Objective:To investigate whether the IgG from patients with antiphospholipid syndrome(APS) is capable of stimulating blood monocytes to express tissue factor(TF) activity and to explore the mechanisms possibly concerned. Methods:Freshly isolated peripheral blood monocytes were cultured in media containing purified APS IgG or normal IgG or RP-1 [a monoclonal anti-?_2 glucoprotein I(?-?_2GPI)antibody] and/or other agonists and the TF activity on monocytes was investigated by measuring factor Ⅶa-dependent generation of factor Xa.Results:APS IgG as well as RP-1 significantly increased monocytes TF activity, comparing to normal IgG;exogenous ?_2GPI enhanced the effects as the intact IgG molecule. Conclusion:That certain antiphospholipid antibodies, mainly anti-?_2GPI antibodies, induce monocytes TF activity in an antigen-specific manner, not via Fc receptor pathway,is contributed to the thrombotic diathesis in APS.

AIM: To observe the mRNA expressions of Insulin-induced gene 2(Insig-2) and Vitamin D receptor(VDR) in HepG2 cell line and investigate the possible mechanism of berberine on regulating blood lipid. METHODS: HepG2 cells were incubated with Bet at different concentrations(1,3,10,30,100 ?mol/L),and incubated with 3 ?mol/L berberine for various times(0,12,24,48,72 h).Total RNA was extracted and the mRNA expressions of Insig-2 and VDR from HepG2 cells treated by berberine were quantified by RT-PCR.(RESULTS): After exposure to gradient concentrations of berberine for 24 h,the mRNA expressions of Insig-2 and VDR in HepG2 cells increased obviously peaked at 3 ?mol/L concentration, then the up-regulating effect declined gradually. The relative coefficient between them was(0.753)(P

OBJECTIVE:To investigate the inhibition effect of berberine on colon carcinoma lovo cell. METHODS:Colon car-cinoma lovo cells were cultured and treated with berberine and meloxicam in various concentrations. At 6 h,12 h and 24 h after treatment, the proliferation of lovo cell was assayed by MTT method. RT-PCR method and Western blot testing were used to eval-uate the mRNA and protein expressions of COX-2 in lovo cell. RESULTS:Berberine with concentration larger than 10-5 mol?L-1 and meloxicam with concentration larger than 10-4 mol?L-1 could inhibit the growth and proliferation of lovo cell and showed concentration and time dependent manners(P

Objective To observe the clinical efficacy of coupled plasma filtration adsorption (CPFA) combined with high volume hemofiltration (HVHF) for severe acute pancreatitis (SAP) and explore the practicality, the safety and the therapeutic mechanism. Methods This was a prospective, randomized clinical trail. A total of 29 patients with SAP were di-vided into two groups:treatment group (n=15, CPFA and HVHF) and control group (n=14, HVHF). The APACHEⅡscore, se-rum triglyceride, liver and kidney function, white blood cell (WBC), heart rate (HR), respiration, mean arterial pressure (MAP) and oxygenation index were compared before and after treatment between two groups. The organ function was as-sessed, and the adverse reactions and prognosis were observed in two groups. Results After treatment, there were signifi-cant decreases in APACHEⅡscore and WBC, and significant increase in PaO2/FiO2 in two groups, especially significant in treatment group. There was a significant decrease in the level of triglycerides in 7-d treatment group than that of 3-d treat-ment group (P<0.05). There were no thrombocytopenia, bleeding, allergies, blood clotting and other adverse reactions in two groups of patients. There were significant decreases in the blood purification time and the length of hospital stay in treatment group than those of control group (P<0.05). There was no significant difference in mortality between two groups. Conclu-sion CPFA combined with HVHF can effectively control inflammation, reduce the serum level of triglyceride and improve the prognosis in patients with SAP.

Mortality is a key index for assessing technical level and treatment quality of hospital. We filtered and analyzed the literatures about the influence of nurse factors on patient mortality out of China in recent years to guide the study within China. We found the nurse factors impacting mortality included workload, configuration of nursing staff, education status and working environment. Those findings point out the direction of the research on relationship between nurse factors and patient mortality in China, which has great significance.

AIM:To observe the changes in heme oxygenase-1(HO-1) expression in the lung after ischmia-reperfusion of hind limbs in rats.METHODS:Hind limbs ischemia was made by clamping infrarenal aorta with a microvascular clip and lung injury was made by following reperfusion. Lung tissue was obtained from the animals subjected to sham operation, 4 h ischemia without reperfusion and 4 h, 8 h, 16 h, 24 h, 48 h reperfusion following 4 h ischemia. The levels of HO-1 mRNA and protein were measured at different times by Northern blot and Western blot. Immunohistochemistry technique was used to determine the cell types responsible for limb ischemic reperfusion induced HO-1 expression. RESULTS:After ischemia-reperfusion of limbs, HO-1 mRNA increased by 4 h, reached a peak at 16 h, and returned toward baseline at 24-48 h. This time course correlated with increased HO-1 protein. Immunohistochemical studies showed HO-1expressed in a variety of cell types, including the airway epithelium, alveolar macrophages and vascular smooth muscular cells. There were no positive signals in sham group and ischemia group both in mRNA levels and protein levels. CONCLUSION:The expression of HO-1 in the lung is not induced by limb ischemia or sham operation, but induced by limb reperfusion after ischemia in rats.

The clinical ability of employees in laboratory medicine refers to their ability to give useful information and helps for clinical diagnosis and treatment. In laboratory medicine education, the cultivation of the undergraduates'clinical ability should be strengthened.Case teaching and clinical signs analysis is practical to the cultivation of clinical ability. In addition,we should carry out the reform of the teaching materials in order to adapt to the requirements of the clinical ability training. Improving the clinical ability should be the students' a lifelong career planning.
