1.Effect of radical surgery for CD4+ CD25+ regulatory T cells in peripheral blood of patients with gastric cancer

Objective To investigate the efforts of CD4+ CD25+ regulatory T (Treg) cell adjustment of Treg cells proportion in peripheral blood of patients with gastric cancer before or after surgery.Methods The peripheral blood samples were collected from 10 healthy volunteers,58 gastric cancer patients 2 days before and 7 days after operation.The proportion of Treg cells in peripheral blood were detected by flow cytometry.Results CD4+ CD25+ Treg cell in peripheral blood of patients with gastric cancer increased significantly compared with that of healthy control group (1.1±0.4) % / (1.5±0.5) % (P < 0.05),and further increased after operation (1.5±0.55) % / (1.9±0.8) % (P < 0.01).Conclusion The anti-tumor activity in gastric cancer patients is inhibited and surgery may be involved immune defection to decrease this effort.

The growth associated protein(GAP 43) and neural cell adhesion molecule (NCAM) play an important role both in promoting neurite outgrowth and synaptic plasticity. GAP 43 is a neuron specific phosphoprotein that is highly expressed during the development of the central nervous system, nerve regeneration and the modulation of synaptic function, and it is frequently used as a marker for sprouting. GAP 43 may be involved in G protein interaction, membrane binding, calmodulin binding and protein kinase C phosphorylation. The various interactions, specified by the structural domains, are thought to underlie the role of GAP 43 in synaptic plasticity, participating in membrane extension during neuritogenesis, in neurotransmitter release and long term potentiation. NCAMs involve in the intracellular signaling cascades and stimulate the axonal growth, GAP 43 function is essential for NCAM stimulated neurite outgrowth and that much of this appears because of the phosphorylation of GAP 43 via the FGF receptor dependent stimulation of arachidonic acid. NCAM can accelerate the phosphorylation of GAP 43.

Misoprostol, a prostaglandin E1 analogue, have variety clinical applications. In early 80's, misoprostol is currently licensed primarily to prevent non-steroidal anti-inflammatory drug-associated gastric and duodenal ulcers. Further work shows the drugs have protection to hepatic and renal and prevention of inflammatory and allergic disorders. When combined with mifepristone,complete abortion rate is 95%. On the other hand, misoprostol is also an effective agent for cervical ripening and labor induction. It have no serious side effect, as a result, misoprostol is a drug of high effect and low toxicity, deserving popularization, but man and pregnant woman must prudent.
4.Effect and mechanism of sildenafil on rabbit corpus cavernosum in vitro

AIM To study the effect and mechanism of sildenafil on rabbit corpus cavernosum in vitro . METHOD Rabbit corpus cavernosum(RCC) segments were incubated with various concentrations of sildenafil. The formation of cGMP was stimulated with 1 ?mol?L -1 sodium nitroprusside(SNP), and the cyclic GMP concentration was measured by radioimmunoassy technology. In the organ bath, strips of RCC were precontracted with 10 ?mol?L -1 phenylephrine. Sildenafil were added with increasing doses gradually, graded relaxations were induced using various concentrations of sodium nitroprusside (SNP) and methacholine (MCH). RESULT In the presence of SNP, the cGMP concentration was increased in rabbit penile tissue, the increase of cGMP was greater when sildenafil was added. Sildenafil 10、100 nmol L -1 、1 ?mol?L -1 dose-dependently relaxed RCC strips. CONCLUSION Sildenafil increases cGMP concentration in RCC is attributed to the augmentation of NO/cGMP pathway, and the accumulation of cGMP improves the relaxation of RCC.

Object It was reported that salvianolic acid A from Salvia miltiorrhiza Bge could improve memory dysfunction of mice with cerebral ischemia reperfusion injury The effects of total salvianolic acids (Sal), including salvianolic acid A and B, rosmaric acid and protocatechic aldehyde was now studied to observe its effect on cerebral ischemia reperfusion injury in hope of obtaining a preliminary view on its mechanism of action Methods The effects of Sal on lactic dehydrogenase (LDH), superoxide dismutase (SOD) and malondialdehyde (MDA) of cerebral ischemia reperfusion injured mouse and rat models were observed Results Sal 10, 20 mg/kg iv and 5, 10 mg/kg iv could markedly increase the activities of LDH and SOD and lower the production of MDA in the injured cerebral tissue On focal cerebral ischemia reperfusion injured rat, 24 h reperfusion after 2 h ischemic injury, 5, 10 mg/kg iv Sal could decrease infarct size, ameliorate neurological deficit with obvious inhibition of the decrease of LDH and SOD activities and the over production of MDA Conclusion Sal showed obvious protective effects on cerebral ischemia reperfusion injuries of mice and rats via attenuating oxygen free radical
6.EFFECT OF GINSENOSIDE Rg1 ON LEARNING AND MEMORY IMPAIRMENT INDUCED BY β-AMYLOID PEPTIDE(25-35) AND ITS MECHANISM OF ACTION

