1.Effects of a-linolenic acid on insulin signal transduction protein and glucose transport protein 4 in rat skeletal muscle cells

Objective: To study the effect of a-linolenic acid on insulin resistance and its mechanism concerning the insulin signal transduction,providing a theoratic basis for applying plant oil rich in a-linolenic acid in preventing and treating diabetes. Methods :The effect of a-linolenic acid on protein kinase B(PKB) signal transduction and expression of glucose transport protein 4(GLUT4) in rat skeletal muscles was detected by primary cell culture and Western blot techniques. Results:At the concentration of 0. 125-1. 0?mol/L, a-linolenic acid promoted the PKB phosphorylation, but didnot influence the expression of PKB. The expression of GLUT4 protein was dose-dependent and time-dependent. Adding PI3K inhibitor LY294002(15 ?mol/L) in a-linolenic acid cultured skeletal muscle cells made no significant difference in the level of PKB phosphorylation and GLUT4 protein. Conclusion:a-linolenic acid can improve the level of GLUT4 protein by promoting skeletal muscle PI3K/ PKB signal transduction,and increase the sensitivity of skeletal muscle on insulin, which can alleviate or avoid insulin resistance and glycometabolic disturbance.

Objective To investigate the effect of laparoscopic surgery on the change of hepatic function, in terms of serum liver enzymes and to explore its possible mechanism. Methods The blood samples from 286 patients before and after LC were evaluated for liver enzyme levels. The results were compared with those taken from 40 open cholecystectomy (OC) patients. In order to evaluated the possible effect of CO2 pneumoperitoneum on serum liver enzyme levels, same blood tests were also performed in two groups of color-ectal cancer patients undergoing either laparoscopic resectormy( LCR,n =68) or open resection( OCR,n = 23). 3.Study on PHBHHx co-cultured with mouse induced pluripotent stem cells for use as myocardial patches in vitro

Objective To study the effect of polymer material on adhesion , proliferation and differentiation of mouse in-duced pluripotent stem cells via co-culturing in vitro, to find a suitable polymeric biomaterials for miPSCs attachment , prolifera-tion and differentiation , forming myocardial patches as a new repair method for myocardial infarction .Methods miPSCs were recovered, passaged and cultured with PHBHHx two-dimension films and PHBHHx three-dimension films together.The mor-phology of miPSCs attached on the films was visualized under scanning electron microscope ( SEM) .The mouse induced pluri-potent stem cells were induced by differentiation medium that containing vitamin C .Control group did not add any inducer.The survival and differentiation of miPSCs were observed through immunofluorescence and flow cytometry .Results MiPSCs can grow, proliferate and differentiate on PHBHHx films both two-dimension and three-dimension.Vitamin C, as a favourable in-ducer, can markedly improve the efficiency of miPSCs differentiation of cardiomyocytes on PHBHHx films .Immunofluorescence results demonstrated positive cTnT expression.Flow cytometry measured cTnT expression: Vitamin C inducer group(47.54 ± 1.46)%>without any inducer group(7.02 ±0.95)%(P<0.05).In the process of miPSCs differentiating into myocardial cell, after adding Vitamin C, the expression quantity of cTnT:PHBHHx three-dimensional films(60.32 ±1.76)%>PHBHHx two-dimensional films(53.31 ±1.41)%>traditional cell culture(47.54 ±1.46)%(P<0.05).Conclusion iPSCs can ad-here, survive and differentiate on the PHBHHx film.Vitamin C, as a favourable inducer, can markedly improve the efficiency of miPSCs differentiation of cardiomyocytes .Relative to PHBHHx two-dimensional film culture and traditional culture , the three-dimensional PHBHHx film culture has a great advantage in the process of miPSCs differentiating into myocardial cells .
4.Expression and significance of p27kip1, CyclinD1, Cyclin E in transitional cell carcinoma of bladder

Objective To investigate the expression of p27kip1, CyclinE, CyclinD1 in bladder transitional cell carcinoma (BTCC), and evaluate their relationship with turnout cell proliferation. Methods To study the expression of p27kip1, CyclinE and CyclinD1 protein in 41 cases of paraffin-embedded BTCC tissue and in 12 cases of cystitis tissues by immunohistochemical method SP. Results There was a negative relationship between p27kip1 expression and clinical stage and pathological grade.There was no relationship with lymph node metastasis (P>0.05). There was a positive relationship between CyclinE expression and clinical stage, pathological grade and lymph node metastasis (P<0.05). There was no relationship between CyclinD1 expression and clinical stage, pathological grade and lymph node metastasis (P>0.05). Conclusion The results of this study showed that the abnormal expression of p27kip1, CyclinE, may play important roles in the genesis and progression of BTCC. CyclinD1 may take activation in the early term of BTCC.

