1.The roles of hypoxia inducible factor-1 in apoptosis after cerebral ischemia

Hypoxia-inducible factor-1 (HIF-1) is an inportant regulatory factor in adapting to hypoxia in the hypoxic environment for cells.The changes of the numbers of HIF-1 in cells may regulate hypoxia-responsive gene transcription.HIF-1 may inhibit neuronal apoptosis and thus play a ncuroptotective role,and may also show a neurotoxic effect by inducing neuronal apoptosis.
2.Construction of TH-GDNF vector and its expression in a dopaminergic cell line MES23.5

Objective To construct a vector carrying tyrosine hydroxylase (TH) and glial cell line-derived neurotrophic factor (GDNF) in order to establish a new gene therapy method in Parkinson's disease. Methods Human TH gene fragment from the plasmid pWAV2-TH was cloned into pIRES to construct pIRES-TH. The mouse GDNF gene, amplified by PCR was inserted into pIRES-TH to construct pIRES-TH-GDNF. Restriction analysis and nucleotide sequencing were used to confirm the structure of pIRES-TH-GDNF. Then MES23.5 cells were transfected with this eukaryon vector using Lipofectamine TM2000. The expression of TH and GDNF in mRNA and protein levels were detected by RT-PCR and immunofluorescence after G418 selection. Results The 2 objective fragments were inserted into pIRES correctly. RT-PCR and immunofluorescence results showed that TH and GDNF were highly expressed in MES23.5 cells. Conclusion The plasmid pIRES-TH-GDNF is constructed successfully and can express TH and GDNF in vitro.
3.The renal damage in the neonatal hyperbilirubinemia and the effects of early treatment

Objective To investigate the renal function damage in newberns ruth hyperbilirubinemia and the effects of early treatment. Methods 100 newberns with hyperbilirubinemia were taken as treatment group. Ser-um bilirubin, malondialdehyde (MDA), β_2-MG, and urine-minim protein (β_2-MG, α_1-MG, ALB) were measured within 24 hours after charge in. 50 healthy newborns had been chosen as controls. Anti-oxidate (vitamin E and Glu-tathion) as weel as regular method were given to the treatment group. All the above biochemical indexes were tested in the 3td and 6th day. Results When bilirubin level was in 205.0~256.5 μmol/L, β_2-MG、α_1 -MG in urine and MDA in blood were higher in treatment group than in control group(P < 0.05);when unconjugated bilirubin(UCB) was in in 256.6~342.0 μmol/L, β_2-MG was also raised (P < 0.01, P < 0.05);and when UCB greater than in 342.0 μmol/L, serum β_2-MG and urine ALB raised significantly (P <0.01,P <0.05). After early treatment, bil-irubin decreaed, and, β_2-MG, urine-minim protein declined to normal level, with a faster recovery in treatment group than control group (P < 0.05). Conclusions Hyperbilirubinemia may damage the glomerular filtration and renal tubule's re-absorption function in neonatals. Lipid peroxidation activated by UCB in Hyperbilirubinemia may cause thease damages. Antioxidant combined with regular treatment could lead better results.
4.Evaluation of the application effect of cleaning packaging of surgical medical apparatus and instruments in sterilization and supply center of hospital

Objective To investigate the application effect of cleaning packaging of surgical medical apparatus and instruments in sterilization and supply center of hospital. Methods The first-time qualification rate of cleaning,packaging and sterility test and incidence rate of nosocomial infection before and after the application of cleaning packaging of surgical medical apparatus and instruments in sterilization and supply center of hospital.The application effect was comprehensively evaluated. Results The first-time qualification rate of cleaning,packaging and sterility test after the application of cleaning packaging of surgical medical apparatus and instruments in sterilization and supply center were significantly increased.The incidence rate of nosocomial infection was 1.5% before and 5.5% after the application of cleaning packaging of surgical medical apparatus and instruments,the difference was significant. Conclusions Application of cleaning packaging of surgical medical apparatus and instruments in sterilization and supply center can effectively control the cleaning and packaging quality,guarantee the thoroughness of subsequent disinfection and sterilization,decrease the incidence rate of nosocomial infection caused by poor quality of surgical medical apparatus and instruments.It creates reliable medical environment for patients and obtain good social and economic benefit,which is worthy of wide application.
5.The effects of serum uric acid on expression of APP and BACE1 in rats

Objective To observe the effects of the different serum uric acid levels on expression of Alzheimer’s disease biomarkers (APP and BACE1) in rats. Methods Intraperitoneal injection of oxygen of oxazine acid potassium was used to produce HUA models in rats. H&E staining was used to detect the morphological changes of the hippocampus. Western blot was used to detect the protein levels of APP and BACE1 of the hippocampus. Results Compared with nor-mal control group, the serum uric acid and the protein levels of APP and BACE1 in the hippocampus was obviously in-creased at OAPS treatment group (P<0.01). Compared with low dose OAPS treatment group, the serum uric acid level was significantly increased whereas the protein levels of APP and BACE1 in the hippocampus were significantly decreased at the middle and high dose group (P<0.01). Compared with middle dose group, the serum uric acid level was increased at high dose group (P<0.05) and the expression of APP were decreased in the hippocampus rats but the expression of BACE1 remained unchanged (P>0.05). Conclusion The higher level of serum uric acid may be a protective factor of AD. The higher serum uric acid levels, the lower the risk of AD.
6.Production of single chain antibody-alkaline phosphatase fusion protein

