Objective To determine the content of gentiopicroside and to evaluate the quality of Radix Gentianae Macrophyllae in Gansu province.Methods The HPLC method was performed on Waters C18 column.Methanol-water(3 ∶7) were used as the mobile phase,the flow rate was at 1.0 mL?min-1 with the detection wavelength being 254 nm.Results For gentiopicroside,the linear range was 1.7352～17.3520 ?g and the average recovery was 97.06 %with RSD=2.5 %.Conclusion The content of gentiopicroside in all of the samples produced in Gansu province is higher than or approach to the standard for Radix Gentianae Macrophyllae in Chinese Pharmacopoeia.

Objective To improve detection rate and diagnostic accuracy of single ventricle(SV) in obstetric fetal echocardiography,to investigate the common types and complicated malformations of SV in the fetus,and to summarize the differential announcements in diagnosing fetal SV.Methods In 345 fetal hearts which were diagnosed as congenital heart disease by fetal echocardiography in our hospital,73 cases diagnosed as SV,including 3 cases appeared as ones of twins,were included in this study.Systemic scanning and multiple-views fetal echocardiography were used to examine these enrolled fetuses.Results In all 73 SV eases,3 cases were diagnosed as simple SV,the others were diagnosed as SV accompanied with other abnormalities,among them 44 cases accompanied with single atrium,18 cases with single atrium and persistent truncus arteriosus,2 cases with pulmonary atresia,20 cases with pulmonary artery stenosis,4 cases with partial atrioventricular septal defect,3 eases with aorta dysplasia or aortic valve dysplasia.SV types were classified as 24.7％ in type A,13.7％ in type B,46.6％ in type C and 15.0％ in type D respectively.68.2％ of the cases were diagnosed with aortic D-transposition,and 45.2 ％ with common inlet,42.5 ％ with single inlet and 12.3 ％ with double inlet respectively.42 SV cases were executed termination of pregnancy which 11 cases were confirmed by pathology and the other 31 cases were out of following-up.Conclusions Most cases of fetal SV were accompanied other abnormalities and simple SV was rare.Type C in which ventricular structure was combined with left and right ventricle was the most common type.To avoid the false diagnosis,much attention must be paid to distinguish big papillary muscle and abnormal muscle bundle from interventricular septum during ultrasonic examination.

Objective To evaluate the effects of matrine on the inflammatory,oxidative and migratory responses of HaCaT keratinocytes in an in vitro atopic dermatitis(AD)model.Methods HaCaT cells were divided into the control group(no treatment)and the model group(0.05,0.1 and 0.2 mmol/L).HaCaT cells in the model group were stimulated with 10 μg/L tumor necrosis factor-α(TNF-α)/interferon-γ(IFN-γ)for 24 h.Cells in the model group were pretreated with 0.05,0.1 and 0.2 mmol/L matrine for 1 h and then stimulated with 10 μg/L TNF-α/IFN-γ for 24 h.MTT assay was used to assess the cytotoxicity of matrine.Enzyme-linked immunosorbent(ELISA)assay was performed to determine levels of cytokines(IL-6,IL-8 and IL-1β)and chemokines(TARC,MDC and RANTES).mRNA expression levels of antioxidant genes were detected by real-time quantitative PCR(RT-qPCR).Cell migration was analyzed by scratch closure assay.Immunohistochemistry was commended to analyze the nuclear translocation of NF-κB p65.Western blot assay was used to detect the phosphorylation levels of p38,ERK and p65 proteins.Results Compared with the model group,matrine 0.1 and 0.2 mmol/L significantly decreased the expression levels of IL-6,IL-1β,IL-8 and chemokines,and matrine significantly enhanced the expression levels of superoxide dismutase-1(SOD1),SOD2,catalase(CAT)and glutathione peroxidase(GPx),and promoted scratch wound healing(P＜0.05).In addition,matrine 0.1 and 0.2 mmol/L inhibited the phosphorylation levels of p38,ERK and p65 proteins and the nuclear translocation of p65.Conclusion Matrine possesses anti-inflammatory and antioxidant properties,and stimulates wound closure in eratinocyte AD model,which may be related to the inhibition of the activation of MAPK and NF-κB pathways.

Objective To understand outpatients' mastering situation on cognition and maintenance knowledge of venous indwelling needle, to promote the proper use and maintenance of venous indwelling needle in outpatients. Methods A questionnaire survey was done among 135 patients with venous indwelling needle. Results Patients showed higher cognition on venous indwelling needle,poorer maintenance knowledge andmisunderstanding,and higher expectations on nurses' health education.Conclusions Nurses should do a goodjob of health education on the knowledge and maintenance guidance of venous indwelling needle to outpatients,to further promote its application,and to ensure the correct use of indwelling needle.

Objective:To evaluate the effects of Huoxue Bushen Formula Granules on endometrial receptivity (ER) in patients with polycystic ovary syndrome (PCOS) in ovulation induction cycle.Methods:A randomized controlled trial was conducted. Totally 50 patients with PCOS infertility in Xiyuan Hospital of China Academy of Chinese Medical Sciences from January 2019 to January 2023 were selected as the observation objects, and were divided into observation group (29 cases) and control group (21 cases) according to the random number table method. Both groups were given oral clomiphene citrate on the 5th day of menstrual cycle or progesterone withdrawal. On this basis, the observation group was given Huoxue Bushen Formula Granules on the 5th day of menstruation, while the control group was given estradiol valerate tablets. 7 days after taking medication and injecting hCG from the 10th day of menstruation, ovulation was monitored and sexual intercourse was guided. Both groups were treated for 3 menstrual cycles and followed up for 3 months. B-ultrasound was performed on 6-8 days after ovulation to obtain uterine artery pulsatility index (PI) and blood flow resistance index (RI). TCM syndrome scores were evaluated before and after treatment, and the endometrial receptivity score (Salle score) was used to evaluate the level of ER. The pregnancy and adverse reactions during treatment were recorded in the two groups.Results:After treatment, TCM syndrome score in the observation group [14.0 (13.0, 15.0) vs. 16.0 (14.5, 19.5), Z=-3.23] was lower than that of the control group ( P<0.001). The Salle score of the observation group after treatment [12.0 (9.5.0, 13.5) vs. 10.0 (9.0, 11.0), Z=-3.84] was higher than that before treatment ( P<0.001). The PI (0.68±0.52 vs. 0.81±0.06, t=18.25), RI (1.65±0.36 vs. 2.24±0.45, t=22.78) after treatment in the observation group and the control group were lower than those before treatment ( P<0.001), without statistical significance ( t values were -0.39, -1.37, respectively, P>0.05). The total effective rate of the observation group was 75.86% (22/29), and that of the control group was 71.43% (15/21), without statistical significance ( χ2=0.12, P=0.724). The pregnancy rate of the observation group was 82.76% (24/29), and that of the control group was 57.14% (12/21), with statistical significance ( χ2=3.96, P=0.046). The incidence of adverse reactions of the observation group was 3.45% (1/29) and that of the control group was 9.52% (2/21), without statistical significance ( χ2=0.80, P=0.372). Conclusion:Huoxue Bushen Formula Granules can improve the ER of PCOS infertility patients with kidney deficiency and blood stasis syndrome, improve the pregnancy rate, reduce the TCM syndrome score, improve the quality of life of patients with high safety.
