2.Expression and significance of matrix metalloproteinase-2 and basic fibroblast growth factor in human glioma

Objective To explore the expression and significance of matrix metalloproteinase-2 (MMP-2) and basic fibroblast growth factor (bFGF) in human glioma.Methods The expression levels of bFGF and MMP-2 in 56 cases of human glioma and 9 cases of normal brain tissue specimen were detected by immunohistochemistry technique.Furthermore,the correlation between the expression of the bFGF and MMP-2 was compared.Results In normal brain tissue,grade Ⅰ,Ⅱ and grade Ⅲ,Ⅳ,the positive expression of bFGF were (9.51 + 2.69)%,(24.78 ±- 7.57 )%,(41.23 ± 12.35 )%,respectively,and MMP-2were (7.18 ± 2.12)%,(21.56 ± 3.85 )%,(54.32 + 7.32)%,respectively.The differences were significant among them (P <0.05).bFGF and MMP-2 were positive correlated (r =0.71,P <0.01).Conclusions There is an intrinsic correlation between the expression of the bFGF and MMP-2 and histological grade of human gliomas.These findings suggest that the synergistic interaction of bFGF and MMP-2 play an important role during the tumor invasion.
3.Effects of polysaccharide sulfate on the growth of vascular endothelial cell cultured in vitro and the adhesion of vascular endothelial cell to polymorphonuclear neutrophils

Objective To investigate the effects of polysaccharide sulfate(PSS)on the growth of vascular endothelial cell cultured in vitro and the adhesion of vascular endothelial cell to polymorphonuclear neutrophils(PMN).Methods ECV-304,a kind of human umbilical vein endothelial cells,was cultured in vitro with different concentrations of PSS(0,50,100,150,200,250 g?mL-1),the growth of ECV-304 was observed by means of MTT.Hypoxia/ reoxygenation(H/R) model of ECV-304 was established,the adhesion rate of PMN from patients with cerebral infarction to cultured ECV-304 was detected with or without treatment of PSS and H/R.Results The growth of ECV-304 in PSS-treated subgroups(final concentrations: 50,100,150,200 g?mL-1) were much better than in non-PSS subgroup,especially in subgroup with PSS final concentration of 100 g?mL-1.The adhesion rate of cultured ECV-304 to PMN from patients with cerebral infarction was higher significantly than that from the healthy subject,which was increased when ECV-304 being treated with H/R.The adhesion rate of cultured ECV-304(whether treated with H/R or not) to PMN was decreased significantly when ECV-304 being treated with PSS(100 g?mL-1).Conclusion The experiments in vitro showed that PSS could promote the growth of cultured ECV-304 and decrease the adhesion rate of H/R ECV-304 to PMN.
4.CT and MRI appearances of peripheral primitive neuroectodermal tumors

Objective To explore CT and MRI features of peripheral primitive neuroectodermal tumors (pPNET) .Methods CT and MRI findings were retrospectively analyzed in 9 cases with pPNET confirmed by pathology .Of them ,8 patients underwent CT imaging ,and 5 patients underwent MR imaging .Results The lesions occurred in head and neck in 2 cases ,chest in 3 cases ,scrotum in 1 case ,right scapular region in 1 case ,and lower extremity in 2 cases .The lesions were more solitary (7/9) with round ,lobular or irregular shape .The longest diameter was between 1 .6 cm -13 .8 cm ,and the median diameter was 6 .2 cm .8 cases had blurred boundary .On CT plain scanning ,the lesions were mixed density with predominant isodensity .On MR imaging ,the lesions were slightly T1 hypointensity and heterogeneous T2 hyperintensity .Enhancement patterns were varied ,and the common way was mild‐to‐moderate heterogeneous enhancement .3 cases of them were closely associated with bone ,and showed osteolytic bone destruction . Conclusion The CT and MRI features of pPNET has lack characteristics ,CT and MR imaging have some value for localization diagnosis .
5.Treatment of open pelvic fracture concomitant with perineal laceration using external fixation combined with limited percutaneous internal fixation plus colostomy

