1.Clinical observation on hormone replacement in treatment of perimenopausal syndrome

ObjectiveTo study the changes of related hormone levels using hormone replacement therapy in patients with perimenopausal syndrome,and its clinical significance.Methods Eighty-five patients with perimenopausal syndrome received continuous sequential estrogen and progesterone treatment.Daily oral conjugated estrogen 0.625 mg,28 days as one cycle.In each cycle the 19-28th d combined with medroxyprogesterone acetate 4 mg,once a day,a total of 3 cycles.Observed the changes of hormone levels before and after treatment.ResultsThe levels of estradiol and progesterone were obviously increased after treatment compared with those before treatment [ (83.6 ± 1.9) ng/L vs.(10.0 ± 1.1 ) rng/L,(0.32 ± 0.10)μ g/L vs.(0.22 ± 0.20) μ g/L,P < 0.05 ],and follicles stimulating hormone,luteinizing hormone,total cholesterol and triglyceride were obviously decreased [ (40.8 ± 11.0) U/L vs. (57.1 ± 10.8) U/L, (24.9 ±13.6) U/L vs.(46.2 ± 13.8) U/L,(4.6 ±0.9) mmol/L vs.(6.1 ± 1.0) mmol/L,(1.1 ±0.2) mmol/L vs.( 1.6 ± 0.1 ) mmol/L,P < 0.05 ].Body mass index was no obviously different[ (23.9 ± 1.9) kg/m2 vs.(22.5 ± 2.2)kg/m2,P > 0.05 ].ConclusionApplication of hormone replacement in treatment of perimenopausal syndrome is safe.
2.Social function cognition for bedside care service at patient's home in the residents aged over 55 years in Jingshan Community of Bering

Objective An investigation on social function cognition for bedside care service at patient's home was conducted in the residents aged over SS years in Jingshan Community,Dongcheng District,Beijing,to lay a scientific basis for provision of it.Methods Ninety-seven residents aged over 55 years were selected from 58 families with systematic sampling in Jingshan Community by a questionnaire interview,including history of illness,income,medical expense,awareness of bedside service at patient's home,etc.Results Totally,98.9 % of the residents suffered from at least one kind of chronic disease,98.9 % of them never received bedside care service at their homes,94.9% of them only knew a little of such kind of health care service,91.85% of them knew some social function of it,and 84.5 % of them did not know the criteria of payment for the service.Conclusions Publicity of bedside care service at patient' s home should be strengthened,focusing on its social function,inexpensiveness,convenience,etc.Function of bedside service should be perfected further,with quality care,to make it an important part of community health care.
3.Protective effect of paeonol on human umbilical vein endothelial cells injured by hyperlipidemic serum

OBJECTIVETo investigate protective effect of paeonol (Pae) on human umbilical vein endothelial cells (HUVECs)injured by hyperlipidemic serum and explore its possible mechanism.METHODS The injury model was induced by 20% hyperlipidemic serum incubating HUVECs for 24 h.Pae 124,247 and 495.μmol· L-1 were given followed by administration of hyperlipidemic serum for 24 h.The morphological changes were observed under inverted microscope,cell survival rate was evaluated by MTT method,the nitric oxide (NO) content was measured by nitric acid reductase method and the endothelial nitric oxide synthase (eNOS) mRNA expression was determined by RT-PCR.RESULTSCompared with normal contorl group,most cells in model group split and exfoliated.However,the morphology was tending to normal level after intervention with Pae.Pae significantly improved cell viability(P <0.01).Compared with model group,the survival rate increased from (53.0 ±10.1 ) % to ( 68.4 ± 9.1 ) %,( 84.5 ± 6.7 ) % and (98.1 ± 7.5 ) %,respectively.The NO content and eNOS mRNA expression both increased greatly in Pae groups(P < 0.01 ).Compared with model group,content of NO increased from 54 ± 4 to 79 ± 6,115 ± 5 and ( 136 ± 6) μmol · L- 1,respectively.The expression level of eNOSmRNA improved from 0.215 ± 0.060 to 0.451 ± 0.045,0.563 ± 0.013,0.704 ± 0.068,respectively.CONCLUSIONPae exerts protective effect on HUVECs injured by hyperlipidemic serum by increasing eNOS mRNA expression,which might therefore improve the content of NO.
4.Clinical significance of level of CD4+CD25HiCD127Low regulatory T cells in the peripheral blood of patients with esophageal cancers

