1.Detection of HLA-DRB 1*12 by use of SYBR Green I PCR

Objective To detect the HLA DRB1*12 by use of a new genotyping method for HLA SYBR Green Ⅰ PCR. Methods 1 HLA DRB1*12 sample and 12 unknown clinical sample were collected. The typing of these samples were determined by PCR, which includes a fluorescence dye, SYBR GreenⅠ,and the sequence specific primer. SYBR GreenⅠcould bind to the dsDNA to exhibit fluorescence. The fluorescence enhancement depends on the accumulation of the amplified product. Results 3 of the 12 unknown sample was proved to be DRB1*12 by the analysis of the melting curve of the amplified products. After purification, the type of these products were confirmed DRB1*12 by DNA based sequencing. Conclusions The results of SYBR GreenⅠPCR demonstrate that it can be a new strategy for HLA typing because of its convenience and accuracy.
2.Effects of mild hypothermia on proNGF apoptosis pro-signal following cerebral ischemia and reperfusion

Objective To observe the expressional changes of pro nerve growth factor(proNGF)apoptosis pro-signal factors after cerebral ischemia and reperfusion and discuss the protective effect of mild hypothermia on the brain injury. Methods The randomly chosen female SD rats weighing(220±10)g were divided into normal control group(Group A),cerebral isehemia and reperfusion group (Group B)and mild hypothermia group(Group C).Groups B and C were divided into 3,6 and 9 hours subgroups(eight rats per group).TUNEL methods was used to detect the apoptotic cells in the cortex after cerebral ischemia and reperfusion and treatment with mild hypothermia.The reverse transcription polymerase chain reaction(RT-PCR)was used to detect the expression changes of proNGF,Sortilin and P75gene in the cortex. Results (1)The apoptotic cells were increased with time in the Group B(P<0.05)and were more than those in the Group C in the same time point(P<0.05).(2)There found expression of all factors at all time points in three groups,which was increased with time(P<0.05).(3)The content of every factor at each time point in the Group C was less than that in the Group B(P<0.05). Conclusions (1)Mild hypothermia can decrease the apoptosis after ischemia and reperfusion injury.(2)The cell apoptosis induced by proNGF plays a very important role in the COUrse of cerebral ischemia and reperfusion injury.(3)Mild hypothermia can decrease the expression of apoptosis pro-signal to alleviate the cell apoptosis and protect the brain from ischemia and reperfusion.
3.STUDIES OF METHOD OF THE SPORANGIA PRODUCTION AND DIL UTION OF THE ZOOSPORE SUSPENSIONS FROM PHYTOPHTHORA BOEHMERIAE

A simple method for in vitro production of sporangia by Phytophthora boehmeriae causing cotton boll blight in China was developed, and a method for dilution of zoospore suspensions was presented as well, Large amounts of sporangia were obtained when mycelial mats, produced from Potato extract-Dextrose Liquid (PDL) or Bean extract-Dextrose Liquid (BDL) at 24 ℃ for 48—72 hr ,were rinsed with Mineral Salt Solution (MSS) four times at 20—22℃ for 10—12 h with continuous fluorescent light (cool white), and incubated for additional 12 h after drained off MSS. Darkness favored oospore whereas suppressed sporangial production by P. boehmeriae. BDB is superior to PDB in the production of oospores and sporangia by P. boehmeriae as evidenced by the amount of sporangia or oospores and in particular the fact that the color change of mycelial mats from light orange to purple was synchronous with the sporulation of P. boehmeriae. Tris-succinate buffer(pH6.8, 5.0mmol/L, TSB) was the quantitatively excellent diluent for preserving motility of zoospores of P. boehmeriae in the dilution process.

6.Bibliometric study of the articles on environment enrichment in china

Objective To analyze the literature characteristics on environmental enrichment published in China , and provide reference for researchers .Methods The papers relevant to environmental enrichment published in CNKI database and Wanfang database were statistically analyzed from the aspects of publishing year, journal of publication, author, institution and subjects covered by the article with bibliometrics method.Results There were 422 papers published before Dec 31,2015, in which 273 papers published in 126 kinds of journals.The number of papers increased gradually, especially after 2006.The top 3 journals with the most articles published were Chinese Journal of Wildlife(28),Chinese Journal of Rehabilitation Medicine(15) and Chinese Journal of Behavioral Medicine and Brain Science(12).Xinqiao Hospital of TMMU and Fujian Medical University Union Hospital accounted for the top 2 total number of the papers, from which Yan-hui Chen and Cong-min Zhao published the more articles.Those papers covered 17 subjects, neurology, pathergasiology, biology and pediatrics were the main subjects. There were 3 kinds of animal that got enriched, the first was laboratory animals(69.6%), then zoo animals(22.9%),and farm animals(7.5%).Conclusions The research of environmental enrichment was relatively concentrated in terms of neurology and psychiatry.To improve animal welfare was mainly in the zoo animals;laboratory animal research in this direction was less .It is necessary to strengthen the work in this regard.
7.Clinical Observation of Amlodipine Besylate Combined with Lisinopril and Hydrochlorothiazide,Atorvas-tatin in the Treatment of Severe Primary Hypertension Complicating with Carotid Atherosclerosis

