Analysis of 412 Cases

OBJECTIVE:To explore the factors,general rule and characteristics of adverse drug reactions(ADR)for references of safe and rational clinical medication.METHOD:A total of 412 ADR reports were collected and statistically analyzed in respect of age and sex of patients,route of administration,ADR degree,drug category,ADR-involving organs and clinical manifestations etc.RESULTS:ADR-involving drugs totaled 412 kinds,of which anti-infective agents occupied the first place,followed by Chinese patent medicines.274 ADR cases(66.50%)were induced by intravenous route.ADR occurred systemically and were characterized predominantly by allergic reactions of skin and its appendants,followed by damage of nervous system.70 were new but common ADR cases,while 2 were new severe cases.CONCLUSION:It is necessary to strengthen ADR monitoring and publicizing of ADR-related knowledge to guide the clinical rational use of drugs and reduce the repetitive occurrence of ADR.
6.Effects of Silibinin Capsules Combined with Routine Therapy on Serum Oxidative Injury and Liver Function of Patients with Alcoholic Liver Disease Complicated with Early Liver Fibrosis

OBJECTIVE: To investigate the effects of Silibinin capsules combined with routine therapy on serum oxidative injury and liver function of patients with alcoholic liver disease (ALD) complicated with early liver fibrosis.METHODS: The patients with ALD complicated with early liver fibrosis selected from our hospital during Apr. 2013-Jan. 2015 were divided into control group and study group according to random number table, with 38 cases in each group. Control group received routine symptomatic treatment [temperance, oral administration of Inosine tablets (0. 4 g/times, bid), vitamin and microelement supplementation, etc. ]. Study group was additionally given Silibinin capsule (70 mg/time, bid) orally, for consecutive 48 weeks, on the basis of control group. The clinical efficacies and the incidence of ADR were compared between 2 groups. The changes of serum oxidative injury indexes (SOD, MDA), liver fibrosis indexes [laminin (LN), hyaluronic acid (HA), type IV collagen (IV-C), type Ⅲ procollagen (PC Ⅲ)] and liver function indexes (ALT, AST) were recorded before and after treatment. RESULTS: Before treatment, there was no statistical significance in baseline information between 2 groups (P>0. 05). After treatment, total response rate of study was significantly higher than that of control group (P<0. 05), but there was no statistical significance in the incidence of ADR between 2 groups (P>0. 05). Compared with before treatment, serum levels of MDA, liver fibrosis indexes and liver function indexes in 2 groups were decreased significantly (P<0. 05), while SOD levels were increased significantly (P<0. 05); the improvement of study group was more significant than control group (P<0. 05). CONCLUSIONS: Silibinin capsules combined with routine treatment can enhance clinical efficacy of ALD patients with early liver fibrosis, mainly manifesting as improving oxidative stress, regulating liver function and inhibiting the development of liver fibrosis.
7.Curative effect analysis of different surgical methods in the treatment of adrenal tumors in children

Objective:To investigate the clinical efficacy of laparoscopic adrenalectomy and traditional open adrenalectomy for adrenal tumors in children.Methods:In Department of Pediatric Surgery, Fujian Provincial Maternity and Children′s Hospital from June 2008 to June 2016, the clinical data of 31 pediatric adrenal tumors was retrospectively analyzed.According to different surgical methods, they were divided into traditional open adrenalectomy group and laparoscopic adrenalectomy group.Bleeding loss, operation duration, incision length, abdominal drainage time, postoperative hospital stay, postoperative complications and recurrence were compared between the 2 groups.Results:There was no significant difference in age, sex, weight, and tumor size between the 2 groups (all P>0.05). The intraoperative bleeding loss was (18.06±4.86) mL for open adrenalectomy group and (15.20±4.48) mL for laparoscopic adrenalectomy group, and there existed no significant difference in the amount of bleeding between the 2 groups( T=-1.702, P>0.05). The operation duration was (137.44±19.32) min for open adrenalectomy group versus (134.80±6.21) min for laparoscopic adrenalectomy group, and there was no significant difference in operation duration between the 2 groups ( T=-0.504, P>0.05). The length of incision was (7.94±1.34) cm for open adrenalectomy group versus (3.44±0.21) cm for laparoscopic adrenalectomy group, the length of incision in laparoscopic group was shorter than that in open adrenalectomy group.There was significant difference in the length of incision between the 2 groups ( T=-12.843, P<0.001). The hospitalization time was (9.63±2.55) d for open adrenalectomy group versus (7.20±1.37) d for laparoscopic adrenalectomy group, the hospitalization time in laparoscopic group was shorter than those in open adrenalectomy group.The difference of operation time between the 2 groups was statistically significant ( T=-3.261, P=0.003). The average indwelling time of abdominal drainage tube was(5.94±1.53) d for open adrenalectomy group versus (4.80±1.74) d for laparoscopic adrenalectomy group, with no significant difference in postoperative abdominal drainage time between the 2 groups( T=-1.938, P>0.05). There was 1 case of retroperitoneal hematoma in laparoscopic adrenalectomy group and 2 cases of complications in open adrenalectomy group.There was no significant difference in the incidence of complications between the 2 groups ( P=1.000). There were 3 cases of distant metastasis and 1 case of recurrence in laparoscopic group, and 2 cases of distant metastasis and 1 case of recurrence in open adrenalectomy group.There was no significant difference in recurrence between the 2 groups ( P=1.000). Conclusions:Compared with open adrenalectomy surgery, laparoscopic adrenalectomy in children has various advantages, including beautiful incision, less trauma and fast recovery.However, it is necessary to select the appropriate cases.
8.Application of Ion Torrent PGMTM System in Detection of Fetal DNA in Maternal Plasma