Objective To introduce the methods and key points for the external fixation combinel with limited percutaneous internal fixation plus colostomy in the treatment of open pelvic fracture concomitant with perineal laceration.Methods Nine patients with open pelvic fractures concomitant with perineallaceration were treated by external fixator combined with limited percutaneous internal fixer plus colostomy. Data including injury details, management and outcomes were collected for comparison.Results All the patients survived and attained bony union except for two patients with local infection that was healed several days after repeated wound debridement and use of wide spectrum antibiotics. All the patients had good extremity function at the latest follow-up. Conclusion External fixation combined with limited percutaneous internal fixation plus colostomy is a reliable, safe and less invasive procedure for the treatment of open pelvic fractures concomitant with perineal open wound.
6.Effect of recombinant human erythropoietin pretreatment on PI3K-Akt-GSK-3β signaling pathway in human renal tubular epithelial cells apoptosis induced by ischemia-reperfusion injury

Objective To study the role of PI3K-Akt-GSK-3β signaling in the apoptosis of renal tubular cells after ischemia-reperfusion injury and the protective mechanism of recombinant human erythropoietin(rHuEPO). Methods The human kidney tubular epithelial cells(HK-2)were cultured in vitro in different conditions as control group with serum, ischemia-reperfusion(IR)group, LY294002 group with LY294002(AKT inhibitor)10 μmol/L 30 minutes before IR treatment, LiCl group with LiCl(GSK-33 inhibitor)20 μtmol/L 30 minutes before IR treatment, rHuKPO group with EPO 20 U/ml 30 minutes before IR treatment, rHuEPO + LY294002 group with EPO 20 U/ml and in the presence of LY294002(10 μmol/L)30 minutes before IR treatment, rHuEPO +LiCl group with EPO 20 U/ml and in the presence of LiCl(20 μmol/L)30 minutes before IR treatment. Akt, GSK-33 and caspase-3 activation were measured by Western blotting. The apoptotic ratio of HK-2 cells was measured by flow cytometry. Cell viability was detected by MTT. Results In comparison with the control group, the apoptotic ratio raised up to 15.20%±1.43%, the expression of Akt activity decreased, GSK-33 activity and caspase-3 activity markedly elevated in IR group(P<0.05). LY294002 group up-regulated the apoptotic ratio(18.20%±2.06%), decreased the expression of Akt activity, increased GSK-33 activity and caspase-3 activity, however, LiCl group down-regulated the apoptotic ratio(12.30%±0.85%), increased the expression of Akt activity, decreased GSK-33 activity and caspase-3 activity compared with IR group(P<0.05). rHuEPO group remarkably decreased the apoptotic ratio(11.10%±1.62%), increased the expression of Akt activity, decreased GSK-33 activity and caspase-3 activity compared with IR group(P<0.05). rHuEPO+LY294002 group elevated the apoptotic ratio(13.40%±1.94%), decreased the expression of Akt activity, increased GSK-33 activity and caspase-3 activity, meanwhile, rHuEPO +LiCl group down-regulated the apoptotic ratio(7.50%±1.31%), increased the expression of Akt activity, decreased GSK-33 activity and caspase-3 activity compared with rHuEPO group(P<0.05). Conclusions PI3K-Akt-GSK-3β signaling pathway is involved in HK-2 cells apoptosis induced by ischemia-reperfusion injury and rHuEPO may be used as a new therapy.
7.Effect of inhalation of enflurane in early pregtancy on the expression of NR2B in the hippocampus of offsprings of rats

Objective To investigate the effect of inhalation of enflurane on the expression of N-methyl-D-aspartate receptor subunit 2B (NR2B) in the hippocampus of the offsprings of rats.Methods Thirty SD rats pregnancy 8-10 day weighing 200-250 g were randomly divided into 3 groups ( n =10 each):control group (Group C),4 h inhalation of enflurane group ( group E1 ) and 8 h inhalation of enflurane group ( group E2 ).Group E1 and E2inhaled 1.7 % enflurane (in O2 2 L/min) for 4 and 8 h respectively,while group C inhaled oxygen 2 L/rin for 8 h.The learning and memory functions of the offsprings were assessed at 20 and 30 days after birth by Morris maze test.The expression of the NR2B mRNA were examined by RT-PCR,NR2B protein were examined by mmunohistochemistry.Results Compared with group C,the escape latency was significantly prolonged,the frequency of crossing the original platform was significantly decreased,the staying time at the original platform quadrant was significantly shortened at 3-5 days after the test in group E1 and E2 (.P < 0.05 ),the expression levels of NR2B mRNA and protein were significantly decreased at 20 and 30 days after birth in group E1 and E2 ( P < 0.05).There were no significant differences in the indexes mentioned above between groups E1 and F2 ( P > 0.05).Conclusion Inhalation enflurane in the early pregnancy can result in cognition dysfunction through inhibiting NR2B expression in the hippocampus of the offsprings of the rats.
8.Significance and alterations of p14~(ARF) gene of CDKN2A locus in gastric cancer

