1.Efficacy analysis of Jin Long Capsule (JLC) in neoadjuvant chemotherapy of breast cancer

Objective:To analyze the therapeutic efficacy and adverse reaction induced by Jin Long Capsule (JLC) combined with TEC regimen (docetaxel+epirubicin+cyclophosphamide) in neoadjuvant chemotherapy of breast cancer. Methods:A total of 64 patients who had stagesⅡtoⅢbreast cancer were divided into two groups randomly:JLC combined with neoadjuvant chemotherapy group (JLC group, n=32) and conventional neoadjuvant chemotherapy group (control group, n=32). The chemotherapy cycle was 21 d equal-ly. Evaluations of therapeutic effects were made after three cycles of neoadjuvant chemotherapy and consequent operation. Results:The effective powers (complete response+partial response) of the JLC group and the control group were 84.38% (27/32 cases) and 56.25%(18/32 cases), respectively. The difference was statistically significant (P<0.05). The main adverse effects were gradesⅡtoⅢgastrointestinal tract indisposition and myelosuppression. WBC decrease and incidence rate of nausea and vomiting were obviously low-er in the JLC group than in the control group (P<0.05). The life quality of patients in the JLC group was greatly improved than that in the control group (P<0.05). Conclusion:The application of JLC combined with neoadjuvant chemotherapy can improve therapeutic ef-ficacy and reduce the poisonous side effects of chemotherapy, thereby improving the life quality of patients.
2.Progress of sentinel lymph node detection in breast cancer

Sentinel lymph node (SLN) is the first group of lymph nodes to receive breast lymph nodes and the first group of lymph node metastasis. It can accurately predict the metastatic status of axillary lymph nodes, which is an important reference factor in the diagnosis and treatment of breast cancer. It has become the first choice for early breast cancer staging. The rapid and accurate intraoperative localization and diagnosis of SLN is of great significance for the choice of operation mode and the improvement of therapeutic effect. SLN location and diagnostic methods are still needed to be investigated. This article reviews various methods in clinic recently.
3.Study of using ?-cyanoacrylate to establish the orthotopic transplantation tumor model from the subcutaneous model of human hepatocellular carcinoma in nude mice

Objective Using ?-cyanoacrylate to establish the orthotopic transplantation tumor model from the subcutaneous model of human hepatocellular carcinomas(HCC)in nude mice. Methods The subcutaneous animal tumor model was established by transplanting BEL-7402 cell into the subcutaneous of nude mice. Then the orthotopic transplantation tumor model was established by injecting the subcutaneous transplanted tumor into liver(indirect orthotopic model). Results Using ?-cyanoacrylate to establish the indirect orthotopic model successfully and the survival rates of the transplantation tumor were 95.8 %. Conclusion It was compared with the traditional methods that the indirect orthotopic transplantation tumor model by using ?-cyanoacrylate to establish is an easy method with high successful rate and easy to spread.
4.Biological characteristics of Chinese hamster infected with Babesia

Objective To establish a Chinese hamster model of babesia infection, to find the changing pattern of organs and biochemical parameters in Chinese hamster infected with Babesia, and to promote the detection and treatment of babesiosis.Methods Healthy 5-week old Chinese hamsters were infected by intraperitoneal injection of blood containing Babesia.Blood samples were collected at 0, 2, 4, 6, 8, 10, 12, 14, 16, 23, 30, and 37 days after infection from 5 hamsters at each time point.Blood smears were prepared to detect the parasites using Giemsa staining.ELISA assay was employed to test the IL-2 concentration.The blood biochemical indexes were detected using an automatic biochemical analyzer.DNA was extracted from the whole blood and REAL-TIME RCR was performed to determine the reproduction of Babesia.Aftert the animals were sacrificed, the heart, lung, spleen, liver, and kidney were taken to analyze the changes of organ coefficients.Results The highest level of Babesia in the hamsters occurred on day 4 after the Babesia injection, and then showing a decreasing tendency.However, there was a transient increase on the 12th day after infection.The liver and spleen displayed most extensive response to the infection showing hepatomegaly and splenomegaly, but the variation of heart and kidneys coefficients was within the norm.There were prominent changes of blood cells, especially leucocytes, with two peaks at day 10 and 23 after the Babesia infection.The peak changes of blood biochemical indexes occurred at day 12 after infection.The concentration of serum IL-2 reached a peak on the 10th day after infection.Conclusions The Chinese hamsters display typical characteristics of tick-borne diseases such as hepatomegaly and splenomegaly.The immunological system is activated along with the infection and reaches a highest stage in the second week.Afterwards the Babesia can live in the hamster body for a long period of time.The results of this study provide useful information supporting further studies on the detection, treatment and prevention of Babesiosis.
5.Risk factors analysis of persistent frequency after transurethral resection of the prostate

