1.Effect of inhalation of enflurane in early pregnancy on cognitive function in offsprings of rats

Objective To investigate the effect of inhalation of enflurane in the early pregnancy on the cognitive function in the offsprings of rats. Methods Thirty 8-10 day pregnant SD rats were randomly divided into 3 groups ( n = 10 each): control group (group C), 4 h inhalation of enflurane group (group E1 ) and 8 h inhalation of enflurane group (group E2). Group E1 and E2 inhaled 1.7% entlurane (in O2 2 L/min) for 4 and 8 h respectively, while group C inhaled oxygen 2 L/min for 8 h. The learning and memory functions of the offsprings were assessed at 20 and 30 days after birth by Morris water maze test. Results Compared with group C, the escape latency was significantly prolonged, the frequency of crossing the original platform was significantly decreased and the staying time at the original platform quadrant was significantly shortened at 3-5 days after the test in group E1 and E2 ( P < 0.05). There was no significant difference in the indexes metioned above between group E1 and E2 ( P > 0.05). Conclusion Inhalation of enflurane in the early pregnancy can result in cognitive dysfunction in the offsprings of the rats.
2.Effects of polysaccharide sulfate on the growth of vascular endothelial cell cultured in vitro and the adhesion of vascular endothelial cell to polymorphonuclear neutrophils

Objective To investigate the effects of polysaccharide sulfate(PSS)on the growth of vascular endothelial cell cultured in vitro and the adhesion of vascular endothelial cell to polymorphonuclear neutrophils(PMN).Methods ECV-304,a kind of human umbilical vein endothelial cells,was cultured in vitro with different concentrations of PSS(0,50,100,150,200,250 g?mL-1),the growth of ECV-304 was observed by means of MTT.Hypoxia/ reoxygenation(H/R) model of ECV-304 was established,the adhesion rate of PMN from patients with cerebral infarction to cultured ECV-304 was detected with or without treatment of PSS and H/R.Results The growth of ECV-304 in PSS-treated subgroups(final concentrations: 50,100,150,200 g?mL-1) were much better than in non-PSS subgroup,especially in subgroup with PSS final concentration of 100 g?mL-1.The adhesion rate of cultured ECV-304 to PMN from patients with cerebral infarction was higher significantly than that from the healthy subject,which was increased when ECV-304 being treated with H/R.The adhesion rate of cultured ECV-304(whether treated with H/R or not) to PMN was decreased significantly when ECV-304 being treated with PSS(100 g?mL-1).Conclusion The experiments in vitro showed that PSS could promote the growth of cultured ECV-304 and decrease the adhesion rate of H/R ECV-304 to PMN.
3.Relationship between apolipoprotein E ε4,ε2 alleles and intrauterine growth

Objective To investigate the relationship between apolipoprotein E (ApoE) ε4,ε2 alleles and intrauterine growth.Methods ApoE genotypes of 1418 people born in Peking Union Medical College Hospital were analyzed,allele frequencies were calculated and their parameters at birth were collected.To compare ApoE ε4,ε2 alleles with parameters at birth through single factor analysis and Logistic regression analysis.Results The alleLic frequencies of e2,83 and 84 were 8.11%,83.39% and 8.50% in the group.The results of single factor analysis showed that there was significant difference between the distribution of ponderal index (PI) in the ApoE ε2 allele group(χ2=4.87 ,P=0.027).While there was no significant difference between the distribution of head circumference at birth,placental weight and gestational age in the ApoE ε2 allele group.ApoE ε2 allele showed negative correlation with small PI in the logistic regression analysis(χ2=5.077 ,P=0.024),after adjusted for gender,age,head circumference at birth,placental weight,gestational age,parity and maternal age at delivery.No association between ApoE ε4 allele and parameters at birth was found.Conclusions ApoE ε2 allele may have protective effect on PI.No association was found between ApoE ε4 allele and intrauterine growth.

