1.Identification of Lycium barbarism and Sulfuring Lycium barbarism

OBJECTIVE:To establish a rapid and accurate method for the identification of Lycium barbarism.METHO-DS:The Lycium barbarism in different processing method was studied in respect of 1 000-seed weight,acidity and basicity,UV spectra and TLC etc.RESULTS:Striking differences were noted between Lycium barbarism and Sulfuring Lycium barbarism in 1000-seed weight and all the other indexes.CONCLUSION:UV and TLC were proved to be simple and reliable methods for the identification of Lycium barbarism,which thus can be considered as the quality control methods for Lycium barbarism and its Chinese patient drugs.
2.Diagnosis and treatment of breast cancer during pregnancy: a review of twelve cases

Objective To investigate the principles of diagnosis and treatment of breast cancer during pregnancy.Methods Clinical data of twelve patients with breast cancer during pregnancy admitted to Peking Union Medical College Hospital between January 1995 and October 2015 were analyzed retrospectively.Follow-up studies on the prognosis of the mothers and infants and the long-term survival were carried out,and data were analyzed with descriptive statistical analysis.Results Of these twelve patients,eleven were diagnosed with breast cancer preliminarily by ultrasound.In the remaining one who presented with paraneoplastic neurological syndrome,breast tumor was discovered by the whole-body positron emission tomography/computed tomography (PET-CT) examination after emergent cesarean section.The mean gestational age at ultrasonic diagnosis in the eleven patients was (26.3 ±3.0) weeks,and all of them received surgical treatment during pregnancy.The mean gestational age at surgery was (28.2±4.4) weeks.Ten patients with epithelial tumor underwent modified radical mastectomy.One patient with breast malignant phyllodes tumor received local extended lumpectomy.Postoperative pathological staging revealed one patient with stage Ⅰ a,two with stage Ⅱ a,one with stage Ⅱ b,six with stage Ⅲ a,and one with stage Ⅲ b.The patient with paraneoplastic neurological syndrome was diagnosed as having stage Ⅳ breast cancer,and received chemotherapy,radiotherapy and endocrinotherapy.One patient received chemotherapy during pregnancy.None of the patients received radiotherapy during pregnancy.All twelve patients,except one with breast malignant phyllodes tumor,received postpartum adjuvant therapy.All patients were followed up regularly.Five patients were followed up for more than five years,the progressionfree survival rate was 4/5 and the overall survival was 5/5.The other seven patients were followed up for less than five years,the progression-free survival rate was 6/7 and the overall survival was 7/7.Eleven of the twelve patients had live birth.Eleven cases received cesarean section at (36.5±2.8) gestational weeks,and the other one had mid-trimester induction of labor at 27 gestational weeks.Of four premature infants,three were sent to neonatal intensive care unit because of neonatal respiratory distress syndrome,and one died of intracranial hemorrhage.The prognosis of the ten survived infants was good.Conclusions Breast cancer is one of the most common malignant tumors during pregnancy,and it often presents in more advanced stages compared with that in non pregnant women.Multidisciplinary cooperation and detailed assessment of maternal-fetal risks and benefits are necessary.
3.Qualitative Identification of 4 Constituents in Qindan Granules by TLC

OBJECTIVE:To establish the quality standard for Qindan granules. METHODS: The constituents including Radix Scutellariae, Radix Astragali, Cortex Moutan and Fructus Gardeniae in Qindan granules were identified respectively by TLC. RESULTS: The test samples and their respective reference substances were alike in that the same color chromatographic spots were noted at the corresponding places. The color spectra of the samples were clear yet without interference of surrounding impurity. CONCLUSION: The established method has high specificity and can be used for the quality control of Qindan granule.
4.Expression and clinical significance of SALL4 expression in gastric carcinoma tissues

