1.Expression of multiple resistance-associated protein and P-glycoprotein in oral squamous cell carcinoma for prediction of response to neoadjuvant chemotherapy

Objective:To investigate the expression of Multiple resistance-associated protein-1(MRP1) and P-glycoprotein(P-pg) in oral squamous cell carcinoma(OSCC) before and after neoadjuvant chemotherapy(NACT) and its relation with tumor chemosensitivity and related clinical pathological factors.Methods:The specimens of 106 patients with OSCC before and after NACT were examined by SP immunohistochemistry.The correlation between P-pg and MRP1 expression and therapeutic effect was analyzed.Results:(1)P-pg positive expression rate in OSCC after NACT was 74.53%,which was significantly higher than thatbefore NACT(41.51%)(P﹤0.05);the effective rate of patient with P-pg negative expression before chemotherapy was 56.45%,which was significantly higher than that of patient with P-pg positive expression(34.09%)(P﹤0.05);P-pg expression levels were positively relative to the differentiation of the tumor(P﹤0.05),but not relative to clinical stages and cervical lymph node metastasis(P﹥0.05);(2)MRP1 positive expression rates in OSCC before and after NACT were 49.06%、and 57.55% respectively and the difference was not significant(P﹥0.05);the effective rates of patient with MRP1 negative expression in OSCC before chemotherapy were 50.00% and 44.23% respectively and the difference was not significant(P﹥0.05);MRP1 expression levels were not relative to clinical stages,differentiation of the tumor or cervical lymph node metastasis(P﹥0.05);Conclusion:P-pg and MRP1 have high expression in primary oral squamous cell carcinoma,but only P-pg expression levels are significantly associated with response to chemotherapy and it can be used as a predictive marker of the sensitivity to chemotherapy in patients with OSCC.
2.The detection of the nucleation shifting process of smad4 signal protein in rat dental papillae cells with quantum dots-smad4 monoclonal antibody fluorescent probes

Objective:To prepare semiconductor quantum dots (QDs)-Smad4 monoclonal antibody Fluorescent probes ,and to detect nucleation shifting process of Smad4 signal protein in Rat dental papillae cells(RDPCs) stimulated by TGF-?1 with the Fluorescent probes. Methods:①QDs were chemically modified with Smad4 proteins to prepare water soluble QDs-Smad4 monoclonal antibody Fluorescent probes which were purified after the preparation. ②The properties of these probes were studied through absorption band analyses,fluorescence spectra,fluorescence spectrum analyses and confocal laser scanning fluorescence microscopy. ③The location of Smad4 proteins in RDPCs was studied with SP anti-Smad4 immunocytochemical method,QDs direct mark method,Smad4 monoclonal antibody direct mark method,and direct immunofluorescence imaging.And the properties of Quantum Dots-Smad4 monoclonal antibody Fluorescent probes in RDPCs were detected. ④After RDPCs ware cultivated by TGF-?1 for 0 hours,12 hours,and 14 hours,the dynamic nucleation shifting process of Smad4 signal protein in RDPCs was separately detected with SP anti-Smad4 immunocytochemical method and direct immunofluorescence imaging.Results:QDs and monoclonal antibody ware covalently bonded to form the fluorescent probes which could specifically and effectively recognize Smad4 proteins in RDPCs .It is to demonstrate the dynamic nucleation shifting process of Smad4 signal protein in RDPCs. These Fluorescent probes still had good properties,including broad excite spectra,narrow emission spectra,high fluorescence intensity,and photostability. Conclusion:QDs and monoclonal antibody could be covalently bonded to form the fluorescent probes with distinct optics character,special immunity recognition capability and the ability of imaging mark the protein in cells for long time,which provides the scientific evidence that QDs can visually research the movement and interaction of protein in living cell .

How the hosts recognize and clear invading viruses is one of the key issues in molecular immunology. Previous studies uncovered that many early antiviral proteins, such as Type Ⅰ interferons and PKR, are strongly induced upon virus infection. These proteins not only limit virus replication and spread or cause infected cells to undergo apoptosis, but also induce consequently expression of cytokines and chemokines to initiate acquired immunity. However, the immediate-early signaling events among host and virus interaction were largely unknown. In the past few years, there are great breakthroughs in this rapidly evolving field. TLR3 and RIG-I/MDA5 signaling pathways were shown to play a crucial regulatory role in antiviral processes. These pathways are essential for the vertebrate immune system to recognize and clear RNA virus with different strategies, which are integral parts of innate immune response and directly affect later-stage acquired immunity. The recent know-how on TLR3 and RIG-I/MDA5 signal transduction pathways and their roles in antiviral immunity were summarized.

7.Visual imaging of quantum dots for in vivo malignancy cancer

Quantum dots (QD),a kind of nanocrystal,are made from Ⅱ-Ⅵ or Ⅲ-Ⅴ group elements.It has been reported that compared with current conventional fluorescent markers,QD have excellent optical properties such as strong fluorescence,photochemical stability and can be used for simultaneous multi-channel imaging.Meanwhile,as nanoparticle,QD can be easily surface modified with a variety of biological molecular and can reach cancer cells easily by penetrating the tumor angiogenesis.Therefore,QD have unique advantages in targeted real-time visual imaging of in vivo tumor and have great prospects in the individual diagnosis and treatment of tumor.However,the long-term biosafety after QD into the body still needs further study.

Our knowledge of molecular events and interactions in cell biology and molecular biology are mostly based on findings in nonliving body condition while ignoring the whole process of the events,which inevitably involves false assumption that all molecules behave in the same environment and in the same way. With the emergence of new fluorescent molecular probes prepared through linking new fluorescent material such as quantum dots (QDs) to micromolecule antibody such as antibody simulacrum, the development of methods for transducing extrinsic fluorescent probes into live cells,the application of laser technology, the finer fluorescent micro-imaging system, and the fast and sensitive fluorescence collection system in the application of life sciences, it is now possible to image the trajectory, kinetic behavior, and interaction of biomolecules in live cells. Visualizing single molecule in live cells has opened a new chapter for cell biology and molecular biology. Recent research developments and foreground of single molecule visualizing are discussed in this article.

This review is to provide an overview of current situation and advance of abdominal compartment syndrome. Progress has been made in diagnosis and therapy of abdominal compartment syndrome. At present patients who are diagnosed as abdominal compartment syndrome are associated with a high mortality rate. Therefove, it is important to diagnoze and treat the disease early. Surgical treatment of increased intraabdominal pressure leads in most instances to a rapid and profound correction of the physiological abnormalities. Operative treatment is the unique and effective approach of abdominal compartment syndrome.
