1.Effects of Cimetidine with or without Concomitant Vitamin C Intake on Intragastric Milieu

The intragastric changes especially the changes of nitrite level after the cimetidine treatment with or without concomitant administration of vitamin C were studied in 34 patients with peptic ulcer. The pH value, nitrite level, and bacterial count of the gastrie juice after cimetidine treatment were significantly higher than those prior to the treatment (P0.05). Besides,the reduction of BAO,PAO,and MAO was not influenced by vitamin C intake. It is believed that vitamin C can reduce the nitrite concentration which was elevated by cimetidine treatment, but it will not affect the acid-inhibitory function of cumetidine. This fact is of importance for the safe application of cimetidine
2.A study on p53 and Rb genes in gastric cancer and juxta-cancerous tissue

The deletion and rearrangement of p53 and Rb genes were studied in gastric cancer with Southern blot hybridization and the point mutation in 248 and 249 codons of p53 was studied with PCR-RFLP.It was found that deletion or rearrangement of p53 was seen in 9 cases out of 30 cases of gastric cancer and point mutation at 248 codon of p53 in 2 cases out of 42 cases,and deletion and rearrangement of Rb gene were seen in 2 cases out of 15 cases.
3.A study on the amplification, rearrangement and deletion of ras-oncogenes in gastric cancerous and juxtacancerous tissues

The amplification,rearrangement and deletion of c-Ha-ras,K-ras and N-ras were studied in 33 cases of gastric cancerous and juxtacancerous tissues.It was found that the amplification rearangement,and deletion of c-Ha-ras were shown in 25.8% (8/33) of cases and the ampli-cation of K-ras in 3.8% (1/26).No such changes was found for N-ras.
4.A study on the activation of oncogene and inactivation of suppressor gene in human gastric cancer

The changes of several oncogenes and suppressor genes in the specimens of cancerous and juxtacancerous tissues of 42 cases of gastric cancer were studied with Southern blot hybridization and PCR-RELP method.The probes used were c-Ha-ras,K-ras,N-ras,N-myc,c-myc,hst,EGFR,c-erbB-2,p53 and Rb.Amplification,rearrangement,and deletion of c-Ha-ras were detected in 8/33 (25.8%) cases of gastric cancer (amplification or rearrangement of hst and c-erbB-2 in 11/42 (26.6%) and 12/42 (29.2%) cases respectively (amplification of EGFR in 21.4% of cas-esideletion or rearrangement of p53 and Rb in S/30 (30%) and 2/15 (13%) cases respectively) and amplification or rearrangement of N-ras (0/33),K-ras (1/26),N-myc (1/26),and c-myc (1/ 35) was only rarely encounted.The point mutation in codon 248 and 249 of p53 in gastric cancer was analyzed.2 cases of the 42 harbored point mutation in codon 248 of p53.These findings suggest that e-Ha-ras,hst,c-erbB-2,EGFR and p53 may be the hot point genes in the occurrence and development of gastric cancer.
5.A study on point mutation of ras oncogenes

It is known that no restriction endonuclease cleavage site exists at 12 codon in N-ras gene and at 12 and 13 codon in K-ras gene.Amismatched base was incorporated at the 3' end primer to creat a kind of restriction endonuclease cleavage site.Then it was possible for us to analyze the point mutation of the above mentioned codons with the polymerase chain reaction-restriction fragment length polymorphism.In this paper,the study of the point mutation at 12 and 61 codon in c-Ha-ras,at 12 codon in N-ras,and at 12 and 13 codon in K-ras was reported.
6.Application of service-learning pattern in clinical nursing teaching in Obstetric department

Objective To investigate the application effect of service-learning pattern in clinical nursing teaching in Obstetric department. Methods A total of 113 nursing students in Obstetric department were randomly divided into two groups:57 were trained with traditional clinical nursing teaching method as the control group, 56 with service-learning teaching method as the experimental group. All students were examined for comprehensive assessment, including professional theory, professional operation and comprehensive quality after their training. At the same time, the clinical teaching satisfaction of nursing students were surveyed. Results There was no statistical difference in exam results between two groups of nursing students after training(P>0.05). However, the experimental group had higher achievements in professional operation and comprehensive quality after their training than the control group (35.24 points vs. 32.10 points, 16.27 points vs. 14.05 points), as well as, the clinical teaching satisfaction of the experimental group was higher than that of the control group(96.42% vs. 89.60% ), the differences were statistically significant (P<0.05). Conclusions Service-learning pattern can significantly improve the clinical teaching effect of undergraduate nursing students in Obstetric department.
7.Relationship of compression area, time and weight with crush injury/crush syndrome in rats

