1.Effect ofbuyang huanwu decoction combined with bone marrow mesenchymal stem cell transplantation on cerebral ischemia-reperfusion injury

BACKGROUND:When acute cerebral ischemia attacks, matrix metaloproteinases (MMPs) lead to the occurrence of cerebral edema through degrading the extracelular matrix, breaking the close connection between endothelial cels, increasing the permeability of capilaries, and destroying the blood brain barrier.
OBJECTIVE: From the aspects of MMPs and extracelular matrix, to discuss the therapeutic effects ofbuyang huanwu decoction combined with bone marrow mesenchymal stem cel (BMSCs) transplantation on cerebral ischemia-reperfusion injury.
METHODS:A total of 96 Sprague-Dawley rats were randomly divided into model group, tissue inhibitor of MMPs-1 (TIMP-1) group, TIMP-1+BMSCs group (BMSCs group) andbuyang huanwu decoction+BMSCs+TIMP-1 group (combined group that was divided into four subgroups, 12-, 24-, 36-, 48-hour groups). Rat models of cerebral ischemia-reperfusion were constructed, and TIMP-1 and BMSCs were injected to the brain of rats by a microinjector in a stereotaxic apparatus. Rats in the combined group were given buyang huanwu decoction (10 mL/kg), and rats in the other groups were given the same volume of normal saline at 7 days before surgery. After 10 days of administration, serum samples and brain tissues were colected to determine MMP-2 and MMP-9 levels using ELISA and to detect MMP-9 activity using gelatinases spectrometry method.
RESULTS AND CONCLUSION:Compared with the model group, the levels of MMP-2 and MMP-9 in the serum and MMP-9 activity in the brain were decreased in the other groups to different extents, especialy the levels of MMP-9 (P < 0.05). Compared with the BMSCs group, the levels of MMP-2 and MMP-9 in serum as wel as activities of MMP-9 and pro-MMP-9 in the brain were decreased significantly in the combined group at 36 and 48 hours after treatment (P< 0.01). The results show that thebuyang huanwu decoction can be mutualy cooperated with TIMP-1 to inhibit the degradation of extracelular matrix induced by MMP-2 and MMP- 9, repair the damaged blood brain barrier, prevent and cure cerebral edema after ischemia.
2.Effects of Hedysari radix polysaccharide on the behavior and central neurotransmitters in SAMP8

Objective To investigate the improving effect of Hedysari radix polysaccharide on the behavior and its influence on cerebral central neurotransmitters in senescence accelerated mouse prone 8 (SAMP 8).Methods The senescence accelerated mice 8 were randomly divided into a model group,a Hedysari radix polysaccharide (HRP) group and a aricept group.SAMRl were served as the control group.After intragastrical treatment for 3 months,the behavior changes and the levels of norepinephrine (NE),dopamine (DA),5-hydroxytryptamine (5-HT) and 5-hydroxyindoleacetic acid (5-HIAA) were observed respectively by applying Morris water maze and high performance liquid chromatography (HPLC).Results In the SAMP8 model group,the hidden platform test showed that the latency to find the hidden platform was remarkably prolonged,the latency range from third day to fifth day was (55.31 ±9.62) s,(58.67± 10.89) s,(58.45± 10.53) s,respectively.The level ofNE、DA、5-HT and 5-HIAA was (4.97± 1.74) mmol/L,(1.63±0.57)mmol/L,(4.29±0.94)mmol/L,(2.91±0.85)mmol/L respectively,significantly reduced (P<0.01).Compared with model group,the latency of hidden platform test was (44.73 ± 9.83) s,(40.53±8.37)s,(44.75±9.16)s respectively,which were remarkably shortened.The expression level ofNE,DA,5-HT and 5-HIAA in the HRP treatment group was (7.58± 1.62) mmol/L,(3.25 ±0.70) mmol/L,(7.04±0.95) mmol/L,(4.62±0.79) mmol/L respectively,which obviously increased (P< 0.05).Conclusion HRP could improve learning and memory ability of SAMP8 and the effect may related to up-regulate the expression level of cerebral Central neurotransmitters.
3.Reflections on hospital information systems and health information legislation

The authors explained the urgency and significance of health information legislation in the current practice of hospital information systems and pointed out specific manifestations of the involvement of legal basis. Their recommendations include: ①establishing and perfecting regulations on the management of local networks; ②establishing and perfecting a system of regulations on health information; ③expanding the development, accreditation and spread of techniques for guaranteeing network safety; ④doing a good job of the standard application and management of electronic medical records in the transition period.
4.Analysis of AmpC beta-lactamase gene in Pseudomonas aeruginosa .

