1.Treatment experience of traumatic splentic rupture in 32 cases

Objective To discuss the remedy and clinical effect of traumatic splentic rupture.Methods Clinical data of 32 cases of traumatic splentic rupture were retrospective analyzed.Results No dead cases,6 cases were cured by conservative treatment,18 cases were explored by laparoscope,and 14 cases remained spleen,4 cases were converted to traditionary splenectomy.Traditionary splenectomy was done on 8 cases directly.Conclusions Ⅰ,Ⅱ grade of traumatic splentic rupture may choose conservation treatment or remain spleen by laparoscope operation.Splenectomy should be done on Ⅲ,Ⅳ grade of traumatic splentic rupture in time.
2.The effect of hepatitis B virus on the expression of kinesin family member 4

Objective To investigate the differences of kinesin family member 4 (KIF4A) expression between hepatic carcinoma and adjacent tissue in patients with chronic hepatitis B virus (HBV) infection,and to understand the effect of HBV on the expression of KIF4A.Methods Reverse transcriptase-polymerase chain reaction and Western blot were used to measure the expression of KIF4A in hepatic carcinoma and adjacent tissues. HepG2 cells were co-transfected with KIF4A promoter containing the luciferase gene and HBV infectious clone pHBV1.3,and luciferase activity was measured.Expression of KIF4A in HepG2 cells was measured after tranfected with different doses of pHBV1.3.The student's t-test was used for statistic analysis.Results Expression of KIF4A was much higher in hepatic carcinoma than that in adjacent tissues.HBV enhanced KIF4 A gene promoter activity and the luciferase activities were increased as the concentration of pHBV1.3 increased ( 0,0.2,0.4,0.6 and0.8 μg/mL),which were (126.8± 13.4),(219.8±16.7),(387.6±21.5),(586.5 ± 228.9 ) and (657.6 ± 35.5 ) RUL/μg protein,respectively,while the luciferase activities were (123.6± 13.8),(131.8± 14.6),(129.7-13.5),(135.3± 13.4) and (127.1± 12.7) RUL/μg protein,respectively with different doses of control plasmids transfected,and statistical analysis showed significant difference between them (t=4.875,P=0.006).And HBV upregulated KIF4A mRNA and protein expressions in HepG2 cells in a concentration-dependent manner.Conclusion Expression of KIF4A is enhanced in hepatic carcinoma and HBV can upregulate KIF4A expression.
3.Operation indications for polypoid lesions of gallbladder

Objective To investigate the operative indications for polypoid lesions of gallbladder(PLG) and avoid cholecystectomy for PLG without operation signs.Methods Retrospective analysis of 828 cases of PLG confirmed by pathologic examination was made.Results (1)Cancer should be suspected when a patient is older than 50 years or has a polypoid lesion larger than 1.0 cm.(2)The cholecystectomies for PLG accounted for 2.7%-7.1%of all cholecystectomies in the corresponding period,and cholesterol polyps accounted for 86.11%of all PLG,and carcinoma of gallbladder accounted for 1.92%of all PLG.Conclusion At present most of PLG accepting cholecystectomy are cholesterol polyps,so the high-risk factors of the neoplastic polyps and the operative indications for PLG should be considered deliberately.
4.Application of cardiac pacemaker in laparoscopic cholecystectomy—an analysis of 215 cases

ObjectiveTo evaluate the influence of electrotome on permanent and temporary cardiac pacemaker in laparoscopic cholecystectomy ( LC),and the application of cardiac pacemaker to the cases of cholecystolithiasis combined with bradyarrhythmia.MethodsClinical data of 215 patients with permanent or temporary cardiac pacemaker who underwent were studied for the preoperative and postoperative variation of pacemaker function,and for the influence of electricity coagulation during the operation on cardiac pacemaker function.ResultsLC was successfully completed in all 215 patients.The function of cardiac pacemaker was not obviously interfered during the operation,and the parameters of cardiac pacemaker did not remarkably change after the operation.ConclusionCardiac pacemaker is slightly interfered when electrotome and electrocoagulation were used in LC; LC is feasible and safe for patients with bradyarrhythmia by placement of cardiac pacemaker.
5.Screening and characterization of lipase from a metagenome library of dairy rumen microflora.

