Objective To investigate the relationship between polymorphisms in the thymidylate synthase(TS) promoter enhancer region and invasion of bladder transitional cell carcinoma(BTCC).Methods The TS gene promoter and TS protein expression were detected by PCR and immunohistochemical Streptavidin-Peroxidase staining method respectively in 87 cases of BTCC and the immunoreactivity score(IRS) was estimated.Results The IRS of TS in BTCC was(3.38?2.15).The IRS of TS protein in stage of T_(2-4) was significantly higher than that in stage of T_(a-1),and the IRS was elevated with the increase of pathological grade of BTCC(all P

Objective To investigate the relationship between polymorphisms in the thymidylate synthase promoter enhancer region(TSER) and clinical pathological characteristics of primary colon cancer.Methods TSER polymorphisms and thymidylate synthase(TS) protein were detected by PCR and immunohistochemical staining respectively in primary foci.Results There were three genotypes of TSER including homozygous with double tandem repeats(2R/2R),homozygous with triple tandem(repeats)(3R/3R) and heterozygous with both alleles(2R/3R)in 68 cases of colon cancer(6,40,22 cases respectively).The ratio of 3R/3R was significantly higher in stage of Dukes C than stages in Dukes B and Dukes A.(Dukes A16%;Dukes B30%;Dukes C 93%,?~2=28.56,P
3.Serum proteomic analysis of acute pancreatitis in rats

Objective To investigate the difference of the protein expression in serum of rats with acute edematous pancreatitis (AEP) and acute necrotizing pancreatitis (ANP),and to investigate serum marker for acute pancreatitis severity.Methods The model of AEP and ANP was induced by retrograde injection of 4% or 5% sodium taurocholate into the biliopancreatic duct.Weak cation exchange (WCX2) and surface enhanced laser desorption/ionization time of flight mass spectrometry ( SELDI TOF MS) was used to investigate the difference of the protein expression in serum of rats with AEP and ANP.Results Thirty-eight spectral peak clusters which had significantly different signal intensities between AEP and ANP sera at mass charge ratio between 1000-50 000(P <0.05) were detected.The peak clusters at 9500 and 9700 in the sera of AEP were higher than that in ANP rata.Conclusions Serum analysis with SELDI-TOF MS can detect the difference of the protein expression in rats with AEP and ANP.The decreased expression of the protein of molecular weight of 9500 and 9700 may be a signal of AEP transition into ANP.
4.Serum proteomic analysis of patient with severe acute pancreatitis before and after treatment

Objective To evaluate the different expression of the serum proteome of patient with severe acute pancreatitis (SAP) before and after treatment,and search for the protein that can predict SAP improvement.Methods SELDI-TOF-MS was used to analyze the expression of serum protein spectrum of 20 SAP patients before treatment and 7 days after treatment,and make the APACHE Ⅱ score at the same time.Results The APACHE Ⅱ score was 10.70 ± 3.47 in patients with SAP 7 days after treatment,which were significantly lower than that before treatment ( 13.00 ± 2.21,P <0.05 ).One hundred thirty-two spectral peak clusters were detected before and after treatment,there were 27 proteins that had significantly different signal intensities had been detected from 2000 Da to 50000 Da (P < 0.05 ).The peak cluster at 47 000 Dawas significantly down-regulated after SAP attenuation.While the peak clusters at 1l700 Da and 15900 Dawere significantly up-regulated after treatment.Conclusions Serum analysis with SELDI-TOF MS can detect the different expression of the SAP serum proteomics before and after treatment,the increased expression of the protein with molecular weight of 11500 Da and 15900 Da and the decreased expression of 4700 Da may be asignal of improvement of SAP.
