1.Effects of soil factors on yield and quality of Angelica dahurica var. formosana

Objective To study the effects of the soil factors on the yield and quality of Angelica dahurica originated from Sichuan Province.Methods The yield and contents of imperatorin,isoimperatorin,and other 11 different inorganic elements of A. dahurica,gathered from 15 different habitats of Suining in Sichuan Province were determined.And the basic soil nutrients and other 11 different mineral elements of the soil were analyzed.The effects of the soil factors on the yield and quality were analyzed through correlation analysis.Results The weight,length,circumference,and the contents of imperatorin and isoimperator in the roots of A.dahurica from different habitats were different.And the basic soil nutrients and other 11 different mineral elements of soil were different.The positive correlation between root length of A.dahurica,and soil available P content,root circumference of A.dahurica,and soil organic matter content,root weight of A.dahurica,and soil available P and K contents were all significant.The soil total nitrogen content had negatively significant correlation with the content of isoimperatorin.The correlation among the imperatorin,isoimperator contents of A.dahurica,and 11 other soil inorganic elements was not significant.Conclusion The soil factors have effects on the yield and quality of A.dahurica to some degree.
2.Anti-metastasis effect of curcumin on human pancreatic cancer

Objective To investigate whether curcumin could inhibit the metastasis of pancreatic cancer in vitro and in vivo and its mechanism.Methods Pancreatic cancer AsPC-1 cells were treated with different concentrations of curcumin.After 72 hours,cell survival rate were detected by MTT and the appropriate concentration of curcumin was selected.After treated with 6 μmol/L curcumin,early apoptosis of AsPC-1 cells were detected by Annexin V-FITC/PI double staining flow cytometry,and the effects of curcumin on the migration and invasion of AsPC-1 cells were observed.After establishment of the orthotopic nude mouse model of pancreatic cancer,the mice were orally treated with low dose (20 mg/kg body weight) and high dose (40 mg/kg body weight) of curcumin respectively.Eight weeks later mice were sacrificed to observe the tumor metastasis,immunohistochemical methods were used to detect the positive expressions of CD34,NF-κB and MMP-9 in the tumor tissue.Results Curcumin of different concentrations could inhibit the proliferation of AsPC-1 cells and induce its apoptosis in a dose dependent manner,and 6.μmol/L was the best dose of curcumin.After 6 μmol/L curcumin treatment,the migration coverage of AsPC-1 decreased from 70% to 10%,the number of penetrating cells decreased from 136/200x magnification to 17/200x magnification,and the difference between the two groups was statistically significant (P <0.05).In the nude mouse model of pancreatic cancer,the size of the tumor decreased from (97.8 ± 15.3) mm3 to (44.3 ± 9.7) mm3 in high dose group and (28.1 ±7.1)mm3 in low dose group,the number of distant metastasis decreased from 108.3 ±6.7 to 29.5 ±4.5 and 8.9 ± 2.4,the expression of CD34 decreased from 20.5 ± 2.3 to 10.3 ± 1.2 and 7.9 ± 3.2,and the expression of MMP-9 decreased from 85.2 ± 2.3 to 53.2 ± 1.2 and 34.2 ± 3.1,and the difference was statistically significant (P < 0.05).Conclusions Curcumin can inhibit the metastasis of pancreatic cancer both in vitro and in vivo,and its effect may be related to the inhibition of expressions of CD34 and MMP-9.
3.An experimental vaccine against tetrodotoxin with longer term of validity

Objective:To explore the possibility of immune prevention and cure for tetrodotoxin (TTX) intoxication and develop an antitoxin vaccine against TTX.Methods:TTX was conjugated to Tachypleus tridentatus hemocyanin(TTH) in presence of formaldehyde and this conjugate(TTX-TTH) was used as an immunogen to immunize Balb/C mice.The quality of antisera in animal was detected on schedule by enzyme linked immunosorbent assay(ELISA) and competition-inhibited enzyme immunoassay(CIEIA).Mice immunized with TTX-TTH were challenged intraperitoneally with lethal doses of TTX(1LD=13.5 ?g/kg).Results:The high titer and affinity of antisera could last for a time as long as more than one year.The immunized mice were ip challenged with 1?LD of TTX once and again at a fixed period,there was a affirmative antitoxic effect in about 12 months(total 15?LD),and a partial effect in following time;about one fourth of animal survived till 24 months post initial immunization(total 26?LD),and which was at a stage of senescence in mice.The anti-TTX poisoning effect of animal was approximately consistent with the antisera quality tested.Conclusion:The experimental vaccine of TTX could effectively protect animal from TTX intoxication and its effect was of longer duration of validity.Immunoprophylaxis would be the hopeful means for detoxification of TTX. [
4.Parathyroid Hormone Promotes the Proliferation and Invasion of Chondrosarcoma Cells by Regulating the Expression of Primary Cilia