Using lipase segregation agar containing trioleoylglycerol, we obtained 18 lipase positive clones by screening from a metagenome library of dairy rumen microflora containing 15,360 clones. The average insert size of lipase positive clones was about 60 kb. Lipase enzyme activity analysis by p-NPP method indicated that Lipase6, Lipase7 and Lipase8 had higher lipolytic activities to substrates of p-nitrophenyl palmitate (C16), p-nitrophenyl alaurate (C12) and p-nitrophenyl palmitate (C16) respectively. The optimum pH of Lipase 6, Lipase 7 and Lipase 8 were 7.5. The halflife period of Lipase 8 with the value of 15 min in 70 degrees C decreased with the increase of temperature. In conclusion, the lipases screened in this study had different substrates specificity and good thermo stability, which laid a basis for large-scale industrial application.

6.Inhibitory effect of lncRNA 5033413D16Rik on the activity of T helper 17 cells in experimental autoimmune uveitis

Objective:To investigate the effect of long noncoding RNA (lncRNA) 5033413D16Rik (lncRNA 5033413D16Rik) on the activity of interphotoreceptor retinoid-binding protein 1-20 (IRBP) 1-20-specific T helper 17 (Th17) cells in experimental autoimmune uveitis (EAU). Methods:Twelve SPF grade C57BL/6 female mice aged 8 to 10 weeks were selected and divided into EAU group and normal control group using the random number table method, with 6 mice in each group.Mice in the normal control group received no treatment.Mice in the EAU group were immunized with IRBP 1-20 emulsified in complete Freund's adjuvant to induce EAU.On day 13 after immunization, fundus examination and inflammation scoring were performed, and retinal histopathological changes were observed with hematoxylin and eosin staining.T cells were isolated from the spleen and lymph nodes of EAU mice, and the relative expression of lncRNA 5033413D16Rik was detected by real-time fluorescence quantitative PCR (qRT-PCR).In addition, the isolated T cells were divided into short hairpin RNA (shRNA)-5033413D16Rik transfected group and shRNA-negative control (NC) transfected group, and transfected with corresponding shRNAs, respectively.The knockdown efficiency of shRNA-5033413D16Rik was verified by qRT-PCR.T cells in the two groups were co-cultured with bone marrow-derived dendritic cells (BMDCs) under Th17 polarizing conditions.The relative expression of retinoic acid-related orphan receptor (RORγt), interleukin 17 (IL-17), IL-23 receptor (IL-23R), and granulocyte-macrophage colony-stimulating factor (GM-CSF) mRNA in co-cultured cells were analyzed by qRT-PCR.The IL-17 concentration in co-culture supernatants was assayed by ELISA and percentages of Th17 cells were determined by flow cytometry.The use and care of experimental animals complied with the Regulations for the Administration of Affairs Concerning Experimental Animals promulgated by the State Science and Technology Commission.The study protocol was reviewed and approved by the Experimental Animal Ethics Committee of Tianjin Medical University (No.TJYY2019110117). Results:EAU model was established successfully.qRT-PCR analysis showed that the expression of lncRNA 5033413D16Rik was significantly decreased in T cells from EAU mice compared with normal control mice ( t=-13.332, P<0.001).Compared with the shRNA-NC transfected group, the relative expression of lncRNA 5033413D16Rik in T cells of the shRNA-5033413D16Rik transfected group was significantly reduced ( t=-6.338, P<0.01).In co-cultured T cells, the expression levels of RORγt, IL-17, IL-23R, and GM-CSF mRNA in shRNA-5033413D16Rik transfected group were 1.61±0.13, 1.51±0.13, 1.85±0.33 and 1.45±0.11, which were significantly higher than 1.00±0.01, 1.00±0.01, 1.00±0.01 and 1.00±0.01 in shRNA-NC-transfected group ( t=-6.839, -8.221, -4.538, -4.189; all at P<0.05).ELISA analysis revealed that IL-17 concentraion in shRNA-5033413D16Rik transfected group was (2 350.39±367.02)pg/ml, which was significantly higher than (1 513.31±310.37)pg/ml ( t=-3.016, P=0.039).Flow cytometry showed that the percentage of Th17 cells in shRNA-5033413D16Rik transfected group was (17.20±0.44)%, which was higher than (14.10±0.84)% in normal control group ( t=-3.264, P=0.031). Conclusions:LncRNA 5033413D16Rik can inhibit the expression of pathogenicity related genes in antigen-specific Th17 cells and negatively regulates IRBP 1-20-specific Th17 cell responses.