Objective To study the expression and clinical significance of the SALL4 in human gastric carcinoma tissues. Methods The expression of SALL4 in 91 samples of gastric carcinoma and 37 samples of normal gastric tissues was detected by RT-PCR,Western blot and immunohistochemistry,and its relationship with the clinical data were analyzed statistically.Results The positive expression rate of SALL4 in gastric carcinoma(74.7%)was significantly higher than that(18.9%)in normal gastric mucosa tissues(P <0.05).Moreover,with the decreased with the differentiation of gastric carcinoma,the positive expression rate of SALL4 was increased.The expression of SALL4 mRNA and protein in gastric carcinoma tissues were significantly higher than that in normal gastric tissues(P <0.050).The expression levels of SALL4 were relevant to lymph node metastasis(P =0.001),infiltra-tion depth(P =0.029)and the differentiation degree of gastric carcinoma(P =0.050).Conclusion SALL4 was highly expressed in gastric cancer tissues and relevant to lymph node metastasis,infiltration depth and the differentiation degree,which may have play an important role in the development of gastric cancer.
5.Effect of Rumex.japonicus Houtt extracts on hemopoietic system of model mice with thrombocytopenia

Objective To study the treating effect of middle and high doses of acetone and aether extracts from Rumex.Japonicus Houtt on mouse thrombocytopenia.Methods 80 BACB/C mice were randomly divided into 5 groups(n=16): negative control group(salad oil),middle dose(0.5 g?kg-1) and high dose(5 g?kg-1)of acetone and aether extracts groups.All mice in various group were injected with cyclophos-phamide(0.3 mL/20 g) by hypodermic for 3 d to set up the thrombocytopenic model;after mouse thrombocytopenic model was successfully set up,the mice were given salad oil,middle and high doses of acetone and aether extracts,respectively,by gastric perfusion,20 d later the mice were executed,the numbers of the platelet,red cells and white cells were determined.Results ①The numbers of platelet numbers of all groups on the 14th day after making model decreased significantly(P0.05).Conclusion Middle and high doses of acetone and aether extracts have obviously curative effect on mouse thrombocytopenia.
6.Experimental study on treatment of glioma by embryonic neural stem cell transplantation in rats.

The neural stem cells in Wistar rats were cultured in vitro, purified, and transplanted into C6 glioma model in order to observe their biological characters and provide a basic foundation for treatment of neurological diseases by neural stem cell transplantation. The cells at hippocampal area from gestation 15-day rats were cultured in vitro, and frozen and preserved in liquid nitrogen. C6 tumor-bearing models (n=25) and neural stem cells transplantation models (n=35) were established. When the tumor grew to 3 to 4 weeks, 5 rats in each group were randomly selected for MRI examination. At different intervals, the rats were perfused and sampled for HE staining, GFAP and BrdU immunohistochemical staining. The results showed that after resuscitation of neural stem cells at 1-4 passages, the cell viability was 40%-63% with the difference being not significant. The cells could proliferate, passage, and most cells transplanted into glioma model survived. The mean survival time in neural stem cell transplantation group and control was 4.28 and 3.88 weeks respectively, and the average tumor size in the former was smaller than in the latter. It was concluded that embryonic neural stem cells in rats could proliferate and differentiate, and after resuscitation the biological characteristic and viability of the cells were not influenced. Neural stem cells had inhibitory effects on the growth of glioma cells and could prolong the survival of rat model.

8.Retrospective analysis of external quality control of blood cotting from 2009 to 2013

Objective To know about recent experience of the quality control of blood clotting,to analyze the reasons for its out-of-control,and then to explore the influencing factors of coagulation tests.Methods According to the testing standards of coagula-tion quality control products of Clinical Laboratory Center,Ministry of Health,each batch of products were taken standardized tests.The external quality control results of blood clotting from 2009 to 2013 were analyzed and counted up combined with the in-ternal quality control and daily work performance.Results The overall performance of external quality control was good (from 2009 to 2013).The total scores in 2009 were 90%,95%,were 100% and 100% in 2010,were 95% and 95% in 2011,were 100%and 85% in 2012,and were 100% and 100% in 2013.Conclusion Laboratory internal quality control should be guaranteed.The quality control of pre-analysis,post and analysis and analysis-in-course can improve clinical coagulation testing quality.
9.Effects of reactive by burn rat serum oxygen species on apoptosis of pulmonary microvascular endothelial cells induced.