Objective To discuss the risk of factors influencing persistent frequency after transurethral resection of the prostate (TURP).Methods The clinical data of 119 post-TURP patients treated from January 2014 to June 2015 was retrospectively analyzed.The age was (72.1 ±2.3)years old.There were 15 cases with hypertension,23 cases with diabetes and 6 cases with heart disease.The preoperative IPSS score of 119 cases was (22.1 ± 5.9) points,with (10.2 ± 1.8) points in urinary storage period and (11.8 ± 4.7) points in urination period.Urination frequency was (10.8 ± 2.6) times per day and there were (3.8 ± 0.8) times of nocturnal urination.B-ultrasound:residual urine volume was (38.1 ± 9.1) ml and prostate volume was (34.1 ± 4.2) ml.Preoperative maximum urine flow rate was (8.8 ± 3.9) ml/s.The detrusor pressure at maximum urinary flow rate was (43.9 ± 14.1) cm H2O (1 cmH2O =0.098 kPa),maximum detrusor pressure was (99.7 ± 12.2) cmH2O and effective bladder volume was (217.5 ± 14.8) ml.Contraction of bladder weakened in 40 cases (33.6%) and 36 cases (30.2%) had detrusor overactivity.According to whether continuous urinary frequency was developed,the patients were divided into frequency-positive group and frequency-negative group.The differences between the patients in two groups were compared and univariate analysis was performed.A multivariate logistic regression was performed on statistically significant indicators.Results Among the 119 patients,21 were frequency-positive and 98 were frequency-negative.Univariate analysis showed that age,IPSS score,preoperative urinary storage score,detrusor pressure at maximum urinary flow rate,maximum detrusor pressure,effective bladder volume,contraction decrease of bladder,preoperative detrusor activity were important indicators affecting the condition of postoperative urinary frequency (all P < 0.05).Multivariate analysis showed that old age (OR =3.842,P =0.021),high total IPSS score (OR =5.109,P =0.011),low maximum detrusor pressure (OR =3.477,P =0.039),low effective volume of bladder (OR =4.051,P =0.017) and detrusor overactivity (OR =3.662,P =0.025) were independent risk factors for urinary frequency after TURP.Conclusions The age,the high IPSS score before operation,low maximal detrusor pressure,low effective bladder capacity and the bladder detrusor activity could be independent predictive factors of persistent frequency after TURP.
6.In Vitro Activity of the Novel Tetracyclines, Tigecycline, Eravacycline, and Omadacycline, Against Moraxella catarrhalis

Background:
Tigecycline, eravacycline, and omadacycline are recently developed tetracyclines. Susceptibility of microbes to these tetracyclines and their molecular mechanisms have not been well elucidated. We investigated the susceptibility of Moraxella catarrhalis to tigecycline, eravacycline, and omadacycline and its resistance mechanisms against these tetracyclines.
Methods:
A total of 207 non-duplicate M. catarrhalis isolates were collected from different inpatients. The minimum inhibitory concentrations (MICs) of the tetracyclines were determined by broth microdilution. Tigecycline-, eravacycline-, or omadacycline-resistant isolates were induced under In Vitro pressure. The tet genes and mutations in the 16S rRNA was detected by PCR and sequencing.
Results:
Eravacycline had a lower MIC50 (0.06 mg/L) than tigecycline (0.125 mg/L) or omadacycline (0.125 mg/L) against M. catarrhalis isolates. We found that 136 isolates (65.7%) had the tetB gene, and 15 (7.2%) isolates were positive for tetL; however, their presence was not correlated with high tigecycline, eravacycline, or omadacycline ( ≥ 1 mg/L) MICs.Compared with the initial MIC after 160 days of induction, the MICs of tigecycline or eravacycline against three M. catarrhalis isolates increased ≥ eight-fold, while those of omadacycline against two M. catarrhalis isolates increased 64-fold. Mutations in the 16S rRNA genes (C1036T and/or G460A) were observed in omadacycline-induced resistant isolates, and increased RR (the genes encoding 16SrRNA (four copies, RR1-RR4) copy number of 16S rRNA genes with mutations was associated with increased resistance to omadacycline.
Conclusions
Tigecycline, eravacycline, and omadacycline exhibited robust antimicrobial effects against M. catarrhalis. Mutations in the 16S rRNA genes contributed to omadacycline resistance in M. catarrhalis.
