1.Determination of the Content of Caffeic Acid in Chanhou Tieshijin by HPLC

Objective To establish HPLC method for the determination of the content of caffeic acid in Chanhou Tieshijin. Methods The determination was performed on HangBang C18 column (200 mm?4.6 mm, 5 ?m). The mobile phase was methanol-buffer phosphate (23∶77) at a flow rate of 1.0 mL/min. The UV detection wavelength was set at 323 nm and the sample size was 10 ?L. Results The linear range of caffeic acid was 0.052~0.26 ?g (r=0.999 8), with average recovery rate of 97.44% (RSD=0.78%). Conclusion The established method is simple, rapid and accurate, and suitable for the quality control of Chanhou Tieshijin.
2.The value of anti-deiminated recombinant rat filaggrin antibody in predicting disease activity of rheumatoid arthritis

Objective To investigate the value of deiminated recombinant rat filaggrin antibodies (ArFA)in predicting the activity of rheumatoid arthritis (RA).Methods ArFA was detected with enzyme linked immunosorbent assay(ELISA)in 61 patients with active RA and 48 patients with inactive RA.The ESR,CRP,RF were measured,while tender ioint count,swollen joint count and patient's general assessment were recorded.The level of ArFA in 20 RA patients was measured before and after treatment.Results The level of ArFA in patients with active RA was higher compared with patients with inactive RA [(130±35)U/L cs(66±25)U/L,P=0.004 ],the level of ArFA after treatment was reduced (79.8 U/ml vs 118.2 U/ml,P=0.000).Conclusion The level of ArFA can predict disease activity of RA.
3.The relationship study on psychological contract and turnover intention of neurosurgery nurses

Objective To investigate the neurosurgery nurses turnover intention and the psychological contract status to analyze the relationship between them. Methods The Departure Will Scale and Questionnaire of Psychological Contract were used to survey 128 neurosurgery nurses from seven general hospitals in Lishui city. Results Of neurosurgery nurses turnover intention scale was (2.24±0.71) points, whose leaving intend was stronger;Psychological contract items was (4.17±0.67) points, which organization responsibility score wasbelow employees responsibility; Hierarchical regression analysis showed that the psychological contract in the organization responsibility (organization specification responsibility, organization, personal responsibility and organizational development responsibility) could predict neurosurgery nurses′ willingness to leave, which could be able to explain their willingness to leave 13.5%of the variation. Conclusions Neurosurgery nurses′willingness to leave is strong, and psychological contract is an important factors affecting the will of their departure.

Purpose:To explore the effects of stem cell factor(SCF) in combination with erythropoietin (EPO) on erythropoiesis in patients with anemia associated with non-Hodgkin's lymphoma (NHL). Objective To investigate surgical outcomes of treatment of pilon fractures of Ruedi -Allgower types Ⅱ and Ⅲ by raft screws fixation combined with bone grafting.Methods 21 patients with pilon fractures of Ruedi -Allgower types II and Ⅲ received surgery were selected.They were 15 men and 6 women,23 to 68 years of age(average 38.2 years).16 cases were closed and 5 cases open.12 cases were type II and 9 cases type III.Intervals between injury and operation ranged from 7 to 14 days,with 8.5 days on average.All of them were treated with raft screws fixation by plate type L combined with bone grafting.Results All cases were followed up for an average of 12 months(range from 8 to 30 months).Fractures healed from 16 to 24 weeks,with 18.2 weeks on average.According to the Burwell -Charnley radiological criteria,anatomical reduction was achieved in 17 cases,and fair reduction in 4.According to the Mazur clinical criteria,ankle functions were rated as excellent in 8 cases,good in 10 cases,fair in 2 cases and poor in 1 case.The rate of excellent and good was 85.7%.Conclusion Raft screws fixation combined with bone grafting are effective for the pilon fractures of Ruedi -Allgower types II and Ⅲ,preventing articular surface collapse and leading to anatomic reduction,and has stable fixation and early functional rehabilitation.
6.Inhibitory effects and mechanism of arsenic trioxide on the growth of melanoma B16 cells