Minocycline is the second generation semisynthetic tetracycline antibiotics.A variety of animal models and clinical trials have shown that it has neuroprotective effect.Its mechanism is associated with inhibiting apoptosis, alleviating inflammatory reaction, reducing infarct volume, and alleviating vascular injury.This article reviews the neuroprotective effect of minocycline in preclinical phase and early clinical trials of acute ischemic stroke.
5.Determination of ?-schizandrin in Shuangjia Wuling Capsules by RP-HPLC

6.Clinical significance of changes of cardiac troponin T and other biochemical markers in acute ische-mic stroke patients

Objective Through observing the change of serum cTnT level and other biochemical markers such as T3 and TNF-αin acute ischemic stroke patients , to investigate the correlation between the indictors change and short-term prognosis and stroke severity .Methods This study observed 68 patients (male 35 cases ,mean age 68 ±11.4 years; female 33 cases, mean age 69 ±10.3 years).All cases were collected fasting blood sample to test cTnT ,T3 and TNF-α.Death and other unexpected events were recorded in the form of telephone follow-up within one month in survival patients during and discharge from hospital.Results According to cTnT detection level ,all cases were divided into two groups , normal group (54 cases,79.4%) and positive group(14 cases,20.6%).There was statistical significance between posi-tive cTnT and NIHSS score neurological deficits ( p <0.01).cTnT was negratively correlated with T3( r =-0.324 , P <0.05 ) ,but positively with TNF-α( r =0.67 , P =0.017 ) .All patients were followed up for one month.14.7%patients died including 11.7%during hospital and 3% after discharge.After age, NIHSS adjustment cTnT remains an independent risk factor for death ( RR=2.34 ,95%CI=1.22-5.02 , P<0.01 ) .Conclusion After acute ischemic stroke , cTnT is correlated with dropping T 3 level and in-creasing TNF-αlevel, suggesting that both stress and inflammatory response may be involved in heart dam-age.Abnormal elevation of cTnT influenced short-term prognosis of acute ischemic stroke , which can be used as short-term indicators of poor prognosis in the clinical observations .
7.The association of duration of hypertension and changes in cognitive function in hypertension patients

Objective To investigate the relationship between duration of hypertension and cognitive function in adult hypertension patients.Methods A total of 224 subjects with normal blood pressure were enrolled in group A,and 1 296 patients with poorly controlled hypertension were further divided by the duration of hypertension into group B (≤5 years),group C (6-10 years),group D (11-20years) and group E (> 20 years).Face-to-face surveys were conducted in all the subjects by trained physicians using Mini-Mental-State-Examination (MMSE) and Clock-Drawing-Test (CDT).The incidences of cognitive impairment were compared among the five groups and the relationship between duration of hypertension and cognitive function were analyzed by SPSS 18.0 software.Results (1) Compared with the normotensive group,the hypertensive group performed worse in the scores of MMSE,CDTs,the memory type cognitive function and the non-memory type cognitive function (MMSE:Z =-2.585,P =0.010;C DTs:Z =-3.689,P < 0.001 ; memory type cognitive function:Z =-2.718,P =0.007 ; non-memory type cognitive function:Z =-1.994,P =0.046).(2) The incidences of cognitive impairment in the five groups were 3.6% (8/224),6.6% (26/393),16.6% (72/433),19.7% (55/279) and 33.5%(64/191),respectively.(3) Compared with the group A,the cognitive function was significantly worse in the group E (MMSE:Z =61.314,P < 0.001 ; CDTs:Z =44.642,P < 0.001 ; memory type cognitive function:Z =35.703,P < 0.001 ; non-memory type cognitive function:Z =54.440,P < 0.001).Conclusions Hypertension is a risk factor for the cognitive dysfunction.The incidence of cognitive dysfunction and the severity are positively associated with the duration of hypertension.In those with hypertension over 20 years,the cognitive dysfunction occurs much more obviously.

Objective To investigate the effect of inhalation of enflurane on the expression of N-methyl-D-aspartate receptor subunit 2B (NR2B) in the hippocampus of the offsprings of rats.Methods Thirty SD rats pregnancy 8-10 day weighing 200-250 g were randomly divided into 3 groups ( n =10 each):control group (Group C),4 h inhalation of enflurane group ( group E1 ) and 8 h inhalation of enflurane group ( group E2 ).Group E1 and E2inhaled 1.7 % enflurane (in O2 2 L/min) for 4 and 8 h respectively,while group C inhaled oxygen 2 L/rin for 8 h.The learning and memory functions of the offsprings were assessed at 20 and 30 days after birth by Morris maze test.The expression of the NR2B mRNA were examined by RT-PCR,NR2B protein were examined by mmunohistochemistry.Results Compared with group C,the escape latency was significantly prolonged,the frequency of crossing the original platform was significantly decreased,the staying time at the original platform quadrant was significantly shortened at 3-5 days after the test in group E1 and E2 (.P < 0.05 ),the expression levels of NR2B mRNA and protein were significantly decreased at 20 and 30 days after birth in group E1 and E2 ( P < 0.05).There were no significant differences in the indexes mentioned above between groups E1 and F2 ( P > 0.05).Conclusion Inhalation enflurane in the early pregnancy can result in cognition dysfunction through inhibiting NR2B expression in the hippocampus of the offsprings of the rats.
9.MiR-24 improves beta-like globin gene expression through targeting Sp1.

