1.The plasma levels change of TNF-α,IL-6,IL-10,PLA2 and organ lesion analysis of sever burn injury patients with sepsis

Objective To study plasma levels of tumor necrosis factor-α(TNF-α) ,IL-6 ,IL-10 ,serum phospholipase A2(PLA2) and organ lesion of sever burn injury patients .Methods 57 sever burn injury patients got treated in our department from March 2010 to March 2011 ,in which 33 of them had been diagnosed with sepsis and 19 of which developed into multiple organ dysfunction syndrome(MODS) ,the levels of IL-6 ,IL-10 ,TNF-α,PLA2 were compared among sepsis group(group N) ,MODS group(group M) and negative group(group N) .The organ disfunction and fatality rate were also analyzed .Results The PLA2 ,TNF-α,IL-6 levels of patients in group S and M were significantly higher than group N ,and showed no apparent decline during the first 14 day ;the change trend of IL-10 had no statistically significant difference among the 3 groups(P<0 .05) .The group S and M showed larger burned area and deeper burned depth ,the organ failure and mortality rate were significantly higher than group N ,And with the most common respiratory function failure .Conclusion The PLA2 ,TNF-α,IL-6 levels in sever burn injury patients might be closely relat-ed to the degree of burn .High plasma level of PLA2 ,TNF-α and IL-6 might be closely associated with occurrence of sepsis and MODS and their outcome .

3.Clinical Observation of Tympanic Administration through Eustachian Tube in Treating Middle Ear Adhesion

30 dB and little was 23、14、8、18 ears, respectively. The effective rate was 71.4%. No adverse reaction was seen. Conclusion the technique was non - traumatic, economical, practical and effective.
4.Application of Responsibility Index in Medical Students′Professionalism Evaluation

For the cultivation of medical students medical professionalism and evaluation has become a hot is-sue in the medical colleges medical education at home and abroad, but for junior medical students′professional spirit evaluation research is less.This paper introduces the basic concept of responsibility index and application by introducing foreign medical schools responsibility index for evaluation in school doctor students physician profession-alism, analysis the advantages and disadvantages of the method.Think the responsibility index is based on the spe-cific behavior evaluation, the result is more accurate, is suitable for junior medical students′professional spirit e-valuation system, compared with other evaluation system, responsibility index, pay more attention to guidance and specification in advance.But responsibility index also has some disadvantages.Such as responsibility index evalua-tion process must face some ethical challenges, lack of integrity and evaluation framework design.

7.The effects of 5-fluorouracil on hypertrophic scar of rabbit ears

Objective To investigate the effect of 5‐fluorouracil on hypertrophic scar in rabbit ears .Methods The hyper‐trophic scar models in rabbit ears were reproduced and randomly divided into 3 groups (group A ,group B and group C) .On postop‐erative 60th days ,Hematoxylin‐eosin and Masson staining staining was applied to observe themorphological and collagen change of scars under light microscope .Real‐time PCR and ELISA were conducted to detect the expression of TGF‐β,PDGF ,bFGF and INF‐γ .Results HE and Masson staining showed that thinner dermis ,fewer fibroblasts and collagen fibers ,collagen fibers arranged reg‐ularly and less collagen nodule was observed group B compared with the group A and group C .Furthermore ,we found that TGF‐β, PDGF ,bFGF ,INF‐γmRNA and protein expression levels were significantly lower in group B and group C than the group A ,and group B was more effective than group C .Conclusion 5‐fluorouracil is effective for the treatment of hypertrophic scars ,and the lo‐calized injection of 5‐fluorouracil is more effective than the ointment group .
8.Effects on gene expressions of G protein alpha subunits by activation of non-neuronal muscarinic receptor

Aim To study the subtypes distribution of G-proteins and modulation by carbachol and pilocarpine on gene expression in endothelial cells.Methods The rat aorta endothelial cells were cultivated and incubated with carbachol and pilocarpine at the dose of 10~(-4) mol?L~(-1)for 6 h.Then the total RNA was extracted.The mRNA levels of G-protein ? subunits was measured by RT-PCR.Results Gq/11,Gs and Gi mRNA was detected in rat aorta endothelial cells,while G12/13 mRNA was not detected.Carbachol and pilocarpine treatment induced no changes in Gs,Gi and G11 mRNA.Gq mRNA was 72.7% up-regulated by carbachol and unchanged by pilocarpine.Conclusion In all G-protein ? subunits,only Gq mRNA was changed after activation of non-neuronal muscarinic receptor by carbachol.We can conclude that Gq-protein may play an important role in signal transduction of nonneuronal muscarinic receptor.

How the hosts recognize and clear invading viruses is one of the key issues in molecular immunology. Previous studies uncovered that many early antiviral proteins, such as Type Ⅰ interferons and PKR, are strongly induced upon virus infection. These proteins not only limit virus replication and spread or cause infected cells to undergo apoptosis, but also induce consequently expression of cytokines and chemokines to initiate acquired immunity. However, the immediate-early signaling events among host and virus interaction were largely unknown. In the past few years, there are great breakthroughs in this rapidly evolving field. TLR3 and RIG-I/MDA5 signaling pathways were shown to play a crucial regulatory role in antiviral processes. These pathways are essential for the vertebrate immune system to recognize and clear RNA virus with different strategies, which are integral parts of innate immune response and directly affect later-stage acquired immunity. The recent know-how on TLR3 and RIG-I/MDA5 signal transduction pathways and their roles in antiviral immunity were summarized.
10.Expression and significance of E-cadherin in the serum and transplanted tumor of nude mouse model of low rectal cancer with lymph node metastasis

Objective To establish a nude mouse model of low rectal cancer with lymph node metastasis, to detect the expression of E-calcium (E-cad) in the serum and transplanted tumor tissue of the mice, and to analyze the role of E-cad in lymph node metastasis of rectal cancer.Methods Human colorectal adenocarcinoma cells (SW-480) were inoculated into the rectal submucosa of nude mice to establish a model of low rectal cancer with lymph node metastasis.120 nude mice were randomly divided into three groups (40 mice in each group).Group A (rectal cancer model group) was injected with dimethylhydrazine and implanted with SW480 cells.Group B (dimethylhydrazine control group) was injected with dimethylhydrazine alone.The group C (control group) received no treatment.Serum E-cad was detected by ELISA.The mRNA and protein expression of E-cad in the tumor tissue was detected by RT-PCR and western blot, respectively.Results The serum level of E-cad in group A was 19.48±1.25 mg/L, significantly higher than those in groups B (2.36±0.18 mg/L) and C (2.15±0.12 mg/L) (t =8.28, 9.01, P < 0.05).The serum levels of E-cad were significantly higher in the nude mice with lateral lymph node metastasis than those without lymph node metastasis (t =10.28, P < 0.05).The protein expression of E-cad in the group A was lower than that in the groups B and C (t =9.81, 7.69, P < 0.05).In the group A, the protein expression of E-cad in the nude mice with lateral lymph node metastasis was significantly lower than that without lateral lymph node metastasis (t =9.36, P < 0.05).The mRNA expression of E-cad in the group A was significantly lower than that in the groups B and C (P < 0.05), and the mRNA expression of E-cad in the nude mice with lateral lymph node metastasis was significantly lower than that in the mice without lateral lymph node metastasis (t =7.85, P < 0.05).Conclusions The serum level of E-cad may be closely associated with the lymphatic metastasis of rectal cancer, and the mRNA and protein expressions of E-cad in colorectal cancer tissue were weak or absent, leading to a decrease of adhesion ability between cancer cells, and promote the invasion and metastasis of cancer cells.