OBJECTIVE:To observe the clinical efficacy and safety of amlodipine besylate combined with lisinopril and hydro-chlorothiazide,atorvastatin in the treatment of severe primary hypertension complicating with carotid atherosclerosis. METHODS:90 patients with severe primary hypertension complicating with carotid atherosclerosis were divided into control group (45 cases) and observation group(45 cases)according to random lottery form. Both groups were given Atorvastatin calcium tablet 20 mg/time orally,qd;control group was additionally given Amlodipine besylate tablet 5 mg/time orally,qd;observation group was additional-ly given Lisinopril and hydrochlorothiazide tablet 10 mg/time orally,qd,on the basis of control group. Both groups were treated for 8 weeks. Clinical efficacies of 2 groups were compared as well as blood pressure level,IMT,PV of carotid atherosclerosis, hs-CRP,TNF-α before and after treatment. The occurrence of ADR was recorded. RESULTS:Total response rate of observation group was significantly higher than that of control group,with statistical significance (P<0.05). Before treatment,there were no significant differences in SBP,DBP,IMT,PV,hs-CRP and TNF-α level(P>0.05). After treatment,the levels of SBP,DBP, IMT,PV,hs-CRP and TNF-α level in 2 groups were significantly lower than before;the observation group was significantly lower than the control group,with statistical significance (P<0.05). There was no statistical significance in the incidence of ADR be-tween 2 groups (P>0.05). CONCLUSIONS:Amlodipine besylate combined with lisinopril and hydrochlorothiazide,atorvastatin in the treatment of primary hypertension complicating with carotid atherosclerosis can effectively control the blood pressure level, delay the progression process of carotid atherosclerosis,reduce the inflammatory reaction degree,but dose not increase the occur-rence of ADR with good safety.

AIM:To study the chemical constituents of the root of Myrica nana cheval.METHODS:The constituents were separated and purified by different methods of chromatography,and their structures were elucidated by UV,IR,GM and NMR.RESULTS:Seven compounds were isolated from the root of Myrica nana cheval.Their structures were identified as ?-sitosterol(1),myricanol(2),myricanone(3),ursolic acid(4),quercetin(5),myricetin(6)and myricitrin(7).CONCLUSION:Except for 5,6 and 7,the other compounds were obtained from this plant for the first time.
9.Discussion on the fundamental pathogenesis of Parkinson’s disease (PD):kidney deficiency and blood stasis

Objective:To investigate the TCM pathogenesis of Parkinson’s disease(PD).Methods: Correlation of kidney deficiency and blood stasis and invasion of PD is analyzed based on such points as age of onset,symptoms,location,course of disease,mechanism of molecular biology and treatment.Results: It proved that deficiency of kidney and debility of marrow is the internal condition,and blood stasis is the necessary factor.Pathogenesis of PD includes the characteristic of kidney deficiency and blood stasis,deficiency correlated to stasis.In addition,Chinese herb formula with function of strengthening kidney and activating circulation shows the anti-brain-aging and neuroprotective effect to PD.Conclusion: Kidney deficiency and blood stasis is the fundamental pathogenesis of PD.
10.CT Observation of the Medial Soft Tissue ofStyloid Process in Nasopharyngeal Carcinoma after Radiotherapy

Objective To improve the cognition of abnormal changes of the medial soft tissue of styloid process (MSTSP) in nasopharyngeal carcinoma after radiotherapy. Methods CT scans of nasopharynx in 39 cases with NPC that had change at the MSTSP in NPC after radiotherapy were performed.CT findings of recurrence and non-recurrence at MSTSP were studied and compared carefully.Results In the recurrence cases, the MSTSP was thickening and compactness in all cases, in the non-recurrence cases ,only 37.04% (10/27) were thickening and compactness at MSTSP. In all the cases which had thickening and compactness of the MSTSP, the borders were slightly protruding in 9 cases and the borders were straight in 3 cases in the recurrence cases, while the borders were straight in 7 cases and the borders were slightly hollow in 3 cases,and there wasn't any case appeared as protruding of borders in the non-recurrence cases.