6.The value of detection of serum myocardial enzyme spectrum and cardiac troponin I in neonatal hypoxic ischemic encephalopathy

Objective To discuss the value of analysis of serum myocardial enzyme spectrum and cardiac troponin I(cTnI) in neonatal hypoxic ischemic encephalopathy(HIE) .Methods Serum level of myocardial enzyme spectrum in each people was deter-mined with Hitachi 7602 ,Serum levels of cTnI was determined by chemoluminescence method .Serum levels of myocardial enzyme spectrum and cTnI were collect from 65 neonates with HIE(HIE group) and 68 cases of helthy volunteers(control group) .Results The levels of glutamic oxalacetic transaminase(AST) ,creatine kinase(CK) ,creatine kinase MB isoenzyme(CK-MB) ,lactic dehy-drogenase(LDH) and alpha-hydroxybutyric dehydrogenase(HBDH) in HIE group were(98 .9 ± 9 .6)U/L ,(385 .4 ± 15 .5)U/L , (89 .3 ± 9 .6)U/L ,(300 .8 ± 19 .8)U/L ,(399 .1 ± 17 .9)U/L ,which were higher than those in control group(P<0 .05) .The level of cTnI was(1 .4 ± 0 .4)ng/mL in HIE group ,which was higher than that in control group(P<0 .05) .CK-MB sensitivity was 88 .6% , cTnI speciality rate was 86 .3% .Conclusion The assay of myocardial enzyme spectrum and cTnI is significant in diagnosis of neo-natal HIE .
7.Study on the nutrition status of 6 to 24 months old infants in poverty areas

Objective Discuss the studies of infant nutrition problems of 6 to 24 months old infants in poverty areas and make reasonable suggestions.Methods Collect research reports about child malnutrition at home and abroad,and analyze the monitoring data of nutrition improvement program for children in poor areas carried out by Natiend Institate for Nutrition and Health for of Chinese Center for Disease Control and Prevention.Discuss the nutritional status and related risks of infants aged 6-24 months in poor areas.Results The nutritional problems of rural children in poor areas were prominent.The growth retardation rate of infants aged 6-24 months was 7.6%.Anemia,overweight and obesity are becoming increasingly prominent.Conclusion The nutrition status of 6-24 month old infants in poor areas in China should be paid attention.
8.Correlation between lipoprotein associated phospholipase A 2 and five index in coronary heart disease

Objective To analyze the correlation between serum lipoprotein associated phospholipase A 2(Lp‐PLA2) with five traditional inflammatory factors of TC ,TG ,LDL‐C ,hs‐CRP and Hcy in the patients with coronary heart disease(CHD) ,and to in‐vestigate the relationship between its concentration with the lesions and severity of CHD .Methods Two hundreds cases of CHD were selected as the lesion group(which was subdivide into single vessel lesion ,double vessel lesion and three vessel lesion) ,at the same time ,100 persons undergoing physical examination were selected as the control group .The correlation between Lp‐PLA2 with five traditional inflammatory factors of TC ,TG ,LDL‐C ,hs‐CRP and Hcy was analyzed .The differences of Lp‐PLA2 in the lesion group and the control group were compared and the relation between Lp‐PLA2 level with lesion vessels number was analyzed .Re‐sults Lp‐PLA2 was significantly correlated with LDL‐C ,Hcy ,TC and hs‐CRP(P<0 .05) and had no relation with TG(P>0 .05) , the level of Lp‐PLA2 in the lesion group was significantly higher than that in the control group (P<0 .01) ,the Lp‐PLA2 level was elevated with the number of coronary lesion vessels ,but the correlation is unobvious(P>0 .05) .Conclusion High concentration of Lp‐PLA2 is a risk factor for coronary atherosclerosis ,but the correlation between the Lp‐PLA2 level and the severity of coronary arterial lesion needs further study .
9.Clinical value of combined detection of of SCC and CEA in the diagnosis of lung cancer

Objective To explore the clinical value of combined detection of SCC and CEA in the diagnosis of lung cancer.Methods 382 cases from January 2015 to April 2016 in the respiratory department of our hospital were selected in this survey.Among which 172 cases with lung cancer,110cases with benign lung diseases and 100 healthy person.The CEA and SCC content in serum of all these cases were determined by chemical chemiluminescence,and the diagnostic efficacy of lung cancer by CEA or SCC alone and CEA and SCC combined examination was compared.Results In lung cancer group,the serum levels of CEA and SCC content were significantly higher than those of the normal control group and lung benign disease group(P<0.05),and benign lung disease group were significantly higher than that of the normal control group(P<0.05).and compared with squamous cell carcinoma and small cell lung cancer,CEA has the highest content in adenocarcinoma(P<0.05).Meanwhile,the content of SCC in squamous cell carcinoma was obviously higher than that of small cell lung cancer and adenocarcinoma(P<0.05),The sensitivity of CEA was significantly higher than that of SCC(P<0.05),and the sensitivity and negative predictive value in combined detection group was significantly higher than that of CEA or SCC(P<0.05).Conclusion SCC and CEA could be used to identify different types of lung cancer,and the combination of the two methods could improve the positive diagnosis rate,facilitate the early detection of lung cancer,early treatment,has the promotion value.
10.Study of working memory in patients with no neurological syphilis disease

Objective To investigate the differences of the working memory tasks' performance between the patients with no neurological syphilis disease and the health controls (HC).Methods 2-back test of N-back task paradigm were applied to measure the performances of working memory.A total of 30 patients with syphilis disease were compared with 35 gender-,age-,years of education-matched healthy persons,then compared the accurate rate and reaction time on the tasks between the two groups.Results Participants with anxiety or depression had to be excluded.The results showed that patients impaired on working memory task.The accurate rate and reaction time of the working memory of the patients was worse than that of the healthy persons ((65.31 ± 8.78) % vs (71.00 ±9.64)% ;(1521.14 ±290.57)ms vs (1296.08 ±253.20)ms respectively) (t=-2.472,P=0.016;t=3.337,P =0.001 respectively).The study indicated the reaction time of the working memory for the patients was negative correlation to the age of final diagnosis (r =-0.362,P =0.049).Conclusion The patients with no neurological syphilis disease show impairment on working memory.This probably because treponema pallidum injures the central nervous system.