AIM To study the effect of ginsenoside Rg1 on the learning and memory impairment in mice induced by aggregated β-AP(25-35). METHODS Mice were administered Rg1 (5, 10 mg*kg-1, ip) for 10 d and control mice received daily ip injections of saline after the intracerebroventricular injection of aggregated β-AP(25-35). After the final treatment, passive avoidance and performance in the Morris water maze (MWM) were assessed. and the activity of cortical and hippocampal ChAT and AchE were detected after the final behavior test. RESULTS Ginsenoside Rg1 (5, 10 mg*kg-1, ip) significantly ameliorated the learning and memory impairment induced by β-AP(25-35). Rg1 (5, 10 mg*kg-1) decreased the latencies and swim distances of mice to reach a hidden platform and improved the corresponding changes in search strategies occurred in the Morris water maze, and Rg1 (10 mg*kg-1, ip), increased step-through latencies also. Biochemical analysis showed that Rg1 (5, 10 mg*kg-1, ip) prevented the cortical and hippocampal ChAT activity decline induced by β- AP(25-35), and showed inhibition of the activity of AchE, although β-AP(25-35) showed no effect on the cortical and hippocampal AchE activity. CONCLUSION These data showed that ginsenoside Rg1 significantly improved the learning and memory impairment induced by β-AP(25-35), and this effect could be attibuted to its inhibition of AchE and increase of ChAT activity.

Objective To investigate the effects of divitamins notonginseng and cinnarizine capsule(DNCC) on acute cerebral ischemia.Methods ICR mice were administered three doses of DNCC(420,210,and 105mg/kg;ig.) for ten days,then the gasping time after decapitation was recorded.After three doses of DNCC(292,146,and 73mg/kg;ig.) were given respectively for ten days in rats,the effects of DNCC on the infarct size,histological changes and neurological function scores caused by focal cerebral ischemia which was induced by middle cerebral artery occlusion were investigated.Results DNCC prolonged the gasping time of mice after decapitation and improved the neurological function scores,cerebral ischemia injury and decreased the infarct size in rats.Conclusion DNCC has a protective effect against acute cerebral ischemia.
8.The relationship between NO and gossypol in decreasing sperm quality in male rats

Aim To elucidate the biochemical mechanism of gossypol in inducing the decline of sperm quality.Methods Gossypol was administered orally at the dose of 50 mg?kg-1/2 d for two weeks.Then,the sperm was collected from the left caudal epididymis and was analysed by CASA.The morphological changeand the concentration of nitric oxide(NO)in testes as well as the level of hormone〔follicle-stimulating hormone(FSH)〕,luteotrophic hormone(LH),testosterone(T)in serum were assayed.Results Gossypol could induce the decrease of sperm number and sperm quality.The concentration of NO in testes increased significantly.Among the three kind of hormone,only the concentration of T showed decrease after the oral administration of gossypol.NA1108 could antagonize the decline of sperm quality damaged by gossypol and decrease the content of NO in testes.Conclusions The concentration of NO in testes beyond normal value was one of the toxic mechanism of gossypol that contributed to the inhibition of spermatogenesis.Some drugs with the ability to reduce NO content in testes could also increase sperm quality.
9.Establishment of a patient-derived xenotransplantation model of gastric cancer and its application in pharmacodynamics study

