1.Expression of Vascular Cell Adhesion Molecule 1 and Intercellular Adhesion Molecule 1 in Endometrium and Serum in Endometriosis

Objective:To study the expressions of Vascular Cell Adhesion Molecule 1 (VCAM-1) and Intercellular Adhesion Molecule 1 (iCAM-1) in patients with endometriosis and to explore its effect on the onset of endometriosis.Methods:VCAM-1 mRNA and ICAM-1 mRNA were detected by real-time PCR method in 15 specimens of eutopic endometrium and 15 specimens of ectopic endometrium from patients with endometriosis(EMT group)and 15 specimens of endometrium from patients without endometriosis(control group).VCAM-1 and ICAM-1 protein were detected by Western-Blot method.Soluble VCAM-1 and soluble ICAM-1 in 44 serum of patients with endometriosis(EMT group) and 28 serum of patients without endometriosis(control group) were tested by ELISA.Meanwhile the correlation of expressions of VCAM-1 and ICAM-1 were analyzed in different groups.Results:①The expressions of VCAM-1 mRNA and ICAM-1 mRNA in ectopic endometrium were significantly higher than those of eutopic endometrium and the controls(P <0.01).There was no significant different mRNA expression between eutopic group and control group(P>0.05).②The protein expressions of VCAM-1 and ICAM-1 in eutopic group were significantly higher than those of ectopic group and the control group(P <0.01).The protein expression of VCAM-1 in eutopic group were higher than those in the controls group(P<0.01).There was no significant different protein expression of ICAM-1 between eutopic group and the controls group(P >0.05).③The concentration of sVCAM-1 and sICAM-1 in serum from patients with EMT were significantly higher than those in the control group(P<0.01).The concentration of slCAM-1 in serum from patients with EMT in stage Ⅲ-Ⅳ were significantly higher than those in stage Ⅰ-Ⅱ (P < 0.01).④There was no significant correlation in the expression of mRNA,protein and serum between VCAM 1 and ICAM-1 in ectopic group and eutopic group(P > 0.05).Conclusions:The abnormal expressions of VCAM-1 and ICAM-1 in ectopic endometrium may play an important role in the development of endometriosis.The signaling pathways and protein expression way may be different between VCAM 1 and ICAM 1 in endometriosis.
2.Clinical Study on Mingmen Eight Array Massage Combined with Compound Lysine Hydrochloride and Zinc Gluconic Granules for Treatment of Infantile Diarrhea

Objective To observe the clinical efficacy of the Mingmen eight array massage combined with compound lysine hydrochloride and zinc gluconate granules for the treatment of infantile diarrhea. Methods Totally 120 children with diarrhea met the inclusion criteria were divided into the treatment group, Western medicine group, and general massage group by random number table method, 40 cases in each group. All 3 groups were given dietary guidance and did moderate exercise. Western medicine group was administered orally for compound lysine hydrochloride and zinc gluconate granules, two bags per day for the aged 1–10, three bags per day for the aged >10. Treatment group was given the Mingmen eight array massage therapy based on Western medicine. General massage group received conventional massage therapy based on Western medicine, with four weeks as treatment course. 3 groups of patients were observed before and after treatment for salivary amylase, blood zinc value, urine D-xylose absorption and excretion test, TCM symptom score, and clinical efficacy were evaluated. Results There was statistical significance in the salivary amylase, blood zinc value, and urinary D-xylose absorption and excretion rate between before and after t reatment(P<0.05); after treatment, treatment group showed higher salivary amylase, blood zinc value, and urinary D- xylose absorption and excretion rate than the Western medicine group and general massage group (P<0.05); scores of diarrhea, loose stool, lassitude, lusterless complexion, and appetite reduction decreased compared with before treatment (P<0.05); after treatment, all integrals of treatment group were lower than Western medicine group and general massage group (P<0.05). The total effective rates of the treatment group, Western medicine group and general massage group were 92.50% (37/40), 72.50% (29/40), and 75% (30/40) respectively. There was statistical significance among the three groups (P<0.05). Conclusion Mingmen eight array massage combined with compound lysine hydrochloride and zinc gluconic granules can effectively alleviate the clinical symptoms of patients with infantile diarrhea, and improve the level of laboratory parameters.
3.Clinical observation on the treatment of premature ventricular beats with Zaobo decoction combined with bisoprolol

