1.MLN64 gene and its research advancement in the field of carcinogenesis and progression of breast cancer

The MLN64 gene,which is localized in q12-q21 of the human chromosome 17,encodes a novel transmembrane protein containing 445 amino acids .The C-terminus of MLN64 shares significant homology with the steroidogenic acute regulatory (StAR)protein,while its N-terminal domain exhibits a function of targe-ting the protein to late endosomes .MLN64 is likely to be involved in cholesterol transport and synthesis of steroid hormones.MLN64 gene,coamplified with C -erbB-2,is overexpressed in certain breast carcinomas and exerts an influence on biological characteristics of breast cancer cells .The high levels of MLN 64 observed in some breast carcinomas could contribute to the growth and progression of these tumors through increased intratumoral steroido -genesis,and is considered as a predictive factor of breast cancer prognosis .
2.Therapeutic effect of rehmannia polysaccharide on diabetic nephropathy rat model and its effects on PPARγsignal pathway

Objective To investigate therapeutic effect and molecular mechanism of RPS on Diabetic Nephropathy rats.Methods DN rats were induced by STZ injection and grouped into model group, low-dose RPS group, middle-dose RPS group, high-dose RPS group, Rosiglitazone group and normal group.24 h urine protein,kidney weight index,blood glucose level and TG, BUN and Scr level in normal and DN rats were detected.TGF-βlevel of serum of rats in all groups by ELISA were detected.mRNA and protein expression level of PPARγ,aP2 and GLUT4 by RT PCR and Western blot were also detected.Results DN rats were induced successfully because the 24 h urine protein, kidney weight index and the levels of blood sugar and TG, BUN,Scr l and TGF-βevels, and the mRNA level of PPARγ,aP2 and GLUT4 level in DN rats increased than normal group(P<0.05).The mRNA level of PPARγ,aP2 and GLUT4 level increased(P<0.05) and other indexes decreased(P<0.05) as the doses of RPS increasing.The therapeutic effects of Rosiglitazone group was better than high dose PRS group.The protein level of PPARγ, aP2 and GLUT4 in DN rat skeletal muscle were significantly lower than normal group(P<0.05), and RPS can increase their expression level obviously.Conclusion RPS has a dose-dependent therapeutic effect on DN rats by improving the expression level of related protein in PPAR gamma signaling pathways.
3.The expression and binding property of melatonin receptor in adult hepatoma tissue

Aim To determine whether there existed melatonin receptor (MR) in adult hepatoma tissue and to observe its property of binding.Methods Immuno-histochemistry was used to detect MR and identify its subcellular distribution. Specific binding and kinetic analyses of melatonin receptor were measured by radioligand binding assay.Results The results of immunohistochemistry showed that MT1 and MT2 existed in membrane, cytosol and nucleus of the hepatoma cells, but the expression of them principally localized in the membrane and cytosol. The specific binding assay properties of MR were presented as follows: the maximum binding capacity (B_(max)) was (0.29±0.07) pmol·g~(-1) protein. Equilibrium dissociation constant (K_d) was (48.7±6.5) pmol·L~(-1). The trait of MR, saturation and reversibility, was detected by ~(125)I-Mel specific binding kinetic analyses.Conclusions MR exists in adult hepatoma cells, furthermore, the subtypes of MR (MT1 and MT2) coexist in the membrane and cytosol, respectively,whose characters of the specific binding sites were low binding capacity,high affinity,saturation and reversibility.
4.LDN-193189 inhibits progression and induces apoptosis in human dedifferentiated chondrosarcoma cell line NDCS-1

Objective:To clarify the effects of the BMP receptor inhibitor LDN-193189 in the dedifferentiated chondrosarcoma (DDCS) cell line NDCS-1 and to explore the anti-tumor mechanism of LDN-193189 in DDCS. Methods:NDCS-1 was treated with 5 nmol/L of LDN-193189. MTT assay and clone formation experiments were used to verify that LDN-193189 suppressed cel proliferation. Transwel and wound healing tests were performed to demonstrate that LDN-193189 inhibited cell invasion. Western blot detection was used to show that LDN-193189 inhibited the suppression of BMPR2, p-Smad1/5, and RUNX2 protein expression. Results:The BMPR2 signaling pathway was inhibited by LDN-193189;thus, cell viability and invasion were significantly suppressed. Conclusion:LDN-193189 induces the inhibition of progression in vitro via the BMPR2-p-Smad1/5-RUNX2 signaling pathway in the human DDCS cell line NDCS-1.
5.Study of appropriateness of laboratory reports for clinical hematology in China

Objective To investigate the status of report appropriateness in Clinical Hematology and to identify its potential influence factors to improve the quality of laboratory reports.Methods 1 120 National External Quality Assessment ( EQA ) participants laboratories were enrolled in this study.The questionnaires were assigned to all the participants in both electronic and printed form.The participants were asked to retrospectively count the total reports and the affected reports while the following quality indicators occurred in 2012:report delay, inconformity of data between instrument and LIS as well as between report and request erroneous report transportation, report recall, and report modification.All quality indicators were evaluated in two ways:percentage and sigma (σ) scale.Mann-Whitney Test and Kruskal-Wallis Test were used to analyze the potential impacts of report appropriateness.Results Totally 609 (54.38%)laboratories submitted the survey results .The sigma metrics of six quality indicators on report appropriateness were more than 4.The main reason for report delay was equipment failure.Group comparison suggested that the report appropriateness of laboratories in tertiary-A hospitals and accredited by ISO 15189 werebetter.Report appropriateness with electronic request, electronic form and electronic transmission were superior than that inother ways. Conclusions Report appropriateness in China differs from laboratory to laboratory. Laboratories should develop emergency action plan in case the instrument breaks down and strengthen information technology .Long-term internal quality control and external quality assessment scheme are very meaningful to identify the insufficient performance and improve laboratory quality .(Chin J Lab Med, 2015,38:632-636)
6.Correlations of enterobacteria DNA load in venous blood determined by real-time quantitative polymerase chain reaction with vital signs and blood cell count in febrile surgical patients

