1.Effect of mycophenolic acid on the rat mesangial cell proliferation and on TGF-β and CTGF expressions induced by high glucose

Objective To examine the effect of mycophenolic acid(MPA)on mesangial cell(MC)proliferation and on expressions of transforming growth factor-β(TGF-β)and connective tissue growth factor(CTGF)induced by high glucose.Methods The proliferation activity of mesangial cells incubated for 24h,48h and 72h in different concentrations of MPA was observed by MTT assay.The expressions of TGF-β and CTGF were measured by Western blot.Results MPA inhibited the MC proliferation and expressions of TGF-β and CTGF induced by high glucose.Conclusions MPA decreases the release of extracellular matrix by inhibiting the MC proliferation and expressions of TGF-β and CTGF,and subsequently slows the development of glomerulosclerosis in diabetic nephropathy.
2.Assessment of efficacy of narrow-band ultraviolet B for the treatment of early-stage mycosis fungoides

Objective To evaluate the efficacy of narrow-band ultraviolet B (NB-UVB) for the treatment of early-stage mycosis fungoides (MF).Methods A retrospective study was performed on 98 patients with early-stage MF treated with NB-UVB from January 2003 to December 2013.The rate of and time to recurrence of MF were assessed,and long-term side effects were evaluated in these patients.Results Among 52 patients achieving complete remission,the median number of sessions of NB-UVB radiation was 30 (range,18-87),the median cumulative radiation dose was 24.16 J/cm2 (range,9.11-108.86 J/cm2),and the median follow-up duration was 41 months (range,11-90 months).Relapse occurred in 14 (26.9%) patients with the median time to recurrence being 8.2 months (range,3-60 months).The patients suffering from relapse achieved complete remission after treatment with NB-UVB again.Adverse reactions mainly included skin erythema and itching at the early stage of radiation,as well as temporary pigmentation after radiation.Cutaneous carcinoma occurred in none of these patients during 11-90 months of follow-up.Conclusions NB-UVB phototherapy appeared to be effective for the treatment of early-stage MF,and no cutaneous carcinoma occurred in these patients during the follow-up.Although MF may relapse in some cases,repeated NB-UVB phototherapy is still effective.
3.Influence of the management of family-doctor responsibility system on drug utilization and compliance of hypertensive patients

Objective To discuss the influence of the management of family-doctor responsibility system on drug utilization and compliance of hypertensive patients.Methods 1039 primary hypertension patients in Zhenhai Lianhua Community who were under the management of the residents electronic health files in June 2012,were implemented with the latest family-doctor responsibility system for eight months.The on-site questionnaire survey and electronic health management platform were carried out to analyze the drug utilization and treatment compliance of hypertensive patients.Results The proportion of taking medicine treatment among 1039 patients with essential hypertension reached 85.2%,including 56.3% patients who took one kind of drug,35.4% patients who took two kinds of drugs and 8.3% patients who took at least three kinds of drugs.The most commonly used drugs were Amlodipine Besylate,Felodipine,Irbesartan and Hydrochlorothiazide tablets,Irbesartan,Levamlodipine Besylate tablets,Metoprolol Tartrate tablets and etc.The rate of patient compliance was 65.5%.After comparing the compliance of any two among three kinds of drug groups,the comparison of compliance between patients who took one kind,two or three kinds of drugs showed statistical significance.The comparison of drug types and compliance showed that compliance with compound preparation was statistically different from compliance with ARB,calcium antagonists,sympatholytic agent,diuretics.The educational level was also related to the compliance.Conclusions After the implementation of the family-doctor responsible management,the treatment rate and compliance rate are high,and the patients with hypertension in community still give priority to long-acting drugs.

5.Characteristics of acid reflux in Barrett's esophagus.

To determine the relationship between Barrett's esophagus (BE) and features of gastroesophageal acid reflux, 24 h esophageal pH monitoring was performed in 90 patients. The patients were divided into 3 groups: 31 subjects with BE, 21 with mild esophagitis and 38 with severe esophagitis. The following parameters were evaluated: the percentage time of pH < 4; the number of reflux episodes over 5 min; the duration of longest episodes and DeMeester score over total period and the auterior three parameters in erect and supine position. All these parameters in BE were significantly different from those with mild esophagitis (P < 0.01) and not significantly different from those with severe esophagitis (P > 0.05). During supine position all the above parameters in BE were significantly different from those with reflux esophagitis (P < 0.05). It is concluded that the quantity of acid reflux is not an important factor in development of BE in gastroesophageal reflux (GER), and the acid reflux in supine position might be important in development of BE in GER.

6.Effects of mycophenolic acid on high glucose-induced expression of TGF-beta and CTGF in mesangial cells.