Objective To investigate the effect of parathyroid hormone(PTH)on the proliferation and invasion of chondrosarcoma cells,and the relationship between PTH and the regulation of primary cilia expression.Methods After stimulation of the chondrosarcoma cell line SW1353 with different concentrations of PTH,induction of the expression of cilia with hypoxia and destruction of cilia structure with chloral hydrate,the cell viability was detected by CCK8 assay,the proliferation and invasion of SW1353 by Western blotting and Transwell method,the primary cilia expression by immunofluorescence assay and the GLI1,PTCH1 and IFT88 expression levels by real-time PCR.Results PTH could promote the proliferation of chondrosarcoma cells in a concentration-dependent manner and this effect was correlated with the structural integrity of the primary cilia.PTH could up-regulate the invasive ability of SW1353 cells and increase the expression levels of MMP9,which was suppressed when the primary cilia structure was damaged.Additionally,it was found that PTH could down-regulate the number of primary cilia,which was related to the structural integrity of the primary cilia.It could also regulate the expression levels of GLI1 and PTCH1,the target genes in Hedgehog pathway,and the expression levels of IFT88,the gene associated with the cilia function.Conclusion PTH can promote the proliferation and invasion of chondrosarcoma cells,down-regulate the expression of primary cilia and the downstream target genes.PTH may regulate the malignant biological features of chondrosarcoma by regulating the primary cilia expression.
5.Effect of Tanshinone IIA in Preventing and Treating Oxaliplatin Induced Peripheral Neuropathy.

OBJECTIVETo observe preventive and therapeutic effects of Tanshinone IIA (T II A) on oxaliplatin induced peripheral neuropathy (OlPN) and to explore its effects on the expression of calcitonin gene related peptide (CGRP) and never growth factor (NGF).
METHODSTotally 36 phase II - III patients with malignant tumor of digestive tract undergoing chemotherapy program with oxaliplatin, were equally assigned to the T II A group (using THA at 80 mg/day 1 day before oxaliplatin chemotherapy for 3 successive days) and the control group (using chemotherapy program with oxaliplatin alone) by segmented randomization. After 4 cycles of chemotherapy, the incidence degree and incidence of OlPN were evaluated. Sensory nerve conduction velocity (SNCV) and motor nerve conduction velocity ( MNCV) were tested by EMG evoked potential device. Serum levels of CGRP and NGF were also detected in the two groups before and after chemotherapy. The correlation of serum levels of CGRP and NGF to OIPN was assessed using linear correlation analysis.
RESULTSAfter chemotherapy the OlPN incidence was 27.8% (5/18 cases) in the T II A group, obviously lower than that in the control group (55.6%, 10/18 cases; P < 0.05). Compared with before treatment in the same group, SNCV and MNCV of common peroneal nerve were slowed down, serum NGF levels decreased, and serum CGRP levels obviously increased in the two groups (all P < 0.05). Compared with the control group after treatment, SNCV and MNCV of common peroneal nerve were obviously accelerated, serum NGF levels increased, and serum CGRP levels obviously decreased in the THA group (all P < 0.05). Results of linear correlation analysis indicated serum NGF level was negatively correlated with peripheral neuropathy (PN), serum CGRP expression was positively correlated with neurotoxicity (P < 0.05).
CONCLUSIONT II A could reduce the incidence of OlPN, which might be associated with inhibiting the expression of CGRP and up-regulating NGF activities.