Objective To determine enterobacteria DNA load in venous blood of febrile surgical patients using real-time quantitative polymerase chain reaction (RQ-PCR), to study the correlations between DNA load and vital signs/blood cell count, and to compare the difference between different detection methods in terms of positive rates.Methods A total of 72 blood samples were obtained for bacterial cuhure and RQ-PCR.The correlations of enterobacteria DNA load with body temperature, heart rate, while blood cell count,and percentages of leukocyte and lymphocyte were then analyzed.Results The enterobacteria positive rate determined by RQ-PCR (63.89%) was significantly higher than that by bacterial culture (9.72%) (F =4.383, P =0.036).The DNA load was significantly correlated with both body temperature and heart rate (P =0.006, r =0.323;P =0.000, r =0.411), but not with white blood cell count, percentages of leukocyte and lymphocyte, and age (P=0.438, r=0.093;P=0.825, r=0.027;P=0.451, r=-0.090;P =0.096, r =0.198).Conclusions RQ-PCR can quickly determine the enterobacteria DNA load in peripheral blood with high sensitivity.Routine blood cell count may not accurately reflect the enterobacteria DNA load in blood.Body temperature and heart rate may be influenced by various factors.
7.Effects of nitric oxide on mitochondrial function in cardiomyocytes:pathophysiological relevance

It is now clear that both endogenous and exogenous sources of nitric oxide(NO) exert important modulatory effects on cardiac mitochondrial function.There is also growing evidence that NO can be produced within the mitochondria themselves.NO can influence respiratory activity,both through direct effects on the respiratory chain or indirectly via modulation of mitochondrial calcium accumulation.At pathological concentrations,NO causes irreversible alterations in respiratory function and also interacts with reactive oxygen species(ROS) to form reactive nitrogen species(RNS),which may further impair mitochondrial respiration and even lead to open the mitochondrial permeability transition pore and induce cell death.Diabetes,aging,myocardial ischemia,and heart failure are all associated with altered ROS generation,which can alter the delicate regulatory balance of effects of NO in the mitochondria.
8.The management of liver cancer with blood supply from parasitic vessels after transcatheter arterial chemoembolization

50% was seen in 7 cases after TACE and PEI(30%),and the 1?3?5 year′s survival rate was 52%?35%? and 9%,respectively.Conclusions Adjuvant chemotherapy was hardly beneficial to the treatment of liver cancer with parasitic blood supply after TACE, while PEI enhanced effect of TACE as a supplementary therapy as judged by long term survival. The change of sIL-2R, AFP level was a sensitive indicator for the outcome of the treatment.
9.Clinical study of 40 cases with chronic hydrocephalus after traumatic subarachnoid hemorrhage

Objective To discuss the clinical features and treatment methods of chronic hydrocephalus after traumatic subarachnoid hemorrhage.Methods A total of 40 cases of chronic hydrocephalus after traumatic subarachnoid hemorrhage were analyzed retrospectively.Results Eight cases improved after treatment of non-operation,the lateral ventricle external drainage was done in 32 cases of chronic hydrocephalus.Shunt tube obstruction occurred in 3 patients,with abdominal,subcutaneous and intracranial infection.The shunting operation device was pulled out and the 3 patients received operation for the second time.Two cases appeared intracranial hemorrhage,and the hemorrhage was absorbed after conservative treatment.Total Glasgow prognosis score was good in 32 cases,with mild disability in 5 cases,moderate disability in 2 cases and severe disability in 1 case.Conclusions Traumatic subarachnoid hemorrhage after chronic hydrocephalus should be early diagnosed and early treated,CT and MRI scan is the effective means to the diagnosis and differentiaion.The lateral ventricle external drainage is good method in treatment.
10.The value of pro-adrenomedullin for predicting the severity and outcome of community-acquired pneumonia

Objective To investigate the value of pro-adrenomedullin (pro-ADM) levels for severity as-sessment of community-acquired pneumonia (CAP).Methods 214 CAP patients who were admitted to the emer-gency department were prospectively studied.The levels of plasma pro-ADM were determined using a new sandwich immunoassay.At the same time, procalcitonin, C-reactive protein, interleukin-6 levels, leukocyte count, clinical varia-bles and the pneumonia severity index (PSI) were measured.Results Pro-ADM levels,in contrast to procalcitonin,interleukin-6 levels, increased with increasing of the severity of CAP, which was classified according to the PSI score (P<0.05).In patients who died during follow-up, pro-ADM levels on admission were significantly higher than that in survivors [2.08 vs.4.94 μg/L,Z=-4.081 ,P<0.001].In a receiver operating characteristic (ROC) analysis for survival, the area under the ROC curve (AUC) for pro-ADM was 0.79, which was significantly higher than that for procalcitonin(0.72), C-reactive protein (0.58), interleukin-6 (0.64 ), and total leukocyte count (0.50) and sim-ilar to the AUC of the PSI (0.75).Conclusion Pro-ADM is a useful biomarker for the risk stratification of CAP patients.