We studied the function and mechanism of miR-24 in regulating beta-like globin gene expression. We first detected the expression of miR-24 during erythroid differentiation and also detected the globin gene expression in miR-24 overexpressing K562 cells through q-PCR. Dual-luciferase reporter assay and Western blotting were used to identify target genes of miR-24. "Rescue experiment" was further used to investigate the regulation of miR-24 on globin gene expression whether depending on targeting Sp1 or not. We found that miR-24 increased during hemin-induced K562 cells and EPO-induced HPCs (hematopoietic progenitor cells) erythroid differentiation. Overexpression of miR-24 in K562 cells promoted the epsilon- and gamma-globin gene expression during hemin-induced erythroid differentiation through targeting the negative globin regulator Sp1. These results suggested that miR-24 can improve the expression of beta-like globin gene through targeting Sp1.

10.Minimally invasive percutaneous anterior column screwing assisted by blocking screws for pelvic and acetabular fractures

Objective:To investigate the feasibility and clinical efficacy of percutaneous anterior column screwing assisted by blocking screws for pelvic and acetabular fractures.Methods:A retrospective analysis was conducted of the 13 patients who had been admitted from July 2019 to April 2022 for pelvic and acetabular fractures. There were 8 males and 5 females with an age of (49.1±13.3) years, 7 acetabular fractures (6 on one side and 1 on both sides; by the Letournel-Judet classification: 5 anterior column fractures on 6 sides, and 2 transverse and posterior wall fractures on 2 sides), and 6 pelvic fractures (5 complicated with pelvic posterior ring fracture; by the Tile classification: 1 case of type B2, 3 cases of type C1, and 2 cases of type C2). According to the anatomic zones of the anterior column, 5 fractures were at zone Ⅲ, 3 ones at zone Ⅳ, and 6 ones at zone Ⅴ. The time from injury to surgery ranged from 3 to 14 days, averaging (8.2±2.9) days. Anterograde anterior column screwing assisted by blocking screws was performed for all the 13 patients; the posterior ring was fixated with percutaneous sacroiliac joint screws for the 5 patients complicated with pelvic posterior ring fracture. The surgical time, intraoperative fluoroscopy frequency, and intraoperative bleeding volume for insertion of anterior column screws, fracture reduction quality, and hip joint function at the last follow-up were recorded.Results:A total of 14 anterior column screws were inserted percutaneously in the 13 patients. For insertion of anterior column screws, the surgical time was (65.0±10.2) min, the intraoperative fluoroscopy frequency (63.5±14.5) times, and the intraoperative bleeding volume for each screw less than 30 mL. All the incisions healed primarily after surgery, without such complications as iatrogenic neurovascular injury or poor wound healing. All the 13 patients were followed up for (11.1±2.2) months after surgery. In the patient with bilateral acetabular anterior column fractures for which 2 anterior column screws had been inserted, one screw had to be removed due to its displacement at 1 month after surgery; no such complications as loosening of internal fixation or fracture re-displacement was found in the other patients. All fractures healed after (10.2±2.1) months. According to the Matta scoring for quality of fracture reduction, 7 sides were excellent, 5 sides good, and 2 sides poor; according to the Majeed scoring for the 6 patients with pelvic fracture at the last follow-up, the efficacy was rated as excellent in 4 cases and as good in 2 ones; according to the modified Merle d'Aubigné & Postel scoring for the 7 patients with 8 acetabular fractures at the last follow-up, the efficacy was rated as excellent in 4 hips, as good in 3 hips, and as fair in 1 hip.Conclusion:For pelvic and acetabular fractures, minimally invasive percutaneous anterior column screwing assisted by blocking screws can result in fine clinical efficacy, in addition to its easy procedures, safety and reliability.