Objective To observe the clinical effects of Zaobo decoction combined bisoprolol treating premature ventricular contractions.Methods 108 patients of ventricular premature beat were recruited intoa control group and an observation group (n=54) according to the random number table method.The control group was treated with bisoprolol, while the observation group was treated with Zaobo decoction on the basis of the control group, and both groups were treated for 8 weeks.24 h dynamic electrocardiogram (ECG), renin activity plasma (PRA), angiotensin Ⅱ (angiotensionⅡ) and ALD (Ang) were observed before and after treatment.The clinical effects were evaluated.Results The total effective rate showed significant difference between the observation group and the control group (75.9% vs.57.4%;x2=4.167, P=0.041) after the treatment.After treatment, Ang-Ⅱ (56.22 ± 12.7 pg/ml vs.68.45 ± 12.7 pg/ml, t=5.004) in the observation group was significantly lower than the control group (P<0.01);24 h sinus RR interval standard deviation (129.16 ± 28.56 ms vs.116.13 ± 17.38 ms, t=2.864), every 5 min sinus RR interval mean standard deviation within 24 h (123.57 ± 25.24 ms vs.112.46 ± 18.23 ms, t=2.622), and within 24 h of sinus RR interval difference rms (31.76 ± 11.42 ms vs.22.64 ± 10.32 ms, t=4.354) in the observation group were significantly higher than the control group (P<0.01).Conclusion Zaobo decoction combined with bisoprolol can effectively improve heart rate variability, regulate rernin vascular angiotensin system, and improve the clinical efficacy of the patients with ventricular premature beat.
4.Acupoint injection of astragalus membranaceus injection on patients with tumor leukopenia after radiotherapy and chemotherapy and its influence on cellular immune function

Objective To observe the effect of Zusanli acupoint injection of Astragalus Membranaceus Injection on patients with tumor leukopenia after radiotherapy and chemotherapy and its influence on cellular immune function.Methods According to clinical case of random table,60 cases of malignant tumor patients in Hubei province Xiangyang city hospital from March 2010 to August 2011 were randomly divided into two groups of 30 patients in each group.The treatment group was treated with Zusanli acupoint injection of Astragalus Injection therapy,while the control group was taken Leucogen,vitaminB4,batyl alcohol orally.Venous blood and peripheral white blood cell were taken in both groups before and after treatment.Direct immunofluorescence staining and flow cytometry were used for the detection of peripheral blood T lymphocyte total number and its subsets.Results ① clinical efficacy:Total effectiveness was 77.5%in the treatment group,and 52.5% the control group,the difference was statistically significant between the two groups (x2=8.027,P<0.05).Comparison of the white blood cell count and T lymphocyte subsets of peripheral blood:the white blood cell count was (3.56~4.21) × 109/L in the treatment group,and (2.89~3.39) × 109/in the control group,there was significant difference between two groups (t=2.435,P<0.05); there was significant elevation of CD3 +,CD4+,CD4+/CD8 + [(62± 1.63) %,(43 ± 0.80) %,(1.48 ± 0.23) in the treatment group after the treatment comparing with (48± 1.25)%,(31 ± 0.74)%,(0.93± 0.12) before the treatment (P<0.05 or 0.01),while there was no such changes in the control group.Conclusion Zusanli acupoint injection of Astragalus Injection may modulate the cellular immune function of patients with cancer.It is effective in the treatment of leukopenia after radiotherapy and chemotherapy.
5.Study on the transcription profiling of biofilm formation by Candida albicans at different phases exposed to tyrosol and farnesol

Objective To investigate the effects of tyrosol and farnesol on the transcription profiling of C.albicans biofilm by microarray analysis.Methods The standard strain of C.albicans,SC5314 were cultured into four groups (tyrosol treated,farnesol treated,tyrosol and farnesol co-treated,and untreated control).The cell suspensions of SC5314 were prepared and dispensed into polystyrene flasks to form biofilm.Then,the biofilms were collected at 6 h and 24 h respectively after culturing.RNA samples were extracted and synthesized into cDNA through reverse transcription.The genome arrays were scanned with a confocal LuxScanTM scanner and the images were then analyzed by using LuxScanTM 3.0 software (both from CapitalBio).Bioinformatics analysis of the data was carried out by comparatively analyzing S.cerevisiae gene in KEGG gene database.Results The cDNA microarray data showed that tyrosol and farnesol regulated biofilm formation by regulating the genes associated with biofilm formation such as hypha genes and yeast genes.Tyrosol positively regulated gene expression of C.albicans biofilm,while farnesol played negative role.There were striking differences in gene expression patterns between tyrosol or farnesol treated groups and the control.Tyrosol had no antagonistic effect to farnesol.Bioinformatics analysis showed that the differential gene expression was involved in biological process,molecular function process and cell component formation.These genes regulated the C.albicans biofilm formation by encoding proteins involved in the biological metabolism.Conclusion Tyrosol and farnesol influenced the formation of C.albicans biofilm through regulating gene expression which showed differences at different phases of biofilm formation.
6.Effect on Immunologic Function of Chronic Abacterial Prostatitis Rats Treated by Electroacupuncture at Sanyin Points

