Journal of Chinese Physician 2001;0(04):-
Objective To explore the role of ? chemotactic factor- MCP-1 in the pathogenesis of congestive heart failure by detecting its expression changes. Methods PBMCs were collected from 25 patients with congestive heart failure and 10 healthy controls. RT-PCR was used to detect the mRNA expression of MCP-1 in PBMCs. Correlation of the clinical indexes of heart failure patients with to their MCP-1 mRNA expression levels were analysed. Results The expression level of MCP-1 in the heart failure patients was higher significantly than that of controls (P

Objective To investigate the inhibitory effect of bone marrow mesenchymal stem cells ( BMMSC) on the inflammatory response of microglial cells .Methods The samples were divided into four groups .Group I:microglial (BV-2) cells were cultured in DMEM (high glucose).Group II: BV-2 cells were cultured in DMEM containing lipopo-lysaccharide (LPS).Group III:BV-2cells and BMMSCs were co-cultured in DMEM.Group IV: BMMSCs were cultured in DMEM containing LPS .The growth state and ultrastructure of BV-2 cells were observed and the changes of TNF-αex-pression were detected .Results Different cell densities of BV-2 cells were observed under the optical microscope in an or-der from high level to low level:group I >group III >group II.The expressions of TNF-αwere:group Ⅱ >groupⅢ>group Ⅰ.Ultrastructural observation of BV-2 cells showed that there were a large number of swollen mitochondria and endoplasmic reticulum , some of them showed vacuolization .No BV-2 cells with multiple hucleoli were found in the group II indicating the absence of active cell growth .At the same time, cytolysis was observed only in the group II .The growth of BV-2 cells in the group III was better than that in the group II .Conclusions BMMSCs can inhibit inflammatory response of microglial cells, therefore, play a neuroprotective role.

Objective To investigate the influence of gene transfection antiangiogenesis on microvessel and relative cytokine of hypertrophic scar of rabbits' ear. Methods The hypertrophic scar of rabbtis' ear was reproduced. On the 10th day after epithelization,Ad-METH-1 was injected into tissue of scar.30 days later, the microvessel of scar-tissue was detected by microcirculation microscope.Meanwhile.H&E and immunohistochemical stains were performed. Then the results were analyzed. Results 30 days after Ad-METH-1 injection.in experimental groups, the microvascular count of scar tissue was 12.38±2.56,the percentage of VEGF positive cells was 17.64%,and the percentage of bFGF positive cell was 18.24%:while in the control groups, the microvascular count of scar tissue was 48.12±6.46.the percentage of VEGF positive cell was 31.34%.and the percentage of bFGF positive eell was 28.26%.Results revealed that the count of microvessel of scar tissue in the experimental groups was lower than that in the control groups, between which there was the difference in statistics(P<0.01).and that the percentage of VEGF and bFGF positive cells of scar tissue in the expenmental groups were lower than that in the control groups.between which there was the difference in statistics(P<0.05).Conclusion Ad-METH-1 has marked inhibitory effects on scar tissue hyperplasia of rabbits' ear, angiogenesis and expression of VEGF and bFGF.Using antiangiogenesis therapy at the early phase could inhibit the formation of hypertrophic scar.Gene transfection antiangiogenesis therapy could bid fair to become an effective method to prevent and treat hypertrophic scar.

Objective To investigate the status and present problems of standardization of outpatient and emergency diagnosis information of a hospital, and discuss the room of improvement. Methods The outpatient and emergency diagnosis information of a tertiary hospital was retrieved, and the data were inspected using tracking method over the whole process from diagnostic information input to data maintenance, for analysis of causes of the nonstandard diagnosis. Results The standard rate of the outpatient and emergency diagnosis of the hospital was 51. 07% (15278/29915). Statistics by the departments visited found the traditional Chinese medicine department with the most nonstandard diagnosis;that by diagnosis names found the most nonstandard diagnosis in such common diseases as hypertension and diabetes. These nonstandard diagnosis may be contributed mainly to the system, the standard dictionary, the doctor and the imperfect maintenance mechanism. Conclusions It is suggested to improve the system maintenance mechanism, streamline the maintenance process, and train the doctors in dictionary use and input, to improve the outpatient and emergency diagnostic information standardization.
5.A Review of Epigenetic Modifications Regulate MicroRNA Expression in Lung Cancer.
Chinese Journal of Lung Cancer 2020;23(7):582-588
Lung cancer is one of the primary causes of cancer-induced death among the world. Although the network of molecular implicated in lung cancer is gradually revealed, the exact molecular mechanism of its occurrence and development has not been fully elucidated. As a class of small and endogenous single-stranded non-coding RNAs, microRNAs (miRNAs) are found in a wide range of organisms from plants, viruses to humans. miRNAs involve various functions in normal lung tissue development. They take part in a large amount of biological processes including cell growth, metabolism, proliferation and differentiation. However, aberrant expression of miRNAs could induce the occurrence, development, invasion and metastasis of lung tumor, so it is deemed the novel biomarkers. Similar to that of protein-coding genes, expression and function of miRNA are regulated by various factors and the epigenetic network which includes DNA methylation and histone modification. Moreover, key enzymes driving epigenetic modifications are regulated by miRNAs. Therefore, better understanding of inextricable linkage between miRNAs and epigenome will provides a basis for the feasibility of miRNA-orientated diagnostic, therapeutic and prognostic strategies related to lung cancer in future.
