1.Optimization of Preparation Technics for Liver-strengthening Cholagogue Oral Preparations by Orthogonal Test

OBJECTIVE:To optimize the extraction technics of liver-strengthening cholagogue oral preparation.METHODS:The effects of the four factors-the quantity of the added water,the decoction time,the decoction times and the alcohol precipitation concentration on the extraction results were determined with content of total flavonoids taken as index.RESULTS:The best extracting condition was the following,the amount of the added water was 10 times that of the medicinal materials,the decoction duration was 60min,the alcohol precipitation concentration was 50%and the decoction was carried out twice.CONCLUSION:The preparation under this technics is of high clarity and stable contents.
2.Effects of different levels of neuromuscular blockade on transcranial electric motor-evoked potentials during idiopathic scoliosis

Objective To evaluate the effects of different levels of neuromuscular blockade(NMB)on transcranial electric motor-evoked potentials(TCeMEPs)during idiopathic scoliosis.Methods Thirty American Society of Anesthesiologists physical status Ⅰ or Ⅱ patients of both sexes,aged 11-23 yr,weighing 31-62 kg,scheduled for elective idiopathic scoliosis under general anesthesia,were enrolled in the study.NMB was monitored with train of four(TOF)-Watch SX.The levels of partial NMB were classified into 5 states according to TOF ratio(TOFR)and TOF counts:1 or 2 TOF counts(TOF1),3 TOF counts and TOFR≤15%(TOF2),TOFR 16%-25%(TOF3),TOFR 26%-50%(TOF4),TOFR 51%-75%(TOF5) and TOFR>75%(no NMB).Each state was maintained for 10 min.Failure and false-positive findings in TCeMEP monitoring,development of unexpected body movement and satisfaction with NMB were recorded.Results Compared with no NMB,the failure and false-positive rates of TCeMEP monitoring were significantly increased,the incidence of unexpected body movement was decreased,and the rate of satisfactory NMB was increased at TOF1,TOF2 and TOF3(P<0.05),no significant change was found in failure or false-positive rates of TCeMEP monitoring at TOF4 and TOF5(P>0.05),and the incidence of unexpected body movement was decreased and the rate of satisfactory NMB was increased at TOF4,the rate of satisfactory NMB was increased at TOF5(P<0.05),and no significant change was found in the incidence of unexpected body movement at TOF5(P>0.05).Compared with those at TOF4,no significant change was found in the failure or false-positive rates of TCeMEP monitoring(P>0.05),the incidence of unexpected body movement was significantly increased,and the rate of satisfactory NMB was decreased at TOF5(P<0.05).Conclusion Maintaining TOFR at 26%-50% the partial NMB during surgery does not affect TCeMEP monitoring during idiopathic scoliosis and meets the intra-operative NMB requirements simultaneously,and it is the optimum NMB for this type of surgery.
3.Comparative Analysis of the Components of Volatile Oil in Citrus medica from Different Producing Areas

OBJECTIVE:To compare th e difference of the components of volatile oil in Citrus medica from different producing areas. METHODS :The volatile oil of C. medica from 10 different producing areas was extracted with steam distillation ,and the yield was calculated. The components of the volatile oil of C. medica from different producing areas were analyzed by GC-MS. The compounds were retrieved from NIST 14.L mass spectrum database and identified. Relative mass fraction of chemical component was determined by peak area normalization method. Cluster analysis of samples were performed by using SPSS 20.0 software. RESULTS:The yields of volatile oil of C. medica from 10 different producing areas were 0.10%-1.75%,among which sample from Qianwei county in Leshan city of Sichuan province was the highest (1.75%). A total of 66 components were identified in the volatile oil of C. medica from different producing areas ,with a relative molecular weight of 126.20-392.66. The majority was C 10 and C 15 compounds;isomers with relative molecular weight of 136,154 took up the great proportion ,which were mainly cycloalkane monoterpenes. There were 12 common components in the volatile oil of C. medica from different areas ,which were limonene(24.90%),terpinene(14.71%),(-)-4 terpineol(2.88%),citral(2.33%),α-myrrhene(2.33%),geraniol(1.52%), α-pinene(1.37%),trans bergamot olene (1.16%),isoterpinene(1.13%),methyl palmitate (1.12%),linalool(1.09%)and geranyl acetate(1.04%)according to relative mass fraction ;8 of them were monoterpenes ,2 were sesquiterpenes and 2 were esters. There were 4 categories of C. medica from different producing areas ,i.e. S 2,S4,S6 clustered into one ;S1,S3,S7,S8 clustered into one ; S5 and S 10 clustered into one ;S9 as one . CONCLUSIONS : There are some difference of the components in volatile oil of medica from different producing areas ,and the content of the same component also has great difference in the volatile oil of C. medica from different producing areas.
4.Study on the Separation and Purification Technology of Total Flavonoids from Sparganium stoloni by Box-Behnken Design- response Surface Methodology

OBJECTIVE: To optimize the purification technology of total flavonoids from Sparganium stoloniferum. METHODS: Separation and purification by macroporus adsorption resin, using sample solution pH, flow rate and concentration of eluent, the purification rate of total flavonoids as evalution indexes, the purification technology of total flavonoids from S. stoloniferum were optimized by Box-Behnken design-response surface methodology based on single factor test. Validation test was conducted. RESULTS: The optimal purification technology was sample solution pH 4.8, flow rate of eluent 2.0 BV/h, concentration of eluent 72%. The purification rate of total flavonoids in 3 batches of samples was 72.34% (RSD=1.77%, n=3) in validation test, relative errors of which to predicted value (73.99%) was 2.13%. CONCLUSIONS: The optimal purification technology is stable and feasible, and can be used for the purification of total flavonoids from S. stoloniferum.
5.A hierarchical vascularized engineered bone inspired by intramembranous ossification for mandibular regeneration.

Mandibular defects caused by injuries, tumors, and infections are common and can severely affect mandibular function and the patient's appearance. However, mandible reconstruction with a mandibular bionic structure remains challenging. Inspired by the process of intramembranous ossification in mandibular development, a hierarchical vascularized engineered bone consisting of angiogenesis and osteogenesis modules has been produced. Moreover, the hierarchical vascular network and bone structure generated by these hierarchical vascularized engineered bone modules match the particular anatomical structure of the mandible. The ultra-tough polyion complex has been used as the basic scaffold for hierarchical vascularized engineered bone for ensuring better reconstruction of mandible function. According to the results of in vivo experiments, the bone regenerated using hierarchical vascularized engineered bone is similar to the natural mandibular bone in terms of morphology and genomics. The sonic hedgehog signaling pathway is specifically activated in hierarchical vascularized engineered bone, indicating that the new bone in hierarchical vascularized engineered bone underwent a process of intramembranous ossification identical to that of mandible development. Thus, hierarchical vascularized engineered bone has a high potential for clinical application in mandibular defect reconstruction. Moreover, the concept based on developmental processes and bionic structures provides an effective strategy for tissue regeneration.