Objective To study the immunologic mechanism of electroacupuncture at Sanyin points on chronic abacterial prostatitis rats. Methods Forty-eight male rats were divided randomly into normal group, model group, drug group and electroacupuncture group, 12 rats in each group. The animal model was made by SC purified prostate protein twice with immunologic adjuvant, and the rats in electroacupuncture group and drug group were treated by electroacupuncture at Sanyin points and feeding Cernilton for 15 days. The changes of prostate weight, index, pathology and contents of IgG, expressions of IL-2 and IL-8 in prostate homogenate were observed. Results Compared with normal group, the prostate weight and index were significantly increased in model group (P<0.05), the content of IgG was significantly increased (P<0.05), cytokine IL-2 and IL-8 were highly expressed (P<0.05, P<0.01), prostate tissue’s morphology and structure were significantly impaired in model group. Compared with model group, the prostate weight and index were significantly decreased in electroacupuncture group after treatment (P<0.05 or P<0.01), pathological changes reduced significantly, the content of IgG decreased significantly (P<0.05), the expressions of cytokine IL-2 and IL-8 were significantly decreased (P<0.05, P<0.01), and the pathological manifestation of chronic inflammation was significantly improved. Conclusion Electroacupuncture at Sanyin points can reduce the secretion of local prostate antibody IgG, regulate the expression of cytokine IL-2 and IL-8, protect the morphology and structure of prostate tissue from damage. Electroacupuncture at Sanyin points can adjust local immunologic function and promote the recovery of prostate.
7.The Influence of Different Wrap Method on the Intracranial Pressure of Patients after Standard Craniectomy

Objective To explore the influence of different methods of bandaging on the postoperative intracranial pressure of patients with severe brain injury patients after decompression craniectomy. Methods The standard decompressive craniectomy was use for the 36 cases of severe traumatic brain injury patients, and the intracranial pressure monitoring sensor probe was indwelled in operaion. Two different dressing methods of elasticity mesh cap and applicator were used for the patients respectively at 0h, 72h, 120h and 168h after operation, and the value of intracranial pressure was monitored and recorded. Result The intracranial pressure of elastic cap were significantly higher than the applicator respectively in operation immediate postoperative 72h, 120h and 168h (P<0.05), the difference was statistically significant. Conclusions The intracranial pressure of elastic cap is significantly higher than the applicator at different times after the surgery group.
8.Expressions and clinical significances of SMO, STAT3 and MMP-9 in triple negative breast cancer

Objective:To investigate the expressions and clinical significances of SMO , STAT3 and MMP-9 in triple negative breast cancer ( TNBC) .Methods:The expressions of SMO , STAT3 and MMP-9 were immunohistochemically detected in 33 cases of TNBC, 30 cases of mammary hyperplasia and 18 cases of normal breast tissue , and the relationships among SMO , STAT3 and MMP-9 and clinicopathological parameters and prognosis of TNBC patients were analyzed .Results:In TNBC and mammary hyperplasia tissue , the positive expression rates of SMO were 90.9%and 60.0%,the positive expression rates of STAT3 were 96.9% and 73.3%, the positive expression rates of MMP-9 were 90.9% and 36.7%, respectively, however, expressions of them were completely absent in normal breast tissue .The significant correlations were observed between expressions of SMO , STAT3 and MMP-9 in TNBC, mammary hyperplasia and normal breast tissue (P<0.05).The expression of SMO and STAT3 was correlated with superior histologic grade and tumor stage .The expression of MMP-9 was correlated with metastasis of lymph node .The positive correlation was manifested between SMO and STAT3(r=0.361,P<0.05),between SMO and MMP-9 (r=0.633,P<0.01),between MMP-9 and STAT3 (r=0.803,P<0.01) in TNBC.The survival time of TNBC patients was correlated with SMO expression and pTNM (P<0.05).Conclusion:SMO, STAT3 and MMP-9 may play important roles in development of TNBC and may be an important therapeutic target in TNBC .

10.Establishment of blood purification system for rats

Objective To establish blood purification system for rats and provide a safe and reliable experimental platform for further research of blood purification. Methods The right carotid artery and the contralateral jugular vein of adult male Sprague?Dawley rats were cannulated to creat vascular access for blood purification, by which continuous arteriovenous hemofiltration blood purification system was established. Blood flow, substitution fluid flow and ultrafiltration rate were regulated by rotary mini?pumps. Blood purification therapy continued for 4 hours on the basis of maintained anesthesia and effective anticoagulation. The safety of continuous arteriovenous hemofiltration blood purification systems was evaluated by comparing the arterial blood gas, electrolyte indexes and blood glucose during the blood purification therapy. Closely monitoring the vital signs of rats, such as blood pressure and heart rate, and observing whether there were any side effects, such as massive haemorrhage, thrombogenesis and gas embolism in the therapeutic process. Results There were no obvious changes of arterial blood gas, electrolyte indexes and blood glucose during the blood purification therapy (P>0.05). The vital signs did not fluctuate acutely before and after the blood purification therapy (P>0.05). The incidence rate of side effects was very low. Conclusions
Continuous arteriovenous hemofiltration blood purification system had no obvious adverse effects on healthy rats. Our blood purification system for rats appears to be safe and reliable.