6.The effect of different doses of nutrition support on outcomes in trauma patients

Objective:The objective of this study was to evaluate the relationship between the adequacy of nutritional therapy and prognosis in trauma patients.Methods:217 trauma patients were included.The nutritional therapy interventions were divided into three groups:low energy therapy (<50%),moderate energy therapy (50% ~ 80%),high energy therapy (> 80%).General data,hospitalstay,ICU stay,infectious complications and mortality were compared among groups.Results:The protein intake of high energy group was higher than the other two groups (P <0.01).High energy therapy was associated with reduced mortality and decreased length of hospital stay.Kaplan-Meier survival analysis indicated that high energy therapy had a significantly higher 60 days cumulative survival rate compared with other groups.Conclusion:High energy therapy is more effective and associated with improved mortality and decreased length of stay.
7.Investigation on the effect of SaIB on bone marrow-derived mesenchymal stem cells apoptosis induced by hypoxia and serum deprivation

ObjectiveTo investigate the effect of SalB on bone marrow-derived mesenchymal stem cells (BMSCs) apoptosis induced by hypoxia and serum deprivation (hypoxia/SD) in the vitro. Methods BMSCs were cultured in the vitro and randomly divided into control group, hypoxia/SD group and SalB group.SalB group was composed by four groups and were pretreated by complete medium with 0.1、 1、 10、 100 mg/L SalB for 1 hour. And after that they were washed with phosphate buffer for 2 times, added by IMDM with 0.1、1、 10、 100 mg/L SalB and cultured with hypoxia/SD group together in the same condition of hypoxia/SD for 6hours. The control group was cultured for 6 hours in the condition of aerobic and enough serum. Apoptosis was detected by Hoechst33342 staining with inverted phase contrast, fluorescence microscope and Annexin V/PI dual-color flow cytometry. Results Significant apoptosis of BMSCs was induced by hypoxia/SD in the vitro.The early apoptosis of BMSCs induced by hypoxia/SD was significantly decreased by SalB of 0.1、 1、 10 mg/L(P<0.05) . Conclusion0.1、 1、 10 mg/L SalB can decrease the early apoptosis of BMSCs induced by hypoxia/SD.

·Diffusely infiltrating retinoblastoma is a rare histopatho-logic type of retinoblastoma, accounting for approximate-ly 1% to 2% of retinoblastoma cases. We report a case of diffuse infiltrating retinoblastoma presenting as endo-phthalmitis and comment on the histopathologic findings.
9.Establishment of a SNP genetic identification method for frozen embryos and sperm of inbred mice

Objective To establish a rapid SNP( single-nucleotide polymorphism) genetic identification method for the frozen samples, such as frozen embryos and sperm of inbred mice.Methods In this study, the frozen embryos and sperm of inbred mice were provided by Shanghai Lab.Animal Research Center.Whole genome amplification and PCR-LDR genotyping system were used to get the rich DNA sample.Forty-five SNP were genotyped by multiple polymerase chain re-action and ligase detection reaction( PCR-LDR) .Results The electrophoresis results showed that the whole genome am-plification technique could highly increase the total DNA of frozen embryos.PCR-LDR typing method was suitable for the mouse genome typing of 45 SNPs.Ten strains of inbred frozen embryos and sperms of C57BL/6, BALB/c, FVB/NJ mice were genotyping identified, and their SNP loci data obtained by PCR-LDR were as the same as those of database.The num-ber of frozen mouse embryos was proportional to the number of SNPs detected, and when the embryo number reached more than 12, the detection rate of SNP was 100%.Conclusions This method can be used to the genetic quality identification, and rapidly identify the inbreed frozen mouse embryos and sperms.
10.Analysis on influential factors in China's exports of primary and semi-finished products of traditional Chinese medicine to ASEAN.

Two regression models, based on panel data over the period of 2000-2011, are built and used to analyze what factors determine China's exports of primary and semi-finished products of traditional Chinese medicine to ASEAN. The results indicate that, China GDP, the ratio of ASEAN to China GDP per capita, average export price, the ratio of state-owned assets to total assets, have a significant positive influence on the export volumes of primary products of Chinese medicine. At the same time, RMB appreciation, the ratio of three kinds of foreign-invested assets to total assets, China-ASEAN Early Harvest Program, ASEAN-China Free Trade Area have a significant negative influence. In respect of the export volumes of semi-finished products of Chinese medicine, the significant influential factors are ASEAN GDP and the ratio of ASEAN to China GDP per capita. The former is positive and the latter is negative. In order to optimize the commodity composition of experts, it is needed to increase export volumes of both primary and semi-finished products of Chinese medicine. According to the analysis above, some proposals are put forward, such as, improving the performance of foreign capital, playing an exemplary and leading role in technological innovation by state-owned enterprises, taking advantage of bargaining power of suppliers, increasing outward foreign direct investment.