Objective To detect the levels of CD4+CD25HiCD127Low regulatory T cells (Treg) in the peripheral blood and its clinical significance in patients with esophageal cancer. Methods The levels of Treg in the peripheral blood were detected by three-color flow cytometry (FCM) in 80 patients with esophageal cancer and 20 healthy controls. Among the 80 patients, 30 patients were also further studied for preoperative and postoperative comparison after operation. The clinical and pathological data of each patient were collected and analyzed for the correlation with the Treg levels. Results The level of Treg in the peripheral blood of the control group was lower than that of the esophageal cancer patients [(3.36±1.14) % and (5.70±1.96) %, respectively], with significant difference (P <0.01). The levels of Treg in the peripheral blood was higher in the patients with metastasis of lymph node (n=40) than that in the patients without metastasis of lymph node (5.96±1.36) % and (4.23±1.18) %, respectively] (n=30), with significant difference (P <0.01). The levels of Treg in the peripheral blood of the patients were negatively correlated with their TNM classification. As the TNM classification advanced, the level of the Treg in the peripheral blood increased. Conclusion The levels of Treg in the peripheral blood of the patients with esophageal cancer are significantly higher than that of the healthy subjects, which is correlated with the clinical and pathological conditions. The development of esophageal cancer may relate with suppression of immune function.
6.Effect of amygdalin on end-plate chondrocytes apoptosis induced by IL-1βderived from rat intervertebral discs

Aim To observe the anti-apoptotic effect of different concentrations of amygdalin on the endplate chondrocytes induced by IL-1 βderived from rat inter-vertebral discs and explore the possible mechanism fur-ther.Methods Chondrocytes were obtained from endplate of one-month SD rat intervertebral discs and cultured primary chondrocytes.After identifying,they were divided into normal group,induced group and A-mygdalin 1 0 -2 mol·L -1 ,1 0 -3 mol·L -1 ,1 0 -4 mol· L -1 ,1 0 -5 mol · L -1 administration group.Then the apoptosis was detected by flow cytometry (FCM).Re-al-Time PCR was adopted to detect the mRNA expres-sion of Bax and Bcl-2.The protein expression of Bax and Bcl-2 was detected by Western blot.Results The apoptosis of the endplate chondrocytes induced by IL-1 βderived from rat intervertebral discs could be inhib-ited by amygdalin with different concentrations.Amyg-dalin could reduce the apoptotic rate analysed by FCM,down-regulate the Bax mRNA expression of Bax and up-regulate the Bcl-2 mRNA assayed by RT-PCR;compared with the induced group the differences were statistically significant (P <0.05).Besides,observa-tion of the protein expression of Bax and Bcl-2 by Western blot found that amygdalin 1 0 -4 mol · L -1 could inhibit the effect of IL-1 β,which could increase the protein expression of Bax and reduce the protein expression of Bcl-2.Conclusion Amygdalin has an effect on anti-apoptosis of the end-plate chondrocytes induced by IL-1 βderived from rat intervertebral discs and improve the degeneration of intervertebral discs.
7.Reversal of Multidrug Resistance by IFN-? in KBv200 Cell Lines

Objective: To explore the effects of various concentrations IFN-? on human oral epidermic cancer cell line KB and its multidrug resistant counterpart KBv200.Methods: The toxicity of IFN-? and/or VCR was assayed by MTT method; intracellular rhodamin123 concentration and p-glycoprotein expression were measured by flow cytometry; mdr-1 mRNA was assayed by RT-PCR. Results: High-dose IFN-?(10 000 IU/ml) had significant antiproliferative effects on KB/KBv200 cell lines. Low-dose IFN-? did not have any significant effect on growth of KB/KBv200 cells, but did increase the cytotoxic effect of VCR and the accumulation of Rhodamine123 in KBv200 cell line and had no effect on parent VCR-sensitive KB cell line. IFN-? down-regulated expression of P-gp and mdr-1mRNA . Conclusions: IFN-? has significant effect on the reversal of multidrug resistance of KBv200 cell line via a mechanism of P-gp and mdr1 mRNA down-regulation.

9.Adipose derived mesenchymal stem cell transplantation for Alzheimer's disease

Alzheimer's disease is an irreversible neurodegenerative disease characterized by progressive neuronal loss. To date, there has been no effective medicine or therapy for neurodegenerative disease. With development of stem cell technique and theory, neural stem cell transplantation has been found to be prospective in Alzheimer's disease treatment. However, it was challenged by the deficiency of autologous neural stem cell, which can bypass immunological barrier. Compared with neural stem cells, mesenchymal stem cells exhibit extensive resources, such as liver, bone marrow and adipose, and multiple differentiations into bone, muscle or adipose. Considering the easy access, the minor trauma to the patients, and the neuron differentiation potential of adipose derived mesenchymal stem cells (A-MSC), we hypothesize that A-MSC graft is a potential and innovative strategy for the treatment of Alzheimer's disease.
10.A New Design of Impactor for Rat and Monkey's Spine Cord Injury Models

Spinal cord injury is very high morbidity trauma, seriously affecting the survival and health of patients. To establish a standard animal model has a great significance for studying SCI pathogenesis and pathological changes. A spinal cord injury device for experimental animal was presented in this paper. It can implement fixed-point and fixed-quantity impact by new design. It also can create many animal models of different levels injury. More importantly, it has many advantages such as simple to operate, free from human disturbance, high accuracy and reproducibility. Finally, some existing constraints and challenges about the animal spinal cord injury models made by the device were discussed.