Aim The present study was designed to investigate the growth inhibition and anti-angiogenesis of arsenic trioxide (As 2O 3) on B16 cells xenografts in C57BL/6J mice and observe the effects of As 2O 3 on cell proliferation, cell morphology, cell cycle and apoptosis in vitro. Methods Mice melanoma cell line B16 was transplanted into C57BL/6J mice. The weight of tumor and percent of tumor development were examined after As 2O 3 intraperitoneal administration. HE staining and immunohistochemistry for Ⅷ-R Ag were performed to detect the microvessel density in tumor tissues. CellTiter 96 Aqueous One was used to determine the cell proliferation. Giemsa and Feulgen staining were used to observe the morphological changes of the cells. Cell cycle and apoptosis were analyzed by flow cytometry (FCM). Results As 2O 3 significantly inhibited the tumor growth in vivo, with an inhibitory rate of 81.61%. The microvessel density in tumor tissues was obviously reduced after treated with As 2O 3. In order to examine the strong anticancer action and low toxicity of Trichostatin A (TSA), the effect of TSA was examined on the growth inhibition, acetylation of histone H3 and apoptosis in HL-60 cells by employing MTT, immunocytochemical techniques, and Annexin-V-FITC/ PI assay. Our results showed that TSA could inhibit proliferation of HL- 60 cells in a time- and dose-dependent manner, and the IC50 at the 36th h was 100 ng/ml. The apoptosis-inducing effect of TSA on HL-60 cells was also time- and dose-dependent. But it didn't demonstrate apparent apoptosis induction in NPBMNCs within specific dose and time range. Both of the acetylation of histone H3 in HL-60 cells and NPBMNCs increased significantly (P<0.05) after treated with 100 ng/ml TSA for 4 h. However, there was no significant differences between the two groups (P>0.05). It is concluded that TSA can inhibit growth and induce apoptosis of HL-60 cells in a time- and dose-dependent manner, and is able to selectively induce apoptosis in HL-60 cells but does not respond in NPBMNCs under the same conditions. The difference of TSA between HL-60 cells and NPBMNCs can't be explained by the regulation of histone acetylation.

8.Over-expression of human ribonuclease inhibitor suppresses invasion and migration of transplanted bladder cancer

Objective To investigate the effect of over-expression of human ribonuclease inhibitor suppresses invasion and migration of transplanted bladder cancer .Methods The T24 cells were stably transfected with pIRES2-EGFP-RI and pIRES2-EG-FP plasmid respectively .Using the cell transfected with pIRES2-EGFP and untransfected cell as controls .and the positive clones were screened by G418 ,respectively ;Tumor cells of the three groups at 2 × 106 were respectively injected into the back of BALB/C nude mice to establish the xenograft models .Change of micro-blood vessels in tumor tissue and expression of CD31 were detected by Immunohisto-chemical and HE staining .Immunohisto-chemical assay was used to detect the expression of RI ,MMP-2 ,MMP-9 ,E-cadherin ,N-cadherin ,Vimentin ,Snail ,Slug ,Twist in the tumors .Results Animal experiment showed that the T24-RI cells group significantly inhibited the growth of bladder cancer compared with the other two control groups .Compared with the T24 and T24 vector cells groups ,the microvessel density in tumor tissue of T24-RI group was notably reduced and the expressions of MMP-2 , MMP-9 ,N-cadherin ,Vimentin ,Snail ,Slug ,Twist significantly were decreased simultaneously ,while the expressions of RI and E-cadherin were increased .Conclusion up-regulation RI can inhibit the growth of transplanted bladder cancer in nude mice by decrea-sing the expression of invasion protein and EM T protein .
9.COX-2 involved in the antitumor effect of valdecoxib on Lewis tumor

Objective To study the inhibitory effect of valdecoxib on Lewis tumor and the potential relationship with cyclooxygenase-2(COX-2).Methods HE staining was used to observe lymphocyte infiltration in tumor tissue and the cell structure of the stomach and colon.Western blotting was used to detect the expression of COX-2 in tumor tissue.PGE_2 ELISA kit was used to detect the content of PGE_2 in tumor tissue.Results ① Valdecoxib inhibited the growth of the tumor,and the survival rate was increased.②There was lymphocyte infiltration in treatment group and the content of PGE_2 was decreased.③Valdecoxib did not affect cell structure of stomach and colon,bleeding and clotting time.Conclusion Valdecoxib inhibits the growth of the Lewis tumor and enhances the survival rate.The effect of valdecoxib is related to the inhibition of COX-2.
10.Apoptosis was involved in the antitumor effect of valdecoxib on Lewis cancer

Aim To evaluate the effect and the mechanism of valdecoxib on Lewis cancer. Methods Western blot was used to detect the expression of VEGF, MMP-2, Bax, Bcl-2 and Caspase-3 in tumor tissue. Flow cytometry was used to observe the effect of valdecoxib on proliferation, apoptosis and the cell cycle distribution. MTT assay was used to observe the lymphocyte transformation rate. Results ① Valdecoxib inhibited the growth of the tumor and increased the survival rate. ② 10~40 mg?kg -1?d -1 Valdecoxib increased the apoptosis rate from 19.1% in control group to 23.1%~29.1%, and did not affect the distribution of cell cycle. ③ The expression level of Bcl-2 was decreased and expression levels of Bax, COX-2, MMP-2, Caspase-3 and VEGF were not affected. ④ Valdecoxib did not affect the weight, the lymphocyte transformation rate, spleen index, and thyrus index of the mice. Conclusion The inhibitory effect of valdecoxib on Lewis tumor is associated with apoptosis.