OBJECTIVETo observe the level of intracellular reactive oxygen species (ROS) in rats with severe burn and pulmonary microvascular endothelial cells (PMVECs) treated with serum of rat with burn injury, and to investigate the relationship between ROS and apoptosis of PMVECs.
METHODS(1) Twenty-four SD rats were divided into sham injury group ( n = 3) and burn group (n = 21) according to the random number table (the same grouping method below). Rats in burn group were inflicted with 30% TBSA full-thickness scald on the back, and rats in sham injury group were sham injured. Blood samples were collected from abdominal aorta at post injury hour 6, 12, 24, 36, 48, 60, 72 respectively from 3 rats of burn group. The serum content of ROS was assayed by ELISA. The same determination was performed in rats of sham injury group. (2) Five rats were subjected to scald injury as above, and burn serum was prepared 24 hours after injury. Another 5 rats without receiving any treatment were used to prepare normal serum. (3) Marginal pulmonary tissue was harvested from 20 SD young rats. Cells were cultured with tissue block method and indentified with immunohistochemical staining. The third passage of PMVECs in logarithmic phase were inoculated in 6-well plates and 12-well plates. PMVECs in both plates were divided into 4 groups: normal serum group, burn serum group, normal serum + MnTBAP group, and burn serum + MnTBAP group, with 3 wells in each group. Cells in the former 2 groups were respectively cultured with special nutrient solution of endothelial cells without serum added with 15% healthy rat serum or 15% burn rat serum. Cells in the latter 2 groups were cultured with the same culture conditions as in the former two groups correspondingly with addition of 100 µmol/L MnTBAP in the nutrient solution. After being cultured for 24 h, the content of ROS in PMVECs in 6-well plates was detected with flow cytometry. The apoptosis of PMVECs in 12-well plates was observed with acridine orange-ethidium bromide staining, and the apoptosis rate was calculated. Data were processed with one-way analysis of variance and LSD-t test.
RESULTS(1) The serum contents of ROS in rats of burn group were respectively (187 ± 21), (235 ± 22), (231 ± 25), (291 ± 20), (315 ±23) nmol/mL at post injury hour 24, 36, 48, 60, 72, which were significantly higher than that in sham injury group [(141 ± 19) nmol/mL, with t values respectively 7. 86, 9. 57, 13. 87, 14.98, 18.40, P values below 0.01]. (2) Primary cells grew slowly and showed a cobblestone appearance. After passages, cells grew with orderly distribution. The positive rate of coagulation factor VIII of cells was (96 ± 5)% , and thus they were identified as PMVECs. (3) In normal serum group, burn serum group, normal serum + MnTBAP group, and burn serum + MnTBAP group, the contents of ROS in PMVECs were respectively 798 ± 40, 1 294 ± 84, 763 ± 59, 926 ± 42 ( F =93.01, P <0.01), and the apoptosis rates of PMVECs were respectively (6.2 ± 1.3)%, (57.3 ± 6. 7)%, (3.7 ± 0. 8)%, (28.7 ± 5. 7)% (F = 224.50, P <0.01) after being cultured for 24 h. Compared with those of normal serum group, the content of ROS and apoptosis rate of PMVECs in burn serum group increased significantly (with t values respectively 10.40 and 49.06, P values below 0.01). The content of ROS and apoptosis rate of PMVECs in burn serum + MnTBAP group were significantly lower than those in burn serum group (with t values respectively 7.48 and 23.94, P values below 0.01).
CONCLUSIONSSerum content of ROS was increased in severely burned rats. Burn rat serum stimulation on PMVECs can lead to the increase of the intracellular ROS and induce apoptosis. However application of MnTBAP can scavenge ROS and reduce the apoptosis induced by burn rat serum.