The gene and the amino acid sequence of the structural and regulatory region of the Pseudomonas aeruginosa with different resistance patterns were analyzed. Six strains with different resistance patterns were selected and the AmpC beta-lactamase was identified. The objective gene fragment was amplified by colonies PCR. The sequences of the PCR-products were analyzed. The DNA sequence of the structural gene ampC and the regulatory genes ampR, ampD and ampE was detected. The 6 strains and the wild-type Pseudomonas aeruginosa are highly homogeneous in structural and regulatory region. Some new mutant points were found.

5.Mechanical performance of proximal human femur after poly-ether-ether-ketone femoral prosthesis implantation

BACKGROUND: Greatly higher elastic modulus of traditional metal prosthesis than bone elastic modulus is mechanical factor for aseptic loosening of artificial joint, resulting in stress shielding, bone resorption and bone atrophy.OBJECTIVE: To measure femoral stress after prosthetic replacement made of carbon fiber reinforced poly-ether-ether-ketone (CF/PEEK), and compare with domestic AML prosthesis, so as to explore the application prospect of compound material in total hip femoral head prosthesis. DESIGN, TIME AND SETTING: Comparative analysis was performed at Orthopedics Laboratory, First Hospital of Soochow University and Institute of Biomechanical Engineering of Shanghai University from September 2002 to March 2004.MATERIALS: CF/PEEK prosthesis was designed by the authors, composed of long CF/PEEK compound material as inner layer, 100 Gpa and short CF/PEEK compound material as outer layer, 20 Gpa; the layer intensity at stress center was strengthened, and the appearance and size of the CF/PEEK prosthesis simulated domestic AML cobalt alloy prosthesis (Beijing Huajiehao Company). Five pairs (10 femurs) of femurs were from fresh adult cadavers. METHODS: Five pairs of human cadaveric femora were divided equally into left and right groups: one group received a cobalt-chrome molybdenum alloy (CoCrMo) implant and the other received CF/PEEK implant. Strain-gauge rosettes were attached to external surface of each implant and proximal femora. The loading condition simulated single-limb stance. MAIN OUTCOME MEASURES: strains distribution on the normal femur and femoral surface after replacement with two kinds of prostheses; shearing strain at femur-prosthesis interface of two groups.RESULTS: Following prosthesis implantation, the strains exhibited a progressive proximal-to-distal increase, which was similar to that in the intact femur, and the strain was maximum near the distal prosthesis. The strains on the medial and lateral surface of the femur were reduced after implantation of both types of implants. The pattern and magnitude of the strains, however, were closer to those in the intact femur after insertion of CF/PEEK prosthesis than in the CoCrMo one. CONCLUSION: Mechanical performance of CF/PEEK compound prosthesis is similar to bone tissue, and can provide similar strain under loading condition. Therefore, it can provide immediate postoperative stability and proximal loading transfer.
6.Investigation and Analysis on Environmental Pollution Due to a Small Antimony Smeltery in Southwest Guizhou Province

Objective To understand the impacts of a small antimony smeltery on surrounding environment. Methods The samples of raw materials,such as ore and coal,soil and plant around a small antimony smeltery in southwest Guizhou province were analyzed. Results The levels of arsenic,mercury,fluoride ,antimony and selenium in soil around the antimony smeltery significantly increased compared with the average background levels of related elements in nationwide range . The levels of arsenic, mercury,fluoride ,antimony and selenium in soil around the antimony smeltery were 1. 73~4.69,4. 75~237.5,2. 26~3.42,35.6~36. 2,54. 29~57.14 times as much as their related average levels in our country respectively. The levels of arsenic ,mercury ,fluoride and selenium in soil around the smeltery were 1.25~ 3. , 58,1.86~ 93.14,1.38~ 2.08,33. 04~ 34.78 times as much as their related average levels of Guizhou province respectively. The levels of arsenic, mercury, fluoride, antimony and selenium in corn leaves around the smeltery was 67,26.7,1046.4,2220,38. 8 times as much as their related average levels in non-polluted plants respectively. Conclusion The air pollutants,such as sulphur,arsenic,mercury,fluoride,antimony and selenium in industrial waste gas emitted from the antimony smeltery had resulted in heavy pollution to the environment, which was aggravated by the application of coal with high level of sulphur.
7.The tidal breathing lung function changes and clinical significance of severe pneumonia in infants and young children during convalescent period