Objective To explore the feasibility of detecting of Y-STR of fetal DNA in m aternal plasm a using Ion Torrent PGMTM System . Methods A total of 16 fetal DNA sam ples from m aternal plasm as (8 cases from 38 w eeks gestational age and 8 ones from 12 w eeks) w ere prepared and a m ultiplex assay w ith 7 STR loci (DYS390,DYS391,DYS393,DYS438,DYS437,DYS456,DYS635) w as designed for m ul-tiplex-PC R am plification. U sing Ion Torrent PGMTM System , the results of Y-STR sequences and capillary electrophoresis w ere obtained and com pared. Results Y-STR specific alleles w ere detected in the m ater-nal plasm a of all the pregnant w om en having m ale babies of second and third trim ester, w hich w ere higher than that detected by capillary electrophoresis. C onsistent Y-STR genotypes w ere observed betw een fetal DNA from m aternal plasm a and genom ic DNA from the new born babies. Conclusion B ased on Ion Torrent PGMTM System , the prenatal Y-STR detection m ethod m ay provide a high-sensitive and high-throughput choice for prenatal STR detection in forensic testing.
9.The status of glucose intolerance in breast cancer patients without DM history after systemic treatment with surgery and/or chemo-therapy

Objective:To determine the status of glucose intolerance in breast cancer patients without DM history after combined treatment with surgery and/or chemotherapy through an oral glucose tolerance test (OGTT). Methods:All 121 breast cancer patients more than 3 months after combined treatments with surgery and/or chemotherapy and without the diagnosis of diabetes underwent OGTT and fasting. Then, 2 h glucose levels were measured to identify glucose tolerance and diabetes. Meanwhile, six patients with a history of diagnosed diabetes did not undergo OGTT. Results:The median ages of all breast cancer patients and the mean duration after combined treatments with surgery and/or chemotherapy were 50.4 years and 19 months, respectively. Among the 121 breast cancer pa-tients without the history of diabetes, the incidences of diabetes, prediabetes, and normal glucose intolerance were 19.8%(24 cases), 45.5%(55 cases) and 34.7%(42 cases), respectively. Among all breast cancer patients, the incidences of previously diagnosed diabetes, undiagnosed diabetes, and prediabetes were 4.72%, 18.9%, and 43.3%, respectively. The ratio of previously undiagnosed diabetes was about 80%. About 80.0% of undiagnosed diabetes and 74.5% of prediabetes met the criteria for elevated 2 h plasma glucose levels through OGTT instead of elevated fasting glucose levels. Conclusion: Breast cancer patients during follow-up after combined treat-ments with surgery and/or chemotherapy highly suffer from glucose intolerance, with high incidences of undiagnosed diabetes and pre-diabetes. OGTT should be made for breast cancer patients after combined treatments for early diagnosis, prevention, and treatment of di-abetes.

Objective: To observe the targeting therapy of the hdsFv-RC-RNase recombinant single chain immunotoxin on the xenograft of the human hepatocellular carcinoma in nude mice and to explore its clinical potentiality. Methods: The prokaryotic expression vector TIG-hdsFv-RC-RNase was transformed into E.coli BL21(DE3)plys to largely express recombinant single chain immunotoxin hdsFv-RC-RNase against hepatocellular carcinoma induced by IPTG. The expressed product was purified via Ni-NTA affinity chromatography under native conditions and mildly refolded. The ELISA was used to analyze its immunological activity of antigen-binding capability. The xenogrft model of the human hepatocellular carcinoma in nude mice was established and the targeting therapy of hdsFv-RC-RNase was evaluated. Results:After induced by IPTG, a new protein band with M_r 41 000 was found in the supernatant of the bacteria and expressed in a soluble form. The expressed product was purified to homogeneity via Ni-NTA affinity chromatography under native conditions.