Objective To investigate the relationship of compression area, time and weight as risky factors with local injury and systemic pathophysiological responses in rats so as to establish repeatable experimental model of crush syndrome. Methods A total of 144 male SD rats were divided into two groups, ie, mortality investigation group and biochemical indicator investigation group. Every group included the same 18 subgroups based on 18 kinds of combination with different levels of compression area (the right or both hind limbs), time (4, 6, 8 hours) and weight (2, 3, 4 kg). The circumference of the compressed hind limbs of all rats were measured and serum potassium (K+), serum creatine phosphokinase (CK), creatinine and carbamide were measured too before compression and three hours after decompression. Incidence of myoglobinuria of all rats was recorded. Muscles and kidneys were evaluated morphologically. Results The compressed hind limbs of all rats swelled significantly after three hours of reperfusion (P < 0. 05). All serum K + , CK, CR and BUN were increased significantly with the increase and prolongation of the compression area, time and weight (P < 0.05). Signs of direct cellular damage and ischemia-reperfusion injury were found in histology specimens of local compressed muscle.Hyperemia of glomeruli and renal tubule was found in the kidneys. Renal tubular necrosis and renal tubular cast were observed in group with compression weight ≥3 kg and compression duration ≥6 hours.Conclusions Increase and prolongation of the compression area, time and weight can aggravate the severity of crush injury. Compression area is more risky factor of severe crush injury. Both hind limbs ascompression area, compression weight ≥ 3 kg and compression duration ≥ 6 hours can be the effective experimental conditions for establishment of crush syndrome model in rats.
8.The influence of using continuous quality improvement program in the follow-up survey to atherosclerotic renal artery stenosis patients

Objective To improve the compliance and prognosis of patients with atherosclerotic renal artery stenosis(ARAS) by using continuous quality improvement program(CQI).Methods Survey patients by the way of using the four-step problem-solving framework referred as the PDCA cycle,i.e.plan,do,check and act.Compare the results of patients which have obtained before and after CQI.Results The follow-up rate of patients was improved from 46.9% to 83.3%, and most of the patients are willing to enter the follow-up program.The patients are more likely to receive examinations of ARAS,such as blood pressure,renal function,renal artery color duplex sonography and renal scintigraphy.The control rate of blood pressure was also improved from 45.6% to 93.1% Conclusion CQI is a useful method to improve the compliance and prognosis for ARAS patients,and nurses have played an important role during the whole process.
9.Application of serum and urinary endothelin-1, interleukin-6 assay in screening atherosclerotic renal artery stenosis

Objective To investigate the possibility of using serum and urinary endothelin-1 (ET-1), interleukin-6 (IL-6) assay as screening tools for atherosclerotic renal artery stenosis (ARAS). Methods Serum and urinary samples from 49 patients with ARAS, 32 cases with≥2 risk factors for atherosclerosis and 30 normal controls were detected for ET-1 and IL-6 by RIA. The receiver operating characteristic (ROC) curves were then generated to assess their accuracy in screening ARAS by using selective renal angiography as golden standard. Results Urinary ET-1, urinary IL-6 and urinary-serum ratio of ET-1 in ARAS cases were higher than those in two control groups, and were all correlated with degree of renal artery stenosis. The area under the ROC curve of urinary ET-1, urinary IL-6 and urinary-serum ratio of ET-1 was 0.792, 0.756 and 0.779, respectively. The sensitivity and specificity of urinary ET-1 to distinguish ARAS(≥50%) was 80.0% and 72.8% respectively using 6.72 ng/mmol creatinine as the cut-off point. The cut-off value of ET-1 urinary-serum ratio was 12.59 with a sensitivity of 66.7% and a specificity of 61.7%. The sensitivity and specificity of urinary IL-6 were 73.3% and 70.4% respectively using 23.85 ng/mmol creatinine as the cut-off point. The sensitivity and specificity were improved to 80.0% and 77.8% respectively when using 12.60 ng/mmol creatinine as the cut-off point and combined with hypertension to perform series test. Conclusion Urinary ET-1 and urinary IL-6 could be used as screening tools for ARAS.
10.Arsenic trioxide inhibits KM3 cell growth and induces apoptosis and potentiated by bortezomib

AIM: To observe if there is a synergistical effect on induction of apoptosis when arsenic trioxide alone or combination with bortezomib in KM3 cells. METHODS: KM3 cells were treated with arsenic trioxide alone or combined with bortezomib, the numbers of viable cells were determined by trypan blue exclusion. Cell growth inhibition was examined by MTT method. The cells were simultaneously stained with annexin V-FITC and PI and apoptosis was determined by bivariate flow cytometry using a FACScan. Reverse trascriptional-PCR (RT-PCR) method was used to examine the change of p65 mRNA and Western blotting to measure the expression of protein p65, p-p65, caspase-3, -8, -9, and poly ADP-ribose polymerase (PARP). RESULTS: Arsenic trioxide inhibited the cell growth and induced apoptosis. The mechanism was responsible for the activation of caspase-mediated induction of apoptosis. A synergistic effect of combination with bortezomib on apoptosis was observed. CONCLUSION: Arsenic trioxide inhibits KM3 cell growth and induces apoptosis with a synergistical effect when cotreated with bortezomib.