Objective:Making single chain antibody(scFv)-alkaline phosphatase(Ap)fusion protein.Methods:An expression vector pSTE2-C66-Ap was constructed by sequentially inserting the Ap coding region into plasmid pSTE2-8E5 and replacing the VH-VL fragment of 8E5 by VH-VL fragment of C66.The fusion protein scFvC66-Ap was expressed in E.coli.and analysed by SDS-PAGE and immunoblotting.Results:Have obtained the scFvC66-Ap fusion protein with a molecular weight of 75 kD.It bound a 60 kD molecule from KG1a cell proteins on immunoblotting membrane detected directly by Ap enzymatic activity.Conclusion:A method permits the production of scFv-Ap conjugates in E.coli.which can replace conventionally prepared Ap-labeled antibodies in immunoassays.
7.A Comparison of Three Diagnostic Criterions for Metabolic Syndrome Applied in Population of Xilinhaote City

Objective To compare the differences of three diagnostic criterions for metabolic syndrome (MS) proposed by the International Diabetes Federation (IDF) in 2005, the Third Report of National Cholesterol Education Program-Adult Treatment Panel Ⅲ (NCEP-ATPⅢ) in 2005 and Chinese Diabetes Society(CDS)in 2004 in population of Xilinguole city in inner mongolia. Methods 1 060 employees aged more than 20 yrs in enterprises taken health examination in Xilinguole hospital were investigated by epidemiological survey and the prevalence rates of MS and its components were calculated according to these three definitions respectively, and compared the agreement rates and difference between these definitions. Results The age-adjusted MS prevalence rates were 25.0%, 27.9% and 22.5% according to IDF (2005), ATPⅢ (2005) and CDS (2004) respectively. The agreement rate of diagnosis between IDF and ATPⅢ was the highest, and the Kappa value was 0.929. The kappa values between IDF and CDS,ATPⅢ and CDS were 0.726 and 0.763, respectively. There were 4.3% and 10.8% subjects respectively without MS diagnosed by IDF and CDS criterion presented at least 3 risk factors respectively. For Mongolia nationality people, when the waist circumference was 90cm in male and BMI was 25kg/m2 in female, the sensitivity and specificity were relative better, and the distance of ROC curve was the shortest, but for Han nationality, BMI≥25 kg / m2 in male and waist circumference ≥80 cm was better. Conclusion ATPⅢ definition can screen out the highest prevalence of MS and risk factors aggregation among three definitions, and it's best for screening for the high risk population of cardio-cerebrovascular diseases. The cutoff value of waist circumference (WC) in IDF and ATP Ⅲ seems better for Mongolia nationality men; while the cutoff value in CDS was better for Han nationality men. We should combine waist circumference and body mass index to judge the condition of obesity.

9.Vitamin D and the recurrence of rheumatoid arthritis

Objective To identify the correlation between vitamin D prescription and recurrence in rheumatoid arthritis (RA).Methods A randomized controlled trial study was performed in 170 RA patients who were in remission during the past 2 months.According to the level of vitamin D,170 RA patients were divided into normal vitamin D group (84 patients) and deficiency vitamin D group (86 patients).Deficiency vitamin D group were randomly allocated to receive vitamin D treatment(vitamin D treatment group) or without vitamin D treatment (control group).In the 6-month follow-up period,the recurrence status was observed and compared.Results In the 6-month follow-up period,the recurrence rate of RA in normal vitamin D group was 16.7%(14/84),in vitamin D treatment group was 19.0%(8/42) and in control group was 29.5%(13/44),and there was no significant difference (P > 0.05).In vitamin D treatment group,no hypercalcemia and hyperphosphatemia occured.The age,course of disease and remission time in recurrence patients of three groups were no significant difference (P > 0.05).The level of vitamin D in recurrence patients of vitamin D treatment group was higher than that in recurrence patients of control group:(25.5 ± 8.9) ng/L vs.(20.9 ± 8.6)ng/L,and there was significant difference(P< 0.05).Conclusion Vitamin D deficiency is not identified to be a risk factor for RA recurrence.Vitamin D does not reduce the recurrence of RA.
10.CD4+CD25high regulatory cells in peripheral blood of NSCLC patients.

The proportion and changes of CD4+CD25high regulatory T cells (Trs) in peripheral blood of non-small cell lung cancer (NSCLC) patients were analyzed and their clinical significance explored. The peripheral blood was collected from 61 patients with NSCLC and 15 healthy controls. By using monoclonal antibodies, the blood samples were evaluated with the flow cytometry for lymphocyte subsets (CD3+, CD4+ and CD8+) and CD4+CD25high Tr cells. The results showed that the proportion of CD4+CD25high Tr cells in NSCLC group was significantly higher than in control group [(4.36 +/-2.07) % vs (2.04+/-1.03) %, P<0.01]. The proportion of CD4+CD25 high Tr cells in late stage was higher than that in early stage [stages I +II (2.26+/-0.6) %; stage III (3.28+/-1.38) %; stage IV (6.06 +/-4.08) %] (P<0.05). Kaplan-Meier survival analysis revealed that the prognosis of the patients who had higher proportion of CD4+CD25high Tr cells in peripheral blood was worse (P=0.0026). In conclusion, the relative increase in CD4+CD25high Tr cells in peripheral blood may be related to immunosuppression and tumor progression in patients with NSCLC. This finding suggests that CD4+CD25+high Tr cells in peripheral blood of NSCLC may be positive for prognosis analysis. The use of depletion of the CD4+CD25high Tr cell therapy to treat NSCLC patients may be an effective strategy.