Objective:To establish a patient-derived xenotransplantation (PDX) animal model of gastric cancer, and observe the anti-cancer effect of chemotherapeutic drugs on this model.Methods:Human gastric cancer tissues were inoculated into the subcutaneous tissues of both axillaries of NOG mice and were subcultured for 3 generations. The tumor tissues of the third-generation NOG mice were selected and inoculated into the subcutaneous tissues of left axillary of 21 severe combined immunodeficiency-non-obese diabetes mellitus (SCID-NOD) mice to establish PDX mouse model of gastric cancer. The inoculated mice were divided into control group (mice received only 0.9% sodium chloride injection), oxaliplatin group, cisplatin group, paclitaxel group, fluorouracil group, tegafur, gimeracil and oteracil porassium capsules group and capecitabine group, with 3 mice in each group, and the corresponding drugs were given. The mice survival status, tumor volume and tumor weight at different times were recorded. Mice were sacrificed on the 61st day of administration, and the tumor inhibition effects of 6 kinds of chemotherapy drugs on the PDX model of gastric cancer were evaluated.Results:After being subcultured for 3 generations, the stability of tumor transmission in PDX animal model of gastric cancer was improved, and the homogeneity of tumor growth was good at the initial stage. At the early stage of administration, the model was more sensitive to oxaliplatin, fluorouracil and capecitabine, and the tumor growth inhibition (TGI) values on the 31st day were 63.37%, 52.11% and 78.48%, at the end of administration, the model had the best sensitivity to capecitabine with a TGI value of 59.22% and a tumor inhibition rate of 58.65% on the 61st day. The TGI curve after administration showed that paclitaxel had no obvious anti-tumor effect, cisplatin had the worst anti-tumor effect, and the model had poor tolerance to tegafur, gimeracil and oteracil porassium capsules.Conclusion:The PDX animal model of gastric cancer is successfully established, and capecitabine has the best tumor suppressive effect on this model.

Objective To investigate the change of peripheral blooe T cell subsets,NK cells ane serum soluble interleukin-2 receptor(sIL-2R)concentration in patients with gastric cancer before ane after surgery,ane to uneerstane immune function status ane changes of perioperative gastric cancer patients. Methods One huneree ane thirty-five perioperative gastric cancer patients were selectee as our subjects who hospitalizee from May 2009 to May 2011 in Tumor Hospital of Shanxi Province,ane they were servee as treatment group,while 50 healthy subjects were selectee as controls. The number of CD3 + ,CD4 + ,CD8 + T cells,rate of CD4 + / CD8 +ane the NK cell ratio in blooe cells were eetectee by flow cytometry. ELIAS was appliee to measure serum sIL-2R concentration. Observee the above ineexes of control group at the same perioe,ane comparee the ineexes before operation of 1 e of the treatment group. Results The rate of NK cells,CD3 + T cells ane CD4 + / CD8 +ratio in patients at pre-operation were(10. 11 ± 3. 64)% ,(55. 60 ± 9. 61)% ,(30. 22 ± 6. 17)% ,1. 14 ± 0. 35,respectively,lower than that of control group(( 28. 39 ± 5. 81 )% ,( 68. 65 ± 7. 39 )% ,( 47. 87 ± 4. 85)% ,1. 82 ± 0. 24 respectively;t = - 5. 9,8. 6,8. 2,12. 7;P < 0. 01). CD8 + T positive rate of cells increasee from(27. 05 ± 7. 86)% to(34. 26 ± 6. 23)%(t = - 6. 5,P < 0. 01). At 14th eay after surgery,the cell immune function of the patients recoveree graeually,ane there were statistically significant eifferences in the above ineexes comparee with pre-operation(P < 0. 05). The eramatic changes were seen among patients with the late Gastric cancer TNM staging. Comparee with patients with stage Ⅳ,all above ineex were significant eifferent from that of patients with stageⅠ,Ⅱ(P < 0. 05),ane no significant eifferences was seen in patients with stageⅢ(P > 0. 05). The concentration of serum sIL-2R in patients with gastric cancer before operation was(575. 71 ± 34. 77)U/ L,higher than that of healthy persons((428. 26 ± 21. 77)U/ L,t = - 7. 9,P < 0. 01),ane serum sIL-2R levels in patients with stage Ⅲ,Ⅳ was lower than that of patients with stage Ⅰ,Ⅱ patients with low(P< 0. 05). Conclusion The immune function of patients with gastric carcinoma is relatee to tumor loae size ane eifferent pathological staging. The ineex of the ratio of NK cells ane T lymphocyte subsets,serum sIL-2R levels can be servee as ineicators for monitoring perioperative evaluation of prognosis of gastric cancer.