Objective To investigate the changes in tidal breathing lung function of severe pneumonia in re-covery phase and correlation with the condition and prognosis in infants and the young children.Methods Tidal breathing lung function of 44 cases of severe pneumonia infants and 50 cases of mild pneumonia infants in their recovery phase were measured by infantile spirometer made in Germany.According to ratio of exhaled volume at peak tidal expi-ratory flow to total expiratory volume(VPTEF /VE),severe pneumonia infants were divided into normal,mild,moderate and severe groups.All these children were followed up for the frequency of rehospitalization because of lower respiratory tract infections and the frequency of wheezing within 1 year after discharge.Statistical were analyzed by using t test andχ2 test.Results Compared with the mild pneumonia infants,the respiratory rate(RR)of the severe pneumonia infants was significantly increased [(36.31 ±0.78)times/min vs (30.83 ±0.74)times/min](P <0.01 );inspiratory time to expiratory time ratio(TI /TE)(0.65 ±0.01 vs 0.71 ±0.01 ),exhaled time at peak tidal expiratory flow(TPTEF) [(0.24 ±0.08)s vs (0.31 ±0.04)s]were significantly decreased (all P <0.01 );ratio of TPTEF to expiratory time (TPTEF /TE)[(1 9.1 5 ±0.77)% vs (23.00 ±0.51 )%],VPTEF /VE[(21 .28 ±0.86)vs (25.42 ±0.56)%]were significantly decreased (all P <0.01 ).The frequency of rehospitalization because of lower respiratory tract infections (χ2 =30.50,P <0.01 )and the frequency of wheezing within 1 year after discharge(χ2 =23.43,P <0.01 )in the severe pneumonia group were significantly higher than those of the mild pneumonia group;in the groups with different lung function damage by severe pneumonia,the frequency of rehospitalization because of lower respiratory tract infections (χ2 =1 0.64,P =0.033;χ2 =20.81 ,P =0.001 )and the frequency of wheezing (χ2 =1 0.53,P =0.037;χ2 =21 .25, P =0.001 )within 1 year after discharge in mild and severe group were significantly higher than those of the mild group,while there was no significant difference between normal group and mild group (P >0.05).Thirteen cases of se-vere pneumonia infants were followed up for 1 year.Of all these 1 3 cases,pulmonary function in 1 case restored to nor-mal within 3 months;that of 6 cases restored to normal within 6 months;the conditions of 3 cases restored to normal within 1 year;while the pulmonary function of other 3 cases was still abnormal 1 year after discharge.Conclusions Lung function damage of severe pneumonia infants is more serious than the mild pneumonia in infants in their recovery phase.For severe pneumonia infants,the more serious of lung function damages,the more likely to be re -hospitaliza-tion because of lower respiratory tract infections and wheezing in short time.
8.The effects of human hepatocarcinoma HepG2 cell culture supernatant on T lymphocyte activity

Objective To observe the role of human hepatocarcinoma HepG2 cell culture supernatant on T lymphocyte of the activation, proliferation, IL-2 secretion and inducing apoptosis in vitro. To explore the mechanism of immune escape in tumor.Methods Peripheral blood mononuclear cells (PBMC) were cultured with different concentration of tumor cell culture supernatant. Cell proliferation was examined with standard cytometry and MTT. The apoptotic rate and immune phenotype were analyzed by FACS. The level of IL-2 was determined with ELISA.Results Adding HepG2 supernatant, there was no significant influence in activation of lymphocyte, but the proliferation rat and the secretion of IL-2 were obviously decreased. The lymphocyte apoptosis rate was raised.Conclusion The tumor cell may produce a certain soluble protein, thus escape from the immune surveillance of tumor-bearing host.
9.The initial stability of CF/PEEK femoral stem after total hip replacement

Objective To explore the initial stability of a carbon fiber reinforced polyetheretherketone composite (CF/PEEK) stem in human femurs after total hip replacement. Methods 8 pairs of human cadaveric femurs were used to test the CF/PEEK prosthesis versus a conventional cementless stem of cobalt-chrome molybdemum alloy(CoCrMo). The bone-femoral prosthesis interface motion was measured in vitro using displacement transducers. Combined axial and torsional loads and simulated single-leg stance loading were applied. Subsidence, rotation and interface motion were measured with load cycles up to 1 000 N. Results The extent of the micro-motion in the proximal femoral region was larger than that in the distal femoral region. The CF/PEEK composite prosthesis showed less dynamic micro-motion in the proximal region compared with the CoCrMo alloy prosthesis stem, with the average difference being 42%.