The effects of mycophenolic acid (MPA) on high glucose-induced expression of transforming growth factor-beta (TGF-beta) and connective tissue growth factor (CTGF) in mesangial cells (MC) were investigated. Rat MC were cultured in the presence of different concentrations of MPA (1.0 and 10.0 micromol/L) or MPA plus high glucose for 72 h. The expression of TGF-beta and CTGF was detected by Western blot. The results showed that high glucose could induce the expression of TGF-beta and CTGF in MC, but MPA could inhibit this effects. MPA did not influence the expression of TGF-beta and CTGF in normal glucose. It was concluded that MPA might prevent the progression of diabetic nephropathy by inhibiting the expression of TGF-beta and CTGF in MC.
7.Developmental competence of pig oocytes maturation in different culture mediums

Objective To optimize systems of pig embryo in vitro production. Methods Pig oocytes were matured and fertilized in vitro in different culture mediums. Maturation rate and cleavage rate were analyzed. Results Gonadotropin induced maturation of pig oocytes in vitro , and improved the frequence of oocytes developed into M Ⅱ. The highest maturation and cleavage rates, present in medium with 15 IU/ml FSH+30 IU/ml LH, were 62% and 15% respectively. 8.Study on the reversion of drug resistance in clinical isolates of E. Coli by EGS technique

Objective To explore the potential of converting the chloramphenicol resistant clinical E. Coli isolates to chloramphenicol sensitive ones by employing external guide sequence(EGS) technique in vitro. Methods Recombinant plasmids with EGScat 1+2 and tetracycline resistant gene, named PAlterl EGScat1+2, was constructed. Routine Cacl 2 method was used to introduce recombinant plasmid into the chloramphenicol resistant clinical isolates E. Coli 4758. Colony PCR was used to test and A600 was used to detect growth rates in liquid and solid culture of various concentrations of chloramphenicol. Results The chloramphenicol resistant clinical isolates E. Coli 4758 grew well in chloramphenicol(35 ?g/ml, 70 ?g/ml, 105 ?g/ml, 170 ?g/ml) plates whereas the transformants tE4758 with PAlter1 EGScat 1+2 failed to grow in these concentrations, which indicated its resistance to the chloramphenicol was reversed. Conclusions EGS molecules are able to convert the drug resistance in clinical E. Coli isolates in vitro.
9.Influence of two photoactivation modes on the hardness of packable and conventional resin-based composites

BACKGROUND: Soft-start is a newly photoactivation mode, which has certain effect on composite resin. However, previous study mainly concentrated on the conventional resin-based composites, the reports regarding soft-start on packable resin-based composites is poorly understood. OBJECTIVE: It is assumed that soft-start photoactivation had effect on packable resin-based composites, in addition, to investigate its effect on the hardness of packable resin-based composites. DESIGN, TIME AND SETTING: A double factors design. The experiment was performed at the Department of Stomatology, First Affiliated Hospital, Sun Yat-sen University and Chemical Mine Metal Material Test Laboratory, Guangdong Inspection and Quarantine Technology Center in October 2007. MATERIALS: Three packable resin-based composites were Ecusphere-Carat (EC, DMG Company, Germany), Filtek P60 (P60, 3M EPSE Company, USA), Tetric Ceram HB (HB, Ivoclar Vivadent Company, Liechtenstein) and a conventional composite FiltekZ250 (Z250, 3M EPSE Company, USA). The color of composites was A3. METHODS: Three packable resin-based composites and a conventional composite were filling in a cylindrical container (7 mm diameter, 4 mm depth), to obtain 80 samples, and then were divided into different groups according to the composite and photoactivation mode (n=10). In the soft-start photoactivation, samples were irradiated by 300 mW/cm~2 for 10 s, and then 600 mW/cm~2 for 30 s. Standard photoactivation was irradiated with 600 mW/cm~2 for 40 s.MAIN OUTCOME MEASURES: The microhardness of the top and bottom of the specimens was determined by Vickers microhardness tester. RESULTS: Three packable composites had higher hardness values than conventional composite. Though soft-start photoactivation could decrease the hardness of packable composites, the difference had no significant difference to standard mode (P > 0.05). There was significant difference on the top hardness and on the bottom hardness of conventional composite between two photoactivation modes (P < 0.05). CONCLUSION: The soft-start mode and resin-based composites should be selected carefully according to different filing areas.
10.BIODEGRADABLE POLY (D,L-LACTIDE-CO-GLYCOLIC ACID) MICROSPHERESUSED AS ADJUVANT IN DNA IMMUNIZATION OF HEPATITIS B VIRUS S GENE

Study the immunological adjuvant function of biodegradable microspheres for DNA immunization. Empty poly(D,L-lactide-co-glycolic acid) microspheres were prepared using the water-inoil-in-water(w-o-w)technique; A plasmid DNA pRc-CMV encoding hepatitis B virus S antigen was constructed;The mixture of the microspheres and the plasmid DNA was prepared by incubation method. The mixture was administered to Balb/c mice by intramuscular injection. Result: The high antibody titer (1:1600) of intramuscular injection of the mixture of microspheres and the plasmid DNA was obtained, similar to that of intramuscular injection of the mixture of AL(OH)3 and hepatitis B virus S antigen; while intramuscular injection the plasmid DNA elicited no serum antibody respones. Conclusion: biodegradable microspheres may be used as an good adjuvant for DNA immunization.
