Major histocompatibility complex class Ⅰ chain related gene A(MICA) is located within the MHC class Ⅰ region of chromosome 6,including six exons.The MICA locus encodes membrane-bound polypeptides,similar with the classic class Ⅰ molecules,including three extracellular domains (α1,α2 and α3),a transmembrane domain,and a cytoplasmic tail,but is not associated with β2-microglobulin.MICA gene is expressed on endothelial cells,epithelial cells as well as most epithelial tumor cells surfaces,and are involved in the process of cell differentiation,proliferation and infiltration in a variety of tumor cells.Soluble MICA (sMICA) can be used as the prognosis predicting factor for some tumors.MICA gene plays an important role in the pathogenesis of a variety of malignant tumors,which might bring new ideas in prevention and treatment for malignant tumors.

3.Photocytotoxic and apoptosis of hypericin-mediated on nasopharyngeal carcinoma cell line of human in vitro

AIM: To study effects of hypericin associated with light emitting diode irradiation on nasopharyngeal carcinoma cell line of human in vitro. METHODS: CNE-2 cells were exposed respectively to different concentration of hypericin,and compared with hematoporphyrin as positive control group,and incubated for 6 h in the dark, then accumulative radiated energe of yellow and red light irradiation equivalent to 5. 67 J/cm2 were given throughout 90 min,killing effect and apoptosis of CNE-2 cells were detected by MTT assay after incubation of 24 h and flow cytometry. RESULTS: MTT assay showed that cytotoxicity of hypericin and hematoporphyrin with light irradiation presented in dose-dependent way,their IC50 values were 0. 049 and 0. 650 ?g/mL,respectively. Flow cytometry showed that hypericin with light irradiation could block cell growth at S phase and G2 phase,when CNE-2 cells were exposed to hypericin at 0. 20 ?g/mL for 18,28 and 48 h,the apoptosis rate reached at 9. 97% ,72. 19% and 92. 24% . CONCLUSION: Hypericin with light irradiation could obviously inhibit the growth of CNE-2 cells and induce apoptosis.
4.Two new phragmalin-type limonoid orthesters from Chukrasia tabularis var. velutina.

As a part of our ongoing research program to isolate novel phragmalin limonoids from the stem bark of Chukrasia tabularis var. velutina, two new phragmalin limonoids orthesters, tabularin Q (1) and tabulalide O (4), along with two known compounds tabularin C (2) and tabularin H (3) were isolated. Their structures were elucidated by means of extensive 1D and 2D spectroscopic analyses which included HSQC, HMBC, and ROESY experiments and HR-ESI-MS.
5.Effect of acute hypervolemic hemodilution on myocardial damage in severely burned rabbits

Objective To evaluate the effect of acute hypervolemic hemodilution (AHH) on myocardial damage in severely burned rabbits.Methods Eighteen healthy adult rabbits,weighing 2.0-2.5 kg,were randomly divided into 3 groups (n =6 each) using a random number table:control group (group C),burn group (B group) and AHH group.Rabbits were subjected to 3rd degree burn covering 40% of the total body surface area.After the model was established,6% hydroxyethyl starch 130/0.4 was infused intravenously,and the target Hct was 25% in AHH group.Before AHH (To) and at 2,4 and 8 h after AHH (T1-3),left ventricular systolic pressure (LVSP),left ventricular end-diastolic pressure (LVEDP),+dp/dtmax and-dp/dtmax were recorded,and blood samples from femoral veins were taken to determine the concentration of serum cardiac troponin I (cTnI) by ELISA.The rabbits were sacrificed at T3,and myocardial specimens were removed for microscopic examination of pathological changes with light microscope.Results Compared with group C,the serum cTnI concentration and LVEDP were significantly increased,LVSP and +dp/dtmaxwere decreased at T1-3 in B and AHH groups,and-dp/dtmax at T1-3 in group B and-dp/dtmax at T3 in group AHH were decreased.Compared with group B,LVSP,+dp/dtmax and-dp/dtm,x were significantly increased at T1-3,and no significant change was found in serum cTnI concentration and LVEDP in group AHH.There was no significant difference in the pathological changes between group B and group AHH.Conclusion AHH can not aggravate the early myocardial damage in severely burned rabbits.
6.Assessment of 22 tumor promotion marker genes for predicting tumor promotion potential of chemicals in Bhas42 cells

OBJECTIVE To evaluate whether the tumor promoting activity syste m based on the ex-pression of 22 tumor pro motion marker genes in Bhas 42 cell transformation assay can be developed to be a new screening technique for predicting tumor promoting potential of che mical.METHODS Chose the 12-O-tetradecanoylphorbol-13-acetate (TPA)as positive che mical and use the Bhas 42 cell lines as test syste m.Define the day of seeding cells as d 0.On d 4,medium in each well was changed with the medium DF5F containing 0.05 mg·L -1 TPA or 0.5 %DMSO,after treat for 24,48 and 72 hrs,collect the cell samples.Extract the RNA fro m the cell samples and detect the expression of Ccnb1 ,Rif1 , Mc m3,Chek1 ,Jun,Fosl1 ,Hells,Vegfa,St mn1 ,Prl2c3,Scarb1 ,Phex,Orm1 ,Orm2,Nup54, Slc2a1 ,Il1 rl1 ,Rad51 ap1 ,Tfrc,Ab1 ,Car13 and Pik3r5 at different ti mepoints by RT-PCR.RESULTS Co mpared to the negative group,the expression of 3 genes was increased after treat with TPA for 24 hr which are Vegfa,Il1 rl1 and Pik3r5;the expression of 12 genes was increased after treat with TPA for 48 hr which are Chek1 ,Hells,Mc m3,Rad51 ap1 ,Vegfa,Il1 rl1 ,Prl2c3,Slc2a1 ,Tfrc,Ab1 ,St mn1 and Rif1 ;the expression of 10 genes was increased after treat with TPA for 72 hr which are Chek1 ,Hells, Rad51 ap1 ,Vegfa,Il1 rl1 ,Prl2c3,Slc2a1 ,Tfrc,St mn1 and Ccnb1 .CONCLUSION The 22 tumor pro-motion marker genes showed different sensitivity to the positive control che mical at different ti mepoints after treat with TPA.The result was consistent with the pro motion activity of TPA.The 22 tumor pro motion marker genes combined with the Bhas 42 cell transformation assay can be developed to be a new screening technique for predicting tumor promoting potential of che mical.
7.Retroperitoneal iaparoscopic ureterolithotomy for impacted upper-ureteral calculi: a report of 42 cases