AIM: To investigate the significance and changes of p14 ARF gene in gastric cancer.METHODS: The tumors and gastric tissues neighboring carcinoma from 48 patients with gastric cancer were studied. The homozygous deletions, mutations, methylation of the CpG islands, and mRNA expression of p14 ARF gene were assessed by PCR, PCR-SSCP, PCR based methylation assay, and RT-PCR.RESULTS: ①The homozygous deletion rate of p14 ARF was 31 3% (15/48), and no homozygous deletions were examined in all the gastric tissues neighboring tumor. ②There were no point mutations of p14 ARF in 33 gastric cancers without homozygous deletion and in the matched gastric tissues adjacent to tumor. ③Methylation rate of the CpG islands of p14 ARF was significantly higher in gastric cancers(47.9%, 23/48) than that in gastric tissues neighboring cancer (4.2%, 2/48)( P
9.Damage control using percutaneous transhepatic biliary drainage in acute cholangitis of severe type secondary to intrahepatic choledocholithiasis

Objective To study the safety and efficacy of damage control using percutaneous transhepatic biliary drainage (PTBD) in acute cholangitis of severe type (ACST) secondary to intrahepatic choledocholithiasis.Methods The clinical data of 8 patients who received PTBD after hospital admission followed by conventional surgery for ACST when their general condition improved were retrospectively studied.Results All patients received PTBD successfully and the amount of bile drained was 100-400 ml in the first day.The general condition of these 8 patients became better after 24 h and the total bilirubin decreased for about 25-100 mmol/L after 48 h.Three patients with a platelet count of less than 20 × 109/L showed an improved count to more than 50 × 109/L 72 h after PTBD.All patients were operated at different times after the PTBD:2 received T-tube drainage,3 T-tube drainage combined with left hepatectomy,and 3 choledochojejunostomy.Seven patients recovered uneventfully,but 1 developed hepatic failure with the total billurubin rose to more than 200 μmol/L.He was discharged home with the PTBD tube.During the waiting time of 7 days to 3 months before surgery,the tubes were kept patent and no mortality or morbidity such as bleeding,bile leakage,and peritonitis occurred.Conclusions PTBD was a safe and efficacious procedure for patients who were in a serious condition with ACST secondary to intrahepatic choledocholithiasis.It was more likely to be successful as it is minimally invasive and therefore well-tolerented.It reduced the biliary pressure,relieved the ongoing sepsis,and was a good preparatory procedure before any conventional surgery.
10.Pumping performance of a new piezoelectric pump for drug delivery.

A novel double-chamber series piezoelectric pump has been presented and tested. The pump is a multi-layer circular planar structure, consisting of PMMA (polymethyl methacrylate) pump body, two PZT actuator membranes and three cantilever valves. The PZT actuators are driven at a phase difference of 180 degrees, which is equal to two one-chamber pumps running in series. The output performance depends on the geometrical parameters of the actuator membrane. The prototype pump, fabricated with the PZT membrane 0.18 mm in thickness and 50 mm in diameter of 50 mm, can deliver drug in either direct way (pumping liquid drug) or indirect way (pumping air to extrude liquid drug from a sealed container). The frequency-response characteristic of the two handling methods is of difference. The pump obtains optimum performance at low frequency for liquid as medium, and at high frequency for air as medium. For both the direct delivery and indirect delivery, the maximum flowrate achieved reached up to 220 ml/min and 35 ml/min, respectively; and the maximum backpressure obtained amounted to about 14 KPa and 21 KPa, respectively, at the applied voltage of 80 V with frequency of 20 Hz.