The clinical data of 42 patients with impacted upper-ureteral calculi undergoing retroperitoneal laparoscopicureterolithotomy(RPLU)from January 2008toDecember 2010 were retrospectively analyzed.RPLU was successfully performed in 40 cases,and simultaneous laparoscopic electrocoagulation ablation was also performed in 11 cases complicated with ureteral inflammatory polyps.No massive bleeding occurred during the operations.The stone-free rate 3 d after operation was 100%,and no stone left as reviewed by kidney ureter bladder X-ray (KUB) and B-ultrasonography.During the follow-up of 3 to 24 months (mean 11 months),40 cases showed no stone recurrence and no ureteral stricture in KUB and B-ultrasonography.The hydronephrosis of all RPLU patients relieved.Results indicate that RPLU is a safe and effective minimally invasive operation for treatment of impacted upper-ureteral calculi.
8.Effect of special anticoagulation method of hybrid coronary revascularization procedure on postoperative bleeding and clinical outcomes

Objective To investigate the effects of special anticoagulation method of hybrid coronary revascularization procedure on postoperative bleeding and clinical outcomes.Methods One hundred and forty-one ASA Ⅱ or Ⅲ consecutive patients,aged 35-82 yr,weighing 45-105 kg,undergoing one-stop hybrid coronary revascularization procedure from June 2007 to January 2011 in Fuwai cardiovascular hospital were enrolled and served as Hybrid group.Using propensity score methodology,these patients were matched with another 141 patients who had undergone off-pump coronary artery bypass grafting (OPCAB group) during the same period,selecting from the surgical database.These ASAⅡ or Ⅲ 141 patients,aged 43-80 yr,weighing 47-110 kg,who underwent OPCAB,were served as control subjects.In group Hybrid,the left internal mammary artery was isolated and then 100-120 U/kg heparin was administrated to maintain activated clotting time between 250-300 s,and additional heparin was given if needed.Protamine 1 mg/kg was used to reverse the effect of heparin after anastomosis between left internal mammary artery and left anterior descending branch.Before stent implantation,6000-8000 U heparin and 300 mg clopidogrel were administrated.Activated clotting time was maintained between 200-250 s until the end of operation.Clopidogrel 75 mg/d was taken for at least 12 months after operation.The volume of postoperative chest tube drainage,mechanical ventilation time,length of stay in intensive care unit,postoperative transfusion of allogeneic blood,re-intubation,myocardial damage,acute kidney injury,atelectasis,hydrothorax and thromboembolism were recorded.Results No thromboembolism was observed within the stent during stay in hospital after operation in patients.The volume of chest tube drainage was significantly less,mechanical ventilation time and length of stay in intensive care unit were shorter,the incidence of atelectasis and hydrothorax and transfusion of allogeneic blood requirement were lower in Hybrid group than in OPCAB group (P < 0.05).There was no significant difference in the incidences of re-intubation,myocardio damage,and acute kidney injury between the two groups (P >0.05).Conclusion Special anticoagulation method of hybrid coronary revascularization procedure does not increase postoperative bleeding and has a better clinical outcomes during the early period.
9.Changes of Treg/Th17 in mice of β2 glycoprotein 1 induced experimental anti-phospholipid syndrome

Objective:To observe the change of CD4+CD25+regulatory T cells and Th17 cells in mice with experimental anti-phospholipid antibody syndrome ( EAPS ) .Methods: EAPS model was established by immunizing BALB/c mice with recombinant humanβ2 glycoprotein 1 (rhβ2GP1).The levels of serum anti-β2 glycoprotein 1 (anti-β2GP1),anti-cardiolipin antibody (aCA),IL-17,IL-2,IL-6 and TGF-βwere tested by ELISA.The rate of abortion,activated partial thromboplastin time (APTT) and platelet count were also detected.Flow cytometry was applied to detect the percentages of the CD4+CD25+regulatory T cells and Th17 cells in peripheral blood mononuclear cells.Results:Compared with the control group,the levels of anti-β2 GP1,aCA,IL-17,IL-2 and IL-6 were significantly increased,the rate of abortion was increased,APTT time was prolonged and the levels of TGF-βand platelet count were de-creased in model mice (P<0.05).No significant difference was detected of percentage of Treg cells in PBMC at the eighth weeks in model group (P>0.05),but percentage of Treg cells was lower than that in control group after 12 weeks (P<0.05);the percentage of Th17 cells in model group was higher than that in control group (P<0.05).In addition,the ratio of Treg/Th17 cells was lower in model mice than that in control group.Conclusion: The imbalance of CD4+CD25 Treg/Th17 cells may participate in the pathogenesis of EAPS.
